    • The Visual Object Tracking challenge VOT2018 is the sixth annual tracker benchmarking activity organized by the VOT initiative. Results of over eighty trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in the recent years. The evaluation included the standard VOT and other popular methodologies for short-term tracking analysis and a “real-time” experiment simulating a situation where a tracker processes images as if provided by a continuously running sensor. A long-term tracking subchallenge has been introduced to the set of standard VOT sub-challenges. The new subchallenge focuses on long-term tracking properties, namely coping with target disappearance and reappearance. A new dataset has been compiled and a performance evaluation methodology that focuses on long-term tracking capabilities has been adopted. The VOT toolkit has been updated to support both standard short-term and the new long-term tracking subchallenges. Performance of the tested trackers typically by far exceeds standard baselines. The source code for most of the trackers is publicly available from the VOT page. The dataset, the evaluation kit and the results are publicly available at the challenge website (http://votchallenge.net).

Results of 81 trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in the recent years. The evaluation included the standard VOT and other popular methodologies for short-term tracking analysis as well as the standard VOT methodology for long-term tracking analysis. The VOT2019 challenge was composed of five challenges focusing on different tracking domains: (i) VOT-ST2019 challenge focused on short-term tracking in RGB, (ii) VOT-RT2019 challenge focused on "real-time" short-term tracking in RGB, (iii) VOT-LT2019 focused on long-term tracking namely coping with target disappearance and reappearance. Two new challenges have been introduced: (iv) VOT-RGBT2019 challenge focused on short-term tracking in RGB and thermal imagery and (v) VOT-RGBD2019 challenge focused on long-term tracking in RGB and depth imagery. The VOT-ST2019, VOT-RT2019 and VOT-LT2019 datasets were refreshed while new datasets were introduced for VOT-RGBT2019 and VOT-RGBD2019. The VOT toolkit has been updated to support both standard short-term, long-term tracking and tracking with multi-channel imagery. Performance of the tested trackers typically by far exceeds standard baselines. The source code for most of the trackers is publicly available from the VOT page. The dataset, the evaluation kit and the results are publicly available at the challenge website(1).

  • Heterogeneous nuclear ribonucleoprotein L facilitates recruitment of 53BP1 and BRCA1 at the DNA break sites induced by oxaliplatin in colorectal cancer

    • Although oxaliplatin is an effective chemotherapeutic drug for treatment of colorectal cancer (CRC), tumor cells can develop mechanisms to evade oxaliplatin-induced cell death and show high tolerance and acquired resistance to this drug. Heterogeneous nuclear ribonucleoprotein L (hnRNP L) has been proved to play a critical role in DNA repair during IgH class switch recombination (CSR) in B lymphocytes, while, its role in CRC and chemotherapeutic resistance remain unknown. Our study aims to uncover an unidentified mechanism of regulating DNA double-strand breaks (DSBs) by hnRNP L in CRC cells treated by oxaliplatin. In present study, we observed that knockdown of hnRNP L enhanced the level of DNA breakage and sensitivity of CRC cells to oxaliplatin. The expression of key DNA repair factors (BRCA1, 53BP1, and ATM) was unaffected by hnRNP L knockdown, thereby excluding the likelihood of hnRNP L mediation via mRNA regulation. Moreover, we observed that phosphorylation level of ATM changed oppositely to 53BP1 and BRCA1 in the CRC cells (SW620 and HCT116) which exhibit synergistic effect by oxaliplatin plus hnRNP L impairment. And similar phenomenon was observed in the foci formation of these critical repair factors. We also found that hnRNP L binds directly with these DNA repair factors through its RNA-recognition motifs (RRMs). Analysis of cell death indicated that the RRMs of hnRNP L are required for cell survival under incubation with oxaliplatin. In conclusion, hnRNP L is critical for the recruitment of the DNA repair factors in oxaliplatin-induced DSBs. Targeting hnRNP L is a promising new clinical approach that could enhance the effectiveness of current chemotherapeutic treatment in patients with resistance to oxaliplatin.

  • Robot learning towards smart robotic manufacturing : A review

    • Robotic equipment has been playing a central role since the proposal of smart manufacturing. Since the beginning of the first integration of industrial robots into production lines, industrial robots have enhanced productivity and relieved humans from heavy workloads significantly. Towards the next generation of manufacturing, this review first introduces the comprehensive background of smart robotic manufacturing within robotics, machine learning, and robot learning. Definitions and categories of robot learning are summarised. Concretely, imitation learning, policy gradient learning, value function learning, actor-critic learning, and model-based learning as the leading technologies in robot learning are reviewed. Training tools, benchmarks, and comparisons amongst different robot learning methods are delivered. Typical industrial applications in robotic grasping, assembly, process control, and industrial human-robot collaboration are listed and discussed. Finally, open problems and future research directions are summarised.

    • Injectable bioactive ceramics with excellent osteogenesis play important roles in bone regeneration field. In this study, we creatively fabricated the injectable nano-structured silicon-containing hydroxyapatite (Si-HAp) microspheres in diameter of 70–100 µm via hydrothermal treatment of calcium silicate (CaSiO3) microspheres in Na3PO4 aqueous solution. The fabricated Si-HAp microspheres were constructed by nano-rods with a diameter of 100 nm and length up to 1.5 µm. Comparing with the pure HAp microspheres, the obtained nano-structured Si-HAp microspheres exhibited excellent protein loading properties and sustained protein release capacities. Most importantly, the Si-substitution could apparently enhance the proliferation, osteogenic differentiation and the genes expression of angiogenic factors of rat bone marrow mesenchymal stem cells (rBMSCs). These all indicated that the nano-structured Si-HAp microspheres fabricated via hydrothermal transformation method might be used as promising injectable bioactive biomaterials and drug deliveries for bone regeneration.

  • Effects of the Selective MPS1 Inhibitor MPS1-IN-3 on Glioblastoma Sensitivity to Antimitotic Drugs

    • Background Glioblastomas exhibit a high level of chemotherapeutic resistance, including to the antimitotic agents vincristine and taxol. During the mitotic agent-induced arrest, glioblastoma cells are able to perform damage-control and self-repair to continue proliferation. Monopolar spindle 1 (MPS1/TTK) is a checkpoint kinase and a gatekeeper of the mitotic arrest.Methods We used glioblastoma cells to determine the expression of MPS1 and to determine the effects of MPS1 inhibition on mitotic errors and cell viability in combination with vincristine and taxol. The effect of MPS1 inhibition was assessed in different orthotopic glioblastoma mouse models (n = 3-7 mice/group). MPS1 expression levels were examined in relation to patient survival.Results Using publicly available gene expression data, we determined that MPS1 overexpression corresponds positively with tumor grade and negatively with patient survival (two-sided t test, P < .001). Patients with high MPS1 expression (n = 203) had a median and mean survival of 487 and 913 days (95% confidence intervals [CI] = 751 to 1075), respectively, and a 2-year survival rate of 35%, whereas patients with intermediate MPS1 expression (n = 140) had a median and mean survival of 858 and 1183 days (95% CI = 1177 to 1189), respectively, and a 2-year survival rate of 56%. We demonstrate that MPS1 inhibition by RNAi results in sensitization to antimitotic agents. We developed a selective small-molecule inhibitor of MPS1, MPS1-IN-3, which caused mitotic aberrancies in glioblastoma cells and, in combination with vincristine, induced mitotic checkpoint override, increased aneuploidy, and augmented cell death. MPS1-IN-3 sensitizes glioblastoma cells to vincristine in orthotopic mouse models (two-sided log-rank test, P < .01), resulting in prolonged survival without toxicity.Conclusions Our results collectively demonstrate that MPS1, a putative therapeutic target in glioblastoma, can be selectively inhibited by MPS1-IN-3 sensitizing glioblastoma cells to antimitotic drugs.

    • Human-robot collaboration (HRC) is becoming a trend in manufacturing industry. However, the dramatic changes of requirements from the market put a higher demand for the flexibility of manufacturing systems. Cyber-Physical Production System (CPPS) which offers benefits of autonomy, self-organisation, and interoperability can be adopted to increase the flexibility of manufacturing systems. IEC 61499 (International Electrotechnical Commission) function blocks (FBs) are modularised and reusable software components for distributed industrial control. It is a suitable technology to realise a CPPS. Therefore, CPPS and FBs can be combined to realise the HRC system. This paper proposes a framework and the implementation method of IEC 61499 FB based CPPS for physical human-robot interaction (pHRI) which is type of HRC. An industrial robot based CPPS for pHRI is decomposed into modularised FBs that can be networked to fulfil manufacturing tasks. An energy consumption FB based on a novel empirical energy consumption model is also added to the system for energy consumption monitoring of the Robot. An assembly case is used to demonstrate the feasibility of the proposed system. Results show that the FB based CPPS for pHRI possesses the potential capability for HRC based assembly. The future work is also discussed.

    • Human intention prediction is vital for the efficiency of human-robot collaboration (HRC) and is usually modeled based on data-driven methods. However, due to the complexity and diverse nature of HRC, data collection for human intention prediction suffers from low sampling efficiency which restricts the application of HRC in manufacturing. Different from traditional real world data collection, a digital modeling method for HRC is proposed in this paper to generate virtual HRC data. The dynamic musculoskeletal model of human is adopted to simulate the musculoskeletal dynamics of human. The metabolic energy consumption of human is computed and used as an indicator to evaluate the reality of the generated virtual data. The virtual data are used to train human intention prediction model and compared with experimental data. Experimental results show the reality of virtual data and its effectiveness for human intention modeling in human-robot collaborative assembly. The proposed method has potential for reducing the cost of data collection compared with purely experiments.

    • Impaired albumin reabsorption by proximal tubular epithelial cells (PTECs) has been highlighted in diabetic nephropathy (DN), but little is known about the underlying molecular mechanisms. Here we find that ORAI1-3, are preferentially expressed in PTECs and downregulated in patients with DN. Hyperglycemia or blockade of insulin signaling reduces the expression of ORAI1-3. Inhibition of ORAI channels by BTP2 and diethylstilbestrol or silencing of ORAI expression impairs albumin uptake. Transgenic mice expressing a dominant-negative Orai1 mutant (E108Q) increases albuminuria, and in vivo injection of BTP2 exacerbates albuminuria in streptozotocin-induced and Akita diabetic mice. The albumin endocytosis is Ca2+-dependent and accompanied by ORAI1 internalization. Amnionless (AMN) associates with ORAIs and forms STIM/ORAI/AMN complexes after Ca2+ store depletion. STIM1/ORAI1 colocalizes with clathrin, but not with caveolin, at the apical membrane of PTECs, which determines clathrin-mediated endocytosis. These findings provide insights into the mechanisms of protein reabsorption and potential targets for treating diabetic proteinuria.

    • The high efficiency all-small-molecule organic solar cells (OSCs) normally require optimized morphology in their bulk heterojunction active layers. Herein, a small-molecule donor is designed and synthesized, and single-crystal structural analyses reveal its explicit molecular planarity and compact intermolecular packing. A promising narrow bandgap small-molecule with absorption edge of more than 930 nm along with our home-designed small molecule is selected as electron acceptors. To the best of our knowledge, the binary all-small-molecule OSCs achieve the highest efficiency of 14.34% by optimizing their hierarchical morphologies, in which the donor or acceptor rich domains with size up to ca. 70 nm, and the donor crystals of tens of nanometers, together with the donor-acceptor blending, are proved coexisting in the hierarchical large domain. All-small-molecule photovoltaic system shows its promising for high performance OSCs, and our study is likely to lead to insights in relations between bulk heterojunction structure and photovoltaic performance.

    • Relaxation pathways of upper excited electronic states of malachite green (MG) in ethanol and in films are studied by steadystate and time-resolved spectroscopic techniques. In contrast to ethanol, where MG emits weak short-lived spectrally well separated S-2 and S-1 fluorescence with the lifetimes similar to 0.3 and similar to 0.9 ps, MG films show a much stronger broadband fluorescence within 430-700 nm, revealing multiexponential kinetics with the characteristic decay times tau(1) approximate to 1 ps, tau(2) approximate to 10 ps, tau(3) approximate to 0.05-0.8 ns, and tau(4) approximate to 2-3 ns. By the analysis of spectroscopic responses of MG in ethanol and in films as well as by theoretical modeling, we demonstrate that significant increase of fluorescence lifetimes and substantial enhancement of fluorescence intensity in MG films are stipulated by the decrease of efficiency of the S-2 -> S-1 and S-1 -> S-0 internal conversion, which in turn is caused by hindrance of rotation of MG's phenyl rings controlling the S-2/S-1 and S-1/S-0 conical intersections. These findings indicate that MG films may become promising non-Kasha materials (with reasonable S-2 emission) with numerous photophysical and photochemical applications.

    • Recently, sustainable development has obtained increasing attentions from governments, industry, and academia owing to the limited natural resources. In the area of energy consumption, manufacturing accounts for a major portion of the total energy usage in industry. There is a clear necessity for energy-efficient manufacturing by optimizing manufacturing activities. Condition monitoring is the technology that provides runtime information for optimization. This paper aims to provide a better understanding of past achievements and future trends of condition monitoring towards energy-efficient manufacturing. Since there are a variety of sensors and technologies that can be used for condition monitoring towards energy-efficient manufacturing, this paper divides manufacturing activities into three levels, namely unit process level, shop-floor level, and supply chain level, and summarizes and discusses the sensors and technologies required to enable energy-efficient manufacturing on each level. With the advancement of technology, condition monitoring shows the characteristic of intelligence. Intelligent sensors that can be applied to condition monitoring in energy-efficient manufacturing are also reviewed. This paper can be helpful to manufacturers who are willing to improve energy efficiency in own manufacturing practice.
