    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • Despite pluripotent stem cells sharing key transcription factors, their maintenance involves distinct genetic inputs. Emerging evidence suggests that super-enhancers (SEs) can function as master regulatory hubs to control cell identity and pluripotency in humans and mice. However, whether pluripotency-associated SEs share an evolutionary origin in mammals remains elusive. Here, we performed comprehensive comparative epigenomic and transcription factor binding analyses among pigs, humans, and mice to identify pluripotency-associated SEs. Like typical enhancers, SEs displayed rapid evolu-tion in mammals. We showed that BRD4 is an essential and conserved activator for mammalian pluripotency-associated SEs. Comparative motif enrichment analysis revealed 30 shared transcription factor binding motifs among the three species. The majority of transcriptional factors that bind to identified motifs are known regulators associated with pluripotency. Further, we discovered three pluripotency-associated SEs (SE-SOX2, SE-PIM1, and SE-FGFR1) that displayed remarkable conservation in pla-cental mammals and were sufficient to drive reporter gene expression in a pluripotency-dependent manner. Disruption of these conserved SEs through the CRISPR-Cas9 approach severely impaired stem cell pluripotency. Our study provides insights into the understanding of conserved regulatory mechanisms underlying the maintenance of plu-ripotency as well as species-specific modulation of the pluripotency-associated regula-tory networks in mammals.

  • How hydrology and anthropogenic activity influence the molecular composition and export of dissolved organic matter : Observations along a large river continuum

    • Large rivers are the main arteries for transportation of carbon to the ocean; yet, how hydrology and anthropogenic disturbances may change the composition and export of dissolved organic matter along large river continuums is largely unknown. The Yangtze River has a watershed area of 1.80 x 10(6) km(2). It originates from the Qinghai-Tibet Plateau and flows 6300 km eastward through the center of China. We collected samples (n = 271) along the river continuum and analyzed weekly samples at the most downstream situated gauging station in 2017-2018 and gathered long-term (2006-2018) water quality data. We found higher gross domestic product, population density, and urban and agricultural land use downstream than upstream of the Three Gorges Dam, coinciding with higher dissolved organic carbon (DOC), UV absorption (a(254)), specific ultraviolet absorbance (SUVA(254)), parallel factor analysis-derived C1-C5, aliphatic compounds, and lower a(250):a(365) and spectral slope (S275-295). Chemical oxygen demand, humic-like C1-C2 and C6, and protein-like C4 and C7 increased, while dissolved oxygen and ammonium decreased with increasing discharge at most of the sites studied, including the intensively monitored downstream site. The annual DOC fluxes were ca. 1.5-1.8 Tg yr(-1), and 12-18% was biodegradable in a 28-d bio-incubation. Our results highlight that urbanization and stormwater periods enhanced the export of both terrestrial organic-rich substances and household effluents from nearshore residential areas. Our study emphasizes the continued need to protect the Yangtze River watershed as increased organic carbon loading or altered composition and bio-lability may change the ecosystem function and carbon cycling.

The absorptive and reflective behavior of OA components contributes to determining particle optical properties and thus their effects on the radiative budget of the troposphere. There is limited knowledge on the influence of the molecular composition of OA on particle optical properties in the polluted urban environment. In this study, we characterized the molecular composition of oxygenated OA collected on filter samples in the autumn of 2018 in Beijing, China, with a filter inlet for gases and aerosols coupled to a high-resolution time-of-flight chemical ionization mass spectrometer (FIGAERO–CIMS). Three haze episodes occurred during our sampling period with daily maximum concentrations of OA of 50, 30, and 55 µg m−3. We found that the signal intensities of dicarboxylic acids and sulfur-containing compounds increased during the two more intense haze episodes, while the relative contributions of wood-burning markers and other aromatic compounds were enhanced during the cleaner periods. We further assessed the optical properties of oxygenated OA components by combining detailed chemical composition measurements with collocated particle light absorption measurements. We show that light absorption enhancement (Eabs) of black carbon (BC) was mostly related to more oxygenated OA (e.g., dicarboxylic acids), likely formed in aqueous-phase reactions during the intense haze periods with higher relative humidity, and speculate that they might contribute to lensing effects. Aromatics and nitro-aromatics (e.g., nitrocatechol and its derivatives) were mostly related to a high light absorption coefficient (babs) consistent with light-absorbing (brown) carbon (BrC). Our results provide information on oxygenated OA components at the molecular level associated with BrC and BC particle light absorption and can serve as a basis for further studies on the effects of anthropogenic OA on radiative forcing in the urban environment.

    • For organic solar cells, the vertical and lateral micro-/nanometer-scale structure in the active layer largely determines the device performance. In this work, the surface and bulk domain size of the photoactive layer are successfully manipulated with a facile two-step spraying method, that is, an ultrathin active layer by high-pressure spraying is deliberately stacked on top of the thick active layer by ultrasonic spraying. Thus, the morphology is effectively optimized with the comprehensive study of optical and electrical characteristics, such as photon absorption, exciton dissociation efficiency, and bimolecular recombination. Moreover, the novel method can be used not only in the fullerene system but also in the nonfullerene system, demonstrating the remarkable universality through this synergy method. This work provides an easy and reliable strategy to improve photovoltaic device performance in the industrial large-area spray-coating process.

    • With the rapid development of modern HVDC transmission technology, higher insulation properties are put forward on the oil-paper insulation system of the transformer, which determine the transformer service life to a certain extent. Traditional transformer oil-paper insulation is becoming increasingly difficult to meet the demands of insulation system with large capacity and miniaturization at ultra-high voltage level. In order to improve the insulation strength of oil-paper system, the insulation cellulose paper modified by TiO2 nanoparticles of different diameters (5 nm, 10 nm, 20 nm, 30 nm) were prepared, in addition, each of modified cellulose paper has different mass fraction of TiO2 nanoparticles (1%, 3%, 5%, 7% wt.). The partial discharge (PD) detection platform was established, and the partial discharge inception voltage (PDIV) values of the oil-paper insulation system with and without nanoparticles were measured. To investigate the PD characteristics, the PD waveforms and PD frequency spectrums of modified cellulose paper and the unmodified were obtained. The suppression mechanism of TiO2 nanoparticles on PD was explored through scanning electron microscope (SEM) observation. All the experiment results indicate that adding nano-TiO2 is beneficial to enhance the insulation properties of oil-paper insulation, and the optimum diameter and mass fraction of TiO2 nanoparticles to suppress oil-paper PD were obtained.

  • Regulating reconstruction of oxide-derived Cu for electrochemical CO2 reduction toward n-propanol

    • Oxide-derived copper (OD-Cu) is the most efficient and likely practical electrocatalyst for CO2 reduction toward multicarbon products. However, the inevitable but poorly understood reconstruction from the pristine state to the working state of OD-Cu under strong reduction conditions largely hinders the rational construction of catalysts toward multicarbon products, especially C-3 products like n-propanol. Here, we simulate the reconstruction of CuO and Cu2O into their derived Cu by molecular dynamics, revealing that CuO-derived Cu (CuOD-Cu) intrinsically has a richer population of undercoordinated Cu sites and higher surficial Cu atom density than the counterpart Cu2O-derived Cu (Cu2OD-Cu) because of the vigorous oxygen removal. In situ spectroscopes disclose that the coordination number of CuOD-Cu is considerably lower than that of Cu2OD-Cu, enabling the fast kinetics of CO2 reaction and strengthened binding of *C-2 intermediate(s). Benefiting from the rich undercoordinated Cu sites, CuOD-Cu achieves remarkable n-propanol faradaic efficiency up to similar to 17.9%, whereas the Cu2OD-Cu dominantly generates formate.

  • Gene-Viral Cancer Therapy Using Dual-Regulated Oncolytic Adenovirus with Antiangiogenesis Gene for Increased Efficacy.

    • Conditionally replicative adenovirus (CRAD) represents a promising approach for cancer therapy. Several CRADs controlled by the human telomerase reverse transcriptase promoter have been developed. However, because of their replicative capacity, the importance of cancer specificity for CRADs needs to be further emphasized. In this study, we have developed a novel dual-regulated CRAD, CNHK500-mE, which has its E1a and E1b gene controlled by the human telomerase reverse transcriptase promoter and the hypoxia response element, respectively. It also carries a mouse endostatin expression cassette controlled by the cytomegalovirus promoter. These properties allow for increased cancer cell targeting specificity and decreased adverse side effects. We showed that CNHK500-mE preferentially replicated in cancer cells. Compared with a replication-defective vector carrying the same endostatin expression cassette, CNHK500-mE-mediated transgene expression level was markedly increased via viral replication within cancer cells. In the nasopharyngeal tumor xenograft model, CNHK500-mE injection resulted in antitumor efficacy at day 7 after therapy. Three weeks later, it led to significant inhibition of xenograft tumor growth due to the combined effects of viral oncolytic therapy and antiangiogenesis gene therapy. Pathologic examination showed that most cancer cells were positive for adenoviral capsid protein and for apoptotic terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling in the CNHK500-mE-treated tumor tissues, and the microvessels in these tumor tissues were diminished in quantity and abnormal in morphology. These results suggest that, as a potential cancer therapeutic agent, the CNHK500-mE is endowed with higher specificity to cancer cells and low cytotoxicity to normal cells.   • Stalled oligodendrocyte differentiation in IDH-mutant gliomas.

    • Roughly 50% of adult gliomas harbor isocitrate dehydrogenase (IDH) mutations. According to the 2021 WHO classification guideline, these gliomas are diagnosed as astrocytomas, harboring no 1p19q co-deletion, or oligodendrogliomas, harboring 1p19q co-deletion. Recent studies report that IDH-mutant gliomas share a common developmental hierarchy. However, the neural lineages and differentiation stages in IDH-mutant gliomas remain inadequately characterized.Using bulk transcriptomes and single-cell transcriptomes, we identified genes enriched in IDH-mutant gliomas with or without 1p19q co-deletion, we also assessed the expression pattern of stage-specific signatures and key regulators of oligodendrocyte lineage differentiation. We compared the expression of oligodendrocyte lineage stage-specific markers between quiescent and proliferating malignant single cells. The gene expression profiles were validated using RNAscope analysis and myelin staining and were further substantiated using data of DNA methylation and single-cell ATAC-seq. As a control, we assessed the expression pattern of astrocyte lineage markers.Genes concordantly enriched in both subtypes of IDH-mutant gliomas are upregulated in oligodendrocyte progenitor cells (OPC). Signatures of early stages of oligodendrocyte lineage and key regulators of OPC specification and maintenance are enriched in all IDH-mutant gliomas. In contrast, signature of myelin-forming oligodendrocytes, myelination regulators, and myelin components are significantly down-regulated or absent in IDH-mutant gliomas. Further, single-cell transcriptomes of IDH-mutant gliomas are similar to OPC and differentiation-committed oligodendrocyte progenitors, but not to myelinating oligodendrocyte. Most IDH-mutant glioma cells are quiescent; quiescent cells and proliferating cells resemble the same differentiation stage of oligodendrocyte lineage. Mirroring the gene expression profiles along the oligodendrocyte lineage, analyses of DNA methylation and single-cell ATAC-seq data demonstrate that genes of myelination regulators and myelin components are hypermethylated and show inaccessible chromatin status, whereas regulators of OPC specification and maintenance are hypomethylated and show open chromatin status. Markers of astrocyte precursors are not enriched in IDH-mutant gliomas.Our studies show that despite differences in clinical manifestation and genomic alterations, all IDH-mutant gliomas resemble early stages of oligodendrocyte lineage and are stalled in oligodendrocyte differentiation due to blocked myelination program. These findings provide a framework to accommodate biological features and therapy development for IDH-mutant gliomas.

    • Recombinant adeno-associated virus serotype 2 (rAAV2) vector has been widely employed for gene therapy. Recent progress suggests that the new serotypes of AAV showed a better performance than did AAV2 in normal tissues. Here, we evaluate the potential role of human vascular endothelial growth factor (VEGF) gene transfer using rAAV vector pseudotyped with serotype I capsid proteins (rAAV1) in the treatment of muscle ischemia. In ischemic skeletal muscles, the rAAV1-LacZ vector allowed higher level, broader distribution, and long-lasting gene expression compared with the rAAV2-LacZ vector. Muscle VEGF165 production following the rAAV1-VEGF165 vector injection was 5-10 times higher than that following the rAAV2-VEGF165 vector injection. VEGF165 production mediated by the rAAV1-VEGF165 vector stimulated a large set of neovascularization with relatively mature vascular structures and enhanced muscle regeneration in the ischemic skeletal muscles. Thus, the rAAV1-NEGF165 vector mediated gene transfer may be a therapeutic approach to peripheral vascular diseases.

    • Broad Balmer emission lines in active galactic nuclei (AGN) may display dramatic changes in amplitude, even disappearance and re-appearance in some sources. As a nearby galaxy at a redshift of z = 0.0264, Mrk 590 suffered such a cycle of Seyfert type changes between 2006 and 2017. Over the last 50 yr, Mrk 590 also underwent a powerful continuum outburst and a slow fading from X-rays to radio wavelengths with a peak bolometric luminosity reaching about 10 per cent of the Eddington luminosity. To track its past accretion and ejection activity, we performed very long baseline interferometry (VLBI) observations with the European VLBI Network (EVN) at 1.6 GHz in 2015. The EVN observations reveal a faint (∼1.7 mJy) radio jet extending up to ∼2.8 mas (projected scale ∼1.4 pc) toward north, and probably resulting from the very intensive AGN activity. To date, such a parsec-scale jet is rarely seen in the known changing-look AGN. The finding of the faint jet provides further strong support for variable accretion as the origin of the type changes in Mrk 590.

  • Extended Catalog of Winged or X-shaped Radio Sources from the FIRST Survey

    • We present a catalog of 290 ?winged? or X-shaped radio galaxies (XRGs) extracted from the latest (2014 December 17) data release of the ?Very Large Array Faint Images of the Radio Sky at Twenty centimeter.? We have combined these radio images with their counterparts in the TIFR GMRT sky survey at 150 MHz, in an attempt to identify any low surface brightness radio emission present in these sources. This has enabled us to assemble a sample of 106 ?strong? XRG candidates and 184 ?probable? XRG candidates whose XRG designation needs to be verified by further observations. The present sample of 290 XRG candidates is almost twice as large as the number of XRGs currently known. Twenty-five of our 290 XRG candidates (9 ?strong? and 16 ?probable?) are identified as quasars. Double-peaked narrow emission lines are seen in the optical spectra of three of the XRG candidates (two ?strong? and one ?probable?). Nearly 90% of the sample is located in the FR II domain of the Owen?Ledlow diagram. A few of the strong XRG candidates have a rather flat radio spectrum (spectral index ? flatter than ?0.3) between 150 MHz and 1.4 GHz, or between 1.4 and 5 GHz. Since this is not expected for lobe-dominated extragalactic radio sources (like nearly all known XRGs), these sources are particularly suited for follow-up radio imaging and near-simultaneous measurement of the radio spectrum.

  • NGC 5252: a pair of radio-emitting active galactic nuclei?

    • The X-ray source CXO J133815.6+043255 has counterparts in the UV, optical, and radio bands. Based on the multiband investigations, it has been recently proposed by Kim et al. as a rarely seen off-nucleus ultraluminous X-ray (ULX) source with a black hole mass of ≥104 M⊙ in the nearby Seyfert galaxy NGC 5252. To explore its radio properties at very high angular resolution, we performed very long-baseline interferometry (VLBI) observations with the European VLBI Network (EVN) at 1.7 GHz. We find that the radio counterpart is remarkably compact among the known ULXs. It does not show a resolved structure with a resolution of a few milliarcsecond (mas), and the total recovered flux density is comparable to that measured in earlier sub-arcsecond-resolution images. The compact radio structure, the relatively flat spectrum, and the high radio luminosity are consistent with a weakly accreting supermassive black hole in a low-luminosity active galactic nucleus. The nucleus of NGC 5252 itself has similar radio properties. We argue that the system represents a relatively rare pair of active galactic nuclei, where both components emit in the radio.

    • Accretion of black holes at near-Eddington or super-Eddington rates represents the most powerful episode driving black hole growth, potentially occurring across various types of objects. However, the physics governing accretion and jet-disk coupling in such states remains unclear, primarily due to the difficulty in detecting associated jets, which may emit extremely weakly or exhibit episodic behavior. Only a few near/super-Eddington systems have demonstrated radio activity, and it remains uncertain whether jets exist and what their properties are in super-Eddington active galactic nuclei (AGNs) and ultraluminous X-ray sources. This uncertainty stems mainly from the complex radio emission mix, which includes contributions from jets, star formation activity, photoionized gas, accretion disk wind, and coronal activity. In this work, we conducted high-resolution, very long baseline interferometry observations to investigate jets in the highly accreting narrow-line Seyfert I system I Zw 1. Our observations successfully revealed small-scale jets (with a linear size of ∼45 pc) at both 1.5 and 5 GHz, based on the high radio brightness temperature, radio morphology, and spectral index distribution. Additionally, the parsec-scale jet observed in I Zw 1 displays a knotted morphology reminiscent of other sources accreting at similar rates. In summary, the high accretion rates and jet properties observed in the AGN I Zw 1 may support the AGN/X-ray binary analogy in this extreme state.

    • The jet in active galactic nuclei (AGN) is a key ingredient in understanding the co-evolution of galaxies and their central supermassive black holes (SMBHs). Unfortunately, the mechanism of jet launching and collimation is still elusive. The observational evidence of decreasing radio loudness with increasing Eddington ratio implies that the jet should be coupled with the accretion process. To further explore the relationship between the jet and accretion, it is necessary to extend our knowledge of the jet to an extreme end of the Eddington ratio distribution of AGN. Using Very Long Baseline Array (VLBA), we report the detection of the parsec-scale radio structure in Mrk 335, a radio-quiet narrow-line Seyfert 1 galaxy with an Eddington ratio close to/above unity. The VLBA image at 1.5 GHz reveals an elongated structure extending ∼20 pc in north-south direction with a peak flux density of 1.98 ± 0.05 mJy beam-1 and radio brightness temperatures as high as 6 × 107 K. This feature provides a strong evidence of a parsec-scale (bipolar) jet launched from a highly accreting SMBH. We discuss the result by comparing Mrk 335 with other highly accreting systems, e.g. Galactic black holes and tidal disruption events, and recall the discovery of collimated corona in the vicinity of SMBH in Mrk 335 by previous X-ray observations, whose relation to the parsec-scale radio jet should be explored by future simultaneous X-ray spectroscopy and high resolution radio observations.

    • Continuous time (CT) quadrature bandpass sigma delta modulators (QBSDM) are widely used in wireless receivers recently due to its inherent anti-aliasing filtering and complex filtering embedded in the loops to efficiently realize asymmetric noise-shaping. The complex noise-transfer-function (NTF) can be realized with complex coefficients or by implementing the loop as a cascade of complex integrators with only real coefficients, which is desirable due to its lower sensitivity to loop mismatch. In this paper, we will study the effect of different feedback waveforms inside the modulator on the NTF of quadrature sigma delta modulators. Complex NTF design is discussed for different bandpass center frequencies relative to the sampling frequency.

  • Single amplifier sigma delta modulator with input feedforward

    • It has recently become very popular to feedforward the input signal in wideband delta-sigma modulators, so that the integrators only process quantization errors. The advantage is that the actual signal is not distorted by op-amp and integrator nonlinearities. The paper applies this technique to single amplifier based sigma delta modulator in both analog and digital way. Their characteristic has been analyzed and simulation results are given which show the improved performance.

    • The pluripotency gene regulatory network of porcine induced pluripotent stem cells(piPSCs), especially in epigenetics, remains elusive. To determine the biological function of epigenetics, we cultured piPSCs in different culture conditions. We found that activation of pluripotent gene- and pluripotency-related pathways requires the erasure of H3K9 methylation modification which was further influenced by mouse embryonic fibroblast (MEF) served feeder. By dissecting the dynamic change of H3K9 methylation during loss of pluripotency, we demonstrated that the H3K9 demethylases KDM3A and KDM3B regulated global H3K9me2/me3 level and that their co-depletion led to the collapse of the pluripotency gene regulatory network. Immunoprecipitation-mass spectrometry (IP-MS) provided evidence that KDM3A and KDM3B formed a complex to perform H3K9 demethylation. The genome-wide regulation analysis revealed that OCT4 (O) and SOX2 (S), the core pluripotency transcriptional activators, maintained the pluripotent state of piPSCs depending on the H3K9 hypomethylation. Further investigation revealed that O/S cooperating with histone demethylase complex containing KDM3A and KDM3B promoted pluripotency genes expression to maintain the pluripotent state of piPSCs. Together, these data offer a unique insight into the epigenetic pluripotency network of piPSCs.
