    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • To provide higher data rates, as well as better coverage, cost efficiency, security, adaptability, and scalability, the 5G and beyond 5G networks are developed with various artificial intelligence techniques. In this two-part paper, we investigatethe application of artificial intelligence (AI) and in particular machine learning (ML) to the study of wireless propagation channels. It firstly provides a comprehensive overview of ML for channel characterization and ML-based antenna-channel optimization in this first part, and then it gives a state-of-the-art literature review of channel scenario identification and channel modeling in Part II. Fundamental results and key concepts of ML for communication networks are presented, and widely used ML methods for channel data processing, propagation channel estimation, and characterization are analyzed and compared. A discussion of challenges and future research directions for ML-enabled next generation networks of the topics covered in this part rounds off the paper.

    • This two-part paper investigates the application of artificial intelligence (AI) and in particular machine learning (ML) to the study of wireless propagation channels. In Part I, we introduced AI and ML as well as provided a comprehensive survey on ML enabled channel characterization and antenna-channel optimization, and in this part (Part II) we review state-of-the-art literature on scenario identification and channel modeling here. In particular, the key ideas of ML for scenario identification and channel modeling/prediction are presented, and the widely used ML methods for propagation scenario identification and channel modeling and prediction are analyzed and compared. Based on the state-of-art, the future challenges of AI/ML-based channel data processing techniques are given as well.

  • VLBI astrometry on the white dwarf pulsar AR Scorpii

    • AR Scorpii (AR Sco), the only-known radio-pulsing white dwarf binary, shows unusual pulsating emission at the radio, infrared, optical, and ultraviolet bands. To determine its astrometric parameters at the radio band independently, we conducted multi-epoch Very Long Baseline Interferometry (VLBI) phase-referencing observations with the European VLBI Network at 5 GHz and the Chinese VLBI Network plus the Warkworth 30-m telescope (New Zealand) at 8.6 GHz. By using the differential VLBI astrometry, we provide high-precision astrometric measurements on the parallax (pi = 8.52(-0.07)(+0.04) mas) and proper motion (mu(alpha) = 9.48(-0.07)(+0.04) mas yr(-1), mu(delta) = -51.32(-0.38)(+0.22) mas yr (-1)). The new VLBI results agree with the optical Gaia astrometry. Our kinematic analysis reveals that the Galactic space velocities of AR Sco are quite consistent with that of both intermediate polars and polars. Combined with the previous tightest VLBI constraint on the size, our parallax distance suggests that the radio emission of AR Sco should be located within the light cylinder of its white dwarf.

  • A prospective clinical study on the mechanisms underlying critical illness myopathy : A time-course approach

    • Background: Critical illness myopathy (CIM) is a consequence of modern critical care resulting in general muscle wasting and paralyses of all limb and trunk muscles, resulting in prolonged weaning from the ventilator, intensive care unit (ICU) treatment and rehabilitation. CIM is associated with severe morbidity/mortality and significant negative socioeconomic consequences, which has become increasingly evident during the current COVID-19 pandemic, but underlying mechanisms remain elusive.Methods: Ten neuro-ICU patients exposed to long-term controlled mechanical ventilation were followed with repeated muscle biopsies, electrophysiology and plasma collection three times per week for up to 12 days. Single muscle fibre contractile recordings were conducted on the first and final biopsy, and a multiomics approach was taken to analyse gene and protein expression in muscle and plasma at all collection time points.Results: (i) A progressive preferential myosin loss, the hallmark of CIM, was observed in all neuro-ICU patients during the observation period (myosin:actin ratio decreased from 2.0 in the first to 0.9 in the final biopsy, P < 0.001). The myosin loss was coupled to a general transcriptional downregulation of myofibrillar proteins (P < 0.05; absolute fold change >2) and activation of protein degradation pathways (false discovery rate [FDR] <0.1), resulting in significant muscle fibre atrophy and loss in force generation capacity, which declined >65% during the 12 day observation period (muscle fibre cross-sectional area [CSA] and maximum single muscle fibre force normalized to CSA [specific force] declined 30% [P < 0.007] and 50% [P < 0.0001], respectively). (ii) Membrane excitability was not affected as indicated by the maintained compound muscle action potential amplitude upon supramaximal stimulation of upper and lower extremity motor nerves. (iii) Analyses of plasma revealed early activation of inflammatory and proinflammatory pathways (FDR < 0.1), as well as a redistribution of zinc ions from plasma.Conclusions: The mechanical ventilation-induced lung injury with release of cytokines/chemokines and the complete mechanical silencing uniquely observed in immobilized ICU patients affecting skeletal muscle gene/protein expression are forwarded as the dominant factors triggering CIM.

    • Tafazzin (TAZ) is a cardiolipin (CL) biosynthetic enzyme important for maintaining mitochondrial function. TAZ affects both the species and content of CL in the inner mitochondrial membrane, which are essential for normal cellular respiration. In pancreatic beta cells, mitochondrial function is closely associated with insulin secretion. However, the role of TAZ and CL in the secretion of insulin from pancreatic islets remains unknown. Male 4-month-old doxycycline-inducible TAZ knock-down (KD) mice and wild-type littermate controls were used. Immunohistochemistry was used to assess beta-cell morphology in whole pancreas sections, whereas ex vivo insulin secretion, CL content, RNA-sequencing analysis, and mitochondrial oxygen consumption were measured from isolated islet preparations. Ex vivo insulin secretion under nonstimulatory low-glucose concentrations was reduced similar to 52% from islets isolated from TAZ KD mice. Mitochondrial oxygen consumption under low-glucose conditions was also reduced similar to 58% in islets from TAZ KD animals.TAZ deficiency in pancreatic islets was associated with significant alteration in CL molecular species and elevated polyunsaturated fatty acid CL content. In addition, RNA-sequencing of isolated islets showed that TAZ KD increased expression of extracellular matrix genes, which are linked to pancreatic fibrosis, activated stellate cells, and impaired beta-cell function.These data indicate a novel role for TAZ in regulating pancreatic islet function, particularly under low-glucose conditions.

    • The China Initiative Accelerator Driven System (CiADS) was proposed by China Academy of Science since 2015. The subcritical reactor in CiADS is a liquid Lead Bismuth Eutectic (LBE) cooled fast reactor. When the reactor core is in operation, the LBE coolant will directly contact and corrode the inner surface of reactor vessel. Due to the high temperature, the corrosion will be more severe. If the stress on the reactor vessel exceeds the limit, the plastic deformation will occur, leading to the generation and expansion of defects and cracks, and the safety of the reactor will be affected. Therefore, evaluating the stress field of the reactor vessel under different operating conditions is a very important research project. In this paper, the finite element analysis software ADINA was applied to analyze the reactor vessel in CiADS, and the ASME Code was used as stress assessment standards. We can preliminarily prove that the stress assessments of the vessel during the postulated Unprotected Loss of Flow (ULOF) accidents satisfy the requirements of ASME Code. The limit reactivity insertion to protect the vessel from plastic deformation is 0.58$ in the postulated Unprotected Transient over Power (UTOP) accidents based on our current results. Therefore, we can preliminarily conclude that the current material selection and structural design of the reactor vessel in CiADS could survive most of the postulated transient accidents considering the stress effect.

    • LAMOST J024048.51+195226.9 (J0240+1952) was recently identified as the second AE Aquarii (AE Aqr)-type cataclysmic variable, possessing the fastest known rotating white dwarf. We performed a Very Long Baseline Interferometry (VLBI) observation of J0240+1952 utilizing the European VLBI Network at 1.7 GHz, to obtain the first view of the radio morphology on mas scale. Our high-resolution VLBI image clearly shows that the radio emission is compact on mas scale (≲2 AU), with no evidence for a radio jet or extended emission. The compact radio source has an average flux density of ∼0.37 mJy, and its brightness temperature is given at ∼2.3 × 107 K, confirming a non-thermal origin. The emission exhibits irregular variations on a time-scale of tens of minutes, similar to the radio flares seen in AE Aqr. The measured VLBI position of J0240+1952 is consistent with that derived from Gaia. Our results favour the model in which the radio emission is attributed to a superposition of synchrotron radiation from expanding magnetized blobs of this system.

    • PURPOSE: Kashin-Beck disease (KBD) is an endemic osteoarthropathy, and the severe knee pain and functional limitations were seriously affecting the quality of life in patients with end-stage KBD. We retrospectively evaluated the clinical outcomes and the quality of life in KBD patients with total knee arthroplasty (TKA).METHODS: A total of 22 subjects (25 knees) suffered KBD with severe knee pain and underwent primary TKA. Knee pain was measured by visual analogue scale (VAS), and the knee function was evaluated by Knee Society Clinical Rating System Score (KSS). KBD Quality of Life (KBDQOL) was used to evaluate the quality of life in KBD patients before and after TKA.RESULTS: There were no major complications after TKA. The levels of VAS score were obviously deceased in post-operation than that in pre-operation. The levels of KSS score were increased in one year after TKA compared with the pre-operative values, and it maintained a higher level on three years after TKA. The average KBDQOL score level of each domain in pre-operation and one and three years after TKA was increased accordingly. The average scores of physical function, activity limitation, support of society, mental health, and general health in one year after TKA were significantly higher than those in pre-operation.CONCLUSIONS: TKA can reduce knee pain, improve knee function, and improve the quality life in KBD patients. KBDQOL questionnaire may be a promising instrument for assessing the quality life in KBD patients.

    • The Visual Object Tracking challenge VOT2021 is the ninth annual tracker benchmarking activity organized by the VOT initiative. Results of 71 trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in recent years. The VOT2021 challenge was composed of four sub-challenges focusing on different tracking domains: (i) VOT-ST2021 challenge focused on short-term tracking in RGB, (ii) VOT-RT2021 challenge focused on "real-time" short-term tracking in RGB, (iii) VOT-LT2021 focused on long-term tracking, namely coping with target disappearance and reappearance and (iv) VOT-RGBD2021 challenge focused on long-term tracking in RGB and depth imagery. The VOT-ST2021 dataset was refreshed, while VOT-RGBD2021 introduces a training dataset and sequestered dataset for winner identification. The source code for most of the trackers, the datasets, the evaluation kit and the results along with the source code for most trackers are publicly available at the challenge website(1).

    • The Visual Object Tracking challenge VOT2019 is the seventh annual tracker benchmarking activity organized by the VOT initiative. Results of 81 trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in the recent years. The evaluation included the standard VOT and other popular methodologies for short-term tracking analysis as well as the standard VOT methodology for long-term tracking analysis. The VOT2019 challenge was composed of five challenges focusing on different tracking domains: (i) VOT-ST2019 challenge focused on short-term tracking in RGB, (ii) VOT-RT2019 challenge focused on "real-time" short-term tracking in RGB, (iii) VOT-LT2019 focused on long-term tracking namely coping with target disappearance and reappearance. Two new challenges have been introduced: (iv) VOT-RGBT2019 challenge focused on short-term tracking in RGB and thermal imagery and (v) VOT-RGBD2019 challenge focused on long-term tracking in RGB and depth imagery. The VOT-ST2019, VOT-RT2019 and VOT-LT2019 datasets were refreshed while new datasets were introduced for VOT-RGBT2019 and VOT-RGBD2019. The VOT toolkit has been updated to support both standard short-term, long-term tracking and tracking with multi-channel imagery. Performance of the tested trackers typically by far exceeds standard baselines. The source code for most of the trackers is publicly available from the VOT page. The dataset, the evaluation kit and the results are publicly available at the challenge website(1).

    • Most B cell precursor acute lymphoblastic leukemia (BCP ALL) can be classified into known major genetic subtypes, while a substantial proportion of BCP ALL remains poorly characterized in relation to its underlying genomic abnormalities. We therefore initiated a large-scale international study to reanalyze and delineate the transcriptome landscape of 1,223 BCP ALL cases using RNA sequencing. Fourteen BCP ALL gene expression subgroups (G1 to G14) were identified. Apart from extending eight previously described subgroups (G1 to G8 associated with MEF2D fusions, TCF3–PBX1 fusions, ETV6–RUNX1–positive/ETV6–RUNX1–like, DUX4 fusions, ZNF384 fusions, BCR–ABL1/Ph–like, high hyperdiploidy, and KMT2A fusions), we defined six additional gene expression subgroups: G9 was associated with both PAX5 and CRLF2 fusions; G10 and G11 with mutations in PAX5 (p.P80R) and IKZF1 (p.N159Y), respectively; G12 with IGH–CEBPE fusion and mutations in ZEB2 (p.H1038R); and G13 and G14 with TCF3/4–HLF and NUTM1 fusions, respectively. In pediatric BCP ALL, subgroups G2 to G5 and G7 (51 to 65/67 chromosomes) were associated with low-risk, G7 (with ≤50 chromosomes) and G9 were intermediate-risk, whereas G1, G6, and G8 were defined as high-risk subgroups. In adult BCP ALL, G1, G2, G6, and G8 were associated with high risk, while G4, G5, and G7 had relatively favorable outcomes. This large-scale transcriptome sequence analysis of BCP ALL revealed distinct molecular subgroups that reflect discrete pathways of BCP ALL, informing disease classification and prognostic stratification. The combined results strongly advocate that RNA sequencing be introduced into the clinical diagnostic workup of BCP ALL. four decades, most of the recurring chromosomal abnormalities, including aneuploidy, chromosomal rearrangements/gene fusions (e.g., ETV6–RUNX1, BCR–ABL1, and TCF3–PBX1), and rearrangements of KMT2A (previously MLL), were identified by.

  • A Multifunctional Small-Molecule Hole- Transporting Material Enables Perovskite QLEDs with EQE Exceeding 20%

    • Hole-transporting materials (HTMs) play critical roles in the device performance and stability of perovskite quantum dot light-emitting diodes (Pe-QLEDs). However, the development of small-molecule HTMs for achieving high-performance Pe-QLEDs has proven to be very challenging because of their low hole mobility and poor solvent resistance. Herein, we tailor-made a multifunc-tional small-molecule HTM, termed X10, with methoxy as the substituents, for application in Pe-QLEDs. X10 features high hole mobility, good film-forming ability, and strong solvent resistance ability as well as defect passivation effect. Subsequently, Pe-QLEDs employing X10 as HTM presented a promising external quantum efficiency (EQE) of 20.18%, which is 7-fold higher than that of the reference HTM-TCTA-based ones (EQE approximate to 2.88%). To the best of our knowledge, this is the first case in which a small-molecule HTM displays a high EQE over 20% in Pe-QLEDs. Our work provides important guidance for the rational design of multifunctional small-molecule HTMs for high-performance Pe-QLEDs.

  • Population Genomics Reveal Recent Speciation and Rapid Evolutionary Adaptation in Polar Bears

    • Polar bears are uniquely adapted to life in the High Arctic and have undergone drastic physiological changes in response to Arctic climates and a hyper-lipid diet of primarily marine mammal prey. We analyzed 89 complete genomes of polar bear and brown bear using population genomic modeling and show that the species diverged only 479-343 thousand years BP. We find that genes on the polar bear lineage have been under stronger positive selection than in brown bears; nine of the top 16 genes under strong positive selection are associated with cardiomyopathy and vascular disease, implying important reorganization of the cardiovascular system. One of the genes showing the strongest evidence of selection, APOB, encodes the primary lipoprotein component of low-density lipoprotein (LDL); functional mutations in APOB may explain how polar bears are able to cope with life-long elevated LDL levels that are associated with high risk of heart disease in humans.

  • Prey diversity and prey stomach contents affect astaxanthin levels in piscivorous fish

    • We analyzed astaxanthin concentrations and the composition of geometrical (E/Z) astaxanthin isomers in 631 tissue samples from the four chief fish species in the pelagic zone of the brackish Baltic Sea. Salmon and herring showed signs of astaxanthin deficiency, but cod and sprat did not. The isomers were distributed selectively in fish tissues, with highest proportions of all-E-astaxanthin in salmon gonads (71%) and lowest in herring gonads (19%). We discovered that the clupeids are no ideal prey for salmon and cod with respect to their high whole-body concentrations of astaxanthin Z-isomers, which have low bioavailability for salmon and cod. The salmon in the Baltic Sea is entirely dependent on herring and sprat for food intake while cod feeds on a more diverse diet, including crustaceans. This explains the normal low astaxanthin levels in the salmon in the Baltic Sea. Observed decreases in astaxanthin levels in the Baltic salmon during the last 50 years, which are related to a reproductional disturbance (M74 syndrome), can be explained by the here described poor quality of herring as astaxanthin source in combination with recorded changes in the feeding ecology of the Baltic salmon with less sprat and more herring in the diet today. Herring is inferior to sprat as astaxanthin source, especially in autumn when a salmon or cod obtains four times more bioavailable all-E-astaxanthin (by weight) from sprat than from herring. The Baltic herring is starving more than the sprat as a result of competition between the clupeids though fishing mortality and recruitment problems of the cod, their major predator during the last decades. Therefore, less crustacean astaxanthin (mainly all-E) is transferred directly to piscivorous fish from herring stomachs than from sprat stomachs.

    • Atlantic salmon living in the brackish Baltic Sea have lower muscle pigmentation than populations elsewhere. The pigment in question is the antioxidant and vitamin A precursor astaxanthin, which is synthesized by crustaceans from algal carotenoids. Baltic salmon feed nearly exclusively on the clupeids sprat and herring. To evaluate astaxanthin availability to salmon we assessed astaxanthin levels and isomeric composition in their prey fish. We also analyzed astaxanthin dynamics in the dominant piscivorous fish in the Baltic Sea, the Atlantic cod. The geometrical E-(trans-) and Z-(cis-) isomers were distributed selectively in fish tissues, with highest E : Z ratios in salmon gonads (82 : 18) and lowest in herring gonads (24 : 76). Sprat and herring are not ideal prey with respect to their high whole-body concentrations of Z-isomers, which have low bioavailability for salmon and cod. These Z-isomers predominantly accumulate in the clupeid gonads. A crucial mechanism for the transport of astaxanthin from clupeids to piscivores is the direct transfer of crustacean astaxanthin (mainly all-E) from the clupeid stomachs. Low stomach astaxanthin content in clupeids decreases total astaxanthin transfer to higher trophic levels. In autumn, herring stomachs (including contents) had 12.5 times lower astaxanthin concentrations than sprat stomachs, and herring had 2.8 times less whole-body all-E-astaxanthin (by weight) than sprat. These results confirm recent reports of starvation in the Baltic herring, which may further decrease astaxanthin levels in the Baltic salmon. Cod did not have lower astaxanthin levels than their Atlantic counterpart, which may be attributed to their lower need for astaxanthin and higher food diversity.

    • Two recent studies suggest that experimental pain sensitivity is associated with low-grade systemic inflammation. However, only 2 biomarkers have been identified, and the studies were conducted in adult individuals where confounding effects of comorbid diseases cannot be excluded. We therefore tested associations between pain sensitivity and 119 inflammation-related serum biomarkers in 827 healthy adolescents (15-19 years) in the population-based Tromso Study: Fit Futures. The main outcome measure was cold-pressor pain tolerance (CPT), tested by placing the dominant hand in circulating cold (3 degrees C) water for a maximum of 105 seconds. Secondary outcomes were heat and pressure pain threshold and tolerance. Twelve proteins and 6 fatty acids were significantly associated with CPT after adjustment for possible confounding factors and correction for multiple comparisons. Of these, all fatty acids and 10 proteins were protective, ie, higher biomarkers levels were associated with increased CPT, whereas 2 biomarkers were associated with lower tolerance. Taken together, these biomarkers predicted completion of the tolerance test with a C-statistic of 0.65. Results for heat and pressure pain tolerance were remarkably similar, strengthening the generalizability of our findings. In this cohort of young healthy individuals, we found a relationship between inflammation-related biomarkers and pain tolerance and thresholds. Biomarkers with anti-inflammatory and analgesic effects predominated, suggesting that the development of prophylactic dietary or pharmaceutical treatments may be possible.

    • Pulsed corona discharge is an efficient method on NO oxidization, and the investigation of the oxidization process is significant both for model validation and industrial application. In-situ visualization of NO and OH in pulsed corona discharge was performed by planar laser-induced fluorescence (PLIF) in this work. Two dimensional NO oxidization and OH consumption were studied under different conditions. Some significant results were obtained for mixing behaviors of NO oxidization process. The NO oxidization rate and OH consumption increased by 22% and 40%, respectively, as the Re number of additional gas flow increased from 1379 to 4138. The OH utilization ratio was defined to describe the effect of OH radicals on NO oxidization process. It is demonstrated that OH plays an important role on NO oxidization.     • Determination of the structures of materials involving more light elements such as boron-rich compounds is challenging and technically important in understanding their varied compositions and superior functionalities. Here we resolve the long-standing uncertainties in structure and composition about the highest boride (termed MoB4, Mo1-xB3, or MoB3) through the rapid formation of large sized boron-rich molybdenum boride under pressure. Using high-quality single-crystal X-ray diffraction analysis and aberration-corrected scanning transmission electron microscopy, we reveal that boron-rich molybdenum boride with a composition of Mo0.757B3 exhibits P6(3)/mmc symmetry with a partial occupancy of 0.514 in 211 Mo sites (Mol), and direct observations reveal the short-range ordering of cation vacancies in (010) crystal planes. Large anisotropic Young's moduli and Vickers hardness are seen for Mo0.757B3, which may be attributed by its two-dimensional boron distributions. Mo0.757B3 is also found to be superconducting with a transition temperature (T-c) of 2.4 K, which was confirmed by measurements of resistivity and magnetic susceptibility. Theoretical calculations suggest that the partial occupancy of Mo atoms plays a crucial role in the emergence of superconductivity.

    • Background: High doses of doxorubicin put cancer patients at risk for developing dilated cardiomyopathy. Previously, we showed that doxorubicin treatment decreases SIRT3 (sirtuin 3), the main mitochondrial deacetylase and increases protein acetylation in rat cardiomyocytes. Here, we hypothesize that SIRT3 expression can attenuate doxorubicin induced dilated cardiomyopathy in vivo by preventing the acetylation of mitochondrial proteins. Methods: Nontransgenic, M3-SIRT3 (truncated SIRT3; short isoform), and M1-SIRT3 (full-length SIRT3; mitochondrial localized) transgenic mice were treated with doxorubicin for 4 weeks (8 mg/kg body weight per week). Echocardiography was performed to assess cardiac structure and function and validated by immunohistochemistry and immunofluorescence (n=4-10). Mass spectrometry was performed on cardiac mitochondrial peptides in saline (n=6) and doxorubicin (n=5) treated hearts. Validation was performed in doxorubicin treated primary rat and human induced stem cell derived cardiomyocytes transduced with adenoviruses for M3-SIRT3 and M1-SIRT3 and deacetylase deficient mutants (n=4-10). Results: Echocardiography revealed that M3-SIRT3 transgenic mice were partially resistant to doxorubicin induced changes to cardiac structure and function whereas M1-SIRT3 expression prevented cardiac remodeling and dysfunction. In doxorubicin hearts, 37 unique acetylation sites on mitochondrial proteins were altered. Pathway analysis revealed these proteins are involved in energy production, fatty acid metabolism, and oxidative stress resistance. Increased M1-SIRT3 expression in primary rat and human cardiomyocytes attenuated doxorubicin-induced superoxide formation, whereas deacetylase deficient mutants were unable to prevent oxidative stress. Conclusions: Doxorubicin reduced SIRT3 expression and markedly affected the cardiac mitochondrial acetylome. Increased M1-SIRT3 expression in vivo prevented doxorubicin-induced cardiac dysfunction, suggesting that SIRT3 could be a potential therapeutic target for mitigating doxorubicin-induced dilated cardiomyopathy.

    • Environmental pollution is escalating due to rapid global development that often prioritizes human needs over planetary health. Despite global efforts to mitigate legacy pollutants, the continuous introduction of new substances remains a major threat to both people and the planet. In response, global initiatives are focusing on risk assessment and regulation of emerging contaminants, as demonstrated by the ongoing efforts to establish the UN's Intergovernmental Science-Policy Panel on Chemicals, Waste, and Pollution Prevention. This review identifies the sources and impacts of emerging contaminants on planetary health, emphasizing the importance of adopting a One Health approach. Strategies for monitoring and addressing these pollutants are discussed, underscoring the need for robust and socially equitable environmental policies at both regional and international levels. Urgent actions are needed to transition toward sustainable pollution management practices to safeguard our planet for future generations.

    • Sixty genetic loci associated with abdominal obesity, measured by waist circumference (WC) and waist-hip ratio (WHR), have been previously identified, primarily from studies conducted in Europeanancestry populations. We conducted a meta-analysis of associations of abdominal obesity with approximately 2.5 million single nucleotide polymorphisms (SNPs) among 53,052 (for WC) and 48,312 (for WHR) individuals of Asian descent, and replicated 33 selected SNPs among 3,762 to 17,110 additional individuals. We identified four novel loci near the EFEMP1, ADAMTSL3, CNPY2, and GNAS genes that were associated with WC after adjustment for body mass index (BMI); two loci near the NID2 and HLA-DRB5 genes associated with WHR after adjustment for BMI, and three loci near the CEP120, TSC22D2, and SLC22A2 genes associated with WC without adjustment for BMI. Functional enrichment analyses revealed enrichment of corticotropin-releasing hormone signaling, GNRH signaling, and/or CDK5 signaling pathways for those newly-identified loci. Our study provides additional insight on genetic contribution to abdominal obesity.

    • Bismuth-based double perovskite Cs2AgBiBr6 is regarded as a potential candidate for low-toxicity, high-stability perovskite solar cells. However, its performance is far from satisfactory. Albeit being an indirect bandgap semiconductor, we observe bright emission with large bimolecular recombination coefficient (reaching 4.5 +/- 0.1 x 10(-11) cm(3) s(-1)) and low charge carrier mobility (around 0.05 cm(2) s(-1) V-1). Besides intermediate Frohlich couplings present in both Pb-based perovskites and Cs2AgBiBr6, we uncover evidence of strong deformation potential by acoustic phonons in the latter through transient reflection, time-resolved terahertz measurements, and density functional theory calculations. The Frohlich and deformation potentials synergistically lead to ultrafast self-trapping of free carriers forming polarons highly localized on a few units of the lattice within a few picoseconds, which also breaks down the electronic band picture, leading to efficient radiative recombination. The strong self-trapping in Cs2AgBiBr6 could impose intrinsic limitations for its application in photovoltaics.

    • The cathode interface plays a critical role in achieving high-performance fullerene/perovskite planar solar cells. Herein, the simple molecule Isatin and its derivatives are introduced at the back contact [6,6]-phenyl-C61-butyric acid methyl ester (PCBM)/Al as a cathode modification interlayer. It is revealed that the Isatin interlayers facilitate electron transport/extraction and suppress electron recombination, attributed to the formation of negative dipole potential steps and the passivation of the interfacial trap density. The average power conversion efficiencies of the resulting devices are significantly improved by 11% from 17.68% to 19.74%, with an enhancement in all device parameters including short-circuit current, open-circuit voltage, and fill factor. The hysteresis index is found to disappear. In addition, such interlayer enhances device stability under ambient conditions compared to the control devices due to suppression of moisture-induced degradation of the perovskite films. These findings provide a comprehensive understanding of the engineering of the back contact between PCBM and the metal electrode to improve efficiency and stability of perovskite solar cells.

  • Coupling Downscaling and Calibrating Methods for Generating High-Quality Precipitation Data with Multisource Satellite Data in the Yellow River Basin

    • Remote sensing precipitation data have the characteristics of wide coverage and revealing spatiotemporal information, but their spatial resolution is low. The accuracy of the data is obviously different in different study areas and hydrometeorological conditions. This study evaluated four precipitation products in the Yellow River basin from 2001 to 2019, constructed the optimal combined product, conducted downscaling with various machine algorithms, and performed corrections using meteorological station precipitation data to analyze the spatiotemporal trends of precipitation. The results showed that (1) GPM and MSWEP had the best four evaluation indicators, with R2 values of 0.93 and 0.90, respectively, and the smallest FSE and RMSE, with a BIAS close to 0. A high-precision mixed precipitation dataset, GPM-MSWEP, was constructed. (2) Among the three methods, the downscaling results of DFNN showed higher accuracy. (3) The results, after correction with GWR, could more effectively enhance the accuracy of the data. (4) Precipitation in the Yellow River Basin showed a decreasing trend in January, September, and December, while it exhibited an increasing trend in other months and seasons, with 2002 and 2016 being points of abrupt change. This study provides a reference for the production of high-precision satellite precipitation products in the Yellow River basin.

    • Sensorimotor timing behaviors typically exhibit an elusive phenomenon known as the negative asynchrony. When synchronizing movements (e.g. finger taps) with an external sequence (e.g. a metronome), people’s taps precede event onsets by a few tens of milliseconds. We recently reported that asynchrony is less negative in participants with lower asynchrony variability. This indicates an association between negative asynchrony and variability of timing. Here, in 24 metronome-synchronization data sets, we modeled asynchrony series using a sensorimotor synchronization model that accounts for serial dependence of asynchronies. The results showed that the modeling well captured the negative correlation between the mean and SD of asynchrony. The finding suggests that serial dependence in asynchronies is an essential mechanism of timing variability underlying the association between the mean and SD of asynchrony.

    • Researchers have puzzled over the phenomenon in sensorimotor timing that people tend to tap ahead of time. When synchronizing movements (e.g., finger taps) with an external sequence (e.g., a metronome), humans typically tap tens of milliseconds before event onsets, producing the elusive negative asynchrony. Here, we present 24 metronome-tapping data sets from 8 experiments with different experimental settings, showing that less negative asynchrony is associated with lower tapping variability. Further analyses reveal that this negative mean-SD correlation of asynchrony is likely to be observed for sequence types appropriate for synchronization, as indicated by the statistically negative lag 1 autocorrelation of inter-response intervals. The reported findings indicate an association between negative asynchrony and timing variability.

Exploring and analyzing these categories, we present early insights pointing to design implications for future eyes-free interactions.

    • Carbon is one of the most fascinating elements due to its structurally diverse allotropic forms stemming from its bonding varieties (sp, sp2, and sp3). Exploring new forms of carbon has always been the eternal theme of scientific research. Herein, we report the amorphous (AM) carbon materials with high fraction of sp3 bonding recovered from compression of fullerene C60 under high pressure and high temperature previously unexplored. Analysis of photoluminescence and absorption spectra demonstrates that they are semiconducting with a bandgap range of 1.5–2.2 eV, comparable to that of widely used amorphous silicon. Comprehensive mechanical tests demonstrate that the synthesized AM-III carbon is the hardest and strongest amorphous material known so far, which can scratch diamond crystal and approach its strength. The produced AM carbon materials combine outstanding mechanical and electronic properties, and may potentially be used in photovoltaic applications that require ultrahigh strength and wear resistance.

    • A catalyst systern containing an electrophilic center and a sterically hindered nUcleophilic center in one molecule was applied to the cycloaddition reaction of CO2 and epoxides. This intramolecularly two-centered cooperation catalyst showed activity even at a high [epoxide]/[catalyst] ratio up to 50000 under mild conditions such as solvent-free, ambient temperature, and low CO2 pressure. The reaction of CO2 with (S)-propylene oxide at 80 degrees C in the presence of the bifunctional catalyst gives (S)-propylene carbonate in 96% ee with retention of stereochemistry.

    • Background: Necrotizing enterocolitis (NEC) is the leading cause of neonatal gastrointestinal mortality; effective interventions are lacking with limited understanding of the pathogenesis of NEC. The importance of Toll-like receptor 4 (TLR4) signaling in NEC is well documented; however, the potential mechanisms that regulate enterocyte inflammation and apoptosis remain unclear. The aim of this study was to characterize the role of TLR4-mediated inflammation and apoptosis in the development of NEC and to determine the major apoptotic pathways and regulators in the process. Methods: TLR4-deficient C57BL/10ScNJ mice and lentivirus-mediated stable TLR4-silent cell line (IEC-6) were used. NEC was induced by formula gavage, cold, hypoxia, combined with lipopolysaccharide in vivo or lipopolysaccharide stimulation in vitro. Enterocyte apoptosis was evaluated by TUNEL or Annexin analysis. The expression of TLR4, caspase3, caspase8, caspase9, Bip, Bax, Bcl-2, and RIP was detected by Western blot and immunofluorescence. Inflammatory factors such as tumor necrosis factor-a and interleukin-2 were examined by Luminex. Results: Defect of TLR4 led to suppressed enterocytes apoptosis both in vitro and in vivo; the expression of caspase3, caspase8, Bip, and Bax was decreased; and caspase9 and Bcl-2 were increased. NEC severity was attenuated in TLR4-deficient mice compared with wild-type counterparts, and enterocytes apoptosis was correlated with NEC severity. RIP and cytokine level of tumor necrosis factor-a and interleukin-2 were also decreased. Conclusions: TLR4-induced inflammation and apoptosis play a critical role in the pathogenesis of NEC. TLR4 inhibition, combined with extrinsic (caspase8) and/or endoplasmic reticulum stress (Bip) apoptosis signaling blockade could serve as a potential effective treating strategy for NEC.
