    • The novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is the cause of a rapidly spreading illness, coronavirus disease 2019 (COVID-19), affecting more than seventeen million people around the world. Diagnosis and treatment guidelines for clinicians caring for patients are needed. In the early stage, we have issued "A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus (2019-nCoV) infected pneumonia (standard version)"; now there are many direct evidences emerged and may change some of previous recommendations and it is ripe for develop an evidence-based guideline. We formed a working group of clinical experts and methodologists. The steering group members proposed 29 questions that are relevant to the management of COVID-19 covering the following areas: chemoprophylaxis, diagnosis, treatments, and discharge management. We searched the literature for direct evidence on the management of COVID-19, and assessed its certainty generated recommendations using the Grading of Recommendations, Assessment, Development and Evaluation (GRADE) approach. Recommendations were either strong or weak, or in the form of ungraded consensus-based statement. Finally, we issued 34 statements. Among them, 6 were strong recommendations for, 14 were weak recommendations for, 3 were weak recommendations against and 11 were ungraded consensus-based statement. They covered topics of chemoprophylaxis (including agents and Traditional Chinese Medicine (TCM) agents), diagnosis (including clinical manifestations, reverse transcription-polymerase chain reaction (RT-PCR), respiratory tract specimens, IgM and IgG antibody tests, chest computed tomography, chest x-ray, and CT features of asymptomatic infections), treatments (including lopinavir-ritonavir, umifenovir, favipiravir, interferon, remdesivir, combination of antiviral drugs, hydroxychloroquine/chloroquine, interleukin-6 inhibitors, interleukin-1 inhibitors, glucocorticoid, qingfei paidu decoction, lianhua qingwen granules/capsules, convalescent plasma, lung transplantation, invasive or noninvasive ventilation, and extracorporeal membrane oxygenation (ECMO)), and discharge management (including discharge criteria and management plan in patients whose RT-PCR retesting shows SARS-CoV-2 positive after discharge). We also created two figures of these recommendations for the implementation purpose. We hope these recommendations can help support healthcare workers caring for COVID-19 patients.

  • Technology Roadmap for Flexible Sensors

    • Humans rely increasingly on sensors to address grand challenges and to improve quality of life in the era of digitalization and big data. For ubiquitous sensing, flexible sensors are developed to overcome the limitations of conventional rigid counterparts. Despite rapid advancement in bench-side research over the last decade, the market adoption of flexible sensors remains limited. To ease and to expedite their deployment, here, we identify bottlenecks hindering the maturation of flexible sensors and propose promising solutions. We first analyze challenges in achieving satisfactory sensing performance for real-world applications and then summarize issues in compatible sensor-biology interfaces, followed by brief discussions on powering and connecting sensor networks. Issues en route to commercialization and for sustainable growth of the sector are also analyzed, highlighting environmental concerns and emphasizing nontechnical issues such as business, regulatory, and ethical considerations. Additionally, we look at future intelligent flexible sensors. In proposing a comprehensive roadmap, we hope to steer research efforts towards common goals and to guide coordinated development strategies from disparate communities. Through such collaborative efforts, scientific breakthroughs can be made sooner and capitalized for the betterment of humanity.

  • Quantification for total demethylation potential of environmental samples utilizing the EGFP reporter gene

    • Abstract The demethylation potential of pollutants is arguably an innate component of their toxicity in environmental samples. A method was developed for determining the total demethylation potential of food samples (TDQ). The demethylation epigenetic toxicity was determined using the Hep G2 cell line transfected with pEGFP-C3 plasmids containing a methylated promoter of the EGFP reporter gene. The total demethylation potential of the sample extracts (the 5-AZA-CdR demethylation toxic equivalency) can be quantified within one week by using a standard curve of the 5-AZA-CdR demethylation agent. To explore the applicability of TDQ for environmental samples, 17 groundwater samples were collected from heavy polluted Kuihe river and the total demethylation potentials of the sample extracts were measured successfully. Meaningful demethylation toxic equivalencies ranging from 0.00050 to 0.01747 μM were found in all groundwater sample extracts. Among 19 kinds of inorganic substance, As and Cd played important roles for individual contribution to the total demethylation epigenetic toxicity. The TDQ assay is reliable and fast for quantifying the DNA demethylation potential of environmental sample extracts, which may improve epigenetic toxicity evaluations for human risk assessment, and the consistent consuming of groundwater alongside the Kuihe river pose unexpected epigenetic health risk to the local residents.

  • Review on Thermal Performance of Nanofluids With and Without Magnetic Fields in Heat Exchange Devices

    • Addition of nanoparticles into a fluid can improve the heat transfer performance of the base fluid in heat exchangers. In this work, the preparation method and process of nanofluids are introduced, and thermal properties of nanofluids, such as thermal conductivity and viscosity, are discussed deeply. This paper summarizes various theoretical models of thermal conductivity and viscosity of nanofluids. A comprehensive literature survey on applications and limitations of nanofluids has been compiled. This paper also aims to review the natural and forced convective heat transfer characteristics of nanofluids with and without magnetic fields. The discussion for the natural convective heat transfer of nanofluids focuses on the heat transfer performance of non-conventional enclosures and electric heaters. The effects on heat transfer due to variations of heated walls are also investigated. Specific applications of nanofluids in a tube with trapezoidal ribs, double-tube heat exchangers, and plate heat exchangers have been reviewed and presented in a discussion about forced convective heat transfer. The previous results show that the inlet temperature of nanofluids obviously affects the heat transfer characteristics of double-tube heat exchangers, whereas a multi-walled carbon nanotube–water nanofluid shows significant advantages in plate heat exchangers. Finally, this paper studies natural convective heat transfer of magnetic fluids in a square cavity and forced convection heat transfer in a straight tube and a corrugated structure under the action of magnetic fields. It is found that the heat transfer performance of an Fe3O4–water nanofluid is enhanced when a magnetic field is applied to the corrugated plate heat exchangers, and the pressure drop can be reduced by around 10%. It is recommended that natural convection of magnetic fluids needs to be investigated experimentally in a real cavity and a corrugated channel under the influence of a magnetic field. In addition, studies of alternating magnetic field are recommended to reveal any improvements of thermal performance of magnetic fluids in heat exchange devices. This review puts forward an effective solution for improvement of the thermal performance of heat transfer equipment and serves as a basic reference for applications of nanofluids in heat transfer fields.

    • Currently, little is known regarding the role of peroxisome proliferator-activated receptor-beta (PPAR beta) in the vascular endothelial cells (VECs) of colorectal cancers (CRCs). The aim of this study was to investigate the relationship of PPAR beta expression in the VECs of CRCs in terms of the prognosis and clinicopathological features of CRC patients. The expression and localization of PPAR beta in the primary cancers and the matched normal mucosal samples of 141 Swedish CRC patients were analyzed in terms of its correlation with clinicopathological features and the expression of angiogenesis-related genes. This study also included 92 Chinese CRC patients. PPAR beta was predominantly localized in the cytoplasm and was significantly downregulated in the VECs of CRC compared to that of the normal mucosa. The low expression levels of PPAR beta in the VECs of CRC were statistically correlated with enhanced differentiation, early staging and favorable overall survival and were associated with the increased expression of VEGF and D2-40. The patients exhibiting elevated expression of PPAR beta in CRC cells but reduced expression in VECs exhibited more favorable survival compared with the other patients, whereas the patients with reduced expression of PPAR beta in CRC cells but increased expression in VECs exhibited less favorable prognosis. PPAR beta might play a tumor suppressor role in CRC cells in contrast to a tumor promoter role in the VECs of CRCs.

  • Observation of a linked-loop quantum state in a topological magnet

    • Quantum phases can be classified by topological invariants, which take on discrete values capturing global information about the quantum state1–13. Over the past decades, these invariants have come to play a central role in describing matter, providing the foundation for understanding superfluids5, magnets6,7, the quantum Hall effect3,8, topological insulators9,10, Weyl semimetals11–13 and other phenomena. Here we report an unusual linking-number (knot theory) invariant associated with loops of electronic band crossings in a mirror-symmetric ferromagnet14–20. Using state-of-the-art spectroscopic methods, we directly observe three intertwined degeneracy loops in the material’s three-torus, T3, bulk Brillouin zone. We find that each loop links each other loop twice. Through systematic spectroscopic investigation of this linked-loop quantum state, we explicitly draw its link diagram and conclude, in analogy with knot theory, that it exhibits the linking number (2, 2, 2), providing a direct determination of the invariant structure from the experimental data. We further predict and observe, on the surface of our samples, Seifert boundary states protected by the bulk linked loops, suggestive of a remarkable Seifert bulk–boundary correspondence. Our observation of a quantum loop link motivates the application of knot theory to the exploration of magnetic and superconducting quantum matter.

  • Lipid phenotyping of lung epithelial lining fluid in healthy human volunteers

    • Background: Lung epithelial lining fluid (ELF)-sampled through sputum induction-is a medium rich in cells, proteins and lipids. However, despite its key role in maintaining lung function, homeostasis and defences, the composition and biology of ELF, especially in respect of lipids, remain incompletely understood. Objectives: To characterise the induced sputum lipidome of healthy adult individuals, and to examine associations between different ELF lipid phenotypes and the demographic characteristics within the study cohort.Methods: Induced sputum samples were obtained from 41 healthy non-smoking adults, and their lipid compositions analysed using a combination of untargeted shotgun and liquid chromatography mass spectrometry methods. Topological data analysis (TDA) was used to group subjects with comparable sputum lipidomes in order to identify distinct ELF phenotypes.Results: The induced sputum lipidome was diverse, comprising a range of different molecular classes, including at least 75 glycerophospholipids, 13 sphingolipids, 5 sterol lipids and 12 neutral glycerolipids. TDA identified two distinct phenotypes differentiated by a higher total lipid content and specific enrichments of diacyl-glycerophosphocholines, -inositols and -glycerols in one group, with enrichments of sterols, glycolipids and sphingolipids in the other. Subjects presenting the lipid-rich ELF phenotype also had significantly higher BMI, but did not differ in respect of other demographic characteristics such as age or gender.Conclusions: We provide the first evidence that the ELF lipidome varies significantly between healthy individuals and propose that such differences are related to weight status, highlighting the potential impact of (over)nutrition on lung lipid metabolism.

    • Background: Asthma is a chronic respiratory disease with significant heterogeneity in its clinical presentation and pathobiology. There is need for improved understanding of respiratory lipid metabolism in asthma patients and its relation to observable clinical features.Objective: We performed a comprehensive, prospective, cross-sectional analysis of the lipid composition of induced sputum supernatant obtained from asthma patients with a range of disease severities, as well as from healthy controls.Methods: Induced sputum supernatant was collected from 211 adults with asthma and 41 healthy individuals enrolled onto the U-BIOPRED (Unbiased Biomarkers for the Prediction of Respiratory Disease Outcomes) study. Sputum lipidomes were characterized by semiquantitative shotgun mass spectrometry and clustered using topologic data analysis to identify lipid phenotypes.Results: Shotgun lipidomics of induced sputum supernatant revealed a spectrum of 9 molecular phenotypes, highlighting not just significant differences between the sputum lipidomes of asthma patients and healthy controls, but also within the asthma patient population. Matching clinical, pathobiologic, proteomic, and transcriptomic data helped inform the underlying disease processes. Sputum lipid phenotypes with higher levels of nonendogenous, cell-derived lipids were associated with significantly worse asthma severity, worse lung function, and elevated granulocyte counts.Conclusion: We propose a novel mechanism of increased lipid loading in the epithelial lining fluid of asthma patients resulting from the secretion of extracellular vesicles by granulocytic inflammatory cells, which could reduce the ability of pulmonary surfactant to lower surface tension in asthmatic small airways, as well as compromise its role as an immune regulator.

    • Analysis of induced sputum supematant is a minimally invasive approach to study the epithelial lining fluid and, thereby, provide insight into normal lung biology and the pathobiology of lung diseases. We present here a novel proteomics approach to sputum analysis developed within the U-BIOPRED (unbiased biomarkers predictive of respiratory disease outcomes) international project. We present practical and analytical techniques to optimize the detection of robust biomarkers in proteomic studies. The normal sputum proteome was derived using data-independent HDMSE applied to 40 healthy nonsmoking participants, which provides an essential baseline from which to compare modulation of protein expression in respiratory diseases. The "core" sputum proteome (proteins detected in >= 40% of participants) was composed of 284 proteins, and the extended proteome (proteins detected in >= 3 participants) contained 1666 proteins. Quality control procedures were developed to optimize the accuracy and consistency of measurement of sputum proteins and analyze the distribution of sputum proteins in the healthy population. The analysis showed that quantitation of proteins by HDMSE is influenced by several factors, with some proteins being measured in all participants' samples and with low measurement variance between samples from the same patient. The measurement of some proteins is highly variable between repeat analyses, susceptible to sample processing effects, or difficult to accurately quantify by mass spectrometry. Other proteins show high interindividual variance. We also highlight that the sputum proteome of healthy individuals is related to sputum neutrophil levels, but not gender or allergic sensitization. We illustrate the importance of design and interpretation of disease biomarker studies considering such protein population and technical measurement variance.

    • A new species of Papaveraceae, Corydalis sunhangii, in the section Trachycarpae, is described and illustrated from Nyingchi City, Xizang, China. The new species has some resemblance to Corydalis kingdonis, but differs by radical leaves prominent, usually several, blade tripinnate (vs. insignificant, few, blade bi- to triternate); cauline leaf usually one, much smaller than radical leaves, usually situated in lower half of stem (vs. usually two, larger than radical leaves, concentrated in upper third of stem); racemes densely 13-35-flowered (vs. rather lax, 4-11-flowered); claw of lower petal shallowly saccate (vs. very prominently and deeply saccate); capsule oblong, with raised lines of dense papillae (vs. broadly obovoid, smooth). Phylogenetic analysis, based on 68 protein-coding plastid genes of 49 samples, shows that C. sunhangii is not closely related to any hitherto described species, which is consistent with our morphological analysis.

  • Semantic Integration of Plug-and-Play Software Components for Industrial Edges Based on Microservices

    • The industrial cyber-physical system enables collaboration between distributed nodes across industrial clouds and edge devices. Flexibility and interoperability could be enhanced significantly by introducing the service-oriented architecture to industrial edge devices. From the industrial edge computing perspective, software components shall be dynamically composed across heterogeneous edge devices to perform various functionalities. In this paper, a knowledge-driven Microservice-based architecture to enable plug-and-play software components is proposed for industrial edges. These software components can be dynamically configured based on the orchestration of microservices with the support of the knowledge base and the reasoning process. These semantically enhanced plug-and-play microservices could provide rapid online reconfiguration without any programming efforts. The use of the plug-and-play software components is demonstrated by an assembly line example.

    • Metasurface polarization optics that consist of 2D array of birefringent nano-antennas have proven remarkable capabilities to generate and manipulate vectorial fields with subwavelength resolution and high efficiency. Integrating this new type of metasurface with the standard vertical cavity surface-emitting laser (VCSEL) platform enables an ultracompact and powerful solution to control both phase and polarization properties of the laser on a chip, which allows to structure a VCSEL into vector beams with on-demand wavefronts. Here, this concept is demonstrated by directly generating versatile vector beams from commercially available VCSELs through on-chip integration of high-index dielectric metasurfaces. Experimentally, the versatility of the approach for the development of vectorial VCSELs are validated by implementing a variety of functionalities, including directional emission of multibeam with specified polarizations, vectorial holographic display, and vector vortex beams generations. Notably, the proposed vectorial VCSELs integrated with a single layer of beam shaping metasurface bypass the requirements of multiple cascaded optical components, and thus have the potential to promote the advancements of ultracompact, lightweight, and scalable vector beams sources, enriching and expanding the applications of VCSELs in optical communications, laser manipulation and processing, information encryption, and quantum optics.

  • Meta-analysis uncovers genome-wide significant variants for rapid kidney function decline

    • Rapid decline of glomerular filtration rate estimated from creatinine (eGFRcrea) is associated with severe clinical endpoints. In contrast to cross-sectionally assessed eGFRcrea, the genetic basis for rapid eGFRcrea decline is largely unknown. To help define this, we meta-analyzed 42 genome-wide association studies from the Chronic Kidney Diseases Genetics Consortium and United Kingdom Biobank to identify genetic loci for rapid eGFRcrea decline. Two definitions of eGFRcrea decline were used: 3 mL/min/1.73m2/year or more ("Rapid3"; encompassing 34,874 cases, 107,090 controls) and eGFRcrea decline 25% or more and eGFRcrea under 60 mL/min/1.73m2 at follow-up among those with eGFRcrea 60 mL/min/1.73m2 or more at baseline ("CKDi25"; encompassing 19,901 cases, 175,244 controls). Seven independent variants were identified across six loci for Rapid3 and/or CKDi25: consisting of five variants at four loci with genome-wide significance (near UMOD-PDILT (2), PRKAG2, WDR72, OR2S2) and two variants among 265 known eGFRcrea variants (near GATM, LARP4B). All these loci were novel for Rapid3 and/or CKDi25 and our bioinformatic follow-up prioritized variants and genes underneath these loci. The OR2S2 locus is novel for any eGFRcrea trait including interesting candidates. For the five genome-wide significant lead variants, we found supporting effects for annual change in blood urea nitrogen or cystatin-based eGFR, but not for GATM or LARP4B. Individuals at high compared to those at low genetic risk (8-14 vs 0-5 adverse alleles) had a 1.20-fold increased risk of acute kidney injury (95% confidence interval 1.08-1.33). Thus, our identified loci for rapid kidney function decline may help prioritize therapeutic targets and identify mechanisms and individuals at risk for sustained deterioration of kidney function.

  • Effects of Corrosion Products Deposited on 304 Stainless Steel on Reduction of Se (IV/VI) in Simulated Groundwater

    • Selenium (Se) is a key mobile fission product in the geological disposal of nuclear waste. It is necessary to analyze the reductive deposition behavior of iron-based materials to Se(IV) and Se(VI) in groundwater. In the present work, the corrosion behavior of 304 stainless steel in simulated groundwater (SG) and the effects of corrosion products on the dissolution of Se were investigated by electrochemical and immersion tests. Experimental results revealed that passivation films formed on 304 stainless-steel samples were destroyed by polarization measurements, forming corrosion products consisting of Fe(II) compounds, such as Fe3O4 and FeO. Corrosion products deposited on the surface of steel samples previously treated by polarization measurements in SG + CaCl2/Na2CO3/Na2SiO3 solutions effectively reduced soluble Se(IV) and Se(VI) during immersion tests, depositing FeSe2 on sample surfaces.

    • Aims/hypothesis: The study aimed to investigate the effects of HLA class I genes on susceptibility to type 1 diabetes with different onset ages, in addition to the well-established effects of HLA class II genes. Methods: A total of 361 patients with type 1 diabetes (192 patients with onset <18 years and 169 patients with onset ≥18 years) and 500 healthy control participants from China were enrolled and genotyped for the HLA-A, -B, -C, -DQA1, -DQB1 and -DRB1 genes using next-generation sequencing. Results: The susceptible DR3 (β = −0.09, p = 0.0009) and DR4-DQ8 (β = −0.13, p = 0.0059) haplotypes were negatively associated with onset age, while the protective DR11 (β = 0.21, p = 0.0314) and DR12 (β = 0.27, p < 0.0001) haplotypes were positively associated with onset age. After adjustment for linkage disequilibrium with DR-DQ haplotypes, A*11:01:01 was positively associated with onset age (β = 0.06, p = 0.0370), while the susceptible C*15:02:01 was negatively associated with onset age (β = −0.21, p = 0.0050). The unit for β was double square-root (fourth root) transformed years of change in onset age associated with per copy of the HLA haplotype/allele. In addition, B*46:01:01 was protective (OR 0.41, 0.46; pc [corrected for multiple comparisons] = 0.0044, 0.0040), whereas A*24:02:01 (OR 2.71, 2.25; pc = 0.0003, 0.0002) and B*54:01:01 (OR 3.96, 3.79; pc = 0.0018, 0.0004) were predisposing in both the <18 group and the ≥18 group compared with healthy control participants. In the context of DR4-DQ4, A*11:01:01 (61.29% vs 28.26%, pc = 0.0144) was increased while the predisposing A*24:02:01 (19.35% vs 47.83%, pc = 0.0403) was decreased in patients with onset ≥18 years when compared with patients with onset <18 years. Conclusions/interpretation: In addition to DR-DQ haplotypes, novel HLA class I alleles were detected to play a role in susceptibility to type 1 diabetes with different onset ages, which could improve the understanding of disease heterogeneity and has implications for the design of future studies.     • As a typical material of the insert in high-level radioactive waste (HLW) geological disposal canisters, iron-based materials will directly contact with groundwater after the failure of a metallic canister, acting as a chemical barrier to prevent HLW leaking into groundwater. In this paper, anoxic groundwater was simulated by mixing 10 mM NaCl and 2 mM NaHCO3 purged by Ar gas (containing 0.3% CO2) with different added ions (Ca2+, CO32− and SiO32−) and operation temperatures (25, 40 and 60 °C). An electrochemical measurement, immersion tests and surface characterization were carried out to study the corrosion behavior of pure iron in the simulated groundwater. The effects of Ca2+ on the corrosion behavior of iron is negligible, however, Cl− plays an important role in accelerating the corrosion activity with the increased concentration and temperature. With increased concentrations of CO32− and SiO32−, the corrosion resistance of iron is largely improved, which is attributed to the formation of a uniform passivation film. The independent effects of temperature on the corrosion behavior of iron are resulted from the repeated passivation–dissolution processes in the formation of the passivation film, resulting from the synergistic effects of CO32−/SiO32− and Cl−. The formation of ferric silicate is dominant in the passivation film with the addition of SiO32−, which effectively protects the iron surface from corrosion.

    • Cloud detection is a prerequisite in many remote sensing applications, and it has been tackled through different approaches from simple thresholding to complicated deep network training. On the other hand, existing approaches are susceptible to failures while handling thin clouds, largely because of their small sizes, sparse distributions, as well as high transparency and similarity to the non-cloud background regions. This paper presents global context dense block U-Net (GCDB-UNet), a robust cloud detection network that embeds global context dense block (GCDB) into the U-Net framework and is capable of detecting thin clouds effectively. GCDB consists of two feature extraction units for addressing the challenges in thin cloud detection, namely, a non-local self-attention unit that extracts sample correlation features by aggregating the sparsely distributed thin clouds and a squeeze excitation unit that extracts channel correlated features by differentiating their importance. In addition, a dense connection scheme is designed to exploit the multi-level fine-grained representations from the two types of extracted features and a recurrent refinement module is introduced for gradual enhancement of the predicted classification map. We also created a fully annotated cloud detection MODIS dataset that consists of 1192 training images, 80 validation images and 150 test images. Extensive experiments on Landsat8, SPARCS and MODIS datasets show that the proposed GCDB-UNet achieves superior cloud detection performance as compared with state-of-the-art methods.     • Objective: The quality and rationality of many recently registered clinical studies related to coronavirus disease 2019 (COVID-19) needs to be assessed. Hence, this study aims to evaluate the current status of COVID-19 related registered clinical trial. Methods: We did an electronic search of COVID-19 related clinical studies registered between December 1, 2019 and February 21, 2020 (updated to May 28, 2020) from the ClinicalTrials.gov, and collected registration information, study details, recruitment status, characteristics of the subjects, and relevant information about the trial implementation process. Results: A total of 1,706 studies were included 10.0% of which (n=171) were from France, 943 (55.3%) used an interventional design, and 600 (35.2%) used an observational design. Most of studies (73.6%) aimed to recruit fewer than 500 people. Interferon was the main prevention program, and antiviral drugs were the main treatment program. Hydroxychloroquine and chloroquine (230/943, 24.4%) were widely studied. Some registered clinical trials are incomplete in content, and 37.4% of the 1,706 studies may have had insufficient sample size. Conclusion: The quality of COVID-19 related studies needs to be improved by strengthening the registration process and improving the quality of clinical study protocols so that these clinical studies can provide high-quality clinical evidence related to COVID-19.

    • Background: Blood eosinophil counts ≥0.3x109/L are used to define Type-2, eosinophilic asthma. However, differential responses to T2 biologics of patients with eosinophilic asthma suggests that this may be a heterogeneous phenotype with subsets driven by different molecular mechanisms.Methods: Blood transcriptomic data, acquired from 99 severe asthmatics from the U-BIOPRED study (62% female, mean age 54 yr, 41% on oral steroids), were clustered by topological data analysis and cluster boundaries defined by the MORSE method. Gene pathway signatures were identified by Ingenuity Pathway Analysis.Results: Analysis revealed 3 clusters with different modulated gene pathways, i.e. molecular phenotypes. Subtype 1 had high IFN-γ, low IL5, low IL13 and low IL17 gene expression, with reduced glucocorticoid-induced gene expression. Subtype 2 had low IFNγ, high IL5, high IL13 and low IL17 gene expression. Subtype 3 had low IFNγ, high IL5, high IL13 and high IL17 gene expression. Pathway analysis suggested a strong steroid response in Subtypes 2 and 3. Clinically, the three clusters were not different in respect of age, gender, prevalence of atopy, blood or sputum eosinophil counts. Subtype 3 was characterized by high neutrophil counts in blood and bronchial epithelium, frequent sinus disease and asthma exacerbations, OCS treatment, low allergic sensitisation and low exhaled NO. Subtype 1 was characterized by high exhaled NO and more frequent IgE therapy.Conclusion: This study suggests that eosinophilic severe asthma (≥0.3x109/L) can be stratified further into 3 subtypes with distinct gene expression profiles that could be developed as molecular diagnostic biomarkers to guide treatment and thereby improve patient outcomes.

    • Fine particulate matter (PM2.5) can cause brain damage and diseases. Of note, ultrafine particles (UFPs) with an aerodynamic diameter less than or equal to 100 nm are a growing concern. Evidence has suggested toxic effects of PM2.5 and UFPs on the brain and links to neurological diseases. However, the underlying mechanism has not yet been fully illustrated due to the variety of the study models, different endpoints, etc. The adverse outcome pathway (AOP) framework is a pathway-based approach that could systematize mechanistic knowledge to assist health risk assessment of pollutants. Here, we constructed AOPs by collecting molecular mechanisms in PM-induced neurotoxicity assessments. We chose particulate matter (PM) as a stressor in the Comparative Toxicogenomics Database (CTD) and identified the critical toxicity pathways based on Ingenuity Pathway Analysis (IPA). We found 65 studies investigating the potential mechanisms linking PM2.5 and UFPs to neurotoxicity, which contained 2, 675 genes in all. IPA analysis showed that neuroinflammation signaling and glucocorticoid receptor signaling were the common toxicity pathways. The upstream regulator analysis (URA) of PM2.5 and UFPs demonstrated that the neuroinflammation signaling was the most initially triggered upstream event. Therefore, neuroinflammation was recognized as the MIE. Strikingly, there is a clear sequence of activation of downstream signaling pathways with UFPs, but not with PM2.5. Moreover, we found that inflammation response and homeostasis imbalance were key cellular events in PM2.5 and emphasized lipid metabolism and mitochondrial dysfunction, and blood-brain barrier (BBB) impairment in UFPs. Previous AOPs, which only focused on phenotypic changes in neurotoxicity upon PM exposure, we for the first time propose AOP framework in which PM2.5 and UFPs may activate pathway cascade reactions, resulting in adverse outcomes associated with neurotoxicity. Our toxicity pathway-based approach not only advances risk assessment for PM-induced neurotoxicity but shines a spotlight on constructing AOP frameworks for new chemicals.

    • Background: The aim of this study was to investigate differences in clinical features and HLA genotypes between adult-onset and childhood-onset patients with type 1 diabetes in a Chinese population. Materials and Methods: This study enrolled 716 Han Chinese patients with type 1 diabetes from Guangdong (258 childhood-onset and 458 adult-onset) to compare their clinical features. Of them 214 patients with classical type 1 diabetes (100 childhood-onset and 114 adult-onset) were selected for HLA DR and DQ genotyping by next-generation sequencing. Results: Adult-onset patients were characterized by longer duration of symptoms before diagnosis, lower frequency of DKA at disease onset, less frequent autoantibody positivity, higher serum C-peptide concentrations, and better glycemic control. These findings were replicated in the restricted cohort of 214 patients with classical type 1 diabetes. Compared with childhood-onset patients, adult-onset patients had a lower frequency of the DR9 haplotype, as well as lower frequency of high-risk DR3/DR4 and DR3/DR9 genotypes, but higher frequency of DR3/DR3 genotype and DR3/X, DR4/X or DR9/X (X, non-risk) genotypes. Conclusions: Adult-onset type 1 diabetic patients with susceptible haplotypes (DR3, DR4 or DR9) were more likely to carry protective DR-DQ haplotypes than childhood-onset patients, which suggested the association between less risk DR-DQ genotypes and the less severe clinical manifestation in adult-onset patients.

    • Background: Molecular stratification of childhood asthma could enable targeted therapy.Aims: Unbiased analysis of gene expression in paediatric severe (SA) and moderate/mild asthma (MA) blood samples to identify sub-phenotypes.Methods: Transcriptomic profiling by microarray analysis of blood from the U-BIOPRED paediatric cohort (Fleming ERJ 2015), pre- and school-age children, (SApre, n=62; MApre, n=42; SAsc, n=75 and MAsc, n=37). Topological data analysis (TDA) was used for unbiased clustering.Results: Sub-phenotypes, P1, P2, P3 and P4 were identified and are highlighted in the TDA network in the figure and a heatmap of selected variables. P1 (38% of the cohort, median 11 yrs) was characterised by low expression of glucocorticoid receptor (GR) mRNA splice variant with a long 3’ UTR (q = 2.43E-17), but no significant difference in the expression of glucocorticoid receptor (GR) mRNA splice variant with a short 3’ UTR. In P1, COX2 expression was up (q = 1.89E-06) and IFN-γ was down (q = 5.61E-06), characteristics of a decreased steroid response.Conclusion: Unbiased analysis of U-BIOPRED paediatric peripheral blood gene expression identified a sub-phenotype, P1, with an inhibited steroid response. P1 is associated with low expression of a splice variant of GR with a long 3’ UTR.

    • We propose a novel interlayer potential, which is different from usual interatomic potentials. The interlayer potential represents the interaction between atomic layers in a layered material. Based on the Chen-Mobius inversion method in combination with ab initio calculations, the interlayer interactions are obtained for the face centered cubic (fcc) (111) planes. In order to check the validity of our interlayer potential, we calculate the intrinsic stacking fault energy (gamma(sf)) and the surface energy (gamma(s)) of five metals: Al, Ni, Cu, Ag and Au. The predicted gamma(sf) and gamma(s) values are compared with the theoretical results obtained from direct calculations and also with the available experimental data. Using the interlayer potentials, we also investigate the phonon dispersion and phonon density of state in the fcc (111) plane family of the considered metals.

    • Using ab initio alloy theory in combination with the Chen-Mobius inversion method, we obtain the interaction potentials between layers of the face centered cubic (fcc) (111) plane family for elemental Pd and Ag metals and for Pd0.5Ag0.5 random alloy. Based on the interlayer potentials, we investigate properties related to the fcc (111) planes, such as the stacking fault energy, shear elastic constant, surface energy, surface relaxations, and phonon dispersion along the fcc [111] direction. The results agree well with those obtained by fully self-consistent ab initio calculations and also with the available experimental data, indicating that the properties related to the close-packed planes are well described by the interlayer potentials.

    • Precious metal Pt-based electrocatalysts have been widely used in the methanol catalytic oxidation anodes in direct methanol fuel cells. However, decreasing their cost and improving their efficiency and durability have still been challenging. Herein, ternary PtPdCu nanocatalysts are synthesized through a facile one-step hydrothermal synthesis method. When KI is present with a suitable amount in the synthesis, PtPdCu nanospheres with surface-embedded CuI clusters (CuI/PtPdCu) are fabricated. However, without KI, the prepared PtPdCu catalysts show a distinct hollow structure (h-PtPtCu). CuI/PtPdCu displays the highest specific activity with enhancement 4 times higher than commercial Pt/C for the methanol oxidation reaction in an alkaline medium. The superior activity can be attributed to two aspects: 1) the electronic effect originating from the highly alloyed PtPdCu; 2) the synergetic effect resulting from surface inlaid CuI clusters, which can promote the CO intermediate removal. Furthermore, because of the stable Pt-Pd-rich surface and its special linked hollow structure, the h-PtPtCu catalyst exhibits good durability with only a 3.6% decay in the specific activity.

    • Dissociative electron attachment to carbon monoxide, e(-) + CO -> C + O-, at 9.5, 10.0, and 10.6 eV is investigated by using the anion velocity time-sliced map imaging technique. The completely backward scattering distributions of the fast O- fragment are observed at 10.0 and 10.6 eV. The single electron-molecule resonance model fails in interpretation to these unusual angular distributions, while a quantum interference model including two (at 10.0 eV) or three (at 10.6 eV) dissociative outgoing waves is consequently proposed and provides the satisfying results about the experimental data fittings. Moreover, coherent interference among the (2)Pi, (2)Delta, and (2)Phi resonant states of CO- at 10.6 eV could be further established, based on the fact that the sum of the phase-shift fitting values equals pi rad.

    • Due to excellent thermal performance and application prospects, nanofluids are attracting many researchers to pay more attention to new types of heat transfer fluids. However, previous studies have focused on the effects of nanofluids on thermophysical properties without considering the limitation of dispersion stability in practical applications. Excellent nanofluid stability is judged with the zeta potential value above 30 mV. Unstable nanofluids block the fluid flow in heat exchanger channels, which reduces the system performance by over 23 %. Based on a systematic review of improving nanofluid stability, this research discusses the preparation, characterisation, influencing factor, dispersion mechanism, and dispersion method on nanofluid stability. Four methods for improving nanofluid stability are summarised. The nanofluid is stable at pH values between 4 and 9 b y controlling its ionic concentration. The nanofluid concentration is required below 2 % to improve the repulsion between nanoparticles. The stability of the nanofluid is affected by the type and amount of the surfactant, which fails to improve the nanofluid stability at temperature above 60 °C. In addition, the molecular forces between the mixed nanofluids enhance the stability of the nanofluid. This review examines the variation patterns of nanofluid stability and the effect of stability on heat transfer. It is expected to identify some opportunities and demonstrate future challenges in both the in-lab research and the commercialisation of nanofluids.

    • Excitation energies and lifetimes for the 213 lowest states of the n <= 5 configurations in B-like S XII are calculated using highly correlated wave functions, optimized with the fully relativistic multiconfiguration Dirac-Hartree-Fock method. Multipole transition rates and associated radiative data (line strengths and oscillator strengths) for transitions connecting these levels are also reported. The theoretical excitation energies are systematically compared with the NIST Atomic Spectra Database in which misidentifications are pointed out. After eliminating the latter, a mean energy difference with the standard deviation between computed and observed energies of 12 +/- 341 cm(-1) is obtained for the n >= 3 high-lying states. This level of accuracy confirms that elaborate ab initio calculations can assist in the identification of new emission lines in the solar and other astrophysical spectra. The present work provides atomic data of high accuracy for an ion of astrophysical interest, B-like S XII, for which experimental data are scarce.

    • Chronic kidney disease (CKD) is responsible for a public health burden with multi-systemic complications. Through transancestry meta-analysis of genome-wide association studies of estimated glomerular filtration rate (eGFR) and independent replication (n = 1,046,070), we identified 264 associated loci (166 new). Of these,147 were likely to be relevant for kidney function on the basis of associations with the alternative kidney function marker blood urea nitrogen (n = 416,178). Pathway and enrichment analyses, including mouse models with renal phenotypes, support the kidney as the main target organ. A genetic risk score for lower eGFR was associated with clinically diagnosed CKD in 452,264 independent individuals. Colocalization analyses of associations with eGFR among 783,978 European-ancestry individuals and gene expression across 46 human tissues, including tubulo-interstitial and glomerular kidney compartments, identified 17 genes differentially expressed in kidney. Fine-mapping highlighted missense driver variants in 11 genes and kidney-specific regulatory variants. These results provide a comprehensive priority list of molecular targets for translational research.

Based on the experimental results, a mechanism of H2O2 reacting with W powder in the presence of Tris is proposed. The mechanism well explained the relationship between surface reactions and homogeneous Haber-Weiss reactions.

The rolling conditions in floating LNG systems affect the uniformity of gas–liquid distribution in heat exchangers. Thermal and hydraulic performances of printed circuit heat exchangers (PCHEs) with straight, zigzag, and trapezoidal channels are investigated under static and rolling conditions. The results reveal that all channels enhance heat transfer near the pseudo-critical point of LNG at a rolling period of 2 s and a rolling amplitude of 15°. The Nusselt number for the PCHE with a zigzag channel increases by 32.2% and 72.5% compared to those with straight and trapezoidal channels. The highest increase in comprehensive performance is obtained for the zigzag channel with an evaluation index of 1.23 under the rolling condition. Compared with under the static condition, the Nusselt number and Fanning friction factor for the zigzag channel increase by 12% and 28% under the rolling condition. The thermal performance is weakened by the nonuniform flow velocity and improved by the enhancement of flow turbulence. The thermal–hydraulic performance increases with the rolling period from 1 s to 3 s and the rolling amplitude from 15° to 45°. The maximum improvement of 65.3% and 97.2% in Nusselt number and Fanning friction factors is observed at a rolling period of 1 s and a rolling amplitude of 45°. The methods to suppress the deterioration of heat transfer in microfluidic channels under rolling conditions are proposed to satisfy the requirement of LNG with low-resistance and high-efficiency microfluidic structure.

    • In this paper, periodic vortices are generated by a fluid passing a cylindrical obstacle, d, near the micro-channel inlet. Two elastic pillars are arranged on the walls, and the effect of the pillar spacing on heat transfer performance is studied using the Arbitrary Lagrangian-Euler method. With the spacing of 10d, the small pillar amplitude of 2 μm is not conducive to the generation of vortices. The flexible vortex generator has higher heat transfer efficiency and lower pressure loss than the rigid vortex generator. The two pillars with no spacing generate isolated vortices, and the mixing of these vortices is insufficient downstream the pillars. It is found that with the pillar spacing of 5d, the overall performance factor is significantly higher than that with the pillar spacing of 0d and 10d in the Reynolds number range of 800 to 1100. The average Nusselt number with the spacing of 5d increases by 19.2% compared to that with the spacing of 0d at the Reynolds number of 1000. When the Reynolds number is 1100, the overall performance factor is 43% higher than that with a single rigid pillar. The vortices are periodically generated by the two pillars with the 5d spacing, and the disturbance to the boundary layer enhances the heat transfer downstream the region in the micro-channel.

    • In this paper, the numerical dispersion of a hierarchical reconstruction strategy for DG/FV method has been optimized. For the hierarchical strategy, the cell averages and the first derivatives are reconstructed based on the Hermite WENO method; the second derivatives are reconstructed via Green-Gauss integration and WENO reconstruction. Then, the numerical dispersion has been optimized by minimizing error with bandwidth optimization technique. Furthermore, to adjust the loss of compactness due to the reconstructed second derivatives, two modifications were also proposed with optimal weights. Eventually, from the implementations in scalar and compressible Euler equations, the superiority of numerical dispersion and accuracy of present methods could be validated. The shock capturing capacity has also been validated in 1D and 2D cases.

    • Microgels with a thermo-sensitive poly(N-isopropylacrylamide) (polyNIPAm) backbone and bis-imidazolium (VIM) ionic cross-links, denoted as poly(NIPAm-co-VIM), were successfully prepared. The as-synthesized ionic microgels were converted to nanoreactors, denoted as Au@PNI MGs, upon generation and immobilization of gold nanoparticles (Au NPs) of 5-8 nm in size into poly(NIPAm-co-VIM). The content of Au NPs in microgels could be regulated by controlling the 1,6-dibromohexane/vinylimidazole molar ratio in the quaternization reaction. The microgel-based nanoreactors were morphologically spherical and uniform in size, and presented reversible thermo-sensitive behavior with volume phase transition temperatures (VPTTs) at 39-40 degrees C. The Au@PNI MGs were used for the reduction of 4-nitrophenol, of which the catalytic activity could be modulated by temperature.
