    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • Comparative genomics reveals insights into avian genome evolution and adaptation

    • Birds are the most species-rich class of tetrapod vertebrates and have wide relevance across many research fields. We explored bird macroevolution using full genomes from 48 avian species representing all major extant clades. The avian genome is principally characterized by its constrained size, which predominantly arose because of lineage-specific erosion of repetitive elements, large segmental deletions, and gene loss. Avian genomes furthermore show a remarkably high degree of evolutionary stasis at the levels of nucleotide sequence, gene synteny, and chromosomal structure. Despite this pattern of conservation, we detected many non-neutral evolutionary changes in protein-coding genes and noncoding regions. These analyses reveal that pan-avian genomic diversity covaries with adaptations to different lifestyles and convergent evolution of traits.

    • Lithium-ion batteries (LiB) are widely used in electric vehicles (EVs) and battery energy storage systems, and accurate state estimation relying on the relationship between battery Open-Circuit-Voltage (OCV) and State-ofCharge (SOC) is the basis for their safe and efficient applications. To avoid the time-consuming lab test needed for obtaining OCV-SOC curves, this study proposes a data-driven universal method by using operation data collected onboard about the variation of OCV with ampere-hour (Ah). To guarantee high reliability, a series of constraints have been implemented. To verify the effectiveness of this method, the constructed OCV-SOC curves are used to estimate battery SOC and State-of-Health (SOH), which are compared with data from both lab tests and EV manufacturers. Results show that a higher accuracy can be achieved in the estimation of both SOC and SOH, for which the maximum deviations are less than 3.0% and 2.9% respectively.

  • Compact spin-valley-locked perovskite emission

    • Circularly polarized light sources with free-space directional emission play a key role in chiroptics(1), spintronics(2), valleytronics(3) and asymmetric photocatalysis(4). However, conventional approaches fail to simultaneously realize pure circular polarization, high directionality and large emission angles in a compact emitter. Metal-halide perovskite semiconductors are promising light emitters(5-8), but the absence of an intrinsic spin-locking mechanism results in poor emission chirality. Further, device integration has undermined the efficiency and directionality of perovskite chiral emitters. Here we realize compact spin-valley-locked perovskite emitting metasurfaces where spin-dependent geometric phases are imparted into bound states in the continuum via Brillouin zone folding, and thus, photons with different spins are selectively addressed to opposite valleys. Employing this approach, chiral purity of 0.91 and emission angle of 41.0 degrees are simultaneously achieved, with a beam divergence angle of 1.6 degrees. With this approach, we envisage the realization of chiral light- emitting diodes, as well as the on-chip generation of entangled photon pairs.

  • Chromium IV exposure, via Src/Ras signaling, promotes cell transformation

    • Hexavalent chromium [Cr(VI)] is a well-known environment carcinogen. The exposure of Cr(VI) through contaminated soil, air particles, and drinking water is a strong concern for the public health worldwide. While many studies have been done, it remains unclear which intracellular molecules transduce Cr(VI)-mediated carcinogenic signaling in cells to promote cancer. In this study, we demonstrated that upon Cr(VI) treatment, the intracellular receptor src was activated, which further upregulated Ras activity, leading to the augmentation of ROS and onset of ER stress in human lung epithelial BEAS-2B or keratinocytes. These cells were formed colonies in soft agar cultures following the persistent Cr(VI) treatment. Furthermore, anti-apoptotic factor Bcl-2 was upregulated and activated in the colonies. Thus, our study suggests that Cr(VI), though activating the src and Ras signaling axis, perturbs redox state and invokes ER stress for the establishment of carcinogenic actions in the cells. In this process, Bcl-2 appears playing an important role. By uncovering these intracellular targets, our study may help developing novel strategies for better environmental protection, especially in areas contaminated or polluted by Cr(VI) as well as for effective cancer treatments. © 2017 Wiley Periodicals, Inc.

  • Suppression of PKC causes oncogenic stress for triggering apoptosis in cancer cells

    • Gain of functional mutations in ras occurs in more than 30% of human malignancies and in particular 90% of pancreatic cancer. Mutant ras, via activating multiple effector pathways, not only promote cell growth or survival, but also apoptosis, depending upon cell types or circumstances. In order to further study the mechanisms of apoptosis induced by oncogenic ras, we employed the ras loop mutant genes and demonstrated that Akt functioned downstream of Ras in human pancreatic cancer or HPNE cells ectopically expressing mutated K-ras for the induction of apoptosis after the concurrent suppression of PKC α and β. In this apoptotic process, the redox machinery was aberrantly switched on in the pancreatic cancer cells as well as prostate cancer DU145 cells. p73 was phosphorylated and translocated to the nucleus, accompanied with UPR activation and induction of apoptosis. The in vitro results were corroborated by the in vivo data. Thus, our study indicated that PKC a and β appeared coping with oncogenic Ras or mutated Akt to maintain the balance of the homeostasis in cancer cells. Once these PKC isoforms were suppressed, the redox state in the cancer cells was disrupted, which elicited persistent oncogenic stress and subsequent apoptotic crisis. © Ganapathy et al.

  • The development and validation of sensory and emotional scales of touch perception.

    • No comprehensive language exists that describes the experience of touch. Three experiments were conducted to take steps toward establishing a touch lexicon. In Experiment I, 49 participants rated how well 262 adjectives described sensory, emotional and evaluative aspects of touch. In Experiment II, participants rated pairwise dissimilarities of the most descriptive words of the set. Multidimensional scaling (MDS) solutions representing semantic-perceptual spaces underlying the words resulted in a touch perception task (TPT) consisting of 26 'sensory' attributes (e.g., bumpiness) and 14 'emotional' attributes (e.g., pleasurable). In Experiment III, 40 participants used the TPT to rate unseen textured materials that were moved actively or received passively against the index fingerpad, volar forearm, and two underarm sites. MDS confirmed similar semantic-perceptual structures in Experiments II and III. Factor analysis of Experiment III data decomposed the sensory attribute ratings into factors labeled Roughness, Slip, Pile and Firmness, and the emotional attribute ratings into Comfort and Arousal factors. Factor scores varied among materials and sites. Greater intensity of sensory and emotional responses were reported when participants passively, as opposed to actively, received stimuli. The sensitivity of the TPT in identifying body site and mode of touch-related perceptual differences affirms the validity and utility of this novel linguistic/perceptual tool.

  • Changing grains for the prevention and treatment of Kashin-Beck disease in children : a Meta-analysis

    • To evaluate the efficacy of changing grains on the prevention and treatment of Kashin-Beck Disease (KBD) in children, community-based trials were acquired from seven electronic databases (up to July 2014). As a result, the methodological quality of the six trials that have been included into our analysis was low. The pooled ORs favoring the prevention and treatment effects of changing grains were 0.15 (95% CI: 0.03-0.70) and 2.13 (95% CI: 1.44-3.16) respectively by meta-analysis. Subgroup analysis demonstrated the pooled OR favoring treatment effect of exchanging grains rather than drying grains both compared with endemic grains. The results showed that changing grains had obvious effects on the prevention and treatment of KBD in children. However, the evidences were limited by the potential biases and confounders. Large and well-designed trials are still needed.

    • This paper is concerned with model-based dynamic peak-power evaluation for LiNMC and LiFePO4 batteries under different operating conditions. The battery test and our prior study on linear-parameter-varying (LPV) battery modeling are briefly introduced. The peak-power estimation method that incorporates an explicit prediction horizon and design constraints on the battery current, voltage, and SOC are elaborated and its computational load is analyzed. The discharge and charge peak powers are quantitatively assessed under different dynamic characterization tests, in which a comparison with the conventional PNGV-HPPC method and approaches using the less accurate models is conducted. The robustness of the peak-power estimation approach against varying battery temperatures and aging levels is investigated. The methods to improve the credibility of the peak-power assessment in the context of battery degradation are explored.

    • Kashin-Beck disease (KBD) is a chronic endemic osteochondropathy with unclear pathogenesis. It is a degenerative disease similar to osteoarthritis, but with different manifestations of cartilage damage. The aim of this investigation was to show the protein changes in KBD cartilage and to identify the candidate proteins in order to understand the pathogenesis of the disease. Proteins were extracted from the media of primary cell cultures of KBD and normal chondrocytes, and separated by two-dimensional fluorescence difference gel electrophoresis (2-D DIGE). MALDI-TOF/TOF analysis revealed statistically significant differences in 27 proteins from KBD chondrocyte cultures, which consisted of 17 up-regulated and ten down-regulated proteins. The results were further validated by Western blot analysis. The proteins identified are mainly involved in cellular redox homeostasis and stress response (MnSOD, Hsp27, Peroxiredoxin-1, and Cofilin-1), glycolysis (PGK-1, PGM-1, α-enolase), and cell motility and cytoskeletal organization (Actin, Calponin-2, and Keratin). These KBD-associated proteins indicate that cytoskeletal remodeling, glycometabolism, and oxidative stress are abnormal in KBD articular cartilage.

  • Deep neural network-driven in-situ detection and quantification of lithium plating on anodes in commercial lithium-ion batteries

    • Lithium plating seriously threatens the life of lithium-ion batteries at low temperatures charging conditions, but the onboard detection and quantification of lithium plating are severely hampered by the limited available signals and volatile operating conditions in real scenarios. Herein, we propose a detection method to predict the occurrence and quantification of lithium plating under uncertain conditions by only using constant-current curves during charging based on deep learning. A deep neural network (DNN) is developed to extract data-driven features induced by lithium plating from the charge curves, avoiding the challenge of manual feature selection. Only using the most common voltage and current signals as inputs, the network exhibits superior adaptability and accuracy. The detection accuracy of the proposed method is 98.64%, while the quantity of the lithium plating can be accurately predicted with a root-mean-square error <4.1712 mg. Moreover, the generalization ability of the proposed method is verified by its reliable detection accuracy under conditions that are not used in the training dataset. The detection accuracy is 92.39% for brand new charging conditions and 95.53% for brand new aging states. This method shortens the detection time that currently takes more than several hours (the widely used differential curve analysis) to milliseconds and eliminates the need for a rigorous testing environment, showing great potential for onboard application in future battery management systems.

  • Prediction of co-expression genes and integrative analysis of gene microarray and proteomics profile of Keshan disease

    • Keshan disease (KD) is a kind of endemic cardiomyopathy which has a high mortality. However, molecular mechanism in the pathogenesis of KD remains poorly understood. Serum samples were collected from 112 KD patients and 112 normal controls. Gene microarray was used to screen differently expressed genes. Genevestigator was applied to forecast co-expression genes of significant gene. iTRAQ proteomics analysis was used to verify significant genes and their co-expression genes. GO, COG, IPA and STRING were applied to undertake function categorization, pathway and network analysis separately. We identified 32 differentially expressed genes; IDH2, FEM1A, SSPB1 and their respective 30 co-expression genes; 68 differential proteins in KD. Significant proteins were categorized into 23 biological processes, 16 molecular functions, 16 cellular components, 15 function classes, 13 KD pathways and 1 network. IDH2, FEM1A, SSBP1, CALR, NDUFS2, IDH3A, GAPDH, TCA Cycle II (Eukaryotic) pathway and NADP repair pathway may play important roles in the pathogenesis of KD.

    • This study aimed to quantify the relationships between the American Heart Association (AHA) Cardiovascular Health (CVH) metrics, namely AHA Life's Simple 7, and cognitive outcomes. We searched PubMed and Embase (January 1, 2010-August 24, 2022) and finally included 14 longitudinal studies (311654 participants with 8006 incident dementia cases). Random-effects meta-analysis and one-stage linear mixed-effects models were performed. Increased CVH score seemed to associate with decreased risk of incident dementia in a linear manner, but this relationship varied by the measurement age of CVH metrics. That is, midlife CVH tended to have a linear association with late-life dementia risk, whereas a J-shaped association was observed between the late-life CVH score and dementia. In addition, late-life dementia risk was reduced significantly if individuals maintained an ideal level of AHA's CVH guidelines of physical activity, fasting plasma glucose, total cholesterol, and smoking. However, our meta-analysis did not show a significant association between CVH score and global cognitive decline rate. Following AHA's CVH guidelines and maintaining CVH at an optimal level would substantially reduce the late-life dementia risk. More research is required to explore the link between a favorable CVH score and cognitive trajectories among cognitively asymptomatic older populations.

  • Solar Flux Influence on the In-Situ Plasma Density at Topside Ionosphere Measured by Swarm Satellites

    • In this study, we perform the first comprehensive comparison of ion density (Ni) in the topside ionosphere measured by the Langmuir probe (LP) and faceplate (FP) of the thermal ion imager on board Swarm satellites. Our results show a systematic difference between the LP and FP derived Ni values, and the systematic difference shows prominent dependences on solar flux, local time, and season. Although both Ni datasets show generally good linear regression with electron density (Ne) measurements from the incoherent scatter radar (ISR) located at Jicamarca, the Ni derived from LP shows an additional dependence on the solar flux, while such a dependence cannot be seen in the FP-derived Ni. We suggest that the solar flux dependence of LP-derived Ni is related to the ion compositions change at Swarm altitude, which has not been properly accounted for in the LP processing algorithm. More light ions (e.g., H+), diffusing down from the plasmasphere to the Swarm altitude, seem to cause the overestimation of Ni from LP during low solar activity. A linear relation between the Swarm LP-derived Ni and ISR Ne is derived, and such a function is recommended to be implemented into further updates of the Swarm LP plasma density data.

    • BackgroundThe MYC oncoprotein, also known as the master regulator of genes, is a transcription factor that regulates numerous physiological processes, including cell cycle control, apoptosis, protein synthesis and cell adhesion, among others. MYC is overexpressed in approximately 70% of human cancers. Given its pervasive role in cancer biology, MYC down-regulation has become an attractive cancer treatment strategy. MethodsThe CRISPR/Cas9 method was used to produce KO cell models. Western blot was used to analyzed the expressions of MYC and TATA-binding proteinassociated factors 10 (TAF10) in cancer cells (MCF7, A549, HepG2 cells) Cell culture studies were performed to determine the mechanisms by which small molecules (Z363119456, Z363) affects MYC and TAF10 expressions and functions. Mouse studies were carried out to investigate the impact of Z363 regulation on tumor growth. ResultsZ363 activate Thyroid hormone Receptor-interacting Protein 12 (TRIP12), which phosphorylates MYC at Thr58, resulting in MYC ubiquitination and degradation and thereby regulating MYC target genes. Importantly, TRIP12 also induces TAF10 degradation, which reduces MYC protein levels. TRIP12, an E3 ligase, controls MYC levels both directly and indirectly by inhibiting MYC or TAF10 activity. ConclusionsIn summary,these results demonstrate the anti-cancer properties of Z363, a small molecule that is co-regulated by TAF10 and MYC.

  • The Role of Solution Aggregation Property toward High-Efficiency Non-Fullerene Organic Photovoltaic Cells

    • In organic photovoltaic cells, the solution-aggregation effect (SAE) is long considered a critical factor in achieving high power-conversion efficiencies for polymer donor (PD)/non-fullerene acceptor (NFA) blend systems. However, the underlying mechanism has yet to be fully understood. Herein, based on an extensive study of blends consisting of the representative 2D-benzodithiophene-based PDs and acceptor-donor-acceptor-type NFAs, it is demonstrated that SAE shows a strong correlation with the aggregation kinetics during solidification, and the aggregation competition between PD and NFA determines the phase separation of blend film and thus the photovoltaic performance. PDs with strong SAEs enable earlier aggregation evolutions than NFAs, resulting in well-known polymer-templated fibrillar network structures and superior PCEs. With the weakening of PDs' aggregation effects, NFAs, showing stronger tendencies to aggregate, tend to form oversized domains, leading to significantly reduced external quantum efficiencies and fill factors. These trends reveal the importance of matching SAE between PD and NFA. The aggregation abilities of various materials are further evaluated and the aggregation ability/photovoltaic parameter diagrams of 64 PD/NFA combinations are provided. This work proposes a guiding criteria and facile approach to match efficient PD/NFA systems. A systematic study of the representative organic photovoltaic systems shows that the aggregation competition between polymer donor (PD) and non-fullerene acceptor (NFA) is a decisive factor in the phase separation of blend film and thus the photovoltaic performance. Based on 64 PD/NFA combinations, the aggregation ability/photovoltaic parameter heatmaps are plotted, providing a new matching rule for developing high-efficiency PD/NFA systems. image

  • Aging characteristics-based health diagnosis and remaining useful life prognostics for lithium-ion batteries

    • This paper developed methods for improving the practicability of battery health diagnosis and remaining useful life prognostics. Battery state of health was estimated using a feature extraction-based method based on the charging voltage curve. Battery remaining useful life was predicted by identifying recognizable aging stages. Acceleration aging test data for 9 cells at constant current rates including 0.5C, 1C, 1.5C, and 2C, and dynamic current rates were used to validate the developed methods. The capacity estimates were accurate with estimation errors less than 1% at most cycles. The remaining useful life was predicted within 0.3 s at dynamic current rates, with the prediction errors at most cycles less than 10 after 300 cycles and the 95% confidence intervals covering about 20 cycles for each prediction.
