    • Gamma-ray bursts (GRBs) have long been considered a possible source of high-energy neutrinos. While no correlations have yet been detected between high-energy neutrinos and GRBs, the recent observation of GRB 221009A-the brightest GRB observed by Fermi-GBM to date and the first one to be observed above an energy of 10 TeV-provides a unique opportunity to test for hadronic emission. In this paper, we leverage the wide energy range of the IceCube Neutrino Observatory to search for neutrinos from GRB 221009A. We find no significant deviation from background expectation across event samples ranging from MeV to PeV energies, placing stringent upper limits on the neutrino emission from this source.

  • Observation of seasonal variations of the flux of high-energy atmospheric neutrinos with IceCube

    • Atmospheric muon neutrinos are produced by meson decays in cosmic-ray-induced air showers. The flux depends on meteorological quantities such as the air temperature, which affects the density of air. Competition between decay and re-interaction of those mesons in the first particle production generations gives rise to a higher neutrino flux when the air density in the stratosphere is lower, corresponding to a higher temperature. A measurement of a temperature dependence of the atmospheric νμ flux provides a novel method for constraining hadronic interaction models of air showers. It is particularly sensitive to the production of kaons. Studying this temperature dependence for the first time requires a large sample of high-energy neutrinos as well as a detailed understanding of atmospheric properties. We report the significant (>10σ) observation of a correlation between the rate of more than 260,000 neutrinos, detected by IceCube between 2012 and 2018, and atmospheric temperatures of the stratosphere, measured by the Atmospheric Infrared Sounder (AIRS) instrument aboard NASA’s AQUA satellite. For the observed 10 % seasonal change of effective atmospheric temperature we measure a 3.5(3) % change in the muon neutrino flux. This observed correlation deviates by about 2-3 standard deviations from the expected correlation of 4.3 % as obtained from theoretical predictions under the assumption of various hadronic interaction models.

  • The Herschel-Heterodyne Instrument for the Far-Infrared (HIFI)

    • Aims: This paper describes the Heterodyne Instrument for the Far-Infrared (HIFI) that was launched onboard ESA's Herschel Space Observatory in May 2009. Methods: The instrument is a set of 7 heterodyne receivers that are electronically tuneable, covering 480-1250 GHz with SIS mixers and the 1410-1910 GHz range with hot electron bolometer (HEB) mixers. The local oscillator (LO) subsystem comprises a Ka-band synthesizer followed by 14 chains of frequency multipliers and 2 chains for each frequency band. A pair of auto-correlators and a pair of acousto-optical spectrometers process the two IF signals from the dual-polarization, single-pixel front-ends to provide instantaneous frequency coverage of 2 × 4 GHz, with a set of resolutions (125 kHz to 1 MHz) that are better than 0.1 km s-1. Results: After a successful qualification and a pre-launch TB/TV test program, the flight instrument is now in-orbit and completed successfully the commissioning and performance verification phase. The in-orbit performance of the receivers matches the pre-launch sensitivities. We also report on the in-orbit performance of the receivers and some first results of HIFI's operations. Herschel is an ESA space observatory with science instruments provided by European-led Principal Investigator consortia and with important participation from NASA.

  • Tidewater Glaciers and Bedrock Characteristics Control the Phytoplankton Growth Environment in a Fjord in the Arctic

    • Meltwater discharge from tidewater glaciers impacts the adjacent marine environment. Due to the global warming, tidewater glaciers are retreating and will eventually terminate on land. Yet, the mechanisms through which meltwater runoff and subglacial discharge from tidewater glaciers influence marine primary production remain poorly understood, as data in close proximity to glacier fronts are scarce. Here, we show that subglacial meltwater discharge and bedrock characteristics of the catchments control the phytoplankton growth environment inside the fjord, based on data collected in close proximity to tidewater glacier fronts in Kongsfjorden, Svalbard from 26 to 31 July 2017. In the southern part of the inner fjord, glacial meltwater from subglacial discharge was rich in fine sediments derived from erosion of Devonian Old Red Sandstone and carbonate rock deposits, limiting light availability for phytoplankton (0.6 mg m(-3) Chl a on average, range 0.2-1.9 mg m(-3)). In contrast, coarser sediments derived from gneiss and granite bedrock and lower subglacial discharge rates were associated with more favourable light conditions facilitating a local phytoplankton bloom in the northern part of the inner fjord with mean Chl a concentration of 2.8 mg m(-3) (range 1.3-7.4 mg m(-3)). In the northern part, glacier meltwater was a direct source of silicic acid through weathering of the silica-rich gneiss and granite bedrock. Upwelling of the subglacial freshwater discharge plume at the Kronebreen glacier front in the southern part entrained large volumes of ambient, nutrient-rich bottom waters which led to elevated surface concentrations of ammonium, nitrate, and partly silicic acid. Total dissolved inorganic nitrogen transported to the surface with the upwelling of the subglacial discharge plume has a significant potential to enhance summer primary production in Kongsfjorden, with ammonium released from the seafloor being of particular importance. The transition from tidewater to land-terminating glaciers may, thus, reduce the input of nutrients to the surface layer with negative consequences for summer productivity.

  • One thousand plant transcriptomes and the phylogenomics of green plants

    • Green plants (Viridiplantae) include around 450,000-500,000 species(1,2) of great diversity and have important roles in terrestrial and aquatic ecosystems. Here, as part of the One Thousand Plant Transcriptomes Initiative, we sequenced the vegetative transcriptomes of 1,124 species that span the diversity of plants in a broad sense (Archaeplastida), including green plants (Viridiplantae), glaucophytes (Glaucophyta) and red algae (Rhodophyta). Our analysis provides a robust phylogenomic framework for examining the evolution of green plants. Most inferred species relationships are well supported across multiple species tree and supermatrix analyses, but discordance among plastid and nuclear gene trees at a few important nodes highlights the complexity of plant genome evolution, including polyploidy, periods of rapid speciation, and extinction. Incomplete sorting of ancestral variation, polyploidization and massive expansions of gene families punctuate the evolutionary history of green plants. Notably, we find that large expansions of gene families preceded the origins of green plants, land plants and vascular plants, whereas whole-genome duplications are inferred to have occurred repeatedly throughout the evolution of flowering plants and ferns. The increasing availability of high-quality plant genome sequences and advances in functional genomics are enabling research on genome evolution across the green tree of life.

    • Si crystals (n-type, fz) with doping levels between 1.5×1014 and 2×1016 cm-3 containing in addition ∼1018 Sn/cm3 were irradiated with 2-MeV electrons to different doses and subsequently studied by deep level transient spectroscopy, Mössbauer spectroscopy, and positron annihilation. Two tin-vacancy (Sn-V) levels at Ec-0.214 eV and Ec-0.501 eV have been identified (Ec denotes the conduction band edge). Based on investigations of the temperature dependence of the electron-capture cross sections, the electric-field dependence of the electron emissivity, the anneal temperature, and the defect-introduction rate, it is concluded that these levels are the double and single acceptor levels, respectively, of the Sn-V pair. These conclusions are in agreement with electronic structure calculations carried out using a local spin-density functional theory, incorporating pseudopotentials to eliminate the core electrons, and applied to large H-terminated clusters. Thus, the Sn-V pair in Si has five different charge states corresponding to four levels in the band gap.

    • The diatom genus Entomoneis includes species with panduriform frustules characterized by a bilobate, elevated keel, sigmoid raphe canal and numerous open, porous girdle bands. During a phytoplankton survey along the Baltic coast of Sweden, we observed numerous Entomoneis cells, some belonging to the well-known Entomoneis paludosa, while others remained unknown. Morphological and ultrastructural studies of the unknown species were performed with light microscopy (LM), scanning (SEM) and transmission (TEM) electron microscopy, and revealed a unique set of morphological characters. Live cells of Entomoneis grisslehamnensis sp. nov. contain one plate-like plastid and show various degrees of torsion about the apical axis. Microscopic features of cleaned frustules are discussed in comparison with similar species. Most importantly, every 2nd to 5th virga is strongly elevated, uniseriate striae are composed of quadrangular areolae with finely perforated hymenes; the winged keel with a discernible raphe canal is structurally strengthened by raphe fibulae with a flattened wing area, and the main part of the valve bulges towards the margins; striation is decussate on the wings and there is one row of raphe canal areolae at the central area. In addition to describing this new species of an underappreciated, yet cosmopolitan diatom genus, we contribute to the taxonomy of E. paludosa with LM, SEM and TEM details, some of which were not sufficiently noted in original descriptions or re-examination of type material and other specimens made by Dalu et al.     • Hypoxia, a growing worldwide problem, has been intermittently present in the modern Baltic Sea since its formation ca. 8000 cal. yr BP. However, both the spatial extent and intensity of hypoxia have increased with anthropogenic eutrophication due to nutrient inputs. Physical processes, which control stratification and the renewal of oxygen in bottom waters, are important constraints on the formation and maintenance of hypoxia. Climate controlled inflows of saline water from the North Sea through the Danish Straits is a critical controlling factor governing the spatial extent and duration of hypoxia. Hypoxia regulates the biogeochemical cycles of both phosphorus (P) and nitrogen (N) in the water column and sediments. Significant amounts of P are currently released from sediments, an order of magnitude larger than anthropogenic inputs. The Baltic Sea is unique for coastal marine ecosystems experiencing N losses in hypoxic waters below the halocline. Although benthic communities in the Baltic Sea are naturally constrained by salinity gradients, hypoxia has resulted in habitat loss over vast areas and the elimination of benthic fauna, and has severely disrupted benthic food webs. Nutrient load reductions are needed to reduce the extent, severity, and effects of hypoxia.

  • A hypothalamic dopamine locus for psychostimulant-induced hyperlocomotion in mice

    • The lateral septum (LS) has been implicated in the regulation of locomotion. Nevertheless, the neurons synchronizing LS activity with the brain’s clock in the suprachiasmatic nucleus (SCN) remain unknown. By interrogating the molecular, anatomical and physiological heterogeneity of dopamine neurons of the periventricular nucleus (PeVN; A14 catecholaminergic group), we find that Th+/Dat1+ cells from its anterior subdivision innervate the LS in mice. These dopamine neurons receive dense neuropeptidergic innervation from the SCN. Reciprocal viral tracing in combination with optogenetic stimulation ex vivo identified somatostatin-containing neurons in the LS as preferred synaptic targets of extrahypothalamic A14 efferents. In vivo chemogenetic manipulation of anterior A14 neurons impacted locomotion. Moreover, chemogenetic inhibition of dopamine output from the anterior PeVN normalized amphetamine-induced hyperlocomotion, particularly during sedentary periods. Cumulatively, our findings identify a hypothalamic locus for the diurnal control of locomotion and pinpoint a midbrain-independent cellular target of psychostimulants.

    • The Baltic Sea Action Plan (BSAP) requires tools to simulate effects and costs of various nutrient abatement strategies. Hierarchically connected databases and models of the entire catchment have been created to allow decision makers to view scenarios via the decision support system NEST. Increased intensity in agriculture in transient countries would result in increased nutrient loads to the Baltic Sea, particularly from Poland, the Baltic States, and Russia. Nutrient retentions are high, which means that the nutrient reduction goals of 135 000 tons N and 15 000 tons P, as formulated in the BSAP from 2007, correspond to a reduction in nutrient loadings to watersheds by 675 000 tons N and 158 000 tons P. A cost-minimization model was used to allocate nutrient reductions to measures and countries where the costs for reducing loads are low. The minimum annual cost to meet BSAP basin targets is estimated to 4.7 billion a,not sign.

  • A new species of Craspedostauros (Bacillariophyceae) from the west coast of Sweden, with taxonomic and ecological notes on Craspedostauros laevissimus

    • Since its separation from Stauroneis in 1999, several new species of Craspedostauros were discovered in a variety of habitats and geographic locations, adding morphological and phylogenetic data to the investigations of the genus. In a survey of littoral diatoms of Sweden, both on the west and east coasts, two epiphytic stauros-bearing species were encountered and assigned to Craspedostauros following the characteristic features of this genus, including the possession of a stauros narrower than the central area and cribrate areolae. One species is described as new to science; Craspedostauros lateralis sp. nov., and the other is of uncertain identity but bears morphological similarity to C. laevissimus. Caspedostauros lateralis is a marine epiphytic species found in the west coast of Sweden, off Gothenburg city. Based on light and electron microscopy, a detailed description of the morphological and ultrastructural features of these species is given and a comparison of the distinguishing characters with allied species is discussed. Some ecological data and the occurrence of associated species on the host macrophyte are provided.

    • Integral invariants in maximally supersymmetric Yang-Mills theories are discussed in spacetime dimensions 4 <= D <= 10 for SU(k) gauge groups. It is shown that, in addition to the action, there are three special invariants in all dimensions. Two of these, the single-and double-trace F-4 invariants, are of Chern-Simons type in D = 9, 10 and BPS type in D <= 8, while the third, the double-trace of two derivatives acting on F-4, can be expressed in terms of a gauge-invariant super-D-form in all dimensions. We show that the super-ten-forms for D = 10 F-4 invariants have interesting cohomological properties and we also discuss some features of other invariants, including the single-trace d(2)F(4), which has a special form in D = 10. The implications of these results for ultra-violet divergences are discussed in the framework of algebraic renormalisation.

    • Harmful cyanobacterial blooms (=cyanoHABs) are an increasing feature of many waterbodies throughout the world. Many bloom-forming species produce toxins, making them of particular concern for drinking water supplies, recreation and fisheries in waterbodies along the freshwater to marine continuum. Global changes resulting from human impacts, such as climate change, over-enrichment and hydrological alterations of waterways, are major drivers of cyanoHAB proliferation and persistence. This review advocates that to better predict and manage cyanoHABs in a changing world, researchers need to leverage studies undertaken to date, but adopt a more complex and definitive suite of experiments, observations, and models which can effectively capture the temporal scales of processes driven by eutrophication and a changing climate. Better integration of laboratory culture and field experiments, as well as whole system and multiple-system studies are needed to improve confidence in models predicting impacts of climate change and anthropogenic over-enrichment and hydrological modifications. Recent studies examining adaptation of species and strains to long-term perturbations, e.g. temperature and carbon dioxide (CO 2 ) levels, as well as incorporating multi-species and multi-stressor approaches emphasize the limitations of approaches focused on single stressors and individual species. There are also emerging species of concern, such as toxic benthic cyanobacteria, for which the effects of global change are less well understood, and require more detailed study. This review provides approaches and examples of studies tackling the challenging issue of understanding how global changes will affect cyanoHABs, and identifies critical information needs for effective prediction and management. © 2019

    • Deep-water oxygen concentrations in the Baltic Sea are influenced by eutrophication, but also by saltwater inflows from the North Sea. In the last two decades, only two major inflows have been recorded and the lack of major inflows is believed to have resulted in a long-term stagnation of the deepest bottom water. Analyzing data from 1970 to 2000 at the basin scale, we show that the estimated volume of water with oxygen, <2 mL L-1, was actually at a minimum at the end of the longest so-called stagnation period on record. We also show that annual changes in dissolved inorganic phosphate water pools were positively correlated to the area of bottom covered by hypoxic water, but not to changes in total phosphorus load, thus addressing the legacy of eutrophication on a basinwide scale. The variations in phosphorus pools that have occurred during the past decades do not reflect any human action to reduce inputs. The long residence time and internally controlled variation of the large P pool in the Baltic Sea has important implications for management of both N and P inputs into this eutrophicated enclosed basin.

    • Algal community species composition, as estimated by high performance liquid chromatography (HPLC) pigments and microscopy analysis, and trace metal speciation (Cu and Co) and distributions (Fe, Zn, Co and Cu) were measured along a summer transect across the Skagerrak. In waters of Baltic origin, with elevated trace metals levels, but very low macronutrients, a mix of dinoflagellates and haptophytes dominated the low biomass. In the Jutland current, which had high dissolved iron concentrations, a mixed bloom (4-6 mug/l chl a) of diatoms (major species-Leptocylindricus danica) and dinoflagellates (Ceratium sp.) was present. In the waters of the central Skagerrak derived from the North Sea, below the low salinity Baltic water, a large diatom (major species-L. danica) bloom (7.7 mug/l) was present at 35 in. This bloom formed below the pycnocline. and was located at the nutricline for silicate. The lowest concentrations of trace metals were found in the water of North Sea origin. Synechocoecus-like cyanobacteria were observed in the upper waters across the survey area, as were strong binding ligands for Cu, but no clear numerical relationship existed between them, as had been observed by Moffett [Deep-Sea Res. 42 (1995) 1273]m the Sargasso Sea. The [Co]/[Zn] hypothesis of Sunda and Huntsman [Limnol. Oceanogr, 40 (1995) 1404] for coccolithophorids and diatoms was examined using the field data collected. (C) 2002 Elsevier Science B.V.   • Haslea ostrearia-like Diatoms: Biodiversity out of the Blue

    • Diatoms are usually referred to as golden-brown microalgae, due to the colour of their plastids and to their pigment composition, mainly carotenoids (fucoxanthin, diadinoxanthin, diatoxanthin), which mask chlorophylls a and c. The species Haslea ostrearia Gaillon/Bory (Simonsen) appears unique because of its extraplastidial bluish colour, a consequence of the presence of a water-soluble blue pigment at cell apices, marennine. When released in seawater, marenbine can be fixed on gills of oysters and other bivalves, which turn green. This greening phenomenon is economically exploited in Southwestern France, as it gives an added value to oysters. For decades, this singularity ascribed a worldwide distribution to H. ostrearia, first as Vibrio ostrearius, then Navicula ostrearia, last as H. ostrearia, when the genus Haslea was proposed by R. Simonsen (1974). Indeed, this 'birthmark' (presence of blue apices) made H. ostrearia easily recognisable without further scrutiny and identification of the microalga as well as its presence easily deduced from the greening of bivalves. Consequently, the widely admitted cosmopolitan character of H. ostrearia has only been questioned recently, following the discovery in 2008, of a new species of blue diatom in the Black Sea, Haslea karadagensis. The biodiversity of blue diatoms suddenly increased with the finding of other blue species in the Mediterranean Sea, the Canary Islands, etc., the taxonomic characterization of which is in progress. This review thus focuses on the unsuspected biodiversity of blue diatoms within the genus Haslea. Methods for species determination (morphometrics, chemotaxonomy, genomics), as well as a new species, are presented and discussed.

    • We present the first study examining mtDNA transmission in diatoms, using sexual progeny of the pennate species Has lea ostrearia (Naviculaceae). A fragment of the cytochrome oxidase subunit I gene (cox1) with 7 nucleic substitutions between parental clones was used as a parental tracer in 16 F1 clones obtained from two pairs of mating crosses. Each cross involved a parental clone isolated from France (Bay of Bourgneuf) and Sweden (Kattegat Bay). We determined that all progeny possessed only one cox1 parental haplotype. These results suggest that the mitochondrial DNA transmission in H. ostrearia is uniparental. Implications and new topics of investigation are discussed. (C) 2013 Elsevier GmbH. All rights reserved.

    • Mg-protoporphyrin monomethyl ester (MPE) is a biosynthetic intermediate of chlorophyll and converted by MPE cyclase to protochlorophyllide. Limited availability of MPE has so far hampered cyclase research. In a new, simplified, method MPE was prepared from freeze dried bchE mutant Rhodobacter capsulatus DB575 cells by extraction with acetone/H2O/25% NH3. Isolated MPE was identified by absorption and fluorescence spectroscopy, and its purity was analyzed by HPLC. The extracted MPE was dried and redissolved in buffered DMSO and its substrate activity is shown by enzymatic cyclase assays. A linear time course was observed for MPE conversion to protochlorophyllide by enzymes from barley etioplasts. Our innovation of freeze drying the R. capsulatus cells before extraction provides a high yield method for MPE, which is significantly faster and more reproducible than previous extraction methods.

    • Angiopoietin-like 4 (ANGPTL4) is an endogenous inhibitor of lipoprotein lipase that modulates lipid levels, coronary atherosclerosis risk, and nutrient partitioning. We hypothesize that loss of ANGPTL4 function might improve glucose homeostasis and decrease risk of type 2 diabetes (T2D). We investigate protein-altering variants in ANGPTL4 among 58,124 participants in the DiscovEHR human genetics study, with follow-up studies in 82,766 T2D cases and 498,761 controls. Carriers of p.E40K, a variant that abolishes ANGPTL4 ability to inhibit lipoprotein lipase, have lower odds of T2D (odds ratio 0.89, 95% confidence interval 0.85-0.92, p = 6.3 × 10-10), lower fasting glucose, and greater insulin sensitivity. Predicted loss-of-function variants are associated with lower odds of T2D among 32,015 cases and 84,006 controls (odds ratio 0.71, 95% confidence interval 0.49-0.99, p = 0.041). Functional studies in Angptl4-deficient mice confirm improved insulin sensitivity and glucose homeostasis. In conclusion, genetic inactivation of ANGPTL4 is associated with improved glucose homeostasis and reduced risk of T2D.

  • The c-axis thermal conductivity of graphite film of nanometer thickness measured by time resolved X-ray diffraction

    • We report on the use of time resolved X-ray diffraction to measure the dynamics of strain in laser-excited graphite film of nanometer thickness, obtained by chemical vapour deposition (CVD). Heat transport in the CVD film is simulated with a 1-dimensional heat diffusion model. We find the experimental data to be consistent with a c-axis thermal conductivity of similar to 0.7 Wm(-1) K-1. This value is four orders of magnitude lower than the thermal conductivity in-plane, confirming recent theoretical calculations of the thermal conductivity of multilayer graphene.     • A kappa-symmetric action for coincident D-branes is presented. It is valid in the approximation that the additional fermionic variables, used to incorporate the non-abelian degrees of freedom, are treated classically. The action is written as a Bernstein-Leites integral on the supermanifold obtained from the bosonic worldvolume by adjoining the extra fermions. The integrand is a very simple extension of the usual Green-Schwarz action for a single brane; all symmetries, except for kappa, are manifest, and the proof of kappa-symmetry is very similar to the abelian case.

    • Bioactivity of cod (Gadus morhua) and chicken (Gallus domesticus) protein hydrolysates before and aft er in vitro gastrointestinal (GI) digestion was investigated using yeast Saccharomyces cerevisiae as a model organism. Both hydrolysates were exposed to in vitro GI digestion prior to cellular exposure to simulate digestion conditions in the human body and therefore investigate the role of modulations in the GI tract on the cell response. The effect of digested and undigested hydrolysates on intracellular oxidation, cellular metabolic energy and proteome level was investigated. No diff erence in the eff ect on intracellular oxidation activity was obtained between cod and chicken hydrolysates, while higher aff ect on intracellular oxidation was provided by digested hydrolysates, with relative values of intracellular oxidation of cod of (70.2 +/- 0.8) and chicken of (74.5 +/- 1.4) % than by undigested ones, where values of cod and chicken were (95.5 +/- 1.2) and (90.5 +/- 0.7) %, respectively. Neither species nor digestion had any eff ect on cellular metabolic energy. At proteome level, digested hydrolysates gave again signifi cantly stronger responses than undigested counterparts; cod peptides here also gave somewhat stronger response than chicken peptides. The knowledge of the action of food protein hydrolysates and their digests within live cells, also at proteome level, is important for further validation of their activity in higher eukaryotes to develop new functional food ingredients, such as in this case chicken and cod muscle-derived peptides.

    • We have employed time-resolved X-ray diffraction with picosecond temporal resolution to measure the time-dependent rocking curves of laser-irradiated asymmetrically cut single InSb crystals. Coherent acoustic phonons were excited in the crystals by irradiation with 800-nm, 100-fs laser pulses at irradiances between 0.25 and 12 mJ/cm(2). The induced time-dependent strain profiles (corresponding to the coherent phonons) were monitored by diffracting collimated, monochromatic pulses Of X-rays from the irradiated crystals. Recording of the diffracted radiation with a fast low-jitter X-ray streak camera resulted in an overall temporal resolution of better than 2 ps. The strain associated with the coherent phonons modifies the rocking curve of the crystal in a time-dependent manner, and the rocking curve is recorded by keeping the angle of incidence of the X-rays upon the crystal fixed, but varying the energy of the incident X-rays around a central energy of 8.453 keV (corresponding to the peak of the rocking curve of the unperturbed crystal). The observed time-dependent diffraction from the irradiated crystals is in reasonable agreement with simulations over a wide range of energies from the unperturbed rocking-curve peak.

  • The Role of Daylight for Humans : Gaps in Current Knowledge

    • Daylight stems solely from direct, scattered and reflected sunlight, and undergoes dynamic changes in irradiance and spectral power composition due to latitude, time of day, time of year and the nature of the physical environment (reflections, buildings and vegetation). Humans and their ancestors evolved under these natural day/night cycles over millions of years. Electric light, a relatively recent invention, interacts and competes with the natural light-dark cycle to impact human biology. What are the consequences of living in industrialised urban areas with much less daylight and more use of electric light, throughout the day (and at night), on general health and quality of life? In this workshop report, we have classified key gaps of knowledge in daylight research into three main groups: (I) uncertainty as to daylight quantity and quality needed for "optimal" physiological and psychological functioning, (II) lack of consensus on practical measurement and assessment methods and tools for monitoring real (day) light exposure across multiple time scales, and (III) insufficient integration and exchange of daylight knowledge bases from different disciplines. Crucial short and long-term objectives to fill these gaps are proposed.

    • Thioredoxin and thioredoxin reductase can regulate cell metabolism through redox regulation of disulfide bridges or through removal of H(2)O(2). These two enzymatic functions are combined in NADPH-dependent thioredoxin reductase C (NTRC), which contains an N-terminal thioredoxin reductase domain fused with a C-terminal thioredoxin domain. Rice NTRC exists in different oligomeric states, depending on the absence or presence of its NADPH cofactor. It has been suggested that the different oligomeric states may have diverse activity. Thus, the redox status of the chloroplast could influence the oligomeric state of NTRC and thereby its activity. We have characterized the oligomeric states of NTRC from barley (Hordeum vulgare L.). This also includes a structural model of the tetrameric NTRC derived from cryo-electron microscopy and single-particle reconstruction. We conclude that the tetrameric NTRC is a dimeric arrangement of two NTRC homodimers. Unlike that of rice NTRC, the quaternary structure of barley NTRC complexes is unaffected by addition of NADPH. The activity of NTRC was tested with two different enzyme assays. The N-terminal part of NTRC was tested in a thioredoxin reductase assay. A peroxide sensitive Mg-protoporphyrin IX monomethyl ester (MPE) cyclase enzyme system of the chlorophyll biosynthetic pathway was used to test the catalytic ability of both the N- and C-terminal parts of NTRC. The different oligomeric assembly states do not exhibit significantly different activities. Thus, it appears that the activities are independent of the oligomeric state of barley NTRC.

  • External nutrient loads to the Baltic Sea, 1970-2006

    • Any research related to nutrient biogeochemistry of the Baltic Sea, especially studies of eutrophication requires knowledge of the long-term dynamics of external nutrient inputs. Information accumulated in the HELCOM’s pollution load compilations is too aggregated and, until recently, covered only specific years. On the other hand, national datasets with higher resolutions are often difficult to access. Therefore, over the years considerable efforts have been put into a compilation of consistent estimates of nutrient input to the entire Baltic Sea.Excluding exchange with the Skagerrak from this report, the external nutrient input is considered here as consisting of three component parts: waterborne land loads, direct point sources at the coasts, and atmospheric depositions.The present reconstruction of time series of these three components is based on three major sources. The compilation of the land loads database has started within the project “Large-scale Environmental Effects and Ecological Processes in the Baltic Sea, 1990-1995” (Wulff et al., 2001c), continued during the MARE (“Marine Research on Eutrophication, 1999-2006”) project (Wulff et al., 2001a, Eriksson Hägg et al., 2010), and was most recently updated and expanded in connection with the latest HELCOM’s activities on the pollution load compilation (HELCOM, 2011) and the revision of the Baltic Sea Action Plan (BSAP, Wulff et al., 2009). However, there is an important difference between information contained in the HELCOM publications and the data presented here.By its very international nature HELCOM has to deal with whatever data are officially provided by the contracting parties, ending up with certain gaps and inconsistencies in the data sets (e.g. see discussion in HELCOM, 2011). On the other hand, considering eutrophication as an imbalance in the large-scale nutrient cycles, whereby more nutrients come into the system than leave it (e.g. Savchuk and Wulff, 2009 and references therein), we need to know the total amounts of external input as close to the reality as possible. Therefore, in our reconstructions we have been trying to both fill such gaps in and correct possible sources of inconsistencies. The reconstructed data sets have extensively been used by ourselves for various nutrient budget estimates (e.g. Wulff et al., 2001b, Savchuk, 2005) and as the boundary conditions for biogeochemical models (e.g. Savchuk and Wulff, 2007, 2009) including development of the eutrophication segment of BSAP (Wulff, 2007), as well as by many researchers around the Baltic Sea, for instance, within several projects of the BONUS+ research programme (e.g.Eilola et al., 2011; Eriksson Hägg et al., 2011; Meier et al., 2011).The implemented nutrient inputs have naturally been described in these publications, but briefly. In order to facilitate afurther distribution of reconstructed inputs and their usage, here we describe the process of reconstruction in more detail and make available the full data sets in digital form.The data used for the reconstruction have kindly been provided by several institutions and agencies around the Baltic Sea (see below) as well as directly by HELCOM during preparation of PLC-4 and PLC-5 (see HELCOM, 2004, 2011). Since not all data providershad given the permission to distribute the original raw measurements, we have here 3 aggregated all the inputs according to the spatial segmentation of the Baltic Sea (Fig.1) currently implemented in the biogeochemical model BALTSEM (BAltic sea Long-Term large Scale Eutrophication Model). Note also that some part of riverine inputs is available in a decision support system Baltic Nest with a much higher spatial resolution, as is further explained below.

    • Management of eutrophication in marine ecosystems requires a good understanding of nutrient cycles at the appropriate spatial and temporal scales. Here, it is shown that the biogeochemical processes controlling large-scale eutrophication of the Baltic Sea can be described with a fairly aggregated model: simple as necessary Baltic long-term large scale (SANBALTS). This model simulates the dynamics of nitrogen, phosphorus, and silica driven by the external inputs, the major physical transports, and the internal biogeochemical fluxes within the seven major sub-basins. In a long-term hindcast (1970–2003), the model outputs reasonably matched observed concentrations and fluxes. The model is also tested in a scenario where nutrient inputs are reduced to levels that existed over 100 years ago. The simulated response of the Baltic Sea trophic state to this very large reduction is verified by a similar simulation made with a much more complex process-oriented model. Both models indicate that after initial, rather rapid changes the system goes into much slower evolution, and nutrient cycles would not become balanced even after 130 years.

    • “Pre-industrial” trophic conditions in the Baltic Sea were simulated with SANBALTS (Simple As Necessary BAltic Long-Term large Scale) model. External nutrient inputs to the major basins of the Baltic Sea a century ago were reconstructed from various literature and data sources. The reconstructed input of total nitrogen was less than a half and that of total phosphorus was about a third of their contemporary values. The simulated “pre-industrial” conditions are validated by comparison to actual historical data on the water transparency, oxygen concentration, primary production, and net sediment accumulation. The “pre-industrial” trophic state could have been more phosphorus limited than today because simulated basin-wide annual averages of dissolved inorganic phosphorus concentrations of 0.06–0.3 µM P are about 40–80% of their present day values, while dissolved inorganic nitrogen concentrations of 2–4 µM N are almost the same as today or even slightly higher.

    • Rising atmospheric carbon concentrations affect global health, the economy, and overall quality of life. We are fast approaching climate tipping points that must be addressed, not only by reducing emissions but also through new innovation and action toward carbon capture for sequestration and utilization (CCSU). In this perspective, we delineate next-generation biotechnologies for CCSU supported by engineering design principles derived from ecological processes inspired by three major biomes (plant-soil, deep biosphere, and marine). These are to interface with existing industrial infrastructure and, in some cases, tap into the carbon sink potential of nature. To develop ecosystem-inspired biotechnology, it is important to identify accessible control points of CO2 and CH4 within a given system as well as value-chain opportunities that drive innovation. In essence, we must supplement natural biogeochemical carbon sinks with new bioengineering solutions. © 2020 The Authors Atmospheric carbon emissions are driving global tipping points that must be addressed by reducing emissions in combination with carbon capture for sequestration and utilization. We outline that there is much to be gained by translating ecological processes that underpin global carbon cycles into engineering principles that harness plant-soil, deep biosphere and marine microbiomes for creation of new value chains and access to Earth's major carbon storage pools. Rapid innovation is required, and biotechnology has a certain role to play. © 2020 The Authors
