Pleiotropic Effects of GIP on Islet Function Involve Osteopontin

- OBJECTIVE-The incretin hormone GIP (glucose-dependent insulinotropic polypeptide) promotes pancreatic beta-cell function by potentiating insulin secretion and beta-cell proliferation. Recently, a combined analysis of several genome-wide association studies (Meta-analysis of Glucose and Insulin-Related Traits Consortium [MAGIC]) showed association to postprandial insulin at the GIP receptor (GIPR) locus. Here we explored mechanisms that could explain the protective effects of GIP on islet function. RESEARCH DESIGN AND METHODS-Associations of GIPR rs10423928 with metabolic and anthropometric phenotypes in both nondiabetic (N = 53,730) and type 2 diabetic individuals (N = 2,731) were explored by combining data from 11 studies.Insulin secretion was measured both in vivo in nondiabetic subjects and in vitro in islets from cadaver donors. Insulin secretion was also measured in response to exogenous GIP. The in vitro measurements included protein and gene expression as well as measurements of beta-cell viability and proliferation. RESULTS-The A allele of GIPR rs10423928 was associated with impaired glucose- and GIP-stimulated insulin secretion and a decrease in BMI, lean body mass, and waist circumference. The decrease in BMI almost completely neutralized the effect of impaired insulin secretion on risk of type 2 diabetes. Expression of GIPR mRNA was decreased in human islets from carriers of the A allele or patients with type 2 diabetes. GIP stimulated osteopontin (OPN) mRNA and protein expression. OPN expression was lower in carriers of the A allele. Both GIP and OPN prevented cytokine-induced reduction in cell viability (apoptosis). In addition, OPN stimulated cell proliferation in insulin-secreting cells. CONCLUSIONS-These findings support beta-cell proliferative and antiapoptotic roles for GIP in addition to its action as an incretin hormone. Identification of a link between GIP and OPN may shed new light on the role of GIP in preservation of functional beta-cell mass in humans. Diabetes 60:2424-2433, 2011

- Neurochemical evidence indicates that cognitive impairment in dementia of Alzheimer type (DAT) is related to degeneration of cholinergic neurons in the brain. A pharmacological approach is treatment with a cholinesterase inhibitor such as tetrahydroaminoacridine (THA). THA treatment of 17 patients with DAT was studied with a double-blind crossover design with three types of treatment, THA + lecithin, THA + placebo and placebo + placebo. Each treatment period was 6 weeks with wash out periods of 2 weeks. The treatment was evaluated with clinical ratings, psychometric testing, EEG and regional cerebral blood flow (rCBF) measurements. No significant clinical differences between treatment periods were found in the total sample, but marked individual differences were observed. The patients were subdivided into three outcome groups based on four clinical measures: 6 patients improved (responders), 5 patients were mainly unchanged, and 6 patients showed further deterioration during the trial period of 26 weeks. Pretreatment rCBF in responders differed significantly from that of the deteriorated patients. EEG showed more high frequency activity among responders. Hepatotoxic side effects were observed in several cases. Three subjects showed marked increases of liver enzymes, with normalization following dose reduction. The majority of patients who improved or remained unchanged during the study chose to continue THA treatment in an open trial.

- This article presents a specific example of how the work of an editor of the comprehensive historical Swedish dictionary, published by the Swedish Academy, (SAOB) proceeds. The example concerns the Swedish preposition under ’under’ and accurately describes the process of writing that SAOB entry. Drawing from this particular word, it is shown how etymology governs also the semantic description, on what principles the different meanings of a word is either divided into different main sections or kept together within one and the same main section, how SAOB deals with borderline cases, how the description interacts with already written parts of the dictionary, how the main sections may be grouped together in bigger subgroups, how subordinate sections are distinguished and furthermore how the use of a word in compounds may influence on the description of the simplex word.

- During the 16th century, and thus late in a european perspective, the Scandinavian language usage went from normally giving dates according to the church year calendar (each day having a name of its own, mostly referring to a saint) to normally giving them according to our modern calendar (numbering the days within each named month). The present investigation shows how this transition more exactly proceeded within Danish, Swedish and Icelandic – how the new time expressions first were used only by a few pioneers in the beginning of the century, most likely influenced by the renaissance, how they then gained ground in central parts and among the élite of Sweden (rather abruptly) and Denmark (more successively) some decades later, and how they conquered more periphere areas only in the late 16th century. It is also shown how the two types of time expressions were structured in detail – how the old church year ones never gained a standardised format, different genres displaying somewhat different structural tendencies, but how the modern ones on the other side were rapidly shortened to that standard format we now are used to, though by no means present in their earliest Scandinavian usage.

- In the narrative parts of the Greek New Testament, we find instances of historical present. This article aims to show how this is reflected in the Nordic translation history. In Swedish translations of the New Testament, historical present is altogether absent, be it the early translations in the 16th century or the modern translations. The same holds true for some Danish and Faroese translations. However, in the early Danish and Icelandic translations from the Reformation era, there are some instances of historical present (except in one Danish translation), but these are much fewer than in the Greek source text and also somewhat fewer than in the translation by Luther. In Icelandic and Danish translations from the first part of the 20th century, as well as in a contemporary Faroese translation, the strategy is clear: wherever the Greek text has historical present, so do the translations (with only a few exceptions). In the Norwegian translations, the use of historical present is more temperate and furthermore concentrated to specific chapters, where it is even a bit more generous than inthe source text. This strategy is also applied in a later Danish translation. In a later Icelandic translation, there are also fewer instances than in earlier translations, but there is not this tendency of concentration in particular chapters. Thus, two traditions exist in the Nordic bible translations regarding historical present – one where it is totally unknown, and one where it is used more or less frequently, although always inspired by instances in the source text and never as an independent stylistic device.

- The thesis accounts for a comparison between the two different parts of The National Law of King Christopher (1442): the parts taken over from the older national law and the parts rewritten och new written. I also compare with those parts of The Urban Law of King Magnus Eriksson that are independent of the national law. The investigations concern legal case clauses, legal enactment modes, syntactic complexity, word order, vocabulary and alliteration and show that the new parts in most respects rather correspond stylistically with the urban law than with the older national law. Often these correspondencies have a more explicitly generalized style in common, but also foreign influence plays a major part.

- In a linguistic description there is normally a more or less conspicious lack of balance between the space a phenomenon requires to be fully described and its frequence in actual language usage, here called a lack of iconicity. The article investigates to what degree some Danish and Swedish dictionaries are iconical in this sense, i.e. whether the space of a word in the dictionary corresponds with its actual frequence. As expected, iconicity is rather meager, the most frequent words being treated much shorter than there actual frequence would suggest and the less frequent ones the other way around, though the more comprehensive the dictionary the more iconical it normally is regarding the first category. Further, frequent nouns and adjectives can be shown to exhibit a more iconical description than frequent verbs, and compounds and derivations treated together with their base words often exhibit a (shorter and thus) more iconical description than those treated as their own entries (often iconically much too extensive). Iconicity is of course not the overall ideal state of good dictionary entries: frequent form words are often best treated rather generalized (and thus rather shortly), some less frequent words can only be described accurately if described rather extensively. Yet, it is argued that iconicity should not be dismissed alltogether but to be considered as one factor (among many other ones) when deciding upon how much space a word is worth in a dictionary.

- The notion ‘textual history’ (Sw. texthistoria) has often been used in a narrow philological sense denoting the history of version of a single work. Today it is however, at least in the Nordic languages, used in other connections as well, not least within discourse analysis. The article shows that textual history theoretically could be understood as (1) the history of text(s) as language in use, whether concerning the text linguistic resources as such or a specific category of texts or – as often within philology – just one work in its different versions, (2) the external history of the production and use of text(s) as language in use (i. a. literacy), (3) the history of texts as part of the outer course of events (e. g. historical discourse analysis), and (4) the external history of text production and reception within this outer course of events (i. a. traditional history of literature). The notion is thus most diversified and not readily understandable if used in one more specific meaning. It is however suggested that perhaps the most expediant usage is that of type (3): ‘history of texts as external events’, though then in obvious contrast to that widespread philological one within type (1). Textual history would then also be something different than (a part of) language history, a research field of its own.
