  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • The Visual Object Tracking challenge VOT2018 is the sixth annual tracker benchmarking activity organized by the VOT initiative. Results of over eighty trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in the recent years. The evaluation included the standard VOT and other popular methodologies for short-term tracking analysis and a “real-time” experiment simulating a situation where a tracker processes images as if provided by a continuously running sensor. A long-term tracking subchallenge has been introduced to the set of standard VOT sub-challenges. The new subchallenge focuses on long-term tracking properties, namely coping with target disappearance and reappearance. A new dataset has been compiled and a performance evaluation methodology that focuses on long-term tracking capabilities has been adopted. The VOT toolkit has been updated to support both standard short-term and the new long-term tracking subchallenges. Performance of the tested trackers typically by far exceeds standard baselines. The source code for most of the trackers is publicly available from the VOT page. The dataset, the evaluation kit and the results are publicly available at the challenge website (http://votchallenge.net).

The VOT2019 challenge was composed of five challenges focusing on different tracking domains: (i) VOT-ST2019 challenge focused on short-term tracking in RGB, (ii) VOT-RT2019 challenge focused on "real-time" short-term tracking in RGB, (iii) VOT-LT2019 focused on long-term tracking namely coping with target disappearance and reappearance. Two new challenges have been introduced: (iv) VOT-RGBT2019 challenge focused on short-term tracking in RGB and thermal imagery and (v) VOT-RGBD2019 challenge focused on long-term tracking in RGB and depth imagery. The VOT-ST2019, VOT-RT2019 and VOT-LT2019 datasets were refreshed while new datasets were introduced for VOT-RGBT2019 and VOT-RGBD2019. The VOT toolkit has been updated to support both standard short-term, long-term tracking and tracking with multi-channel imagery. Performance of the tested trackers typically by far exceeds standard baselines. The source code for most of the trackers is publicly available from the VOT page. The dataset, the evaluation kit and the results are publicly available at the challenge website(1).

    • Mycotoxin T-2 exerts a causative role in Kashin-Beck disease (KBD) suffering chondrocyte apoptosis and cartilage matrix homeostasis disruption. Recent research corroborated the aberrant levels of pro-inflammatory cytokine IL-1ß in KBD patients and mycotoxin environment. In the present study, we investigated the relevance of IL-1ß in T-2 toxin-evoked chondrocyte cytotoxic injury and aberrant catabolism. High levels of IL-1ß were detected in serum and cartilages from KBD patients and in T-2-stimulated chondrocytes. Moreover, knockdown of IL-1ß antagonized the adverse effects of T-2 on cytotoxic injury by enhancing cell viability and inhibiting apoptosis. However, exogenous supplementation of IL-1β further aggravated cell damage in response to T-2. Additionally, cessation of IL-1β rescued T-2-elicited tilt of matrix homeostasis toward catabolism by elevating the transcription of collagen II and aggrecan, promoting release of sulphated glycosaminoglycans (sGAG) and TIMP1, and suppressing matrix metalloproteinases production including MMP-1, MMP-3 and MMP-13. Conversely, IL-1β stimulation deteriorated T-2-induced disruption of matrix metabolism balance toward catabolism. Mechanistic analysis found the high activation of Wnt/β-catenin in KBD patients and chondrocytes upon T-2. Furthermore, this activation was mitigated after IL-1β inhibition, but further enhanced following IL-1β precondition. Importantly, blocking this pathway by transfection with β-catenin alleviated the adverse roles of IL-1β on cytotoxic injury and metabolism disorders under T-2 conditioning. Together, this study elucidates a new insight into how T-2 deteriorates the pathological progression of KBD by regulating inflammation-related pathways, indicating a promising anti-inflammation strategy for KBD therapy.

  • Optical characterization of Chinese hybrid rice using laser-induced fluorescence techniques—laboratory and remote-sensing measurements

    • Chinese hybrid rice of different varieties, growing in paddies in the Pingqiao district, north of Xinyang city, Henan province, China, was studied in detailed spectroscopic characteristics using laser-induced fluorescence. The base for the studies was the new South China Normal University mobile lidar laboratory, which was dispatched on site, providing facilities both for laboratory studies using a 405 nm excitation source as well as remote sensing measurements at ranges from around 40 m–120 m, mostly employing the 532 nm output from a Nd:YAG laser. We, in particular, studied the spectral influence of the species varieties as well as the level of nitrogen fertilization supplied. Specially developed contrast functions as well as multivariate techniques with principal components and Fisher’s discriminate analyses were applied, and useful characterization of the rice could be achieved. The chlorophyll content mapping of the 30 zones was obtained with the remote sensing measurements.

    • None-fullerene solar cells with ternary architecture have attracted much attention because it is an effective approach for boosting the device power conversion efficiency. Here, the crystalline polymer N2200 as the third component is integrated into J71: ITIC bulk heterojunction. A series of characterizations indicate that N2200 could increase photo-harvesting, balanced hole and electron mobilities, enhanced exciton dissociation, and suppressed charge recombination, which result in the comprehensive improvement of open circuit voltage, short circuit current and fill factor in the device. Moreover, after introduction of N2200, the morphology of the ternary active layer is optimized, and the film crystallinity is improved. This work demonstrates that adding a small quantity of high crystallization acceptor into non-fullerene donor: acceptor mixture is a promising strategy toward developing high-performance organic solar cells.

  • Management of Real-Time Data Consistency and Transient Overloads in Embedded Systems

    • This thesis addresses the issues of data management in embedded systems' software. The complexity of developing and maintaining software has increased over the years due to increased availability of resources, e.g., more powerful CPUs and larger memories, as more functionality can be accommodated using these resources.In this thesis, it is proposed that part of the increasing complexity can be addressed by using a real-time database since data management is one constituent of software in embedded systems. This thesis investigates which functionality a real-time database should have in order to be suitable for embedded software that control an external environment. We use an engine control software as a case study of an embedded system.The findings are that a real-time database should have support for keeping data items up-todate, providing snapshots of values, i.e., the values are derived from the same system state, and overload handling. Algorithms are developed for each one of these functionalities and implemented in a real-time database for embedded systems. Performance evaluations are conducted using the database implementation. The evaluations show that the real-time performance is improved by utilizing the added functionality.Moreover, two algorithms for examining whether the system may become overloaded are also outlined; one algorithm for off-line use and the second algorithm for on-line use. Evaluations show the algorithms are accurate and fast and can be used for embedded systems.

  • Steric hindrance induced low exciton binding energy enables low-driving-force organic solar cells

    • Exciton binding energy (Eb) has been regarded as a critical parameter in charge separation during photovoltaic conversion. Minimizing the Eb of the photovoltaic materials can facilitate the exciton dissociation in low-driving force organic solar cells (OSCs) and thus improve the power conversion efficiency (PCE); nevertheless, diminishing the Eb with deliberate design principles remains a significant challenge. Herein, bulky side chain as steric hindrance structure was inserted into Y-series acceptors to minimize the Eb by modulating the intra- and intermolecular interaction. Theoretical and experimental results indicate that steric hindrance-induced optimal intra- and intermolecular interaction can enhance molecular polarizability, promote electronic orbital overlap between molecules, and facilitate delocalized charge transfer pathways, thereby resulting in a low Eb. The conspicuously reduced Eb obtained in Y-ChC5 with pinpoint steric hindrance modulation can minimize the detrimental effects on exciton dissociation in low-driving-force OSCs, achieving a remarkable PCE of 19.1% with over 95% internal quantum efficiency. Our study provides a new molecular design rationale to reduce the Eb.

    • PURPOSE: Kashin-Beck disease (KBD) is an endemic osteoarthropathy, and the severe knee pain and functional limitations were seriously affecting the quality of life in patients with end-stage KBD. We retrospectively evaluated the clinical outcomes and the quality of life in KBD patients with total knee arthroplasty (TKA).METHODS: A total of 22 subjects (25 knees) suffered KBD with severe knee pain and underwent primary TKA. Knee pain was measured by visual analogue scale (VAS), and the knee function was evaluated by Knee Society Clinical Rating System Score (KSS). KBD Quality of Life (KBDQOL) was used to evaluate the quality of life in KBD patients before and after TKA.RESULTS: There were no major complications after TKA. The levels of VAS score were obviously deceased in post-operation than that in pre-operation. The levels of KSS score were increased in one year after TKA compared with the pre-operative values, and it maintained a higher level on three years after TKA. The average KBDQOL score level of each domain in pre-operation and one and three years after TKA was increased accordingly. The average scores of physical function, activity limitation, support of society, mental health, and general health in one year after TKA were significantly higher than those in pre-operation.CONCLUSIONS: TKA can reduce knee pain, improve knee function, and improve the quality life in KBD patients. KBDQOL questionnaire may be a promising instrument for assessing the quality life in KBD patients.

    • In this study, two iPP resins with similar molecular weight and average isotacticity but different uniformities of stereo-defect distribution were used in the production of cast films. The crystalline morphology and orientation, and tensile behavior during room temperature stretching of the cast films were investigated by scanning electronic microscopy (SEM), 2D-wide angle X-ray diffraction (2D-WAXD) and 2D-small-angle X-ray scattering (2D-SAXS). It was found that under fixed processing condition, iPP with more uniform stereo-defect distribution (PP2) favors the formation of row nucleated lamellar structure during cast film production, and therefore exhibiting hard-elastic deformation behavior during stretching; meanwhile, the sample with less uniform stereo-defect distribution (PP1) shows un-oriented spherulitical morphology in the cast film, and typical plastic deformation behavior during stretching. The crystallization and rheological analysis on the iPP raw materials revealed that, the uniformity of stereo-defect distribution determines the ability of nucleation and crystallization, and further influences the response of the crystalline morphology to the elongation effect of cast film production process, which is of great importance in the structure-property design in the production of iPP membranes.

    • The aerodynamic loads on the overhead bridge bottom surface induced by train passage are reported in this paper. Both moving model test and numerical simulation approaches at the 1:20 scale are used. The numerical work is validated through both mesh independence tests and comparison with experimental data. Typical pressure variation curves are plotted and compared with previous studies. The peak pressure values’ dependence on the Reynolds number is considered through four sets of experiments with different train running speeds. The peak pressure coefficient distribution law for the bridge bottom surface is presented. Differences in the pressure distribution in different bridge bottom areas are explained based on more detailed flow field information. The influence of the bridge height on the aerodynamic load magnitude and time interval is presented. Moreover, the application of the CEN Standard to practical engineering issues is discussed.

    • Recently, communications with the aid of reconfigurable intelligent surface (RIS), which operates with the aim of enhancing the system communication performance, have aroused extensive researches. Furthermore, the use of RIS for positioning has been considered. Therefore, we focus on a practical structure of indoor positioning assisted by distributed RISs through utilizing their ability to manipulate multipath signals, through the developed quasi-static and dynamic modes. Specifically, in the quasi-static mode, for reducing the implementation cost, the reflection coefficients for each RIS are preset and remain constant. In the dynamic mode, the reflection coefficients can be timely updated with a two-step positioning approach toward more accurate positioning performance. Furthermore, the Cramer-Rao lower bound of the developed positioning scheme is quantified through theoretical analysis. Both theoretical analysis and simulation results demonstrate that RIS has the potential to realize accurate positioning even with a single access point, due to its ability to mark the channel and replace traditional active positioning anchors. Meanwhile, we also show that the developed two-step positioning scheme can achieve considerable performance gain in accurate positioning.

    • As a revolutionary technology for future wireless communications, reconfigurable intelligent surface (RIS), characterized by an efficient way of manipulating wireless signals, has been widely investigated in recent years toward enhancing signal quality, energy efficiency, throughput, and so on. However, in RIS-assisted Internet of Things (IoT), a new issue as multipath separation emerges, especially, when deploying multiple RISs to assist communication, since the devices may have limited signal processing capabilities. For alleviating this problem, we conceive a novel RIS-enabled code-division multiple access (CDMA) structure, where each RIS holds a specified time-varying coefficient to tag the channel. Moreover, multipath extraction is further considered, including a practical channel estimation approach along with theoretical derivations in terms of Cramér-Rao lower bound, mean-square error, as well as ergodic channel capacity. Simulation results corroborate the feasibility of the conceived RIS-CDMA structure and the effectiveness of the proposed multipath extraction approach.

    • Wireless powered hybrid backscatter-active communications are recognized as the promising technique to provide the sustainable communication service for wireless devices. In this paper, we consider an intelligent reflecting surface-aided wireless powered hybrid backscatter-active communication network, where multiple sensors devices powered by a power beacon, transmit their information to an access point through backscatter and active communications. In particular, an intelligent reflecting surface is properly deployed to improve the communication quality between sensor devices and access point by utilizing the composite channel gain and phase beamforming gain. Furthermore, we formulate a system energy efficiency maximization problem under the minimum throughput requirements of sensor devices, to jointly optimize the phase shifts of intelligent reflecting surface, time allocation for backscatter communications and active transmissions, and transmit power of sensor devices and power beacon. To solve the formulated non-convex problem, we first unveil the optimal phase shifts of intelligent reflecting surface, and then utilize the variable substitutions and Dinkelbach-based method to develop an iterative method to solve the energy efficiency maximization problem. Simulation results reveal that the proposed method outperforms the existing several benchmark methods in terms of system energy efficiency.

    • Ambient energy harvesting has great potential to contribute to sustainable development and address growing environmental challenges. Converting waste energy from energy-intensive processes and systems (e.g. combustion engines and furnaces) is crucial to reducing their environmental impact and achieving net-zero emissions. Compact energy harvesters will also be key to powering the exponentially growing smart devices ecosystem that is part of the Internet of Things, thus enabling futuristic applications that can improve our quality of life (e.g. smart homes, smart cities, smart manufacturing, and smart healthcare). To achieve these goals, innovative materials are needed to efficiently convert ambient energy into electricity through various physical mechanisms, such as the photovoltaic effect, thermoelectricity, piezoelectricity, triboelectricity, and radiofrequency wireless power transfer. By bringing together the perspectives of experts in various types of energy harvesting materials, this Roadmap provides extensive insights into recent advances and present challenges in the field. Additionally, the Roadmap analyses the key performance metrics of these technologies in relation to their ultimate energy conversion limits. Building on these insights, the Roadmap outlines promising directions for future research to fully harness the potential of energy harvesting materials for green energy anytime, anywhere.

    • OBJECTIVE: To investigate the differences between the gene expression profiles in peripheral blood mononuclear cells (PBMC) from normal controls and patients with Kashin-Beck disease (KBD).METHODS: Twenty KBD patients and 12 normal subjects were selected from a KBD-endemic area and divided into four pairs of KBD vs. control (KBD, n = 5 per pair; control, n = 3 per pair). RNAs were respectively isolated from KBD PBMCs and normal PBMCs. Gene expression profiles were analyzed by oligonucleotide microarray. The gene expression profiles in PBMCs from KBD patients and normal controls were compared and the differentially expressed genes were identified. The obtained microarray data was further confirmed by using quantitative real-time reverse transcription polymerase chain reaction (qRT-PCR).RESULTS: Approximately 501 genes, corresponding to 2.4% of the total probe transcripts, showed a 2-fold change in differential expression. 19.4% (97 out of 501)of the differentially expressed genes were commonly detected in all the four pairs. Among the 97 differentially expressed genes, 83 genes were up-regulated and 14 genes were down-regulated, compared with those in the normal controls. Some differentially expressed genes were found to be related to functions such as immunity, metabolism, apoptosis, cystoskeleton and cell movement, and extracellular matrix. The validity of our microarray data were supported by the results of qRT-PCR assay.CONCLUSION: Differences in the PBMC gene expression profile between the KBD patients and the normal controls exhibited a similar pattern among all the four pairs of microarrays examined, indicating that the suppressed immunity may play an important role in the pathogenesis of KBD.

    • Kashin-Beck disease (KBD) is an endemic degenerative osteoarticular disorder associated with physical disability and a heavy economic burden. Contamination by mycotoxin deoxynivalenol (DON) and selenium deficiency have been proposed to be key etiological factors for KBD, and can work together to aggravate the progression of KBD. Nevertheless, the mechanism of DON in KBD remains elusive. In the present study, exposure to DON dose-dependently suppressed cell viability and expression of pro-proliferation marker PCNA in human chondrocytes, whereas it enhanced lactate dehydrogenase release, cell apoptosis, and caspase-3/9 activity. In addition, DON incubation shifted metabolism homeostasis towards catabolism by suppressing the transcription of collagen II and aggrecan, and the production of sulphated glycosaminoglycans and TIMP-1, while increasing matrix metalloproteinase levels (MMP-1 and MMP-13). Mechanistically, DON exposure induced the activation of Wnt/β-catenin signaling. Intriguingly, blocking this pathway reversed the adverse effects of DON on cytotoxic damage and metabolism disruption to catabolism. Notably, supplementation with selenium reduced DON-induced activation of the Wnt/β-catenin pathway. Moreover, selenium addition abrogated cytotoxic injury and excessive pro-catabolic gene expression in chondrocytes upon DON conditions. These findings confirm that DON may facilitate the development of KBD by inducing cell injury, inhibiting matrix synthesis, and increasing cellular catabolism by activating the Wnt/β-catenin signaling, which were partially reversed by selenium supplementation. Thus, the current study may presents a new viewpoint for how selenium supplementation ameliorates the development of KBD by inhibiting DON-induced cytotoxic injury and metabolism imbalance in chondrocytes.

    • Perovskite solar cells are a highly competitive candidate for next-generation photovoltaic technology. Defects in the perovskite grain boundaries and on the film surfaces however have significant impacts on both the device efficiency and environmental stability. Herein, a strategy using organic dyes as additives to passivate the defect states and produce more n-type perovskite films, thereby improving charge transport and decreasing charge recombination, is reported. Based on this strategy, the power conversion efficiency of the perovskite solar cell is significantly increased from 18.13% to 20.18% with a negligible hysteresis. Furthermore, the rich hydrogen bonds and carbonyl structures in the organic dye can significantly enhance device stability both in terms of humidity and thermal stress. The results present a promising pathway using abundant and colorful organic dyes as additives to achieve high-performance perovskite solar cells.
