    • Humans rely increasingly on sensors to address grand challenges and to improve quality of life in the era of digitalization and big data. For ubiquitous sensing, flexible sensors are developed to overcome the limitations of conventional rigid counterparts. Despite rapid advancement in bench-side research over the last decade, the market adoption of flexible sensors remains limited. To ease and to expedite their deployment, here, we identify bottlenecks hindering the maturation of flexible sensors and propose promising solutions. We first analyze challenges in achieving satisfactory sensing performance for real-world applications and then summarize issues in compatible sensor-biology interfaces, followed by brief discussions on powering and connecting sensor networks. Issues en route to commercialization and for sustainable growth of the sector are also analyzed, highlighting environmental concerns and emphasizing nontechnical issues such as business, regulatory, and ethical considerations. Additionally, we look at future intelligent flexible sensors. In proposing a comprehensive roadmap, we hope to steer research efforts towards common goals and to guide coordinated development strategies from disparate communities. Through such collaborative efforts, scientific breakthroughs can be made sooner and capitalized for the betterment of humanity.

  • Female chromosome X mosaicism is age-related and preferentially affects the inactivated X chromosome

    • To investigate large structural clonal mosaicism of chromosome X, we analysed the SNP microarray intensity data of 38,303 women from cancer genome-wide association studies (20,878 cases and 17,425 controls) and detected 124 mosaic X events >2 Mb in 97 (0.25%) women. Here we show rates for X-chromosome mosaicism are four times higher than mean autosomal rates; X mosaic events more often include the entire chromosome and participants with X events more likely harbour autosomal mosaic events. X mosaicism frequency increases with age (0.11% in 50-year olds; 0.45% in 75-year olds), as reported for Y and autosomes. Methylation array analyses of 33 women with X mosaicism indicate events preferentially involve the inactive X chromosome. Our results provide further evidence that the sex chromosomes undergo mosaic events more frequently than autosomes, which could have implications for understanding the underlying mechanisms of mosaic events and their possible contribution to risk for chronic diseases.

    • Elemene (ELE), a natural plant drug extracted from Curcumae Rhizoma, has been widely used for cancer treatment in China for more than 20 years. Although it is reported to be a broad-spectrum anticancer drug, the mechanism underlying the action of ELE in the treatment of breast cancer remains to be fully elucidated. Heparanase, a mammalian endo-D-glucuronidase, is involved in degradation of the extracellular matrix (ECM), and thus promotes tumor progression and metastasis. The downregulation of heparanase can effectively reduce tumor malignant behaviors. In the present study, the inhibitory effects of ELE were evaluated in breast cancer cells using a Cell Counting kit 8 assay. The migratory and invasive capabilities of cancer cells were investigated using a wound healing assay, real-time cell analysis and a Transwell assay. In addition, western blot analysis was used to assess alterations in the expression levels of key proteins. The present results confirmed the antiproliferative and antimetastatic effects of ELE, using low-molecular weight heparin (LMWH) as a positive control. In addition, ELE was demonstrated to downregulate the expression of heparanase, and decrease the phosphorylation of extracellular signal-regulated kinase and AKT. These findings suggested that ELE may be a promising agent targeting heparanase in the treatment of breast cancer.

  • Substantial increase in future fluvial flood risk projected in China’s major urban agglomerations

    • Urban land will face high fluvial flood risk against the background of climate change and urban expansion. The effect of urban spatial expansion, instead of densification of assets within existing urban cells, on flood risk has rarely been reported. Here, we project the future flood risk of seven urban agglomerations in China, home to over 750 million people. The inundated urban land areas in the future are projected to be 4 to 19 times that at present. Without considering the urban spatial expansion, the inundated urban land areas will be underestimated by 10-50%. Urban land is more likely to be inundated than non-urban land, and the newly-developed urban land will be inundated more easily than the historical urban land. The results demonstrate the urgency of integrating climate change mitigation, reasonable urban land expansion, and increased flood protection levels to minimize the flood risk in urban land.

    • All existing work on real-time scheduling of parallel task graph models with shared resources assumes that a critical section must be contained inside a single vertex. However, this assumption does not hold in many realistic parallel real-time software. In this work, we conduct the first study on real-time scheduling and analysis of parallel task graphs where critical sections are allowed to cross different vertices. We show that allowing this may potentially lead to deadlocks and the so-called resource unrelated blocking time problem. We formalize the conditions for the deadlocks and resource unrelated blocking time to happen, and propose two different solutions to address them and develop corresponding schedulability analysis techniques. We conduct comprehensive experiments to evaluate our method. The results indicate that there is a significant impact to the system schedulability when tasks incur deadlock and resource unrelated blocking. Moreover, the schedulability can benefit from the execution of workload in parallel with critical sections if tasks can be carefully designed so that all deadlocks and resource unrelated blocking time can be avoided, and our methods are efficient to determine the schedulability of systems where critical sections across different vertices exist.

    • The integrated sensing and communication (ISAC) has been envisioned as a promising technology to simultaneously perform high-capacity communication and high-accuracy sensing with efficient resource utilization. Nevertheless, due to the severe mutual constraints between communication and sensing functions, the performance of previously reported ISAC demonstrations notably trails that of individual communication or sensing systems. In this work, an integrated LFM-QAM waveform is proposed and theoretically analyzed by combining linear frequency modulation (LFM) and quadrature amplitude modulation (QAM) formats. Employing the proposed waveform and optical wavelength division multiplexing (WDM) technique, we experimentally implement a multi-channel photonic terahertz (THz)-ISAC wireless system operating at 275 GHz band, simultaneously achieving a communication data rate up to 120 Gbps and a range resolution as high as 2.5 mm. To the best of our knowledge, this is the first time to demonstrate beyond 100 Gbps wireless data rate and mm-scale resolution based on an integrated waveform in the THz region, revealing the potential of THz-ISAC performance.

Up to date, previously reported demonstrations regarding radar sensing performance lack sufficient support in a complex environment with a strong target masking effect. This work tacks this problem by proposing a radar-centric waveform combining linear frequency modulation (LFM) waveform and phase shift keying (PSK). We first derive sensing metrics of the LFM-PSK waveform through theoretical analysis, including range resolution, peak sidelobe ratio (PSLR), and Cramér-Rao lower bound (CRLB). Then a proof-of-concept experiment on a photonics-assisted integrated sensing and communication (ISAC) system operating at 330 with 18 GHz bandwidth is conducted to verify the performance of the proposed LFM-PSK waveform. In the experiment, the proposed waveform can reach a PSLR of up to 20.9 dB and a range resolution of 1.3 cm, simultaneously accommodating a data transmission of 6 Gbit/s. In addition, the effect of embedding symbols on sensing metrics is also discussed, and by comparing the range solution and PSLR with various data rates, around $\sim$ 6 dB gain in the PSLR without any deterioration of range resolution is observed.

  • Characterization of Large Structural Genetic Mosaicism in Human Autosomes

    • Analyses of genome-wide association study (GWAS) data have revealed that detectable genetic mosaicism involving large (>2 Mb) structural autosomal alterations occurs in a fraction of individuals. We present results for a set of 24,849 genotyped individuals (total GWAS set II [TGSII]) in whom 341 large autosomal abnormalities were observed in 168 (0.68%) individuals. Merging data from the new TGSII set with data from two prior reports (the Gene-Environment Association Studies and the total GWAS set I) generated a large dataset of 127,179 individuals; we then conducted a meta-analysis to investigate the patterns of detectable autosomal mosaicism (n = 1,315 events in 925 [0.73%] individuals). Restricting to events >2 Mb in size, we observed an increase in event frequency as event size decreased. The combined results underscore that the rate of detectable mosaicism increases with age (p value = 5.5 x 3 10(-31)) and is higher in men (p value = 0.002) but lower in participants of African ancestry (p value = 0.003). In a subset of 47 individuals from whom serial samples were collected up to 6 years apart, complex changes were noted over time and showed an overall increase in the proportion of mosaic cells as age increased. Our large combined sample allowed for a unique ability to characterize detectable genetic mosaicism involving large structural events and strengthens the emerging evidence of non-random erosion of the genome in the aging population.

  • Analysis of Heritability and Shared Heritability Based on Genome-Wide Association Studies for 13 Cancer Types

    • Background: Studies of related individuals have consistently demonstrated notable familial aggregation of cancer. We aim to estimate the heritability and genetic correlation attributable to the additive effects of common single-nucleotide polymorphisms (SNPs) for cancer at 13 anatomical sites. Methods: Between 2007 and 2014, the US National Cancer Institute has generated data from genome-wide association studies (GWAS) for 49 492 cancer case patients and 34 131 control patients. We apply novel mixed model methodology (GCTA) to this GWAS data to estimate the heritability of individual cancers, as well as the proportion of heritability attributable to cigarette smoking in smoking-related cancers, and the genetic correlation between pairs of cancers. Results: GWAS heritability was statistically significant at nearly all sites, with the estimates of array-based heritability, h(l)(2), on the liability threshold (LT) scale ranging from 0.05 to 0.38. Estimating the combined heritability of multiple smoking characteristics, we calculate that at least 24% (95% confidence interval [CI] = 14% to 37%) and 7% (95% CI = 4% to 11%) of the heritability for lung and bladder cancer, respectively, can be attributed to genetic determinants of smoking. Most pairs of cancers studied did not show evidence of strong genetic correlation. We found only four pairs of cancers with marginally statistically significant correlations, specifically kidney and testes (rho = 0.73, SE = 0.28), diffuse large B-cell lymphoma (DLBCL) and pediatric osteosarcoma (rho = 0.53, SE = 0.21), DLBCL and chronic lymphocytic leukemia (CLL) (rho = 0.51, SE = 0.18), and bladder and lung (rho = 0.35, SE = 0.14). Correlation analysis also indicates that the genetic architecture of lung cancer differs between a smoking population of European ancestry and a nonsmoking Asian population, allowing for the possibility that the genetic etiology for the same disease can vary by population and environmental exposures. Conclusion: Our results provide important insights into the genetic architecture of cancers and suggest new avenues for investigation.

    • Genome-wide association studies (GWAS) have mapped risk alleles for at least 10 distinct cancers to a small region of 63 000 bp on chromosome 5p15.33. This region harbors the TERT and CLPTM1L genes; the former encodes the catalytic subunit of telomerase reverse transcriptase and the latter may play a role in apoptosis. To investigate further the genetic architecture of common susceptibility alleles in this region, we conducted an agnostic subset-based meta-analysis (association analysis based on subsets) across six distinct cancers in 34 248 cases and 45 036 controls. Based on sequential conditional analysis, we identified as many as six independent risk loci marked by common single-nucleotide polymorphisms: five in the TERT gene (Region 1: rs7726159, P = 2.10 × 10(-39); Region 3: rs2853677, P = 3.30 × 10(-36) and PConditional = 2.36 × 10(-8); Region 4: rs2736098, P = 3.87 × 10(-12) and PConditional = 5.19 × 10(-6), Region 5: rs13172201, P = 0.041 and PConditional = 2.04 × 10(-6); and Region 6: rs10069690, P = 7.49 × 10(-15) and PConditional = 5.35 × 10(-7)) and one in the neighboring CLPTM1L gene (Region 2: rs451360; P = 1.90 × 10(-18) and PConditional = 7.06 × 10(-16)). Between three and five cancers mapped to each independent locus with both risk-enhancing and protective effects. Allele-specific effects on DNA methylation were seen for a subset of risk loci, indicating that methylation and subsequent effects on gene expression may contribute to the biology of risk variants on 5p15.33. Our results provide strong support for extensive pleiotropy across this region of 5p15.33, to an extent not previously observed in other cancer susceptibility loci.

  • Coherent control of single spins in silicon carbide at room temperature

    • Spins in solids are cornerstone elements of quantum spintronics(1). Leading contenders such as defects in diamond(2-5) or individual phosphorus dopants in silicon(6) have shown spectacular progress, but either lack established nanotechnology or an efficient spin/photon interface. Silicon carbide (SiC) combines the strength of both systems(5):it has a large bandgap with deep defects(7-9) and benefits from mature fabrication techniques(10-12). Here, we report the characterization of photoluminescence and optical spin polarization from single silicon vacancies in SiC, and demonstrate that single spins can be addressed at room temperature. We show coherent control of a single defect spin and find long spin coherence times under ambient conditions. Our study provides evidence that SiC is a promising system for atomic-scale spintronics and quantum technology.

    • High blood glucose has long been established as a risk factor for tumor metastasis, yet the molecular mechanisms underlying this association have not been elucidated. Here we describe that hyperglycemia promotes tumor metastasis via increased platelet activity. Administration of glucose, but not fructose, reprogrammed the metabolism of megakaryocytes to indirectly prime platelets into a prometastatic phenotype with increased adherence to tumor cells. In megakaryocytes, a glucose metabolism-related gene array identified the mitochondrial molecular chaperone glucose-regulated protein 75 (GRP75) as a trigger for platelet activation and aggregation by stimulating the Ca2+-PKC alpha pathway. Genetic depletion of Glut1 in megakaryocytes blocked MYC-induced GRP75 expression. Pharmacologic blockade of platelet GRP75 compromised tumor-induced platelet activation and reduced metastasis. Moreover, in a pilot clinical study, drinking a 5% glucose solution elevated platelet GRP75 expression and activated platelets in healthy volunteers. Platelets from these volunteers promoted tumor metastasis in a plateletadoptive transfer mouse model. Together, under hyperglycemic conditions, MYC-induced upregulation of GRP75 in megakaryocytes increases platelet activation via the Ca2+-PKC alpha pathway to promote cancer metastasis, providing a potential new therapeutic target for preventing metastasis. Significance: This study provides mechanistic insights into a glucose-megakaryocyte-platelet axis that promotes metastasis and proposes an antimetastatic therapeutic approach by targeting the mitochondrial protein GRP75.

    • Fungal growth-dependent gene coregulation is strongly implicated in alteration of gene-encoding target proteases ruling with an antifungal resistance niche and biology of resistant mutants. On the basis of multialterative processes in this platform, the resistance-modifying strategy is designed in ketoconazole resistant Candida albicans and evaluated with less selective Momordica charantia protein and allosterically phosphorylated derivatives at the Thr102, Thr24 and Thr255 sites, respectively. We demonstrate absolutely chemosensitizing efficacy regarding stepwise-modifying resistance in sensitivity, by a load of only 26.23-40.00 mu g/l agents in Sabouraud's dextrose broth. Five successive modifying-steps realize the decreasing of ketoconazole E-test MIC50 from 11.10 to a lower level than 0.10 mg/l. With the ketoconazole resistance-modifying, colony undergoes a high-frequency morphological switch between high ploidy (opaque) and small budding haploid (white). A cellular event in the first modifying-step associates with relatively slow exponential growth (ie, a 4-h delay)-dependent action, mediated by agents adsorption. Moreover, multiple molecular roles are coupled with intracellularly and extracellularly binding to ATP-dependent RNA helicase dbp6; the 0.08-2.45 fold upregulation of TATA-box-binding protein, rRNA-processing protein and translation initiation factor 5A; and the 7.52-55.33% decrease of cytochrome P450 lanosterol 14 alpha-demethylase, glucan 1, 3-beta glucosidase, candidapepsin-1 and 1-acylglycerol-3-phosphate O-acyltransferase. Spatial and temporal gene coregulation, in the transcription and translation initiation stages with rRNA-processing, is a new coprocessing platform enabling target protease attenuations for resistance-impairing. An updated resistance-modifying measure of these agents in the low-dose antifungal strategic design may provide opportunities to a virtually safe therapy that is in high dose-dependency.

    • Gubenyiliu II (GYII), a Traditional Chinese Medicine (TCM) formula used in our hospital, has shown beneficial effects in cancer patients. In this study, we investigated the molecular mechanisms underlying the beneficial effects of GYII on murine breast cancer models. GYII showed significant inhibitory effects on tumor growth and metastasis in the murine breast cancer model. Additionally, GYII suppressed the proliferation of 4T1 and MCF-7 cells in a dose-dependent manner. A better inhibitory effect on 4T1 cell proliferation and migration was found in the decomposed recipes (DR) of GYII. Moreover, heparanase expression and the degree of angiogenesis were reduced in tumor tissues. Western blot analysis showed decreased expression of heparanase and growth factors in the cells treated with GYII and its decomposed recipes (DR2 and DR3), and thereby a reduction in the phosphorylation of extracellular signal-regulated kinase (ERK) and serine-threonine kinase (AKT). These results suggest that GYII exerts anti-tumor growth and anti-metastatic effects in the murine breast cancer model. The anti-tumor activity of GYII and its decomposed recipes is, at least partly, associated with decreased heparanase and growth factor expression, which subsequently suppressed the activation of the ERK and AKT pathways.

    • Pathogenic variants in MYH3 cause distal arthrogryposis type 2A and type 2B3 as well as contractures, pterygia and spondylocarpotarsal fusion syndromes types 1A and 1B. These disorders are ultra-rare and their natural course and phenotypic variability are not well described. In this study, we summarize the clinical features and genetic findings of 17 patients from 10 unrelated families with vertebral malformations caused by dominant or recessive pathogenic variants in MYH3. Twelve novel pathogenic variants in MYH3 (NM_002470.4) were identified: three of them were de novo or inherited in autosomal dominant way and nine were inherited in autosomal recessive way. The patients had vertebral segmentation anomalies accompanied with variable joint contractures, short stature and dysmorphic facial features. There was a significant phenotypic overlap between dominant and recessive MYH3-associated conditions regarding the degree of short stature as well as the number of vertebral fusions. All monoallelic variants caused significantly decreased SMAD3 phosphorylation, which is consistent with the previously proposed pathogenic mechanism of impaired canonical TGF-β signaling. Most of the biallelic variants were predicted to be protein-truncating, while one missense variant c.4244T>G,p.(Leu1415Arg), which was inherited in an autosomal recessive way, was found to alter the phosphorylation level of p38, suggesting an inhibition of the non-canonical pathway of TGF-β signaling. In conclusion, the identification of 12 novel pathogenic variants and overlapping phenotypes in 17 affected individuals from 10 unrelated families expands the mutation and phenotype spectrum of MYH3-associated skeletal disorders. We show that disturbances of canonical or non-canonical TGF-β signaling pathways are involved in pathogenesis of MYH3-associated skeletal fusion (MASF) syndrome.

Due to the spatial separation between antenna arrays, the large-scale propagation effect is different from a user to different antenna arrays in a cell, which makes power control a challenging problem as compared to conventional massive MIMO. We assume that the BS in each cell obtains the channel estimates via uplink pilots. Based on the channel estimates, the BSs perform maximum ratio transmission for the downlink. We then derive a closed-form spectral efficiency (SE) expression, where the channels are subject to correlated fading. Utilizing the derived expression, we propose a max-min power control algorithm to ensure that each user in the network receives a uniform quality of service. Numerical results demonstrate that, for the network considered in this work, optimizing for max-min SE through the max-min power control improves the sum SE of the network as compared to the equal power allocation.

    • In this paper, we investigate optimal downlink power allocation in massive multiple-input multiple-output (MIMO) networks with distributed antenna arrays (DAAs) under correlated and uncorrelated channel fading. In DAA massive MIMO, a base station (BS) consists of multiple antenna sub-arrays. Notably, the antenna sub-arrays are deployed in arbitrary locations within a DAA massive MIMO cell. Consequently, the distance-dependent large-scale propagation coefficients are different from a user to these different antenna sub-arrays, which makes power control a challenging problem. We assume that the network operates in time-division duplex mode, where each BS obtains the channel estimates via uplink pilots. Based on the channel estimates, the BSs perform maximum-ratio transmission in the downlink. We then derive a closed-form signal-to-interference-plus-noise ratio (SINR) expression, where the channels are subject to correlated fading. Based on the SINR expression, we propose a network-wide max-min power control algorithm to ensure that each user in the network receives a uniform quality of service. Numerical results demonstrate the performance advantages offered by DAA massive MIMO. For some specific scenarios, DAA massive MIMO can improve the average per-user throughput up to 55%. Furthermore, we demonstrate that channel fading covariance is an important factor in determining the performance of DAA massive MIMO.

    • Advances in imaging and display engineering have given rise to new and improved image and videoapplications that aim to maximize visual quality under given resource constraints (e.g., power, bandwidth).Because the human visual system is an imperfect sensor, the images/videos can be represented in a mathematicallylossy fashion but with enough fidelity that the losses are visually imperceptible—commonly termed“visually lossless.” Although a great deal of research has focused on gaining a better understanding ofthe limits of human vision when viewing natural images/video, a universally or even largely accepted definitionof visually lossless remains elusive. Differences in testing methodologies, research objectives, and targetapplications have led to multiple ad-hoc definitions that are often difficult to compare to or otherwise employ inother settings. We present a compendium of technical experiments relating to both vision science and visualquality testing that together explore the research and business perspectives of visually lossless image quality,as well as review recent scientific advances. Together, the studies presented in this paper suggest that a singledefinition of visually lossless quality might not be appropriate; rather, a better goal would be to establish varyinglevels of visually lossless quality that can be quantified in terms of the testing paradigm.

  • Observation of a linked-loop quantum state in a topological magnet

    • Quantum phases can be classified by topological invariants, which take on discrete values capturing global information about the quantum state1–13. Over the past decades, these invariants have come to play a central role in describing matter, providing the foundation for understanding superfluids5, magnets6,7, the quantum Hall effect3,8, topological insulators9,10, Weyl semimetals11–13 and other phenomena. Here we report an unusual linking-number (knot theory) invariant associated with loops of electronic band crossings in a mirror-symmetric ferromagnet14–20. Using state-of-the-art spectroscopic methods, we directly observe three intertwined degeneracy loops in the material’s three-torus, T3, bulk Brillouin zone. We find that each loop links each other loop twice. Through systematic spectroscopic investigation of this linked-loop quantum state, we explicitly draw its link diagram and conclude, in analogy with knot theory, that it exhibits the linking number (2, 2, 2), providing a direct determination of the invariant structure from the experimental data. We further predict and observe, on the surface of our samples, Seifert boundary states protected by the bulk linked loops, suggestive of a remarkable Seifert bulk–boundary correspondence. Our observation of a quantum loop link motivates the application of knot theory to the exploration of magnetic and superconducting quantum matter.

    • We report the generation and analysis of functional data from multiple, diverse experiments performed on a targeted 1% of the human genome as part of the pilot phase of the ENCODE Project. These data have been further integrated and augmented by a number of evolutionary and computational analyses. Together, our results advance the collective knowledge about human genome function in several major areas. First, our studies provide convincing evidence that the genome is pervasively transcribed, such that the majority of its bases can be found in primary transcripts, including non-protein-coding transcripts, and those that extensively overlap one another. Second, systematic examination of transcriptional regulation has yielded new understanding about transcription start sites, including their relationship to specific regulatory sequences and features of chromatin accessibility and histone modification. Third, a more sophisticated view of chromatin structure has emerged, including its inter-relationship with DNA replication and transcriptional regulation. Finally, integration of these new sources of information, in particular with respect to mammalian evolution based on inter- and intra-species sequence comparisons, has yielded new mechanistic and evolutionary insights concerning the functional landscape of the human genome. Together, these studies are defining a path for pursuit of a more comprehensive characterization of human genome function.

    • Belonging to the enyne family, enetriynes comprise a distinct electron-rich all-carbon bonding scheme. However, the lack of convenient synthesis protocols limits the associated application potential within, e.g., biochemistry and materials science. Herein we introduce a pathway for highly selective enetriyne formation via tetramerization of terminal alkynes on a Ag(100) surface. Taking advantage of a directing hydroxyl group, we steer molecular assembly and reaction processes on square lattices. Induced by O-2 exposure the terminal alkyne moieties deprotonate and organometallic bis-acetylide dimer arrays evolve. Upon subsequent thermal annealing tetrameric enetriyne-bridged compounds are generated in high yield, readily self-assembling into regular networks. We combine high-resolution scanning probe microscopy, X-ray photoelectron spectroscopy and density functional theory calculations to examine the structural features, bonding characteristics and the underlying reaction mechanism. Our study introduces an integrated strategy for the precise fabrication of functional enetriyne species, thus providing access to a distinct class of highly conjugated pi-system compounds. Enetriynes, which belong to the enyne family, are characterized by a distinct electron-rich carbon-bonding scheme. Here, the authors report the formation of enetriynes with high selectivity by tetramerization of terminal alkynes on Ag(100).

    • To identify susceptibility genes for amyotrophic lateral sclerosis (ALS), we conducted a genome-wide association study (GWAS) in 506 individuals with sporadic ALS and 1,859 controls of Han Chinese ancestry. Ninety top SNPs suggested by the current GWAS and 6 SNPs identified by previous GWAS were analyzed in an independent cohort of 706 individuals with ALS and 1,777 controls of Han Chinese ancestry. We discovered two new susceptibility loci for ALS at 1q32 (CAMK1G, rs6703183, P-combined = 2.92 x 10(-8), odds ratio (OR) = 1.31) and 22p11 (CABIN1 and SUSD2, rs8141797, P-combined = 2.35 x 10(-9), OR = 1.52). These two loci explain 12.48% of the overall variance in disease risk in the Han Chinese population. We found no association evidence for the previously reported loci in the Han Chinese population, suggesting genetic heterogeneity of disease susceptibility for ALS between ancestry groups. Our study identifies two new susceptibility loci and suggests new pathogenic mechanisms of ALS.

    • A complex reaction mechanism of oxidation of the anti-tubercular prodrug isoniazid (isonicotinic hydrazide, INH) by [IrCl6]2− as a model for redox processes of such drugs in biological systems has been studied in aqueous solution as a function of pH between 0 and 8.5. Similar experiments have been performed with its isomer nicotinic hydrazide (NH). All reactions are overall second-order, first-order in [IrCl6]2− and hydrazide, and the observed second-order rate constants k′ have been determined as a function of pH. Spectrophotometric titrations indicate a stoichiometry of [Ir(IV)]:[hydrazide] = 4:1. HPLC analysis shows that the oxidation product of INH is isonicotinic acid. The derived reaction mechanism, based on rate law, time-resolved spectra and stoichiometry, involves parallel attacks by [IrCl6]2− on all four protolytic species of INH and NH as rate-determining steps, depending on pH. These steps are proposed to generate two types of hydrazyl free radicals. These radicals react further in three rapid consecutive processes, leading to the final oxidation products. Rate constants for the rate-determining steps have been determined for all protolytic species I–IV of INH and NH. They are used to calculate reactivity–pH diagrams. These diagrams demonstrate that for both systems, species IV is ca. 105 times more reactive in the redox process than the predominant species III at the physiological pH of 7.4. Thus, species IV will be the main reactant, in spite of the fact that its concentration at this pH is extremely low, a fact that has not been considered in previous work. The results indicate that pH changes might be an important factor in the activation process of INH in biological systems also, and that in such systems this process most likely is more complicated than previously assumed

    • We examine the impact of multiple primary transmitters and receivers (PU-TxRx) on the outage performance of cognitive decode-and-forward relay networks. In such a joint relaying/spectrum-sharing arrangement, we address fundamental questions concerning three key power constraints: 1) maximum transmit power at the secondary transmitter (SU-Tx), 2) peak interference power at the primary receivers (PU-Rx), and 3) interference power at SU-Rx caused by the primary transmitter (PU-Tx). Our answers to these are given in new analytical expressions for the exact and asymptotic outage probability of the secondary relay network. Based on our asymptotic expressions, important design insights into the impact of primary transceivers on the performance of cognitive relay networks is reached. We have shown that zero diversity order is attained when the peak interference power at the PU-Rx is independent of the maximum transmit power at the SU-TX.

    • Transmit antenna selection with receive maximal-ratio combining (TAS/MRC) and transmit antenna selection with receive selection combining (TAS/SC) are two attractive multiple-input–multiple-output (MIMO) protocols. In this paper, we present a framework for the comparative analysis of TAS/MRC and TAS/SC in a two-hop amplify-and-forward relay network. In doing so, we derive exact and asymptotic expressions for the symbol error rate (SER) in Nakagami-mfading. Using the asymptotic expressions, the SNR gap between the two protocols is quantified. Given that the two protocols maintain the same diversity order, we show that the SNR gap is entirely dependent on the array gain. Motivated by this, we derive the SNR gap as a simple ratio of the respective array gains of the two protocols. This ratio explicitly takes into account the impact of the number of antennas and the fading severity parameter m. In addition, we address the fundamental question of “How to allocate the total transmit power between the source and the relay in such a way that the SER is minimized?” Our answer is given in the form of new compact expressions for the power allocation factor, which is a practical design tool that optimally distributes the total transmit power in the network.

    • Thermal management on a variety of length scales is essential for many industrial, solar and computational systems. Here we explore the thermal properties of a nearly perfect graphite absorber with both high emissivity and thermal conductivity which is fabricated by simple one-step etching. The hemispherical reflectance of the absorber is around 1% in the visible range and the normal specular reflectance is less than 1% from 1.5 mu m to 10 mu m. The thermal conductivity of the absorber is measured above 630 Wm(-1) K-1, which is 2.5 times larger than the aluminum used in commercial heat sinks. Heat dissipation testing indicates that a device is 3.7 +/- 2 degrees cooler with the graphite absorber as the heat spreader than a pristine graphite sheet and 9.1 +/- 2 degrees cooler than an Au-coated graphite sheet, primarily due to improved radiation cooling. A theoretical analysis accounts for the results. Further analysis suggests that the radiative heat dissipation is more prominent in the low-convection environment, especially for a convection coefficient below 10 Wm(-2) K-1.

    • Purpose The karst region in southwestern China is undergoing soil erosion and rocky desertification. The different silicon (Si) fractions along the hillslopes in this mountainous region could benefit plant growth and alleviate the ecological deterioration. However, extensive distribution of carbonate rocks may lead to limited plant available Si. The mountainous terrain in karst region also leads to more Si output, which seriously affects the biogeochemical cycle of Si in this area. Yet, the soil Si fractions in the karst region have not been fully evaluated. Methods Soil profiles and their corresponding plants were sampled from two typical karst mountains in Guizhou, China. The different fractions of non-crystalline Si in soil, accounting for the most important pool for Si availability to plants, were analyzed by the improved sequential chemical extraction and Si concentrations in plants grown in this region were also measured. Results The concentration and storage of non-crystalline Si were higher at lower slopes (storage was 2.44, 2.73, and 3.25 kg center dot m(-2) for upper, middle, and lower slopes, respectively) than other slope positions. Grasses dominated at lower slopes and contained significantly higher Si (mean +/- SD: 14.42 +/- 6.63 mg center dot g(-1)) than trees and shrubs (1.94 +/- 1.78 and 1.29 +/- 1.00 mg center dot g(-1), respectively), which were primarily distributed on upper slopes. However, Si concentrations of the same plant species in different slope positions had no significant correlation with soil acid Na acetate-Si, the Si regarded as directly available for plants. Conclusions This study suggests that plant species and soil properties have a significant impact on the soil Si distribution of hillslopes in karst region. Soil erosion may decrease non-crystalline Si concentrations in soils and impair Si uptake in grasses, which need to be considered in ecosystem management in this region.

    • The ability to tune magnetic orders, such as magnetic anisotropy and topological spin texture, is desired to achieve high-performance spintronic devices. A recent strategy has been to employ interfacial engineering techniques, such as the introduction of spin-correlated interfacial coupling, to tailor magnetic orders and achieve novel magnetic properties. We chose a unique polar-nonpolar LaMnO3/SrIrO3 superlattice because Mn (3d)/Ir (5d) oxides exhibit rich magnetic behaviors and strong spin-orbit coupling through the entanglement of their 3d and 5d electrons. Through magnetization and magnetotransport measurements, we found that the magnetic order is interface-dominated as the superlattice period is decreased. We were able to then effectively modify the magnetization, tilt of the ferromagnetic easy axis, and symmetry transition of the anisotropic magnetoresistance of the LaMnO3/SrIrO3 superlattice by introducing additional Mn (3d) and Ir (5d) interfaces. Further investigations using in-depth first-principles calculations and numerical simulations revealed that these magnetic behaviors could be understood by the 3d/5d electron correlation and Rashba spin-orbit coupling. The results reported here demonstrate a new route to synchronously engineer magnetic properties through the atomic stacking of different electrons, which would contribute to future applications in high-capacity storage devices and advanced computing.

  • Cascade photon upconversion amplification for selective multispectral narrow-band near-infrared photodetection

    • Facing the fact that selective detection of multiple narrow spectral bands in the near-infrared (NIR) region still poses a fundamental challenge, we have, in this work, developed NIR photodetectors (PDs) using photon upconversion nanocrystals (UCNCs) combined with perovskite films. In order to conquer the relatively high pumping threshold of UCNCs, we designed a novel cascade amplification strategy for upconversion luminescence (UCL) through cascading the superlensing effect of dielectric microlens arrays and the plasmonic effect of gold nanorods, which readily leads to a UCL enhancement by more than four orders of magnitude under weak light irradiation. By accommodating multiple optical active lanthanide ions in a core-shell-shell hierarchical architecture, the developed PDs on top can detect three well-separated narrow bands in the NIR region, i.e., 808, 980, and 1540 nm, respectively. Due to the large UCL enhancement, the obtained PDs demonstrate extremely high responsivity of 30.73, 23.15, 12.20 A/W and detectivity of 5.36, 3.45, 1.91x10^11 Jones for the 808, 980, and 1540 nm light detection, respectively, together with short response times in the range of 80-120 ms. Moreover, we demonstrate for the first time that the response to the excitation modulation frequency of a PD can be employed to discriminate the incident light wavelength. We believe that our work provides a novel insight for developing NIR PDs, and that it can spur the development of other applications using upconversion nanotechnology.

    • Since selective detection of multiple narrow spectral bands in the near-infrared (NIR) region still poses a fundamental challenge, we have, in this work, developed NIR photodetectors (PDs) using photon upconversion nanocrystals (UCNCs) combined with perovskite films. To conquer the relatively high pumping threshold of UCNCs, we designed a novel cascade optical field modulation strategy to boost upconversion luminescence (UCL) by cascading the superlensing effect of dielectric microlens arrays and the plasmonic effect of gold nanorods, which readily leads to a UCL enhancement by more than four orders of magnitude under weak light irradiation. By accommodating multiple optically active lanthanide ions in a core-shell-shell hierarchical architecture, developed PDs on top of this structure can detect three well-separated narrow bands in the NIR region, i.e., those centered at 808, 980, and 1540 nm. Recently, Hongwei Song and Wen Xu at Jilin University/China, Haichun Liu at KTH Royal Institute of Technology/Sweden, and their co-workers have successfully fabricated a novel multiple NIR bands photo-detectors (PDs) by combining multiple-excitation-bands core-shell upconversion nanocrystals (UCNCs) with MAPbI(3) perovskite photoelectric conversion layer. Through a cascade optical field modulation strategy, a combination of microlenses and gold plasmon nanocrystals, the emission intensity of the UCNCs and the photoelectric signal of the PDs can be enhanced four orders of magnitude. Moreover, the excitation frequency of the PD has been employed to discriminate the wavelength of incident light for the first time. This work provides a novel insight for developing multiple bands NIR PDs, and for applications of upconversion nanotechnology.

  • Genome-wide identification of resistance genes and cellular analysis of key gene knockout strain under 5-hydroxymethylfurfural stress in Saccharomyces cerevisiae

    • In bioethanol production, the main by-product, 5-hydroxymethylfurfural (HMF), significantly hinders microbial fermentation. Therefore, it is crucial to explore genes related to HMF tolerance in Saccharomyces cerevisiae for enhancing the tolerance of ethanol fermentation strains. A comprehensive analysis was conducted using genome-wide deletion library scanning and SGAtools, resulting in the identification of 294 genes associated with HMF tolerance in S. cerevisiae. Further KEGG and GO enrichment analysis revealed the involvement of genes OCA1 and SIW14 in the protein phosphorylation pathway, underscoring their role in HMF tolerance. Spot test validation and subcellular structure observation demonstrated that, following a 3-h treatment with 60mM HMF, the SIW14 gene knockout strain exhibited a 12.68% increase in cells with abnormal endoplasmic reticulum (ER) and a 22.41% increase in the accumulation of reactive oxygen species compared to the BY4741 strain. These findings indicate that the SIW14 gene contributes to the protection of the ER structure within the cell and facilitates the clearance of reactive oxygen species, thereby confirming its significance as a key gene for HMF tolerance in S. cerevisiae.

    • Hierarchical control of chemical reactions is being considered as one of the most ambitious and challenging topics in modern organic chemistry. In this study, we have realized the one-by-one scission of the X-H bonds (X = N and C) of aromatic amines in a controlled fashion on the Cu(lll) surface. Each dehydrogenation reaction leads to certain metal-organic supramolecular structures, which were monitored in single-bond resolution via scanning tunneling microscopy and noncontact atomic force microscopy. Moreover, the reaction pathways were elucidated from X-ray photoelectron spectroscopy measurements and density functional theory calculations. Our insights pave the way for connecting molecules into complex structures in a more reliable and predictable manner, utilizing carefully tuned stepwise on-surface synthesis protocols.

    • Background: Epidemiological studies observed gender differences in COVID-19 outcomes, however, whether sex hormone plays a causal in COVID-19 risk remains unclear. This study aimed to examine associations of sex hormone, sex hormones-binding globulin (SHBG), insulin-like growth factor-1 (IGF-1), and COVID-19 risk. Methods: Two-sample Mendelian randomization (TSMR) study was performed to explore the causal associations between testosterone, estrogen, SHBG, IGF-1, and the risk of COVID-19 (susceptibility, hospitalization, and severity) using genome-wide association study (GWAS) summary level data from the COVID-19 Host Genetics Initiative (N=1,348,701). Random-effects inverse variance weighted (IVW) MR approach was used as the primary MR method and the weighted median, MR-Egger, and MR Pleiotropy RESidual Sum and Outlier (MR-PRESSO) test were conducted as sensitivity analyses. Results: Higher genetically predicted IGF-1 levels have nominally significant association with reduced risk of COVID-19 susceptibility and hospitalization. For one standard deviation increase in genetically predicted IGF-1 levels, the odds ratio was 0.77 (95% confidence interval [CI], 0.61-0.97, p=0.027) for COVID-19 susceptibility, 0.62 (95% CI: 0.25-0.51, p=0.018) for COVID-19 hospitalization, and 0.85 (95% CI: 0.52-1.38, p=0.513) for COVID-19 severity. There was no evidence that testosterone, estrogen, and SHBG are associated with the risk of COVID-19 susceptibility, hospitalization, and severity in either overall or sex-stratified TSMR analysis. Conclusions: Our study indicated that genetically predicted high IGF-1 levels were associated with decrease the risk of COVID-19 susceptibility and hospitalization, but these associations did not survive the Bonferroni correction of multiple testing. Further studies are needed to validate the findings and explore whether IGF-1 could be a potential intervention target to reduce COVID-19 risk.

  • Zircon U-Pb-Hf Isotopes and Whole-Rock Geochemistry of the “Kulumudi Formation” from the Laofengkou Area (West Junggar) : Implications of the Construction of a Juvenile Arc in the Junggar–Balkhash Ocean

    • The properties of ancient magmatic arcs are crucial for understanding the tectonic evolution of the Central Asian Orogenic Belt. The Middle Devonian Kulumudi Formation in the Laofengkou area of West Junggar lacks accurate chronological data constraints, which hampers the knowledge of the nature of the Late Paleozoic magmatic arcs in the West Junggar and circum-Balkhash areas. In this contribution, samples of pyroclastic rocks and sedimentary rocks were collected from the volcano–sedimentary strata of the Kulumudi Formation. Petrography, zircon U-Pb-Hf isotopic analysis and whole-rock geochemistry were carried out to constrain the age and the tectonic setting of the Kulumudi Formation. The zircon U-Pb age of the lithic crystal tuff from the Kulumudi Formation on the northeast side of the Alemale Mountains was 386 ± 2 Ma, accurately indicating that this rock unit formed during the Middle Devonian. However, the fine sandstone near the Huojierte Mongolian Township, originally assigned as the “Kulumudi Formation”, yielded a maximum depositional age of 341 ± 3 Ma. Combined with the stratigraphic contact, this rock unit was redefined to belong to the Lower Carboniferous Jiangbasitao Formation. According to the whole-rock geochemistry study, the lithic crystal tuff of the Kulumudi Formation was characterized as medium potassium–calc–alkaline series rock, which is relatively enriched in light rare earth elements and large ion lithophile elements (i.e., Rb, Ba, K) and depleted in high-field-strength elements (i.e., Nb, Ta, Ti), showing similar geochemical characteristics to the volcanic arc rocks. By contrast, the fine sandstone from the Jiangbasitao Formation had Al2O3/SiO2 (0.25–0.29) and K2O/Na2O (1.29–1.72) ratios close to those derived from the continental arc and active continental margin and was characterized as part of the continental arc field in the La-Th-Sc and Th-Sc-Zr/10 tectonic discrimination diagrams. Zircon Hf isotope analysis showed that the εHf(t) values of the Kulumudi Formation were +5.6–+12.8, and those of the Jiangbasitao Formation were +11.43–+15.48, both of which show highly positive juvenile characteristics. The above data indicate that the Kulumudi Formation and Jiangbasitao Formation both formed in a juvenile arc setting with ocean–continent subduction. Combined with the previous work, it was concluded that the southward subduction of the ocean basin represented by the Darbut–Karamay ophiolitic mélanges beneath the newly accreted arc crustal segments produced a juvenile arc with positive Hf isotope characteristics.

  • Experimental study on effects of ammonia enrichment on the thermoacoustic instability of lean premixed swirling methane flames

    • Ammonia (NH3) has recently emerged as a promising carbon-free energy carrier. Further development and application of NH3 as fuel in the gas turbine industry can significantly reduce the emissions of carbon dioxide (CO2) and contribute to the achievement of a carbon–neutral society. This study experimentally examined the thermoacoustic instability characteristics of a laboratory-scaled lean premixed gas turbine model combustor operated with different NH3 blending ratios with methane (CH4). Experiments conducted under a wide range of inlet velocities and equivalence ratios suggest that NH3 concentration is critical in determining the characteristics of the instability. Specifically, when the NH3 proportion is less than 50 %, the addition of NH3 causes a mode transition of the instability. However, when the content of NH3 is greater than 50 %, it is shown that the instabilities are suppressed, indicating that the addition of a certain amount of NH3 can enhance the stability of CH4 flames. Additional analysis of flame dynamics reveals that the introduction of NH3 causes the lengthening of the flame front and weakens heat release rate fluctuations in the flame root regions. Further Proper Orthogonal Decomposition (POD) analysis of the flow field shows that the instability modes are strongly coupled with periodic vortex motions of the flow dynamics along the shear layers. Finally, the mode shifting phenomena is successfully predicted by low-order thermoacoustic network modeling. It is suggested that the change in convective time delay caused by NH3 addition is responsible for such transitions.

  • Genome Assembly of the Common Pheasant Phasianus colchicus : A Model for Speciation and Ecological Genomics

    • The commonpheasant (Phasianus colchicus) in the order Galliformes and the family Phasianidae, has 30 subspecies distributed across its native range in the Palearctic realm and has been introduced to Europe, NorthAmerica, and Australia. It is an important game bird often subjected to wildlife management as well as a model species to study speciation, biogeography, and local adaptation. However, the genomic resources for the commonpheasant are generally lacking. We sequenced a male individual of the subspecies torquatus of the common pheasant with the Illumina HiSeq platform. We obtained 94.88Gb of usable sequences by filtering out low-quality reads of the raw data generated. This resulted in a 1.02Gb final assembly, which equals the estimated genome size. BUSCO analysis using chicken as a model showed that 93.3% of genes were complete. The contig N50 and scaffold N50 sizes were 178 kb and 10.2 Mb, respectively. All these indicate that we obtained a high-quality genome assembly. We annotated 16,485 protein-coding genes and 123.3 Mb (12.05% of the genome) of repetitive sequences by ab initio and homology-based prediction. Furthermore, we applied a RAD-sequencing approach for another 45 individuals of seven representative subspecies in China and identified 4,376,351 novel single nucleotide polymorphism (SNPs) markers. Using this unprecedented data set, we uncovered the geographic population structure and genetic introgression among common pheasants in China. Our results provide the first high-quality reference genome for the common pheasant and a valuable genome-wide SNP database for studying population genomics and demographic history.

    • The formation, evolution, and potential as a climate archive of Tienchi Pond was evaluated using geomor-phology, sedimentology, geochemistry, and AMS radiocarbon measurements of tree ring cores. Tienchi Pond is a small ephemeral lake situated on the subtropical Lanyu Island in the Western Pacific Ocean about 90 km east of southern Taiwan. The lake is situated on a mountain ridge and is today characterized by numerous dead trees and stumps. The position at the boundary between the two main components of the SE Asian monsoon system, the Asian landmass and the Western Pacific, makes this lake particularly interesting as a potential natural climate archive. Since previous studies have shown that the bedrock underlying the lake formed over 2 my ago, a volcanic crater origin was ruled out. Rather, element ratios of K/Ti and Ca/Fe together with Pb-210 dating suggest a recent origin for the lake, probably as the result of a natural damming event in the southern, narrower, part of the lake basin, or as a result of increased precipitation at the end of the Little Ice Age (or a combination of both). Radiocarbon measurements on a dead tree near the lake perimeter show C-14 levels of more than 1.7% modern radiocarbon near the bark, suggesting that the tree died in the early 1970s. In contrast, by correlating a large number of radiocarbon measurements to a northern hemisphere reference curve, it was shown that trees closer to the center of the lake died in the early 20th century. This agrees with a scenario where a small lake formed by initial damming, and gradually grew larger through erosion of the surrounding shore lines caused by rapid lake level oscillations.

    • Five sediment cores from the Lomonosov Ridge and the Morris Jesup Rise north of Greenland show the history of sea-ice coverage and primary productivity over the last two glacial cycles. Variations in Manganese content, benthic and planktonic foraminifera, bioturbation, and trace fossil diversity are interpreted to reflect differences in sea-ice cover and sediment depositional conditions between the identified interglacials. Marine Isotope Stage (MIS) 1 and MIS 2 are represented by thin (<< 5 cm) sediment units while the preceding interglacial MIS 5 and glacial MIS 6 are characterized by thick (10 -20 cm) deposits. Foraminiferal abundances and bioturbation suggest that MIS 1 was generally characterized by severe sea-ice conditions north of Greenland while MISS appears to have been considerably warmer with more open water, higher primary productivity, and higher sedimentation rates. Strengthened flow of Atlantic water along the northern continental shelf of Greenland rather than development of local polynyas is here suggested as a likely cause for the relatively warmer marine conditions during MIS 5 compared to MIS 1. The cores also suggest distinct differences between the glacial intervals MIS 2 and MIS 6. While MIS 6 is distinguished by a relatively thick sediment unit poor in foraminifera and with low Mn values, MIS 2 is practically missing. We speculate that this could be the effect from a paleocrystic sea-ice cover north of Greenland during MIS 2 that prevented sediment delivery from sea ice and icebergs. In contrast, the thick sequence deposited during MIS 6 indicates a longer glacial period with dynamic intervals characterized by huge drifting icebergs delivering ice rafted debris (IRD). A drastic shift from thinner sedimentary cycles where interglacial sediment parameters indicate more severe sea-ice conditions gave way to larger amplitude cycles with more open water indicators was observed around the boundary between MIS 7/8. This shift is in agreement with a sedimentary regime shift previously identified in the Eurasian Basin and may be an indicator for the growth of larger ice sheets on the Eurasian landmass during the penultimate glacial period.
