  • Robust estimation of bacterial cell count from optical density

    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • The Visual Object Tracking challenge VOT2021 is the ninth annual tracker benchmarking activity organized by the VOT initiative. Results of 71 trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in recent years. The VOT2021 challenge was composed of four sub-challenges focusing on different tracking domains: (i) VOT-ST2021 challenge focused on short-term tracking in RGB, (ii) VOT-RT2021 challenge focused on "real-time" short-term tracking in RGB, (iii) VOT-LT2021 focused on long-term tracking, namely coping with target disappearance and reappearance and (iv) VOT-RGBD2021 challenge focused on long-term tracking in RGB and depth imagery. The VOT-ST2021 dataset was refreshed, while VOT-RGBD2021 introduces a training dataset and sequestered dataset for winner identification. The source code for most of the trackers, the datasets, the evaluation kit and the results along with the source code for most trackers are publicly available at the challenge website(1).

    • This paper proposes a gait recognition method based on the modified OVR-CSP fusion feature of plantar pressure and Long Short-Term Memory classification (referred to as the OVR-CSP-LSTM model). 10 subjects conducted 4 type of gait experiments including normal speed walking, fast walking, slow walking, imitating stroke gait walking in this paper. Transfer the commonly used Common Spatial Pattern (CSP) feature extraction method for EEG to plantar pressure signals, and splice the OVR-CSP features of 2-class, 3-class and 4-class, adopting Long Short Term Memory Network (LSTM) for classification. In this paper, the Intra-patient mode and Inter-patient mode of 10 people are modeled and compared respectively, and the recognition effects under different sensor number and different position sensors' combination are also studied. The experimental results show that the proposed model has good performance for both modes. The method proposed in this article is expected to be applied to multi-sensor signal processing and classification with spatial characteristics.

    • Li[LixNiyMnzCo1−x−y−z]O2 (lithium-rich NMCs) are benchmark cathode materials receiving considerable attention due to the abnormally high capacities resulting from their anionic redox chemistry. Although their anionic redox mechanisms have been much investigated, the roles of cationic redox processes remain underexplored, hindering further performance improvement. Here we decoupled the effects of nickel and cobalt in lithium-rich NMCs via a comprehensive study of two typical compounds, Li1.2Ni0.2Mn0.6O2 and Li1.2Co0.4Mn0.4O2. We discovered that both Ni3+/4+ and Co4+, generated during cationic redox processes, are actually intermediate species for triggering oxygen redox through a ligand-to-metal charge-transfer process. However, cobalt is better than nickel in mediating the kinetics of ligand-to-metal charge transfer by favouring more transition metal migration, leading to less cationic redox but more oxygen redox, more O2 release, poorer cycling performance and more severe voltage decay. Our work highlights a compositional optimization pathway for lithium-rich NMCs by deviating from using cobalt to using nickel, providing valuable guidelines for future high-capacity cathode design.

  • Optical characteristics of graptolite-bearing sediments and its implication for thermal maturity assessment

    • Graptolite reflectance was thought to be one of the most useful thermal maturity indicators for graptolite bearing sediments, however, the relationship between graptolite reflectance and vitrinite reflectance is not well established. Graptolites, especially in the Wufeng–Longmaxi Formations from the Ordovician to Silurian of South China, have been mistaken for vitrinite-like particles or solid bitumen, which results in inconsistent data on the thermal maturity. In this paper, we have employed optical microscope techniques to describe the detailed optical characteristics of graptolites and solid bitumen in the Wufeng–Longmaxi Formations. Laboratory simulation of maturation was used to determine the relationship between graptolite reflectance and vitrinite reflectance. The organic constituents in the Wufeng–Longmaxi Formations are mainly composed of graptolites and solid bitumen. Granular and non-granular graptolites were observed in the Wufeng–Longmaxi Formations, with nongranular as the most common texture. Solid bitumen can be distinguished from non-granular graptolites by its coarse surface, weaker anisotropy, and lower random reflectance. The combination of non-polarized and polarized light is very helpful to distinguish solid bitumen from graptolite. For comparison, organic material from the early Ordovician Alum Shale Formation of Sweden and Estonia was also studied. The macerals of the Alum shales are mainly composed of lamalginites, mineral-bituminous groundmasses, graptolites, and solid bitumen.The major textures of the graptolites in the Sweden and Estonia sediments are non-granular and granular, respectively.Both non-granular graptolite and vitrinite reflectances display a systematic increase with the increase of heating temperature and time. The granular graptolites in the Estonian sample were gradually changed to nongranular graptolites following laboratory simulated maturation, indicating that granular graptolites can transform into non-granular graptolites with maturation. Solid bitumen in the Wufeng–Longmaxi Formations was derived from the solid residue of kerogen and/or post-oil bitumen. The graptolite random reflectance is a better thermal maturity indicator than graptolite maximum reflectance and is more precise due to the smaller standard deviation. Several equations are proposed to determine the thermal maturity of the graptolite-bearing sediments based on graptolite random reflectance, graptolite maximum reflectance and solid bitumen random reflectance.

  • Simultaneously Improved Efficiency and Stability in All-Polymer Solar Cells by a P-i-N Architecture

    • All-polymer organic solar cells offer exceptional stability. Unfortunately, the use of bulk heterojunction (BHJ) structure has the intrinsic challenge to control the side-chain entanglement and backbone orientation to achieve sophisticated phase separation in all-polymer blends. Here, we revealed that the P-i-N structure can outperform the BHJ ones with a nearly 50% efficiency improvement, reaching a power conversion efficiency approaching 10%. This P-i-N structure can also provide an enhanced internal electric field and remarkably stable morphology Sequential deposition under harsh thermal stress. We have further demonstrated generality of the P-i-N structure in several other all-polymer systems. Considering the adjustable polymer molecular weight and solubility, the P-i-N device structure can be more beneficial for all-polymer systems. With the design of more crystalline polymers, the antiquated P-i-N structure can further show its strength in all-polymer systems by simplified morphology control and improved carrier extraction, becoming a more favorite device structure than the dominant BHJ structure.
