    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • Abstract Improved fractionation process conditions for wood dissolution with switchable ionic liquids (SILs) were determined. The short time, high temperature (STHT) system was introduced as a selective and efficient way to extract components from lignocellulosic material. A SIL based on monoethanol amine (MEA) and 1,8-diazabicyclo-[5.4.0]-undec-7-ene (DBU) formed via coupling with SO2, was applied as a solvent in a 1:3 weight ratio with water. In essence, selective dissolution of mainly lignin was achieved by means of the aqueous SIL at 160 °C (∼6.1 bar corresponding to the vapor pressure of water) in 2 h and in a pressure vessel, for both hard- and soft-wood. About 95 wt-% of wood lignin was extracted. The dissolved components in the spent SIL were recovered by the addition of an anti-solvent whereupon over 70% of the dissolved components were recovered; the recovered fraction contained 19 wt-% hemicellulose while the rest of the material was in essence lignin. The non-dissolved, fluffy material contained ∼70 wt-% cellulose and ∼20 wt-% hemicellulose – a consistency resembling that of Kraft pulp.

  • Adaptive migratory orientation of an invasive pest on a new continent

    • Many species of insects undertake long-range, seasonally reversed migrations, displaying sophisticated orientation behaviors to optimize their migratory trajectories. However, when invasive insects arrive in new biogeographical regions, it is unclear if migrants retain (or how quickly they regain) ancestral migratory traits, such as seasonally preferred flight headings. Here we present behavioral evidence that an invasive migratory pest, the fall armyworm moth (Spodoptera frugiperda), a native of the Americas, exhibited locally adaptive migratory orientation less than three years after arriving on a new continent. Specimens collected from China showed flight orientations directed north-northwest in spring and southwest in autumn, and this would promote seasonal forward and return migrations in East Asia. We also show that the driver of the seasonal switch in orientation direction is photoperiod. Our results thus provide a clear example of an invasive insect that has rapidly exhibited adaptive migratory behaviors, either inherited or newly evolved, in a completely alien environment.

    • Recently, a structural battery with multifunctional carbon fibre anode has been reported. The energy density of active material is not fully extracted due to the low ionic conductivity inside the battery. To identify the main region that attributes to the low ion transportation, we assemble a symmetric structural battery with one anode layer in the centre sandwiched between two cathode layers. Such a design can also be treated as a combination of two asymmetric batteries with one full thickness cathode layer plus one half thickness anode layer. Thus, the travelled distance of lithium ions is shortened only in the anode part. It is found that the area energy density of the symmetric structural battery is doubled compared to a reference asymmetric battery. Thus, the additional cathode layer activates the double amount of carbon fibres in the anode. A plausible reason is that only the carbon fibres next to the separator is activated in the battery.

  • Phytochemicals with activity against methicillin-resistant Staphylococcus aureus

    • Background: The evolution of resistance to antimicrobials is a ubiquitous phenomenon. The evolution of antibiotic resistance in Staphylococcus aureus suggests that there is no remedy with sustaining effectiveness against this pathogen. The limited number of antibacterial drug classes and the common occurrence of cross-resistant bacteria reinforce the urgent need to discover new compounds targeting novel cellular functions. Natural products are a potential source of novel antibacterial agents. Anti-MRSA (methicillin-resistant S. aureus) bioactive compounds from Streptomyces and the anti-MRSA activity of a series of plant extracts have been reviewed respectively. However, there has been no detailed review of the precise bioactive components from plants.Purpose: The present review aimed to summarize the phytochemicals that have been reported with anti-MRSA activities, analyze their structure-activity relationship and novel anti-MRSA mechanisms.Methods: Data contained in this review article are compiled from the authoritative databases PubMed, Web of Science, Google Scholar, and so on.Results: This review summarizes 100 phytochemicals (27 flavonoids, 23 alkaloids, 17 terpenes and 33 others) that have been tested for their anti-MRSA activity. Among these phytochemicals, 39 compounds showed remarkable anti-MRSA activity with MIC values less than 10 mu g/ml, 14 compounds with MIC ranges including values <10 mu g/ml, 5 compounds with MIC values less than 5 mu M; 11 phytochemicals show synergism anti-MRSA effects in combination with antibiotics. Phytochemicals exerted anti-MRSA activities mainly by destroying the membrane structure and inhibiting the efflux pump.Conclusions: The 58 compounds with excellent anti-MRSA activity the 11 compounds with synergistic anti-MRSA effect, especially cannabinoids, xanthones and fatty acids should be further studied in vitro. Novel targets, such as cell membrane and efflux pump could be promising alternatives to develop antibacterial drugs in the future in order to prevent drug resistance.

    • Ioncell-F, a recently developed process for the production of man-made cellulosic fibers from ionic liquid solutions by dry-jet wet spinning, is presented as an alternative to the viscose and N-methylmorpholine N-oxide (NMMO)-based Lyocell processes. The ionic liquid 1,5-diazabicyclo[4.3.0]non-5-ene acetate was identified as excellent cellulose solvent allowing for a rapid dissolution at moderate temperatures and subsequent shaping into continuous filaments. The highly oriented cellulose fibers obtained upon coagulation in cold water exhibited superior tenacity, exceeding that of commercial viscose and NMMO-based Lyocell (Tencel) fibers. The respective staple fibers, which have been converted into two-ply yarn by ring spinning technology, presented very high tenacity. Furthermore, the Ioncell yarn showed very good behavior during the knitting and weaving processes, reflecting the quality of the produced yarn. The successfully knitted and woven garments from the Ioncell yarn demonstrate the suitability of this particular ionic liquid for the production of man-made cellulosic fibers and thus give a promising outlook for the future of the Ioncell-F process.
