    • The nucleosynthesis of elements beyond iron is dominated by neutron captures in the s and r processes. However, 32 stable, proton-rich isotopes cannot be formed during those processes, because they are shielded from the s-process flow and r-process beta-decay chains. These nuclei are attributed to the p and rp process. For all those processes, current research in nuclear astrophysics addresses the need for more precise reaction data involving radioactive isotopes. Depending on the particular reaction, direct or inverse kinematics, forward or time-reversed direction are investigated to determine or at least to constrain the desired reaction cross sections. The Facility for Antiproton and Ion Research (FAIR) will offer unique, unprecedented opportunities to investigate many of the important reactions. The high yield of radioactive isotopes, even far away from the valley of stability, allows the investigation of isotopes involved in processes as exotic as the r or rp processes.

    • The strength of the neutron-neutron correlation function from the E=45AMeV58Ni+27Al, natNi, and 197Au reactions depends on the neutron parallel velocity. This indicates the presence of multiple sources of neutron emission. We find these sources consistent with a dissipative, binary reaction mechanism as it is described by, e.g., Boltzmann-Uehling-Uhlenbeck calculations.

    • During heavy-ion operation in several particle accelerators worldwide, dynamic pressure rises of orders of magnitude were triggered by lost beam ions that bombarded the vacuum chamber walls. This ion-induced molecular desorption, observed at CERN, GSI, and BNL, can seriously limit the ion beam lifetime and intensity of the accelerator. From dedicated test stand experiments we have discovered that heavy-ion-induced gas desorption scales with the electronic energy loss (dEe/dx) of the ions slowing down in matter; but it varies only little with the ion impact angle, unlike electronic sputtering.

    • We describe a device to study reactions relevant for the Single Event Effect (SEE) in microelectronics by means of 200A and 300AMeV, inverse kinematics, Si + H and Si + D reactions. The work is focused on the possibility to measure Z = 2-14 projectile fragments as efficiently as possible. During commissioning and first experiments the fourth quadrant of the CELSIUS storage ring acted as a spectrometer to register fragments in two planes of Si strip detectors in the angular region 0 degrees-0.6 degrees. A combination of ring-structured and sector-structured Si strip detector planes operated at angles 0.6 degrees-1.1 degrees. For specific event tagging a Si+ phoswich scintillator wall operated in the range 3.9 degrees-11.7 degrees and Si Delta E-E telescopes of CHICSi type operated at large angles.

    • This paper is the outcome of a community initiative to identify major unsolved scientific problems in hydrology motivated by a need for stronger harmonisation of research efforts. The procedure involved a public consultation through online media, followed by two workshops through which a large number of potential science questions were collated, prioritised, and synthesised. In spite of the diversity of the participants (230 scientists in total), the process revealed much about community priorities and the state of our science: a preference for continuity in research questions rather than radical departures or redirections from past and current work. Questions remain focused on the process-based understanding of hydrological variability and causality at all space and time scales. Increased attention to environmental change drives a new emphasis on understanding how change propagates across interfaces within the hydrological system and across disciplinary boundaries. In particular, the expansion of the human footprint raises a new set of questions related to human interactions with nature and water cycle feedbacks in the context of complex water management problems. We hope that this reflection and synthesis of the 23 unsolved problems in hydrology will help guide research efforts for some years to come.

  • In vitro and in vivo evaluation of a new sublingual tablet system for rapid oromucosal absorption using fentanyl citrate as the active substance

    • Oromucosal delivery of drugs promotes rapid absorption and high bioavailability, with subsequent almost immediate onset of pharmacological effect. However, many oromucosal delivery systems are compromised by the possibility of the patient swallowing the active substance before it has been released and absorbed locally into the systemic circulation. This paper introduces a new tablet system for sublingual administration and rapid drug absorption. The tablet is based on interactive mixtures of components, consisting of carrier particles partially covered by fine dry particles of the drug, in this case fentanyl citrate. In the interests of increasing retention of the drug at the site of absorption in the oral cavity, a bioadhesive component was also added to the carrier particles. Tablets containing 100, 200 and 400 microg of fentanyl were tested both in vitro and in vivo. The tablets disintegrated rapidly and dissolution tests revealed that fentanyl citrate was dissolved from the formulation almost instantly. Plasma concentrations of fentanyl were obtained within 10 min, with no second peak. These results indicated that the bioadhesive component prevented the fentanyl from being swallowed (the fraction swallowed was considered smaller compared to other mucosal delivery systems), without hindering its release and absorption. This new sublingual tablet formulation may also hold potential for other substances where a rapid onset of effect is desirable.

    • We describe a spontaneously derived mouse line that completely failed to induce Ig class switching in vitro and in vivo. The mice inherited abolished IgG serum titers in a recessive manner caused by a spontaneous G -> A transition mutation in codon 112 of the aicda gene, leading to an arginine to histidine replacement (AID(R112H)). Ig class switching was completely reconstituted by expressing wild-type AID. Mice homozygous for AID(R112H) had peripheral B cell hyperplasia and large germinal centers in the absence of Ag challenge. Immunization with SRBCs elicited an Ag-specific IgG1 response in wild-type mice, whereas AID(R112H) mice failed to produce IgG1 and had reduced somatic hypermutation. The phenotype recapitulates the human hyper-IgM (HIGM) syndrome that is caused by point mutations in the orthologous gene in humans, and the AID(R112H) mutation is frequently found in HIGM patients. The AID(R112H) mouse model for HIGM provides a powerful and more precise tool than conventional knockout strategies.

    • By electrochemical deposition in a single nanopore membrane we fabricate Cu/Co layered single nanowires, that exhibit up to 10% magnetoresistance at room temperature. Single nanopore membranes are prepared by irradiating polycarbonate membranes with exactly one swift heavy ion, and by subsequent chemical etching of the single ion track. Both dc and pulsed electrodeposition of single wires consisting of Cu-Co alloy and Cu/Co multilayers respectively, are performed from a bath containing the two metal ions. By sputtering a gold electrode on the upper membrane surface, the single nanowire embedded in the flexible polymer foil is reliably contacted. While alloy wires exhibit anisotropic magnetoresistance (AMR), multilayer wires display current-perpendicular-to-plane giant magnetoresistance (CPP-GMR) behavior. This demonstrates that both the fabrication and contacting methods are very suitable for the investigation of transport properties, without the necessity of lithographic processes and without manipulation of the nanowires. In addition, the method opens up many new possibilities for single nanowire-based sensors.

    • Allergic asthma is a chronic inflammatory disease, characterized by airway hyperresponsiveness (AHR), inflammation, and tissue remodeling, in which mast cells play a central role. In the present study, we analyzed how mast cell numbers and localization influence the AHR in a chronic murine model of asthma. C57BL/6 (wild-type) and mast cell-deficient B6.Cg- KitW−shmice without (Wsh) and with (Wsh+MC) mast cell engraftment were sensitized to and subsequently challenged with ovalbumin for a 91-day period. In wild-type mice, pulmonary mast cells were localized in the submucosa of the central airways, whereas the more abundant mast cells in Wsh+MC mice were found mainly in the alveolar parenchyma. In Wsh+MC, ovalbumin challenge induced a relocation of mast cells from the perivascular space and central airways to the parenchyma. Allergen challenge caused a similar AHR in wild-type and Wsh mice in the resistance of the airways and the pulmonary tissue. In Wsh+MC mice the AHR was more pronounced. The elevated functional responses were partly related to the numbers and localization of connective tissue-type mast cells in the peripheral pulmonary compartments. A mast cell-dependent increase in IgE and IL-33 together with impairment of the IL-23/IL-17 axis was evoked in Wsh and Wsh+MC mice by allergen challenge. This study shows that within the same chronic murine asthma model the development of AHR can be both dependent and independent of mast cells. Moreover, the spatial distribution and number of pulmonary mast cells determine severity and localization of the AHR.

    • The next steps of deep space exploration are manned missions to Moon and Mars. For safe space missions for crew members, it is important to understand the impact of space flight on the immune system. We studied the effects of 21 days dry immersion (DI) exposure on the transcriptomes of T cells isolated from blood samples of eight healthy volunteers. Samples were collected 7 days before DI, at day 7, 14, and 21 during DI, and 7 days after DI. RNA sequencing of CD3(+) T cells revealed transcriptional alterations across all time points, with most changes occurring 14 days after DI exposure. At day 21, T cells showed evidence of adaptation with a transcriptional profile resembling that of 7 days before DI. At 7 days after DI, T cells again changed their transcriptional profile. These data suggest that T cells adapt by rewiring their transcriptomes in response to simulated weightlessness and that remodeling cues persist when reexposed to normal gravity.

    • We sought to identify genes necessary to induce cytoskeletal change in B cells. Using gene expression microarray, we compared B cells stimulated with interleukin-4 (IL-4) and anti-CD40 antibodies that induce B cell spreading, cell motility, tight aggregates, and extensive microvilli with B cells stimulated with lipopolysaccharide that lack these cytoskeletal changes. We identified 84 genes with 10-fold or greater expression in anti-CD40 + IL-4 stimulated B cells, one of these encoded the guanine nucleotide exchange factor (GEF) dedicator of cytokinesis 10 (Dock10). IL-4 selectively induced Dock10 expression in B cells. Using lacZ expression to monitor Dock10 promoter activity, we found that Dock10 was expressed at all stages during B cell development. However, specific deletion of Dock10 in B cells was associated with a mild phenotype with normal B cell development and normal B cell spreading, polarization, motility, chemotaxis, aggregation, and Ig class switching. Dock10-deficient B cells showed lower proliferation in response to anti-CD40 and IL-4 stimulation. Moreover, the IgG response to soluble antigen in vivo was lower when Dock10 was specifically deleted in B cells. Together, we found that most B cell responses were intact in the absence of Dock10. However, specific deletion of Dock10 in B cells was associated with a mild reduction in B cell activation in vitro and in vivo.

    • The Rho GTPases Cdc42, Rac1, and Rac2 coordinate receptor signaling to cell adhesion, migration, and proliferation. Deletion of Rac1 and Rac2 early during B cell development leads to failure in B cell entry into the splenic white pulp. Here, we sought to understand the role of Rac1 and Rac2 in B cell functionality and during the humoral antibody response. To circumvent the migratory deficiency of B cells lacking both Rac1 and Rac2, we took the approach to inducibly delete Rac1 in Rac2(-/-) B cells in the spleen (Rac1(B)Rac2(-/-) B cells). Rac1(B)Rac2(-/-) mice had normal differentiation of splenic B cell populations, except for a reduction in marginal zone B cells. Rac1(B)Rac2(-/-) B cells showed normal spreading response on antibody-coated layers, while both Rac2(-/-) and Rac1(B)Rac2(-/-) B cells had reduced homotypic adhesion and decreased proliferative response when compared to wild-type B cells. Upon challenge with the T-cell-independent antigen TNP-conjugated lipopolysaccharide, Rac1(B)Rac2(-/-) mice showed reduced antibody response. In contrast, in response to the T-cell-dependent antigen sheep red blood cells, Rac1(B)Rac2(-/-) mice had increased serum titers of IgG1 and IgG2b. During in vitro Ig class switching, Rac1(B)Rac2(-/-) B cells had elevated germline gamma 2b transcripts leading to increased Ig class switching to IgG2b. Our data suggest that Rac1 and Rac2 serve an important role in regulation of the B cell humoral immune response and in suppressing Ig class switching to IgG2b.

    • A new method to visualize and quantify grease flow in between two sealing lips or, in general, a double restriction seal is presented. Two setups were designed to mimic different types of seals; that is, a radial and an axial shaft seal. The flow of the grease inside and in between the sealing restrictions was measured using microparticle image velocimetry. The results show that grease flow due to a pressure difference mainly takes place close to the rotating shaft surface with an exponentially decaying velocity profile in the radial direction. Consequently, contaminants may be captured in the stationary grease at the outer radius, which explains the sealing function of the grease.

    • Particle accelerators have, during Operation with heavy ion beams, shown a significant pressure rise when the intensity of the beam is increased. This pressure rise is due to ion induced desorption, which is the result of beam ions colliding with residual gas atoms in the beam pipe, where they undergo charge exchange. This causes them to hit the vacuum chamber after the next dipole magnet and gas to be released. For the upgrade of the SIS18 synchrotron at GSI the intensity has to be a few orders of magnitude higher than it is today at the injection energy of 10 MeV/u. The aim of this experiment is to measure desorption yields, n, (released Molecules per incident ion) from materials commonly used in accelerators: 316LN stainless steel, Cu, etched Cu, gold coated Cu and Ta, using an Ar beam at impact energies in the range of 5-17.7 MeV/u for perpendicular incidence. The measured initial desorption yields vary for the same material from sample to sample: up to 4.5 times for stainless steel and Lip to 3 times for etched Cu. Therefore more samples should be Studied to have better statistics. Beam conditioning at lower energy does not significantly reduce the desorption yield at higher energy. There is a significant difference Of Lip to a few times in desorption yield between flat and tubular samples. The desorption yield from a Cu sample at grazing incident angle of 125 mrad was an order of magnitude larger than at normal incident angle. It Was found that the total number of positively and negatively charged secondary particles, emitted from the Surface bombarded with heavy ions, does not exceed -40 secondary particles per impact heavy ion. The current of negatively charged particles was about 2.3 times larger than the current for positively charged particles. The impact from secondary particles on dynamic gas pressure was not possible to investigate.

    • TiZrV nonevaporable getter (NEG) coated vacuum chambers is a new vacuum technology which is already used in many particle accelerators worldwide. This coating is also of interest for heavy-ion accelerator vacuum chambers. Heavy-ion desorption yields from an activated as well as a CO saturated NEG coated tube have been measured with 5 MeV/u Ar8+ beam. The sticking probability of the NEG film was obtained by using the partial pressure ratios on two sides of the NEG coated tube. These ratios were compared to results of modeling of the experimental setup with test particle Monte Carlo and angular coefficient methods. The partial pressures inside the saturated NEG coated tube bombarded with heavy ions were up to 20 times larger than those inside the activated one. However, the partial pressure of methane remained the same. The value of the total desorption yield from the activated NEG coated tube is 2600 molecules/ion. The desorption yields after saturation for CH4, H-2, and CO2 were found to be very close to the yields measured after the activation, while CO increased by up to a factor of 5. The total desorption yield for the saturated tube is up to 7000 molecules/ion. The large value of the desorption yield of the activated NEG coated tube, an order of magnitude higher than the desorption yield from a stainless steel tube at normal incident angle, could be explained by the grazing incident angle.

    • An experimental study of nuclear reactions between Si-28 nuclei at 200 and 300 MeV/nucleon and hydrogen or deuterium target nuclei was performed at the CELSIUS storage ring in Uppsala, Sweden, to collect information about the reactions responsible for single-event effects in microelectronics. Inclusive data on Si-28 fragmentation, as well as data on correlations between recoils and spectator protons or alpha particles are compared to predictions from the Dubna cascade model and the Japan Atomic Energy Research Institute version of the quantum molecular dynamics model. The comparison shows satisfactory agreement for inclusive data except for He fragments where low-energy sub-barrier fragments and recoiling fragments with very large momenta are produced much more frequently than predicted. The yield of exclusive data are also severely underestimated by the models whereas the charge distributions of recoils in these correlations compare well. The observed enhancement in He emission, which may well be important for the description of single-event effects, is most likely to be attributed to alpha clustering in Si-28 nuclei.

    • For the GSI future project Facility for Antiproton and Ion Research a beam intensity of 10(12)U(28+)ions/s is planned to be extracted from the GSI heavy ion synchrotron SIS18. Measurements performed in 2001 showed that the beam lifetime of the ions in the synchrotron is decreasing with increasing number of injected particles due to vacuum instabilities caused by ion-induced desorption. The injection energy for the SIS18 is about 10 MeV/u and U28+ ions are accelerated to 200 MeV/u limited by the magnetic rigidity for the low charge state. The aim of this work was to measure the desorption yield as a function of the impact energy from injection to extraction of SIS18 at GSI. Low energy yields at 5.0, 9.7, and 17.7 MeV/u were measured at the Cyclotron of The Svedberg Laboratory in Uppsala. High energy yields at 40, 80, and 100 MeV/u were measured at SIS18 of GSI in a different setup. It was found that the desorption yield scales with the electronic energy loss (dE/dx)(el)(n), with n between 2 and 3, decreasing for increasing impact energy above the Bragg maximum.

    • In deep vein thrombosis (DVT) of the lower limb, the location of the thrombosis determines how great the risk of pulmonary embolism is. We set out to determine whether the duration of lower limb DVT symptoms could be a predictive factor for the location of the thrombosis. We retrospectively collected a series of 100 lower limb phlebography examinations that were positive for DVT and divided the group into 2 subgroups proximal disease (ie, above the knee) or distal disease (ie, below the knee). The subgroups were then plotted against the individual duration of the symptoms. Of the 100 DVTs, 58% were proximal, while 42% were distal. The average duration of the symptoms was 6.3 days for patients with proximal disease and 6.2 days for patients with distal disease. We, therefore, found no difference in symptom duration between patients with proximal and distal lower limb DVT. This may imply that the duration of the DVT symptoms does not represent a predictive factor for DVT-caused pulmonary embolism.

  • Wiskott-Aldrich syndrome protein regulates autophagy and inflammasome activity in innate immune cells

    • Dysregulation of autophagy and inflammasome activity contributes to the development of auto-inflammatory diseases. Emerging evidence highlights the importance of the actin cytoskeleton in modulating inflammatory responses. Here we show that deficiency of Wiskott–Aldrich syndrome protein (WASp), which signals to the actin cytoskeleton, modulates autophagy and inflammasome function. In a model of sterile inflammation utilizing TLR4 ligation followed by ATP or nigericin treatment, inflammasome activation is enhanced in monocytes from WAS patients and in WAS-knockout mouse dendritic cells. In ex vivo models of enteropathogenic Escherichia coli and Shigella flexneri infection, WASp deficiency causes defective bacterial clearance, excessive inflammasome activation and host cell death that are associated with dysregulated septin cage-like formation, impaired autophagic p62/LC3 recruitment and defective formation of canonical autophagosomes. Taken together, we propose that dysregulation of autophagy and inflammasome activities contribute to the autoinflammatory manifestations of WAS, thereby identifying potential targets for therapeutic intervention.

  • µPIV measurement of grease velocity profiles in channels with two different types of flow restrictions

    • Grease is commonly used to lubricate various machine components such as rolling bearings and seals. In this paper the flow of lubricating grease passing restrictions is described. Such flow occurs in rolling bearings during relubrication events where the grease is flowing in the transverse (axial) direction through the bearing and is hindered by guide rings, flanges et cetera, as well as in seals where transverse flow occurs, for example during so-called breathing caused by temperature fluctuations in the bearing. This study uses a 2D flow model geometry consisting of a wide channel with rectangular cross-section and two different types of restrictions to measure the grease velocity vector field, using the method of Micro Particle Image Velocimetry. In the case of a single restriction, the horizontal distance required for the velocity profile to fully develop is approximately the same as the height of the channel. In the corner before and after the restriction, the velocities are very low and part of the grease is stationary. For the channel with two flow restrictions, this effect is even more pronounced in the “pocket” between the restrictions. Clearly, a large part of the grease is not moving. This condition particularly applies to the cases with a low-pressure drop and where high consistency grease is used. In practice this means that grease is not replaced in such “corners” and that some aged/contaminated grease will remain in seal pockets.

  • Decline in lichen biodiversity on oak trunks due to urbanization

    • Biodiversity often suffers from urbanization. In the present study, we focused on how the duration of urbanization affects the richness of 17 epiphytic lichen species and their cover on large oaks in urban environments in a city of 100 000 inhabitants in southeast Sweden. We also surveyed trees in adjacent rural areas, selected to have similar distributions of tree trunk circumference and surrounding oak density (within 300 m). Lichen richness and cover were lower on urban trees compared to rural trees. Furthermore, richness and cover decreased with the length of time that urban trees had been surrounded by houses. Most of the species that were analysed demonstrated a decline in occurrence with respect to the duration of housing development. The reduction in the probability of occurrence varied from 60% (Calicium viride, Evernia prunastri), 80% (Chrysothrix candelaris) to 90% (Ramalina spp.) during the considered 160-year period of urbanization. Therefore, even if valuable trees survive over the course of development, their lichen biota is likely to become depleted over time. © 2014 The Authors.

    • Muscle-specific kinase (MuSK) myasthenia gravis (MG) is a neuromuscular autoimmune disease belonging to a growing group of IgG4 autoimmune diseases (IgG4-AIDs), in which the majority of pathogenic autoantibodies are of the IgG4 subclass. The more prevalent form of MG with acetylcholine receptor (AChR) antibodies is caused by IgG1-3 autoantibodies. A dominant role for IgG4 in autoimmune disease is intriguing due to its antiinflammatory characteristics. It is unclear why MuSK autoantibodies are predominantly IgG4. We hypothesized that MuSK MG patients have a general predisposition to generate IgG4 responses, therefore resulting in high levels of circulating IgG4. To investigate this, we quantified serum Ig isotypes and IgG subclasses using nephelometric and turbidimetric assays in MuSK MG and AChR MG patients not under influence of immunosuppressive treatment. Absolute serum IgG1 was increased in both MuSK and AChR MG patients compared to healthy donors. In addition, only MuSK MG patients on average had significantly increased and enriched serum IgG4. Although more MuSK MG patients had elevated serum IgG4, for most the IgG4 serum levels fell within the normal range. Correlation analyses suggest MuSK-specific antibodies do not solely explain the variation in IgG4 levels. In conclusion, although serum IgG4 levels are slightly increased, the levels do not support ubiquitous IgG4 responses in MuSK MG patients as the underlying cause of dominant IgG4 MuSK antibodies.
