  • Analysis of Heritability and Shared Heritability Based on Genome-Wide Association Studies for 13 Cancer Types

    • Background: Studies of related individuals have consistently demonstrated notable familial aggregation of cancer. We aim to estimate the heritability and genetic correlation attributable to the additive effects of common single-nucleotide polymorphisms (SNPs) for cancer at 13 anatomical sites. Methods: Between 2007 and 2014, the US National Cancer Institute has generated data from genome-wide association studies (GWAS) for 49 492 cancer case patients and 34 131 control patients. We apply novel mixed model methodology (GCTA) to this GWAS data to estimate the heritability of individual cancers, as well as the proportion of heritability attributable to cigarette smoking in smoking-related cancers, and the genetic correlation between pairs of cancers. Results: GWAS heritability was statistically significant at nearly all sites, with the estimates of array-based heritability, h(l)(2), on the liability threshold (LT) scale ranging from 0.05 to 0.38. Estimating the combined heritability of multiple smoking characteristics, we calculate that at least 24% (95% confidence interval [CI] = 14% to 37%) and 7% (95% CI = 4% to 11%) of the heritability for lung and bladder cancer, respectively, can be attributed to genetic determinants of smoking. Most pairs of cancers studied did not show evidence of strong genetic correlation. We found only four pairs of cancers with marginally statistically significant correlations, specifically kidney and testes (rho = 0.73, SE = 0.28), diffuse large B-cell lymphoma (DLBCL) and pediatric osteosarcoma (rho = 0.53, SE = 0.21), DLBCL and chronic lymphocytic leukemia (CLL) (rho = 0.51, SE = 0.18), and bladder and lung (rho = 0.35, SE = 0.14). Correlation analysis also indicates that the genetic architecture of lung cancer differs between a smoking population of European ancestry and a nonsmoking Asian population, allowing for the possibility that the genetic etiology for the same disease can vary by population and environmental exposures. Conclusion: Our results provide important insights into the genetic architecture of cancers and suggest new avenues for investigation.

    • Genome-wide association studies (GWAS) have mapped risk alleles for at least 10 distinct cancers to a small region of 63 000 bp on chromosome 5p15.33. This region harbors the TERT and CLPTM1L genes; the former encodes the catalytic subunit of telomerase reverse transcriptase and the latter may play a role in apoptosis. To investigate further the genetic architecture of common susceptibility alleles in this region, we conducted an agnostic subset-based meta-analysis (association analysis based on subsets) across six distinct cancers in 34 248 cases and 45 036 controls. Based on sequential conditional analysis, we identified as many as six independent risk loci marked by common single-nucleotide polymorphisms: five in the TERT gene (Region 1: rs7726159, P = 2.10 × 10(-39); Region 3: rs2853677, P = 3.30 × 10(-36) and PConditional = 2.36 × 10(-8); Region 4: rs2736098, P = 3.87 × 10(-12) and PConditional = 5.19 × 10(-6), Region 5: rs13172201, P = 0.041 and PConditional = 2.04 × 10(-6); and Region 6: rs10069690, P = 7.49 × 10(-15) and PConditional = 5.35 × 10(-7)) and one in the neighboring CLPTM1L gene (Region 2: rs451360; P = 1.90 × 10(-18) and PConditional = 7.06 × 10(-16)). Between three and five cancers mapped to each independent locus with both risk-enhancing and protective effects. Allele-specific effects on DNA methylation were seen for a subset of risk loci, indicating that methylation and subsequent effects on gene expression may contribute to the biology of risk variants on 5p15.33. Our results provide strong support for extensive pleiotropy across this region of 5p15.33, to an extent not previously observed in other cancer susceptibility loci.

  • Characterization of Large Structural Genetic Mosaicism in Human Autosomes

    • Analyses of genome-wide association study (GWAS) data have revealed that detectable genetic mosaicism involving large (>2 Mb) structural autosomal alterations occurs in a fraction of individuals. We present results for a set of 24,849 genotyped individuals (total GWAS set II [TGSII]) in whom 341 large autosomal abnormalities were observed in 168 (0.68%) individuals. Merging data from the new TGSII set with data from two prior reports (the Gene-Environment Association Studies and the total GWAS set I) generated a large dataset of 127,179 individuals; we then conducted a meta-analysis to investigate the patterns of detectable autosomal mosaicism (n = 1,315 events in 925 [0.73%] individuals). Restricting to events >2 Mb in size, we observed an increase in event frequency as event size decreased. The combined results underscore that the rate of detectable mosaicism increases with age (p value = 5.5 x 3 10(-31)) and is higher in men (p value = 0.002) but lower in participants of African ancestry (p value = 0.003). In a subset of 47 individuals from whom serial samples were collected up to 6 years apart, complex changes were noted over time and showed an overall increase in the proportion of mosaic cells as age increased. Our large combined sample allowed for a unique ability to characterize detectable genetic mosaicism involving large structural events and strengthens the emerging evidence of non-random erosion of the genome in the aging population.

    • To investigate large structural clonal mosaicism of chromosome X, we analysed the SNP microarray intensity data of 38,303 women from cancer genome-wide association studies (20,878 cases and 17,425 controls) and detected 124 mosaic X events >2 Mb in 97 (0.25%) women. Here we show rates for X-chromosome mosaicism are four times higher than mean autosomal rates; X mosaic events more often include the entire chromosome and participants with X events more likely harbour autosomal mosaic events. X mosaicism frequency increases with age (0.11% in 50-year olds; 0.45% in 75-year olds), as reported for Y and autosomes. Methylation array analyses of 33 women with X mosaicism indicate events preferentially involve the inactive X chromosome. Our results provide further evidence that the sex chromosomes undergo mosaic events more frequently than autosomes, which could have implications for understanding the underlying mechanisms of mosaic events and their possible contribution to risk for chronic diseases.

    • Vanadium nitride (VN) peas and cubes were synthesized by regulating the molar ratio of the starting materials (urea/VOCl3) via the urea-metal chloride route. The as-prepared VN compounds were subsequently introduced into dye-sensitized solar cells (DSCs) as counter electrode (CE) catalysts for the regeneration of novel organic thiolate/disulfide (T-/T-2) and traditional iodide/triiodide (I-/I-3(-))redox couples. The cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS), and Tafel polarization curve results proved that the catalytic activity of the prepared VN was significantly affected by particle shape and particle size. The VN peas showed the highest catalytic activity, followed by the small VN cubes and large VN cubes. The I-/I-3(-)-electrolyte-based DSCs yielded a high power conversion efficiency (PCE) of 7.29%. The Pt-free VN CE catalysts are more suitable for the new organic redox couples of T-/T-2. The DSCs based on VN peas CE showed a PCE of 5.57%, an enhancement of 40.7% relative to the Pt-CE-based DSCs (3.96%).

  • Expanding the mutation and phenotype spectrum of MYH3-associated skeletal disorders

    • Pathogenic variants in MYH3 cause distal arthrogryposis type 2A and type 2B3 as well as contractures, pterygia and spondylocarpotarsal fusion syndromes types 1A and 1B. These disorders are ultra-rare and their natural course and phenotypic variability are not well described. In this study, we summarize the clinical features and genetic findings of 17 patients from 10 unrelated families with vertebral malformations caused by dominant or recessive pathogenic variants in MYH3. Twelve novel pathogenic variants in MYH3 (NM_002470.4) were identified: three of them were de novo or inherited in autosomal dominant way and nine were inherited in autosomal recessive way. The patients had vertebral segmentation anomalies accompanied with variable joint contractures, short stature and dysmorphic facial features. There was a significant phenotypic overlap between dominant and recessive MYH3-associated conditions regarding the degree of short stature as well as the number of vertebral fusions. All monoallelic variants caused significantly decreased SMAD3 phosphorylation, which is consistent with the previously proposed pathogenic mechanism of impaired canonical TGF-β signaling. Most of the biallelic variants were predicted to be protein-truncating, while one missense variant c.4244T>G,p.(Leu1415Arg), which was inherited in an autosomal recessive way, was found to alter the phosphorylation level of p38, suggesting an inhibition of the non-canonical pathway of TGF-β signaling. In conclusion, the identification of 12 novel pathogenic variants and overlapping phenotypes in 17 affected individuals from 10 unrelated families expands the mutation and phenotype spectrum of MYH3-associated skeletal disorders. We show that disturbances of canonical or non-canonical TGF-β signaling pathways are involved in pathogenesis of MYH3-associated skeletal fusion (MASF) syndrome.

    • Colorectal cancer risk can be impacted by genetic, environmental, and lifestyle factors, including diet and obesity. Gene-environment interactions (G × E) can provide biological insights into the effects of obesity on colorectal cancer risk. Here, we assessed potential genome-wide G × E interactions between body mass index (BMI) and common SNPs for colorectal cancer risk using data from 36,415 colorectal cancer cases and 48,451 controls from three international colorectal cancer consortia (CCFR, CORECT, and GECCO). The G × E tests included the conventional logistic regression using multiplicative terms (one degree of freedom, 1DF test), the two-step EDGE method, and the joint 3DF test, each of which is powerful for detecting G × E interactions under specific conditions. BMI was associated with higher colorectal cancer risk. The two-step approach revealed a statistically significant G×BMI interaction located within the Formin 1/Gremlin 1 (FMN1/GREM1) gene region (rs58349661). This SNP was also identified by the 3DF test, with a suggestive statistical significance in the 1DF test. Among participants with the CC genotype of rs58349661, overweight and obesity categories were associated with higher colorectal cancer risk, whereas null associations were observed across BMI categories in those with the TT genotype. Using data from three large international consortia, this study discovered a locus in the FMN1/GREM1 gene region that interacts with BMI on the association with colorectal cancer risk. Further studies should examine the potential mechanisms through which this locus modifies the etiologic link between obesity and colorectal cancer.SIGNIFICANCE: This gene-environment interaction analysis revealed a genetic locus in FMN1/GREM1 that interacts with body mass index in colorectal cancer risk, suggesting potential implications for precision prevention strategies.

  • Bcl3 Couples Cancer Stem Cell Enrichment With Pancreatic Cancer Molecular Subtypes

    • Background & Aims: The existence of different subtypes of pancreatic ductal adenocarcinoma (PDAC) and their correlation with patient outcome have shifted the emphasis on patient classification for better decision-making algorithms and personalized therapy. The contribution of mechanisms regulating the cancer stem cell (CSC) population in different subtypes remains unknown.Methods: Using RNA-seq, we identified B-cell CLL/lymphoma 3 (BCL3), an atypical nf-κb signaling member, as differing in pancreatic CSCs. To determine the biological consequences of BCL3 silencing in vivo and in vitro, we generated bcl3-deficient preclinical mouse models as well as murine cell lines and correlated our findings with human cell lines, PDX models, and 2 independent patient cohorts. We assessed the correlation of bcl3 expression pattern with clinical parameters and subtypes.Results: Bcl3 was significantly down-regulated in human CSCs. Recapitulating this phenotype in preclinical mouse models of PDAC via BCL3 genetic knockout enhanced tumor burden, metastasis, epithelial to mesenchymal transition, and reduced overall survival. Fluorescence-activated cell sorting analyses, together with oxygen consumption, sphere formation, and tumorigenicity assays, all indicated that BCL3 loss resulted in CSC compartment expansion promoting cellular dedifferentiation. Overexpression of BCL3 in human PDXs diminished tumor growth by significantly reducing the CSC population and promoting differentiation. Human PDACs with low BCL3 expression correlated with increased metastasis, and BCL3-negative tumors correlated with lower survival and nonclassical subtypes.Conclusions: We demonstrate that bcl3 impacts pancreatic carcinogenesis by restraining CSC expansion and by curtailing an aggressive and metastatic tumor burden in PDAC across species. Levels of BCL3 expression are a useful stratification marker for predicting subtype characterization in PDAC, thereby allowing for personalized therapeutic approaches.

    • We prove the integrability of the two-loop open spin chain Hamiltonian from ABJM determinant like operators given in [1]. By explicitly constructing R-matrices and K-matrices, we successfully obtain the two-loop Hamiltonian from the double row transfer matrices. This proves the integrability of our two-loop Hamiltonian. Based on the vacuum eigenvalues of the transfer matrices, we make a conjecture on the eigenvalues of the transfer matrices for general excited states. Bethe ansatz equations are simply obtained from the analytic conditions at the superficial poles of the eigenvalues.

    • Respiration rate is an important healthcare indicator, and it has become a popular research topic in remote healthcare applications with Internet of Things. Existing respiration monitoring systems have limitations in terms of convenience, comfort, and privacy, etc. This paper presents a contactless and real-time respiration monitoring system, the so-called Wi-Breath, based on off-the-shelf WiFi devices. The system monitors respiration with both the amplitude and phase difference of the WiFi channel state information (CSI), which is sensitive to human body micro movement. The phase information of the CSI signal is considered and both the amplitude and phase difference are used. For better respiration detection accuracy, a signal selection method is proposed to select an appropriate signal from the amplitude and phase difference based on a support vector machine (SVM) algorithm. Experimental results demonstrate that the Wi-Breath achieves an accuracy of 91.2% for respiration detection, and has a 17.0% reduction in average error in comparison with state-of-the-art counterparts.

    • We report the generation and analysis of functional data from multiple, diverse experiments performed on a targeted 1% of the human genome as part of the pilot phase of the ENCODE Project. These data have been further integrated and augmented by a number of evolutionary and computational analyses. Together, our results advance the collective knowledge about human genome function in several major areas. First, our studies provide convincing evidence that the genome is pervasively transcribed, such that the majority of its bases can be found in primary transcripts, including non-protein-coding transcripts, and those that extensively overlap one another. Second, systematic examination of transcriptional regulation has yielded new understanding about transcription start sites, including their relationship to specific regulatory sequences and features of chromatin accessibility and histone modification. Third, a more sophisticated view of chromatin structure has emerged, including its inter-relationship with DNA replication and transcriptional regulation. Finally, integration of these new sources of information, in particular with respect to mammalian evolution based on inter- and intra-species sequence comparisons, has yielded new mechanistic and evolutionary insights concerning the functional landscape of the human genome. Together, these studies are defining a path for pursuit of a more comprehensive characterization of human genome function.

    • The effect of post-manufacture heat-treatment on the mechanical strength of an additively-manufactured austenitic stainless steel has been investigated. Microstructural investigations revealed that the as-manufactured material exhibited a multi-scale structure, composed of grains, cells, dislocations and nano-sized particles. Annealing at 400 degrees C resulted in a 10% increase in yield strength, associated with the additional precipitation of a population of nano-sized silicates. Annealing at higher temperatures resulted in a decrease in strength, attributed primarily to the thermal instability of the cell structure in the as-manufactured material. The results demonstrate that by careful control of annealing conditions the structure and mechanical properties of additively-manufactured austenitic stainless steel can be optimized by post-manufacture heat-treatment.

    • A native interfacial doping layer introduced in core-shell type ZnO nanowires by a simple vapor phase re-growth procedure endows the produced nanowires with both excellent electrical and optical performances compared to conventional homogeneous ZnO nanowires.     • Nitrogen (N) and phosphorus (P) limitation constrains the magnitude of terrestrial carbon uptake in response to elevated carbon dioxide and climate change. However, global maps of nutrient limitation are still lacking. Here we examined global N and P limitation using the ratio of site-averaged leaf N and P resorption efficiencies of the dominant species across 171 sites. We evaluated our predictions using a global database of N- and P-limitation experiments based on nutrient additions at 106 and 53 sites, respectively. Globally, we found a shift from relative P to N limitation for both higher latitudes and precipitation seasonality and lower mean annual temperature, temperature seasonality, mean annual precipitation and soil clay fraction. Excluding cropland, urban and glacial areas, we estimate that 18% of the natural terrestrial land area is significantly limited by N, whereas 43% is relatively P limited. The remaining 39% of the natural terrestrial land area could be co-limited by N and P or weakly limited by either nutrient alone. This work provides both a new framework for testing nutrient limitation and a benchmark of N and P limitation for models to constrain predictions of the terrestrial carbon sink.

    • The ability to tune magnetic orders, such as magnetic anisotropy and topological spin texture, is desired to achieve high-performance spintronic devices. A recent strategy has been to employ interfacial engineering techniques, such as the introduction of spin-correlated interfacial coupling, to tailor magnetic orders and achieve novel magnetic properties. We chose a unique polar-nonpolar LaMnO3/SrIrO3 superlattice because Mn (3d)/Ir (5d) oxides exhibit rich magnetic behaviors and strong spin-orbit coupling through the entanglement of their 3d and 5d electrons. Through magnetization and magnetotransport measurements, we found that the magnetic order is interface-dominated as the superlattice period is decreased. We were able to then effectively modify the magnetization, tilt of the ferromagnetic easy axis, and symmetry transition of the anisotropic magnetoresistance of the LaMnO3/SrIrO3 superlattice by introducing additional Mn (3d) and Ir (5d) interfaces. Further investigations using in-depth first-principles calculations and numerical simulations revealed that these magnetic behaviors could be understood by the 3d/5d electron correlation and Rashba spin-orbit coupling. The results reported here demonstrate a new route to synchronously engineer magnetic properties through the atomic stacking of different electrons, which would contribute to future applications in high-capacity storage devices and advanced computing.

We observed mosaic abnormalities, either aneuploidy or copy-neutral loss of heterozygosity, of >2 Mb in size in autosomes of 517 individuals (0.89%), with abnormal cell proportions of between 7% and 95%. In cancer-free individuals, frequency increased with age, from 0.23% under 50 years to 1.91% between 75 and 79 years (P = 4.8 × 10(-8)). Mosaic abnormalities were more frequent in individuals with solid tumors (0.97% versus 0.74% in cancer-free individuals; odds ratio (OR) = 1.25; P = 0.016), with stronger association with cases who had DNA collected before diagnosis or treatment (OR = 1.45; P = 0.0005). Detectable mosaicism was also more common in individuals for whom DNA was collected at least 1 year before diagnosis with leukemia compared to cancer-free individuals (OR = 35.4; P = 3.8 × 10(-11)). These findings underscore the time-dependent nature of somatic events in the etiology of cancer and potentially other late-onset diseases.

    • The electrolysis processes such as hydrogen evolution reaction (HER) require high efficient catalysts with robust surface stability. A high conductivity is also necessary to speed up the charge transport between the catalyst and the electrolyte. Recently, the observation of exceedingly high conductivity in the topological semimetal MoP, has provided a model catalyst to investigate the correlation between the electrical transport and the electrocatalytic activity for the HER. Thus, MoP is encapsulated in a Mo, P codoped carbon layer (MoP@C). This composite material exhibits outstanding HER performance, with an extremely low overpotential of 49 mV at a current density of 10 mA cm(-2) and a Tafel slope of 54 mV dec(-1) in an alkaline medium. In addition, electron transport analysis indicates that MoP exhibits high conductivity and mobility due to the existence of triple-point fermions and a complex Fermi surface. Furthermore, the presence of P-C and Mo-C bonds at the interface between the carbon layer and the MoP particles modulates the band structure of MoP@C and facilitates fast electron transfer, accumulation, and subsequent delocalization, which are in turn responsible for the excellent HER activity.

    • In bioethanol production, the main by-product, 5-hydroxymethylfurfural (HMF), significantly hinders microbial fermentation. Therefore, it is crucial to explore genes related to HMF tolerance in Saccharomyces cerevisiae for enhancing the tolerance of ethanol fermentation strains. A comprehensive analysis was conducted using genome-wide deletion library scanning and SGAtools, resulting in the identification of 294 genes associated with HMF tolerance in S. cerevisiae. Further KEGG and GO enrichment analysis revealed the involvement of genes OCA1 and SIW14 in the protein phosphorylation pathway, underscoring their role in HMF tolerance. Spot test validation and subcellular structure observation demonstrated that, following a 3-h treatment with 60mM HMF, the SIW14 gene knockout strain exhibited a 12.68% increase in cells with abnormal endoplasmic reticulum (ER) and a 22.41% increase in the accumulation of reactive oxygen species compared to the BY4741 strain. These findings indicate that the SIW14 gene contributes to the protection of the ER structure within the cell and facilitates the clearance of reactive oxygen species, thereby confirming its significance as a key gene for HMF tolerance in S. cerevisiae.

    • <i>Objectives:</i> Eight genotypes (A–H) of hepatitis B virus (HBV) are known with variations in nucleotide sequences greater than 8%. Several recent publications found that the clinical course and outcome of antiviral therapy depended on the genotype of the infecting HBV strain. Large epidemiological studies will require the availability of a system which is rapid, reliable and can be performed on a large number of samples. <i>Methods:</i> To establish a simple and accurate genotyping method, the study collected 369 HBV complete genomic sequences from the GenBank database. Type-specific primers were also designed that separated HBV genotypes A to G by multiplex polymerase chain reaction. <i>Results:</i> By comparison with the traditional restriction fragment length polymorphism method, over 93% of 441 samples were accurately genotyped by current assay, with a higher detection rate and sensitivity to detect mixed HBV infections. <i>Conclusions:</i> This methodology can be applied only to areas prevalent with HBV genotypes A to G. However, it provides an efficient alternative for clinical diagnosis and large-scale studies.

    • Reconfigurable Intelligent Surfaces (RISs) have great potential for Internet of Things (IoT) applications due to their cost-effectiveness and energy-efficiency. However, under the application of RIS, current RIS systems cannot actively predict and avoid interference, as evolving interference signals can dynamically change their frequency bands. To address this issue, we develop a learning-based dynamic RIS spectrum access scheme to avoid interference and enhance wireless transmission reliability. An intelligent metasurface-based modulation (MM) is designed for generating and reflecting anti-interference radio-frequency signals. Specifically, a Dyna-Q algorithm is employed at the RIS controller to learn the frequency of interference signals, which is used to determine the frequency of the carrier signals. Experiments are conducted on the proposed RIS demo system to evaluate its anti-interference capability, and the results show that our scheme leads to an increase of at least 50% in the system throughput. These numerical results demonstrate the effectiveness of proposed scheme.

    • Background: Risk variants identified so far for colorectal cancer explain only a small proportion of milial risk of this cancer, particularly in Asians.Methods: We performed a genome-wide association study (GWAS) of colorectal cancer in East Asians, cluding 23,572 colorectal cancer cases and 48,700 controls. To identify novel risk loci, we selected 60 omising risk variants for replication using data from 58,131 colorectal cancer cases and 67,347 controls European descent. To identify additional risk variants in known colorectal cancer loci, we performed nditional analyses in East Asians.Results: An indel variant, rs67052019 at 1p13.3, was found to be associated with colorectal cancer risk P = 3.9 x 10(-8) in Asians (OR per allele deletion = 1.13, 95% confidence interval = 1.08-1.18). This sociation was replicated in European descendants using a variant (rs2938616) in complete linkage sequilibrium with rs67052019 (P = 7.7 x 10(-3)). Of the remaining 59 variants, 12 showed an association P < 0.05 in the European-ancestry study, including rs11108175 and rs9634162 at P < 5 x 10(-8) and o variants with an association near the genome-wide significance level (rs60911071, P = 5.8 x 10(-8); 62558833, P = 7.5 x 10(-8)) in the combined analyses of Asian- and European-ancestry data. In addition, ing data from East Asians, we identified 13 new risk variants at 11 loci reported from previous GWAS.Conclusions: In this large GWAS, we identified three novel risk loci and two highly suggestive loci for lorectal cancer risk and provided evidence for potential roles of multiple genes and pathways in the iology of colorectal cancer. In addition, we showed that additional risk variants exist in many colorectal ncer risk loci identified previously.Impact: Our study provides novel data to improve the understanding of the genetic basis for colorectal ncer risk.

    • Exploring the future hydroclimatic conditions of source region of Yangtze River (SRYaR), an alpine affected by climate change significantly, is essential for basin water resources management and development ss global climate change intensifies and the process of climate warming and humidification in Northwest China. This study proposed a practical framework for assessing water resource response to the context of climate changes in alpine catchments from the respective of both runoff and hydroclimatic conditions. Utilizing Grid-RCCC-WBM driven by corrected climatic forcing from the global climate models, this study estimate the prospective overall warmer and wetter pattern in the source region of Yangtze River. The key results indicated that: (1) Under all future scenarios, both temperature and precipitation within the catchment exhibit a significant upward trend. Projections from multi-model ensembles (MME) suggest that during the mid-term period (2041–2060, MT), temperatures are expected to rise by [0.74 °C, 3.08 °C] compared to the baseline period (1995–2014), with precipitation changes ranging from [4.8%, 21.4%]. (2) Future runoff within the catchment exhibits a consistent increase, with a linear trend rate of 1.1 mm/decade. runoff changes in MT compared to the baseline period vary from [−5.1%, 33.7%]. Runoff decreases in the northern part of the catchment, while notable increases occur in the southeastern and western regions. (3) In the future, the ratio of catchment evaporation capacity to precipitation decreases in comparison to the baseline period with an augmentation in soil moisture, enhancing its capacity for water retention and reducing the conversion of precipitation to evaporation, resulting a wetting trend of the catchment. (4) The future snowpack in the catchment continues to decrease, with a significant reduction in both the proportion of snowfall relative to total precipitation and the proportion of snowmelt runoff relative to total runoff, the risk of water resources crisis in the watershed is escalating.

    • This REgional Carbon Cycle Assessment and Processes regional study provides a synthesis of the carbon balance of terrestrial ecosystems in East Asia, a region comprised of China, Japan, North and South Korea, and Mongolia. We estimate the current terrestrial carbon balance of East Asia and its driving mechanisms during 1990-2009 using three different approaches: inventories combined with satellite greenness measurements, terrestrial ecosystem carbon cycle models and atmospheric inversion models. The magnitudes of East Asia's terrestrial carbon sink from these three approaches are comparable: -0.293 +/- 0.033 PgC yr(-1) from inventory-remote sensing model-data fusion approach, -0.413 +/- 0.141 PgC yr(-1)(not considering biofuel emissions) or -0.224 +/- 0.141 PgC yr(-1) (considering biofuel emissions) for carbon cycle models, and -0.270 +/- 0.507 PgC yr(-1) for atmospheric inverse models. Here and in the following, the numbers behind +/- signs are standard deviations. The ensemble of ecosystem modeling based analyses further suggests that at the regional scale, climate change and rising atmospheric CO2 together resulted in a carbon sink of -0.289 +/- 0.135 PgC yr(-1), while land-use change and nitrogen deposition had a contribution of -0.013 +/- 0.029 PgC yr(-1) and -0.107 +/- 0.025 PgC yr(-1), respectively. Although the magnitude of climate change effects on the carbon balance varies among different models, all models agree that in response to climate change alone, southern China experienced an increase in carbon storage from 1990 to 2009, while northern East Asia including Mongolia and north China showed a decrease in carbon storage. Overall, our results suggest that about 13-27% of East Asia's CO2 emissions from fossil fuel burning have been offset by carbon accumulation in its terrestrial territory over the period from 1990 to 2009. The underlying mechanisms of carbon sink over East Asia still remain largely uncertain, given the diversity and intensity of land management processes, and the regional conjunction of many drivers such as nutrient deposition, climate, atmospheric pollution and CO2 changes, which cannot be considered as independent for their effects on carbon storage.

    • A heterogeneous unmanned aerial vehicle (UAV) network where UAVs carrying different resources form coalition and cooperatively carry out tasks is of crucial importance for fulfilling diverse tasks. However, the existing coalition formation (CF) game model only optimizes the composition of UAVs in a single coalition, which results in disjoined coalitions. In order to tackle this issue, a sequential overlapping coalition formation (OCF) game is proposed by considering the overlapping and complementary relations of resource properties and the task execution order. Moreover, different from the Pareto and selfish orders, a bilateral mutual benefit transfer (BMBT) order is proposed to optimize the cooperative task resource allocation through partial cooperation among overlapping coalition members. Furthermore, using the preference relation between UAVs carrying resources and tasks requiring the same type of resource, a preference gravity-guided tabu search (PGG-TS) algorithm is developed to obtain a stable coalition structure. Numerical results verify that the proposed PGG-TS algorithm increases the average utility of tasks by 12.5% and 38.5% compared with the split-merge preferred OCF algorithm and non-overlapping CF algorithm, respectively. The utility of the proposed BMBT order increases by 25.1% and 34.3% compared with selfish and Pareto orders, respectively.

    • In adversarial scenarios, it is crucial to timely monitor what tactical messages that opponent transmitters are sending to intended receiver(s), and disrupt the transmissions immediately if in need. The issue becomes more challenging in face of an intelligent transmitter. To address the above-stated issue, a full-duplex (FD) technique is utilized to enable simultaneous jamming and eavesdropping (SJE) at a friendly jammer node. In particular, the "Two Birds with One Stone" strategy is utilized at the jammer node to realize effective rate degradation and information eavesdropping. A confrontation game between an intelligence-empowered FD jammer and its opponent is investigated. Specifically, to capture their adversarial relationship in an environment with incomplete information, a power-domain Bayesian-Stackelberg game is proposed. The existence of a Stackelberg equilibrium (SE) power solution is proved. The semi-closed-form solutions of SE are derived, which are proved to be asymptotically optimal (have a gap of less than 1% with the exact utility), and improves the jammer node 10% utility compared with the Nash equilibrium. Additionally, the SJE strategy outperforms the half-duplex (HD) and other benchmark schemes.

    • Background: Menopausal hormone therapy (MHT), a common treatment to relieve symptoms of menopause, is associated with a lower risk of colorectal cancer (CRC). To inform CRC risk prediction and MHT risk-benefit assessment, we aimed to evaluate the joint association of a polygenic risk score (PRS) for CRC and MHT on CRC risk.Methods: We used data from 28,486 postmenopausal women (11,519 cases and 16,967 controls) of European descent. A PRS based on 141 CRC-associated genetic variants was modeled as a categorical variable in quartiles. Multiplicative interaction between PRS and MHT use was evaluated using logistic regression. Additive interaction was measured using the relative excess risk due to interaction (RERI). 30-year cumulative risks of CRC for 50-year-old women according to MHT use and PRS were calculated.Results: The reduction in odds ratios by MHT use was larger in women within the highest quartile of PRS compared to that in women within the lowest quartile of PRS (p-value = 2.7 × 10−8). At the highest quartile of PRS, the 30-year CRC risk was statistically significantly lower for women taking any MHT than for women not taking any MHT, 3.7% (3.3%–4.0%) vs 6.1% (5.7%–6.5%) (difference 2.4%, P-value = 1.83 × 10−14); these differences were also statistically significant but smaller in magnitude in the lowest PRS quartile, 1.6% (1.4%–1.8%) vs 2.2% (1.9%–2.4%) (difference 0.6%, P-value = 1.01 × 10−3), indicating 4 times greater reduction in absolute risk associated with any MHT use in the highest compared to the lowest quartile of genetic CRC risk.Conclusions: MHT use has a greater impact on the reduction of CRC risk for women at higher genetic risk. These findings have implications for the development of risk prediction models for CRC and potentially for the consideration of genetic information in the risk-benefit assessment of MHT use.

    • BACKGROUND: The use of menopausal hormone therapy (MHT) may interact with genetic variants to influence colorectal cancer (CRC) risk. METHODS: We conducted a genome-wide, gene-environment interaction between single nucleotide polymorphisms and the use of any MHT, estrogen only, and combined estrogen-progestogen therapy with CRC risk, among 28 486 postmenopausal women (11 519 CRC patients and 16 967 participants without CRC) from 38 studies, using logistic regression, 2-step method, and 2- or 3-degree-of-freedom joint test. A set-based score test was applied for rare genetic variants. RESULTS: The use of any MHT, estrogen only and estrogen-progestogen were associated with a reduced CRC risk (odds ratio [OR] = 0.71, 95% confidence interval [CI] = 0.64 to 0.78; OR = 0.65, 95% CI = 0.53 to 0.79; and OR = 0.73, 95% CI = 0.59 to 0.90, respectively). The 2-step method identified a statistically significant interaction between a GRIN2B variant rs117868593 and MHT use, whereby MHT-associated CRC risk was statistically significantly reduced in women with the GG genotype (OR = 0.68, 95% CI = 0.64 to 0.72) but not within strata of GC or CC genotypes. A statistically significant interaction between a DCBLD1 intronic variant at 6q22.1 (rs10782186) and MHT use was identified by the 2-degree-of-freedom joint test. The MHT-associated CRC risk was reduced with increasing number of rs10782186-C alleles, showing odds ratios of 0.78 (95% CI = 0.70 to 0.87) for TT, 0.68 (95% CI = 0.63 to 0.73) for TC, and 0.66 (95% CI = 0.60 to 0.74) for CC genotypes. In addition, 5 genes in rare variant analysis showed suggestive interactions with MHT (2-sided P < 1.2 × 10-4). CONCLUSION: Genetic variants that modify the association between MHT and CRC risk were identified, offering new insights into pathways of CRC carcinogenesis and potential mechanisms involved.

    • BACKGROUND: The literature on associations of circulating concentrations of minerals and vitamins with risk of colorectal cancer is limited and inconsistent. Evidence from randomized controlled trials (RCTs) to support the efficacy of dietary modification or nutrient supplementation for colorectal cancer prevention is also limited.OBJECTIVES: To complement observational and RCT findings, we investigated associations of genetically predicted concentrations of 11 micronutrients (β-carotene, calcium, copper, folate, iron, magnesium, phosphorus, selenium, vitamin B-6, vitamin B-12, and zinc) with colorectal cancer risk using Mendelian randomization (MR). METHODS: Two-sample MR was conducted using 58,221 individuals with colorectal cancer and 67,694 controls from the Genetics and Epidemiology of Colorectal Cancer Consortium, Colorectal Cancer Transdisciplinary Study, and Colon Cancer Family Registry. Inverse variance-weighted MR analyses were performed with sensitivity analyses to assess the impact of potential violations of MR assumptions.RESULTS: Nominally significant associations were noted for genetically predicted iron concentration and higher risk of colon cancer [ORs per SD (ORSD): 1.08; 95% CI: 1.00, 1.17; P value = 0.05] and similarly for proximal colon cancer, and for vitamin B-12 concentration and higher risk of colorectal cancer (ORSD: 1.12; 95% CI: 1.03, 1.21; P value = 0.01) and similarly for colon cancer. A nominally significant association was also noted for genetically predicted selenium concentration and lower risk of colon cancer (ORSD: 0.98; 95% CI: 0.96, 1.00; P value = 0.05) and similarly for distal colon cancer. These associations were robust to sensitivity analyses. Nominally significant inverse associations were observed for zinc and risk of colorectal and distal colon cancers, but sensitivity analyses could not be performed. None of these findings survived correction for multiple testing. Genetically predicted concentrations of β-carotene, calcium, copper, folate, magnesium, phosphorus, and vitamin B-6 were not associated with disease risk.CONCLUSIONS: These results suggest possible causal associations of circulating iron and vitamin B-12 (positively) and selenium (inversely) with risk of colon cancer.

However, the LTE network still faces the problem of load imbalance as in GSM/WCDMA networks, and this may cause significant deterioration of system performance. To deal with this problem, mobility load balancing (MLB) has been proposed as an important use case in 3GPP self-organizing network (SON), in which the serving cell of a user can be selected to achieve load balancing rather than act as the cell with the maximum received power. Furthermore, the LTE network aims to serve users with different quality-of-service (QoS) requirements, and the network-wide objective function for load balancing is distinct for different kinds of users. Thus, in this paper, a unified algorithm is proposed for MLB in the LTE network. The load balancing problem is first formulated as an optimization problem with the optimizing variables being cell-user connections. Then the complexity and overhead of the optimal solution is analyzed and a practical and distributed algorithm is given. After that, the proposed algorithm is evaluated for users with different kinds of QoS requirements, i.e., guaranteed bit rate (GBR) users with the objective function of load balance index and non-GBR (nGBR) users with the objective function of total utility, respectively. Simulation results show that the proposed algorithm leads to significantly balanced load distribution for GBR users to decrease the new call blocking rate, and for nGBR users to improve the cell-edge throughput at the cost of only slight deterioration of total throughput.

    • In this letter, we investigate call admission control (CAC) for cumulative rate distribution based scheduling (CS) in wireless communication networks. We first analyze the multi-user diversity gain (MDG) of CS, which is applicable to general channel conditions. Then we propose the minimum rate guaranteed CS/ORR based CAC algorithm, COCAC, which jointly uses CS and opportunistic round robin (ORR) for resource prediction and CAC decision. Furthermore, we evaluate the performance of our COCAC algorithm through simulation. Results show that COCAC can significantly reduce the new call blocking rate, while strictly guarantee the minimum rate requirements of all serving users.

  • Three novel opportunistic scheduling algorithms in CoMP-CSB scenario

    • Coordinated scheduling/beamforming (CSB), which belongs to the coordinated multi-point (CoMP) transmission, has received lots of attention recently due to its great potential to mitigate inter-cell interference (ICI) and to increase the cell-edge throughput, and meanwhile it only requires limited base station cooperation and is easy to implement. However, to the best of our knowledge, there are no effective scheduling algorithms with low complexity and overhead in CoMP-CSB scenario as yet. Thus, in this paper, we propose three novel opportunistic scheduling algorithms in CoMP-CSB scenario. All of them jointly consider the intended channel condition of the scheduled user from its serving cell and the orthogonality between the intended channel and the corresponding interference channels to concurrently scheduled users in nearby cells, thus exploiting multi-user diversity (MUD) and mitigating ICI at the same time. Algorithm 1 cooperatively chooses the most orthogonal user pair within a candidate user set in which all users have a large local channel feedback, while Algorithm 2 concurrently schedules the user pair with the largest ratio between the local channel feedbacks and the aforementioned orthogonality within the same candidate user set. Algorithm 3 performs in the way similar to the proportional fairness scheduling, while making a proper modification for its usage in CoMP-CSB scenario. The performance of the proposed scheduling algorithms are evaluated through simulation. Results show that, they all can significantly enhance the received signal to interference plus noise ratio (SINR) with relatively good fairness guarantee, thus achieving larger throughputs and utilities than several well-known scheduling algorithms. Algorithm 2 even outperforms Algorithm 1 when the aforementioned candidate user set is big enough in size and has a bit more overhead/complexity. Furthermore, Algorithms 3 is the best one among all the three proposed algorithms, but it requires more overhead/complexity than Algorithm 1 and 2. Finally, we give the optimal parameter for all of the three proposed algorithms, which can make a good tradeoff between performance and overhead/complexity.

  • Exome sequencing reveals genetic heterogeneity and clinically actionable findings in children with cerebral palsy

    • Cerebral palsy (CP) is the most common motor disability in children. To ascertain the role of major genetic variants in the etiology of CP, we conducted exome sequencing on a large-scale cohort with clinical manifestations of CP. The study cohort comprised 505 girls and 1,073 boys. Utilizing the current gold standard in genetic diagnostics, 387 of these 1,578 children (24.5%) received genetic diagnoses. We identified 412 pathogenic and likely pathogenic (P/LP) variants across 219 genes associated with neurodevelopmental disorders, and 59 P/LP copy number variants. The genetic diagnostic rate of children with CP labeled at birth with perinatal asphyxia was higher than the rate in children without asphyxia (P = 0.0033). Also, 33 children with CP manifestations (8.5%, 33 of 387) had findings that were clinically actionable. These results highlight the need for early genetic testing in children with CP, especially those with risk factors like perinatal asphyxia, to enable evidence-based medical decision-making. Using exome sequencing data from one of the largest cohorts of children with cerebral palsy, the genetic diagnostic rates of single-nucleotide and copy number variants were assessed and a sizeable fraction found to be clinically actionable.

  • Analysis of expressed sequence tags from Ginkgo mature foliage in China

    • Ginkgo biloba L. is a tree native to China, which has large importance within medicine and horticulture. The extracts from Ginkgo mature leaves with rich flavonoids and terpenoids are commonly used for a variety of folk remedies. We constructed a cDNA library derived from mature leaves of Ginkgo, which consisted of 8.12 x 10(5) clones with the insert length of 500-2,000 bp. We performed an analysis of expressed sequence tags (ESTs) and obtained partial sequences from 2,039 clones, which represented 1,437 unigenes consisting of 249 contigs and 1,188 singletons. The 2,039 ESTs were submitted to GenBank (dbEST) at NCBI and were assigned GenBank accession numbers from GE647881 to GE649919. The 1,235 cDNA clones out of 2,039 (60.1%) were assigned putative functions, and the remaining 804 clones were not similar to any known gene sequences in the databases. The five largest categories of Ginkgo clones were: "energy" (19.4%), "disease/defense" (16%), "metabolism" (11.3%), "unclassified proteins" (12.5%), and "secondary metabolism" (9%). The highly expressed transcripts in the cDNA library were some genes related to photosynthesis, disease/defense, and flavonoid biosynthesis, including ribulose-bisphosphate carboxylase small-chain gene, pathogenesis-related protein gene, light-harvesting chlorophyll a/b binding protein of photosystem gene, catalase gene, and phenylcoumaran benzylic ether reductase gene et al. Many genes with ESTs similar to photosynthesis, secondary metabolism, and stress-response genes were characterized. The analysis of ESTs indicates that it is a useful approach for isolating Ginkgo genes homologous to known genes. Our results provide new information about mature leaf-specific transcripts of Ginkgo.

    • We performed a multistage genome-wide association study including 7,683 individuals with pancreatic cancer and 14,397 controls of European descent. Four new loci reached genome-wide significance: rs6971499 at 7q32.3 (LINC-PINT, per-allele odds ratio (OR) = 0.79, 95% confidence interval (CI) 0.74-0.84, P = 3.0 x 10(-12)), rs7190458 at 16q23.1 (BCAR1/CTRB1/CTRB2, OR = 1.46, 95% CI 1.30-1.65, P = 1.1 x 10(-10)), rs9581943 at 13q12.2 (PDX1, OR = 1.15, 95% CI 1.10-1.20, P = 2.4 x 10(-9)) and rs16986825 at 22q12.1 (ZNRF3, OR = 1.18, 95% CI 1.12-1.25, P = 1.2 x 10(-8)). We identified an independent signal in exon 2 of TERT at the established region 5p15.33 (rs2736098, OR = 0.80, 95% CI 0.76-0.85, P = 9.8 x 10(-14)). We also identified a locus at 8q24.21 (rs1561927, P = 1.3 x 10(-7)) that approached genome-wide significance located 455 kb telomeric of PVT1. Our study identified multiple new susceptibility alleles for pancreatic cancer that are worthy of follow-up studies.

    • High blood glucose has long been established as a risk factor for tumor metastasis, yet the molecular mechanisms underlying this association have not been elucidated. Here we describe that hyperglycemia promotes tumor metastasis via increased platelet activity. Administration of glucose, but not fructose, reprogrammed the metabolism of megakaryocytes to indirectly prime platelets into a prometastatic phenotype with increased adherence to tumor cells. In megakaryocytes, a glucose metabolism-related gene array identified the mitochondrial molecular chaperone glucose-regulated protein 75 (GRP75) as a trigger for platelet activation and aggregation by stimulating the Ca2+-PKC alpha pathway. Genetic depletion of Glut1 in megakaryocytes blocked MYC-induced GRP75 expression. Pharmacologic blockade of platelet GRP75 compromised tumor-induced platelet activation and reduced metastasis. Moreover, in a pilot clinical study, drinking a 5% glucose solution elevated platelet GRP75 expression and activated platelets in healthy volunteers. Platelets from these volunteers promoted tumor metastasis in a plateletadoptive transfer mouse model. Together, under hyperglycemic conditions, MYC-induced upregulation of GRP75 in megakaryocytes increases platelet activation via the Ca2+-PKC alpha pathway to promote cancer metastasis, providing a potential new therapeutic target for preventing metastasis. Significance: This study provides mechanistic insights into a glucose-megakaryocyte-platelet axis that promotes metastasis and proposes an antimetastatic therapeutic approach by targeting the mitochondrial protein GRP75.

    • Dielectric capacitors are very attractive for high power energy storage. However, the low energy density of these capacitors, which is mainly limited by the dielectric materials, is still the bottleneck for their applications. In this work, lead-free single-phase perovskite Srx(Bi1-xNa0.97-xLi0.03)(0.5)TiO3 (x = 0.30 and 0.38) bulk ceramics, prepared using solid-state reaction method, were carefully studied for the dielectric capacitor application. Polar nano regions (PNRs) were created in this material using co-substitution at A-site to enable relaxor behaviour with low remnant polarization (P-r) and high maximum polarization (P-max). Moreover, P-max was further increased due to the electric field induced reversible phase transitions in nano regions. Comprehensive structural and electrical studies were performed to confirm the PNRs and reversible phase transitions. And finally a high energy density (1.70 J/cm(3)) with an excellent efficiency (87.2%) was achieved using the contribution of field-induced rotations of PNRs and PNR-related reversible transitions in this material, making it among the best performing lead-free dielectric ceramic bulk material for high energy storage.

    • Due to the worldwide concerns of environmental protection and sustainable development, lead-free piezoelectric materials are greatly desired for bridging the electrical energy to the mechanical energy. However, their lower energy conversion coefficient compared to the conventional lead-containing piezoelectric materials significantly limits their device applications. Herein, we introduce a novel strategy to increase the strain of lead-free ferroelectric system via material structure design to create polar nano regions (PNRs) and point defects in the material while retaining the global ferroelectric phase. This added short-range structural heterogeneity in the material will facilitate the field-induced phase transition and reversible domain wall switching to enhance the strain. Following this strategy, we demonstrate an ultrahigh strain induced by an electric field in non-textured lead-free Bi0.5Na0.5TiO3 (BNT)-based ceramics. The strain in unipolar mode (Suni) can reach up to 0.74% at 70 kV/cm, making it the highest value in reported lead-free ceramics so far. This puts forward a good route to design high-performance piezoelectric materials by material structure engineering. It also reveals the promising potential of lead-free piezoelectric materials in practical electromechanical device applications.

    • MoSi2 is introduced into dye-sensitized solar cell (DSC) as counter electrode (CE) catalyst for the first time, and the DSC produces power conversion efficiency (PCE) of 4.87%. To improve the catalytic activity, Pt/MoSi2 composite catalyst is synthesized and it is found that 1.13 wt% of Pt loading is enough for achieving high catalytic activity. After optimization, the DSC using the Pt/MoSi2 composite CE shows high PCE of 7.68%, close to the Pt CE based DSC (7.94%).

    • Background: Insular function has gradually become a topic of intense study in cognitive research. Recognition memory is a commonly studied type of memory in memory research. GABA(B)R has been shown to be closely related to memory formation. In the present study, we used intellicage, which is a new intelligent behavioural test system, and a bilateral drug microinjection technique to inject into the bilateral insula, to examine the relationship between GABA(B)R and recognition memory. Methods: Male Sprague- Dawley rats were randomly divided into control, Sham, Nacl, baclofen and CGP35348 groups. Different testing procedures were employed using intellicage to detect changes in rat recognition memory. The expression of GABA(B)R (GB1, GB2) in the insula of rats was determined by immunofluorescence and western blotting at the protein level. In addition, the expression of GABA(B)R (GB1, GB2) was detected by RT-PCR at the mRNA level. Results: The results of the intellicage test showed that recognition memory was impaired in terms of position learning, punitive learning and punitive reversal learning by using baclofen and CGP35348. In position reversal learning, no significant differences were found in terms of cognitive memory ability between the control groups and the CGP and baclofen groups. Immunofluorescence data showed GABA(B)R (GB1, GB2) expression in the insula, while data from RTPCR and western blot analysis demonstrated that the relative expression of GB1 and GB2 was significantly increased in the baclofen group compared with the control groups. In the CGP35348 group, the expression of GB1 and GB2 was significantly decreased, but there was no significant difference in GB1 or GB2 expression in the control groups. Conclusions: GABA(B)R expression in the insula plays an important role in the formation of recognition memory in rats.

  • Diamond Based Optical Metasurfaces for Broadband Wavefront Shaping in Harsh Environment

    • Metasurface enables a new class of “meta-optics” that can manipulate light at subwavelength scale. Despite that versatile metasurfaces have been demonstrated based on a wide range of materials, the vulnerability of conventional materials to harsh environments, i.e., low resistance to corrosion, low transparency at short wavelength, and lack of thermal/mechanical stability, greatly limit their applications in extreme conditions. Diamond is well-known for exceptional properties, including the highest thermal conductivity, high damage resistance, extraordinary hardness, and chemical inertness. Therefore, diamond based metasurface is generally expected to benefit from its material merits for extreme use. However, the performance of diamond metasurface in harsh environments remains unexplored up to date. To address this question, this work is designed to study the suitability of single-crystal diamond based metasurface for broadband applications under harsh environments. As an example, diamond metasurfaces with representative functionalities, including holographic wavefront-shaping, DUV-focusing, are investigated under high-temperature, acid/alkali, and abrasive conditions, respectively. The findings prove the capability of diamond metasurfaces for applications in broadband and harsh conditions, which not only provides a practical and scalable scheme to encode on-demand functionalities into diamond, but also unlocks a capable candidate to develop robust, large bandwidth, and durable meta-optics for advanced wavefront shaping under extreme conditions.

  • Characteristics of health-state utilities used in cost-effectiveness analyses : a systematic review of published studies in Asia

    • Introduction: Cost-utility analysis (CUA) is the preferred form of economic evaluation in many countries. As one of the key data inputs in cost-utility models, health state utility (HSU) has a crucial impact on CUA results. In the past decades, health technology assessment has been expanding rapidly in Asia, yet research examining the methodology and process used to generate cost-effectiveness evidence is scarce. The aim of this study was to examine the reporting of the characteristics of HSU data used in CUAs in Asia and how the characteristics have changed over time.Methods: A systematic literature search was performed to identify published CUA studies targeting Asian populations. Information was extracted for both the general characteristics of selected studies and the characteristics of reported HSU data. For each HSU value identified, we extracted data for four key characteristics, including 1) estimation method; 2) source of health-related quality of life (HRQoL) data; 3) source of preference data; and 4) sample size. The percentage of nonreporting was calculated and compared over two time periods (1990–2010 vs 2011–2020).Results: A total of 789 studies were included and 4,052 HSUs were identified. Of these HSUs, 3,351 (82.7%) were from published literature and 656 (16.2%) were from unpublished empirical data. Overall, the characteristics of HSU data were not reported in more than 80% of the studies. Of HSUs whose characteristics were reported, most of them were estimated using the EQ-5D (55.7%), Asian HRQoL data (91.9%), and Asian health preferences (87.7%); 45.7% of the HSUs was estimated with a sample of 100 or more individuals. All four characteristics showed improvements after 2010.Conclusion: Over the past two decades, there has been a significant increase in CUA studies targeting Asian populations. However, HSU’s characteristics were not reported in most of the CUA studies, making it difficult to evaluate the quality and appropriateness of the HSUs used in those cost-effectiveness studies.
