    • Polygenic risk scores (PRS) have the potential to identify individuals at risk of diseases, optimizing treatment, and predicting survival outcomes. Here, we construct and validate a genome-wide association study (GWAS) derived PRS for nasopharyngeal carcinoma (NPC), using a multi-center study of six populations (6 059 NPC cases and 7 582 controls), and evaluate its utility in a nested case-control study. We show that the PRS enables effective identification of NPC high-risk individuals (AUC = 0.65) and improves the risk prediction with the PRS incremental deciles in each population (Ptrend ranging from 2.79 × 10−7 to 4.79 × 10−44). By incorporating the PRS into EBV-serology-based NPC screening, the test’s positive predictive value (PPV) is increased from an average of 4.84% to 8.38% and 11.91% in the top 10% and 5% PRS, respectively. In summary, the GWAS-derived PRS, together with the EBV test, significantly improves NPC risk stratification and informs personalized screening.

  • Measurement of rainfall interception by xerophytic shrubs in re-vegetated sand dunes

    • More than 40 years of re-vegetation using mainly xerophytic shrubs Artemisia ordosica Krasch. and Caragana korshinskii Kom. at Shapotou Desert Experimental Research Station near Lanzhou, China has resulted in established dwarf-shrub and herbaceous cover on sand dunes. Precipitation, as the sole source of water replenishment in the semiarid area, plays a pertinent role in sustaining the desert ecosystem. A field study was conducted to (a) measure interception loss on shrub canopies during individual rainfall events, (b) determine the canopy storage capacity of individual plants, and (c) explore the relationship between interception and rainfall parameters. The total rainfall and its respective partitions as throughfall were determined and the interception losses in the studied ecosystem were quantified. Interception loss was shown to differ among the xerophyte taxa studied. During the growing seasons, the average shrub community interception loss is 6.9% and 11.7% of the simultaneous overall precipitation, for A. ordosica and C. korshinskii, respectively. Taking into account the observed rainfall conditions and vegetation cover characteristics, it was concluded that the interception loss was 2.7% of the total annual precipitation verified in the period for the A. ordosica community with an average cover of 30%, canopy projection area of 0.8 m(2) and canopy storage capacity of 0.75 mm. In contrast, interception loss for the C korshinskii community was 3.8% with an average cover of 46%, canopy projection area of 3.8 m(2) and canopy storage capacity of 0.71 mm. For individual plants of both shrubs, the proportion of interception loss to gross rainfall decreased notably as the rainfall intensity increased between 0 and 2 min h(-1), while it tended to remain constant at about 0.1-0.2 for A. ordosica and 0.1-0.3 for C. korshinskii when the rainfall intensity was > 2 mm h(-1).

    • The low-lying energy spectrum of the extremely neutron-deficient self-conjugate (N = Z) nuclide Ru-88(44)44 has been measured using the combination of the Advanced Gamma Tracking Array (AGATA) spectrometer, the NEDA and Neutron Wall neutron detector arrays, and the DIAMANT charged particle detector array. Excited states in Ru-88 were populated via the Fe-54(Ar-36, 2n gamma)Ru-88* fusion-evaporation reaction at the Grand Accelerateur National d'Ions Lourds (GANIL) accelerator complex. The observed gamma-ray cascade is assigned to Ru-88 using clean prompt gamma-gamma-2-neutron coincidences in anticoincidence with the detection of charged particles, confirming and extending the previously assigned sequence of low-lying excited states. It is consistent with a moderately deformed rotating system exhibiting a band crossing at a rotational frequency that is significantly higher than standard theoretical predictions with isovector pairing, as well as observations in neighboring N > Z nuclides. The direct observation of such a "delayed" rotational alignment in a deformed N = Z nucleus is in agreement with theoretical predictions related to the presence of strong isoscalar neutron-proton pair correlations.

    • The Monte-Carlo simulated response for γ-ray detection of the FAst TIMing Array (FATIMA) for exploitation within the DEcay SPECtroscopy (DESPEC) experimental system at the FAIR Phase-0 facility at Darmstadt, Germany is presented. In this configuration, FATIMA consisted of 36 LaBr3(Ce) detectors surrounding the AIDA, position-sensitive charged-particle active stopper. The decay of the Iπ=8+ isomer-fed decay cascade in 96Pd, measured in the first DESPEC experiment at the FAIR-0 facility was used to validate the simulations. The experimental data yielded in-situ full-energy peak efficiency values for FATIMA of 11.2(11)%, 6.8(7)%, 3.8(4)% and 2.1(4)% at 106, 325, 684 and 1415 keV respectively, consistent with the values derived from the simulated response.

    • Ever-lasting interest in the structure of Sn-100 and neighbouring nuclei is still well justified by the fact that it is the heaviest doubly-magic nucleus with N=Z. State-of-the-art experimental techniques involving stable and radioactive beam facilities have enabled access to these exotic nuclei. In particular, the analysis of experimental data obtained in two DESPEC experiments at GSI Darmstadt extends the information on the shell structure and its evolution towards N = Z = 50, and allows the study of seniority conservation and proton-neutron interaction in the g(9/2) orbit. Several theoretical approaches for shell-model investigations are discussed and their predictive power assessed. The calculated systematics of the reduced transition probabilities for high- to medium-spin states in N similar to Z isotopes with active g(9/2) orbit is presented for the first time.

    • Somatic genome rearrangements are thought to play important roles in cancer development. We optimized a long-span paired-end-tag (PET) sequencing approach using 10-Kb genomic DNA inserts to study human genome structural variations (SVs). The use of a 10-Kb insert size allows the identification of breakpoints within repetitive or homology-containing regions of a few kilobases in size and results in a higher physical coverage compared with small insert libraries with the same sequencing effort. We have applied this approach to comprehensively characterize the SVs of 15 cancer and two noncancer genomes and used a filtering approach to strongly enrich for somatic SVs in the cancer genomes. Our analyses revealed that most inversions, deletions, and insertions are germ-line SVs, whereas tandem duplications, unpaired inversions, interchromosomal translocations, and complex rearrangements are over-represented among somatic rearrangements in cancer genomes. We demonstrate that the quantitative and connective nature of DNA–PET data is precise in delineating the genealogy of complex rearrangement events, we observe signatures that are compatible with breakage-fusion-bridge cycles, and we discover that large duplications are among the initial rearrangements that trigger genome instability for extensive amplification in epithelial cancers.

  • The Genetic Architecture of the Clustering of Cardiometabolic Risk Factors: A Study of 8- to 17-Year-Old Chinese Twins

    • We explored the genetic architecture of metabolic risk factors of cardiovascular diseases (CVDs) and their clustering in Chinese boys and girls. Seven metabolic traits (body mass index [BMI], waist circumference [WC], systolic blood pressure [SBP], diastolic blood pressure [DBP], total cholesterol [TC], triglyceride [TG], and uric acid [UA]) were measured in a sample of 1016 twins between 8 and 17 years of age, recruited from the Qingdao Twin Registry. Cholesky, independent pathway, and common pathway models were used to identify the latent genetic structure behind the clustering of these metabolic traits. Genetic architecture of these metabolic traits was largely similar in boys and girls. The highest heritability was found for BMI (a2 = 0.63) in boys and TC (a2 = .69) in girls. Three heritable factors, adiposity (BMI and WC), blood pressure (SBP and DBP), and metabolite factors (TC, TG, and UA), which formed one higher-order latent phenotype, were identified. Latent genetic, common environmental, and unique environmental factors indirectly impacted the three factors through one single latent factor. Our results suggest that there is one latent factor influencing several metabolic traits, which are known risk factors of CVDs in young Chinese twins. Latent genetic, common environmental, and unique environmental factors indirectly imposed on them. These results inform strategies for gene pleiotropic discovery and intervening of CVD risk factors during childhood and adolescence.

The constructed level structure exhibits a rotational behavior with a sharp backbending at (h) over bar omega approximate to 0.50 MeV. A decrease in alignment frequency and increase in alignment sharpness in the odd-mass isotonic chains around N = 44 is proposed as an effect of the enhanced isoscalar neutron-proton interactions in odd-mass nuclei when approaching the N = Z line.

    • This study aimed to develop and externally validate a COVID-19 mortality risk prediction algorithm.DesignRetrospective cohort study.SettingFive designated tertiary hospitals for COVID-19 in Hubei province, China.ParticipantsWe routinely collected medical data of 1364 confirmed adult patients with COVID-19 between 8 January and 19 March 2020. Among them, 1088 patients from two designated hospitals in Wuhan were used to develop the prognostic model, and 276 patients from three hospitals outside Wuhan were used for external validation. All patients were followed up for a maximal of 60 days after the diagnosis of COVID-19.MethodsThe model discrimination was assessed by the area under the receiver operating characteristic curve (AUC) and Somers’ D test, and calibration was examined by the calibration plot. Decision curve analysis was conducted.Main outcome measuresThe primary outcome was all-cause mortality within 60 days after the diagnosis of COVID-19.ResultsThe full model included seven predictors of age, respiratory failure, white cell count, lymphocytes, platelets, D-dimer and lactate dehydrogenase. The simple model contained five indicators of age, respiratory failure, coronary heart disease, renal failure and heart failure. After cross-validation, the AUC statistics based on derivation cohort were 0.96 (95% CI, 0.96 to 0.97) for the full model and 0.92 (95% CI, 0.89 to 0.95) for the simple model. The AUC statistics based on the external validation cohort were 0.97 (95% CI, 0.96 to 0.98) for the full model and 0.88 (95% CI, 0.80 to 0.96) for the simple model. Good calibration accuracy of these two models was found in the derivation and validation cohort.ConclusionThe prediction models showed good model performance in identifying patients with COVID-19 with a high risk of death in 60 days. It may be useful for acute risk classification.

This paper reports on first results of an experiment performed in spring 2021, with a focus on beta-decaystudies in the Po-Fr nuclei in the 220 < A <230 island of octupole deformation exploiting the DESPEC setup. Ion-beta correlations and fast-timing techniques are being employed, giving an insight into this difficult-to-reach region.
