    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • Using 14M psi(2S) events taken with the BESII detector, chi(cJ) -> 2(K+K-) decays are studied. For the four-kaon final state, the branching fractions are B(chi(c0,1,2) ->.2(K+K-)) = (3.48 +/- 0.23 +/- 0.47) x 10(-3), (0.70 +/- 0.13 +/- 0.10) x 10(-3), and (2.17 +/- 0.20 +/- 0.31) x 10(-3). For the phi K+K- final state, the branching fractions, which are measured for the first time, are B(chi(c0,1,2) -> phi K+K-) = (1.03 +/- 0.22 +/- 0.15) x 10(-3), (0.46 +/- 0.16 +/- 0.06) x 10(-3), and (1.67 +/- 0.26 +/- 0.24) x 10(-4). For the phi phi final state, B(chi(c0,2) -> phi phi) = (0.94 +/- 0.21 +/- 0.13) x 10(-3) and (1.70 +/- 0.30 +/- 0.25) x 10(-3).

  • Genomic insights into salt adaptation in a desert poplar

    • Despite the high economic and ecological importance of forests, our knowledge of the genomic evolution of trees under salt stress remains very limited. Here we report the genome sequence of the desert poplar, Populus euphratica, which exhibits high tolerance to salt stress. Its genome is very similar and collinear to that of the closely related mesophytic congener, P. trichocarpa. However, we find that several gene families likely to be involved in tolerance to salt stress contain significantly more gene copies within the P. euphratica lineage. Furthermore, genes showing evidence of positive selection are significantly enriched in functional categories related to salt stress. Some of these genes, and others within the same categories, are significantly upregulated under salt stress relative to their expression in another salt-sensitive poplar. Our results provide an important background for understanding tree adaptation to salt stress and facilitating the genetic improvement of cultivated poplars for saline soils.

    • METIS WP4 covers research activities in network-level aspects of the advancement of wireless network technologies towards the year 2020 and beyond. The aim is to develop novel network-level technology concepts to address the challenges foreseen in future scenarios with regard to interference, traffic and mobility management issues. Moreover, another task of this work package is to propose functional enablers which can support the above potential solutions.This document provides* a report of the ongoing progress in WP4 regarding the research topics agreed upon in IR 4.1,* a high level description of the proposed concepts and approaches adopted by different partners.More specifically, the document describes, first set of potential network-level solutions and presents some first research results in order to position them with regards to the state of the art approaches. It also gives an overview of research activities to be considered later in WP4.

    • The pharmacokinetic characteristics of per- and polyfluoroalkyl substances (PFAS) affect their distribution and bioaccumulation in biological systems. The enterohepatic circulation leads to reabsorption of certain chemicals from bile back into blood and the liver and thus influences their elimination, yet its influence on PFAS bioaccumulation remains unclear. We explored the role of enterohepatic circulation in PFAS bioaccumulation by examining tissue distribution of various PFAS in wild fish and a rat model. Computational models were used to determine the reabsorbed fractions of PFAS by calculating binding affinities of PFAS for key transporter proteins of enterohepatic circulation. The results indicated that higher concentrations were observed in blood, the liver, and bile compared to other tissues for some PFAS in fish. Furthermore, exposure to a PFAS mixture on the rat model showed that the reabsorption phenomenon appeared during 8-12 h for most long-chain PFAS. Molecular docking calculations suggest that PFAS can bind to key transporter proteins via electrostatic and hydrophobic interactions. Further regression analysis adds support to the hypothesis that binding affinity of the apical sodium-dependent bile acid transporter is the most important variable to predict the human half-lives of PFAS. This study demonstrated the critical role of enterohepatic circulation in reabsorption, distribution, and accumulation of PFAS.

  • Dynamic analysis of the effect of nose length on train aerodynamic performance

    • The improved delayed detached eddy simulation (IDDES) was used to study the influence of the train’s nose length on its aerodynamic performance. Both the time-averaged and instantaneous near-wake structures and the associated distribution of slipstream velocity are compared for three nose lengths. As the nose length increases, the mean and Std values of the drag and lift force are decreased. The shorter nose-length case results in a higher slipstream velocity. In particular, at the TSI track-side position, the TSI value U_2δ for the 5-m nose length case is 30% and 32% higher than the corresponding values for the 7.5-m and 10-m nose length cases, respectively. The dynamical flow topology in the wake reveals that the flow structures of the 5-m nose length are different from those of the other two cases in the tail streamline surface. As nose length increases, the longitudinal vortices are weaker, and the angle and distance between the longitudinal vortices are smaller. The shear production from the P_xy caused by the separation of the boundary layer at the lateral wall of the tail train is greater than that of the P_xz caused by the separation of the boundary layer at the top and bottom of the tail train.

  • Analysis of Thermal Effect on Residual Stresses of Broached Inconel 718

    • Inconel 718 is a nickel based superalloy that is widely used as a turbine disc material in gas turbine industries. This study details the effect of thermal exposure on the residual stresses produced when broaching Inconel 718. The chosen parameters for broaching in this study are similar to those used when manufacturing turbine discs. The broaching operation produced a high level of tensile residual stresses at the broached surface. A layer with tensile residual stresses was formed in the sub-surface region, followed by a layer several times thicker with compressive residual stresses. Thermal exposure was conducted at 550 °C. The depth distributions of residual stresses after thermal exposure are presented and discussed in this paper. Complete relaxation of the surface tensile residual stresses was observed after 30 h thermal exposure, whereas the 3000 h thermal exposure influenced both the surface and sub-surface residual stress states.

    • Inconel 718 is a Ni-based superalloy that can perform excellently at elevated temperatures. However, surface and subsurface damages in the form of microstructural and property changes and tensile residual stresses are common in a machined Inconel 718 component because of its poor machinability. Such damages have a significant influence on performance and the life time of the part. To characterise microstructural damages and understand how they are correlated to machining conditions are not only important for the evaluation of surface integrity but also for the optimization of machining operations to minimise effects from the machining process. This paper uses the ECCI (electron channelling contrast imaging) and EBSD (electron back scatter diffraction) methods to study the effect of cutting tools and cutting speeds on subsurface plastic deformation of machined Inconel 718. When turning at 200 m/min, a comparable level of plastic deformation was found under the surface machined with uncoated cubic boron nitride (CBN), titanium nitride coated CBN and whisker reinforced alumina (WRA). With an increase in cutting speed, the plastic deformation depth increased, and uncoated CBN tools showed superior performance in term of subsurface microstructure alterations compared to the other tool materials.

    • The novel wrought nickel-based superalloy, AD 730™, is a good candidate material for turbine disc applications at high temperatures beyond 650 °C. The present study focuses on the machining performance of this newly developed alloy under high-speed turning conditions with advanced PCBN tools. Meanwhile, the machined surface integrity as influenced by cutting speed and feed rate was also investigated. The surface integrity was thoroughly characterized in terms of surface roughness and morphology, machining-induced plastic deformation, white layer formation, and residual stresses. It has been found that the cutting speed and feed rate had a strong effect on the cutting forces and resultant surface integrity. The cutting forces required when machining the alloy were gradually reduced with increasing cutting speed, while at 250 m/min and above, the flank tool wear became stronger which led to increased thrust force and feed force. A higher feed rate, on the other hand, always resulted in higher cutting forces. Increasing the cutting speed and feed rate in general deteriorated the surface integrity. High cutting speeds within the range of 200–250 m/min and a low feed rate of 0.1 mm/rev are preferable in order to implement more cost-effective machining without largely reducing the surface quality achieved. The formation of tensile residual stresses on the machined AD 730™, however, could be of a concern where good fatigue resistance is critical.

    • Inconel 718 is a high strength, heat resistant superalloy that is used extensively for components in hot sections of gas turbine engines. This paper presents an experimental study on the thermal stability of broached Inconel 718 in terms of microstructure and nano-hardness. The broaching process used in this study is similar to that used in gas turbine industries for machining fir-tree root fixings on turbine discs. Severe plastic deformation was found under the broached surface. The plastic deformation induces a work-hardened layer in the subsurface region with a thickness of ∼50 μm. Thermal exposure was conducted at two temperatures, 550 ∘C and 650 ∘C respectively, for 300 h. Recrystallization occurs in the surface layer during thermal exposure at 550 ∘C and α-Cr precipitates as a consequence of the growth of recrystallized δ phases. More recrystallized grains with a larger size form in the surface layer and the α-Cr not only precipitates in the surface layer, but also in the sub-surface region when the thermal exposure temperature goes up to 650 ∘C. The thermal exposure leads to an increase in nano-hardness both in the work-hardened layer and in the bulk material due to the coarsening of the main strengthening phase γ′′.

    • The formation mechanism of white layers during broaching and their mechanical properties are not well investigated and understood to date. In the present study, multiple advanced characterization techniques with nano-scale resolution, including transmission electron microscopy (TEM), transmission Kikuchi diffraction (TKD), atom probe tomography (APT) as well as nano-indentation, have been used to systematically examine the microstructural evolution and corresponding mechanical properties of a surface white layer formed when broaching the nickel-based superalloy Inconel 718.TEM observations showed that the broached white layer consists of nano-sized grains, mostly in the range of 20–50 nm. The crystallographic texture detected by TKD further revealed that the refined microstructure is primarily caused by strong shear deformation. Co-located Al-rich and Nb-rich fine clusters have been identified by APT, which are most likely to be γ′ and γ′′ clusters in a form of co-precipitates, where the clusters showed elongated and aligned appearance associated with the severe shearing history. The microstructural characteristics and crystallography of the broached white layer suggest that it was essentially formed by adiabatic shear localization in which the dominant metallurgical process is rotational dynamic recrystallization based on mechanically-driven subgrain rotations. The grain refinement within the white layer led to an increase of the surface nano-hardness by 14% and a reduction in elastic modulus by nearly 10% compared to that of the bulk material. This is primarily due to the greatly increased volume fraction of grain boundaries, when the grain size was reduced down to the nanoscale.

    • The current study focused on the surface integrity issues associated with broaching of Inconel 718 and the structural stability of the broached specimen at high temperatures, mainly involving the microstructural changes and residual stress relaxation. The broaching operation was performed using similar cutting conditions as that used in turbo machinery industries for machining fir-tree root fixings on turbine disks. Thermal exposure was conducted at 723 K, 823 K, and 923 K (450 A degrees C, 550 A degrees C, and 650 A degrees C) for 30, 300, and 3000 hours, respectively. Surface cavities and debris dragging, sub-surface cracks, high intensity of plastic deformation, as well as the generation of tensile residual stresses were identified to be the main issues in surface integrity for the broached Inconel 718. When a subsequent heating was applied, surface recrystallization and alpha-Cr precipitation occurred beneath the broached surface depending on the applied temperature and exposure time. The plastic deformation induced by the broaching is responsible for these microstructural changes. The surface tension was completely relaxed in a short time at the temperature where surface recrystallization occurred. The tensile layer on the sub-surface, however, exhibited a much higher resistance to the stress relief annealing. Oxidation is inevitable at high temperatures. The study found that the surface recrystallization could promote the local Cr diffusion on the broached surface.

  • The JCMT BISTRO-2 Survey: Magnetic Fields of the Massive DR21 Filament

    • We present 850 mu m dust polarization observations of the massive DR21 filament from the B-fields In STar-forming Region Observations (BISTRO) survey, using the POL-2 polarimeter and the SCUBA-2 camera on the James Clerk Maxwell Telescope. We detect ordered magnetic fields perpendicular to the parsec-scale ridge of the DR21 main filament. In the subfilaments, the magnetic fields are mainly parallel to the filamentary structures and smoothly connect to the magnetic fields of the main filament. We compare the POL-2 and Planck dust polarization observations to study the magnetic field structures of the DR21 filament on 0.1-10 pc scales. The magnetic fields revealed in the Planck data are well-aligned with those of the POL-2 data, indicating a smooth variation of magnetic fields from large to small scales. The plane-of-sky magnetic field strengths derived from angular dispersion functions of dust polarization are 0.6-1.0 mG in the DR21 filament and similar to 0.1 mG in the surrounding ambient gas. The mass-to-flux ratios are found to be magnetically supercritical in the filament and slightly subcritical to nearly critical in the ambient gas. The alignment between column density structures and magnetic fields changes from random alignment in the low-density ambient gas probed by Planck to mostly perpendicular in the high-density main filament probed by James Clerk Maxwell Telescope. The magnetic field structures of the DR21 filament are in agreement with MHD simulations of a strongly magnetized medium, suggesting that magnetic fields play an important role in shaping the DR21 main filament and subfilaments.

    • A fused deposition modeling(FDM)system via screw extrusion suitable for feeding granular feedstocks with high solid loading was developed. Key parameters included aspect ratio of the screw, the compression ratio and pitch etc. In order to get constant extruded mass flow and wire diameter a processing window was determined by optimizing the barrel temperature, the nozzle diameter and the screwing speed. Microstructural characterization coupled with flexural strength measurement revealed that a higher printing temperature was beneficial to the inter layer bonding. The sintered zirconia ceramic samples with 99% of theoretical density of the 3 mol% yttria stabilized tetragonal zirconia polycrystal (3Y-TZP) and flexural strength of 890 +/- 60 MPa was obtained. A set of zirconia ceramic parts with complex geometries and controllable dimensional accuracy was also successfully prepared for demonstrating the potential of the technique.

    • PURPOSE: As the field of CEST grows, various novel preparation periods using different parameters are being introduced. At the same time, large, multisite clinical studies require clearly defined protocols, especially across different vendors. Here, we propose a CEST definition standard using the open Pulseq format for a shareable, simple, and exact definition of CEST protocols.METHODS: We present the benefits of such a standard in three ways: (1) an open database on GitHub, where fully defined, human-readable CEST protocols can be shared; (2) an open-source Bloch-McConnell simulation to test and optimize CEST preparation periods in silico; and (3) a hybrid MR sequence that plays out the CEST preparation period and can be combined with any existing readout module.RESULTS: The exact definition of the CEST preparation period, in combination with the flexible simulation, leads to a good match between simulations and measurements. The standard allowed finding consensus on three amide proton transfer-weighted protocols that could be compared in healthy subjects and a tumor patient. In addition, we could show coherent multisite results for a sophisticated CEST method, highlighting the benefits regarding protocol sharing and reproducibility.CONCLUSION: With Pulseq-CEST, we provide a straightforward approach to standardize, share, simulate, and measure different CEST preparation schemes, which are inherently completely defined.

    • The Changsha-Xiangtan-Zhuzhou City Group is a heavy industrial district and accepted as the serious pollution area in the Xiangjiang River basin. In this study, 7 metals (Pb, Hg, Cd, As, Zn, Cu and Se) and the river water quality parameters including pH, dissolved oxygen (DO), Escherichia coli (E. coli), potassium permanganate index (CODMn), dichromate oxidizability (CODCr), five-day biochemical oxygen demand (BOD5), ammonia nitrogen (NH4+-N), total nitrogen (TN), total phosphorus (TP) and fluoride (F-) in 18 sampling sites of the Changsha-Xiangtan-Zhuzhou section are monthly monitored in 2016, which is the year to step into the second stage of the Xiangjiang River Heavy Metal Pollution Control Implementation Plan. It is found that E. coli, TN and TP are the main pollutants in the Changsha-Zhuzhou-Xiangtan section, and the pollution of heavy metal is not serious but As with potential risk to local people especially children should be concerned. In addition, Xiangtan city is mainly featured with heavy metal pollution, while Zhuzhou and Changsha city are both featured with other pollutants from municipal domestic sewage.

    • Genetic variants within nearly 1000 loci are known to contribute to modulation of blood lipid levels. However, the biological pathways underlying these associations are frequently unknown, limiting understanding of these findings and hindering downstream translational efforts such as drug target discovery.To expand our understanding of the underlying biological pathways and mechanisms controlling blood lipid levels, we leverage a large multi-ancestry meta-analysis (N=1,654,960) of blood lipids to prioritize putative causal genes for 2286 lipid associations using six gene prediction approaches. Using phenome-wide association (PheWAS) scans, we identify relationships of genetically predicted lipid levels to other diseases and conditions. We confirm known pleiotropic associations with cardiovascular phenotypes and determine novel associations, notably with cholelithiasis risk. We perform sex-stratified GWAS meta-analysis of lipid levels and show that 3-5% of autosomal lipid-associated loci demonstrate sex-biased effects. Finally, we report 21 novel lipid loci identified on the X chromosome. Many of the sex-biased autosomal and X chromosome lipid loci show pleiotropic associations with sex hormones, emphasizing the role of hormone regulation in lipid metabolism.Taken together, our findings provide insights into the biological mechanisms through which associated variants lead to altered lipid levels and potentially cardiovascular disease risk.

    • We present observations of polarized dust emission at 850 mu m from the L43 molecular cloud, which sits in the Ophiuchus cloud complex. The data were taken using SCUBA-2/POL-2 on the James Clerk Maxwell Telescope as a part of the BISTRO large program. L43 is a dense (N-H2 similar to 10(22) - 10(23) cm(-2)) complex molecular cloud with a submillimeter-bright starless core and two protostellar sources. There appears to be an evolutionary gradient along the isolated filament that L43 is embedded within, with the most evolved source closest to the Sco OB2 association. One of the protostars drives a CO outflow that has created a cavity to the southeast. We see a magnetic field that appears to be aligned with the cavity walls of the outflow, suggesting interaction with the outflow. We also find a magnetic field strength of up to similar to 160 +/- 30 mu G in the main starless core and up to similar to 90 +/- 40 mu G in the more diffuse, extended region. These field strengths give magnetically super- and subcritical values, respectively, and both are found to be roughly trans-Alfvenic. We also present a new method of data reduction for these denser but fainter objects like starless cores.

    • Ambient energy harvesting has great potential to contribute to sustainable development and address growing environmental challenges. Converting waste energy from energy-intensive processes and systems (e.g. combustion engines and furnaces) is crucial to reducing their environmental impact and achieving net-zero emissions. Compact energy harvesters will also be key to powering the exponentially growing smart devices ecosystem that is part of the Internet of Things, thus enabling futuristic applications that can improve our quality of life (e.g. smart homes, smart cities, smart manufacturing, and smart healthcare). To achieve these goals, innovative materials are needed to efficiently convert ambient energy into electricity through various physical mechanisms, such as the photovoltaic effect, thermoelectricity, piezoelectricity, triboelectricity, and radiofrequency wireless power transfer. By bringing together the perspectives of experts in various types of energy harvesting materials, this Roadmap provides extensive insights into recent advances and present challenges in the field. Additionally, the Roadmap analyses the key performance metrics of these technologies in relation to their ultimate energy conversion limits. Building on these insights, the Roadmap outlines promising directions for future research to fully harness the potential of energy harvesting materials for green energy anytime, anywhere.

    • The GABA(B) receptor (GABA(B)R) agonist baclofen has been used to treat alcohol and several other substance use disorders (AUD/SUD), yet its underlying neural mechanism remains unclear. The present study aimed to investigate cortical GABA(B)R dynamics following chronic alcohol exposure. Ex vivo brain slice recordings from mice chronically exposed to alcohol revealed a reduction in GABA(B)R-mediated currents, as well as a decrease of GABA(B1/2)R and G-protein-coupled inwardly rectifying potassium channel 2 (GIRK2) activities in the motor cortex. Moreover, our data indicated that these alterations could be attributed to dephosphorylation at the site of serine 783 (ser-783) in GABA(B2) subunit, which regulates the surface expression of GABA(B)R. Furthermore, a human study using paired-pulse-transcranial magnetic stimulation (TMS) analysis further demonstrated a reduced cortical inhibition mediated by GABA(B)R in patients with AUD. Our findings provide the first evidence that chronic alcohol exposure is associated with significantly impaired cortical GABA(B)R function. The ability to promote GABA(B)R signaling may account for the therapeutic efficacy of baclofen in AUD.

    • AD730® is a new type of nickel-based superalloy, developed by Aubert & Duval, with high temperature properties exceeding conventional superalloys such as Alloy 718. This paper presents the tool life model for this superalloy for the purpose of process optimization and manufacturing cost estimation. The tool life modelling is based on comprehensive machining tests, using the Colding model. The cutting tool material used is CBN 170, a grade specially designed and optimized for machining of nickel-based superalloys.

    • End-to-end autonomous driving provides a simple and efficient framework for autonomous driving systems, which can directly obtain control commands from raw perception data. However, it fails to address stability and interpretability problems in complex urban scenarios. In this paper, we construct a two-stage end-to-end autonomous driving model for complex urban scenarios, named HIIL (Hierarchical Interpretable Imitation Learning), which integrates interpretable BEV mask and steering angle to solve the problems shown above. In Stage One, we propose a pretrained Bird's Eye View (BEV) model which leverages a BEV mask to present an interpretation of the surrounding environment. In Stage Two, we construct an Interpretable Imitation Learning (IIL) model that fuses BEV latent feature from Stage One with an additional steering angle from Pure-Pursuit (PP) algorithm. In the HIIL model, visual information is converted to semantic images by the semantic segmentation network, and the semantic images are encoded to extract the BEV latent feature, which are decoded to predict BEV masks and fed to the IIL as perception data. In this way, the BEV latent feature bridges the BEV and IIL models. Visual information can be supplemented by the calculated steering angle for PP algorithm, speed vector, and location information, thus it could have better performance in complex and terrible scenarios. Our HIIL model meets an urgent requirement for interpretability and robustness of autonomous driving. We validate the proposed model in the CARLA simulator with extensive experiments which show remarkable interpretability, generalization, and robustness capability in unknown scenarios for navigation tasks.

    • A moving model test was carried out to investigate the associated smoke movement characteristics when a subway train on fire runs in a tunnel. Train models of the 1/10 and 1/15 scales were used. The spatial distributions of airflow velocity and smoke concentration were then analyzed, and the differences between moving fire sources and stationary fire sources were discussed. The results show that the smoke movement characteristics of a stationary fire source were greatly different from those of a moving one. Specifically, the smoke movement for the moving fire source was dominated by piston wind. Moreover, the process of the smoke spread could be divided into three stages, during which time the flow direction changed. The peak smoke concentration value occurred after the train tail passed by the measuring point. Besides, the impacts of train speed (60 km/h, 80 km/h, 100 km/h, and 120 km/h) and blockage ratio (0.19 and 0.43) on airflow velocity and smoke concentration were also investigated. With increasing train velocity, the airflow velocity increased, and the smoke concentration decreased. The maximum airflow velocity was approximately linear with the train velocity. Furthermore, the increasing blockage ratio enhanced the piston effect in the tunnel, thus increasing the airflow velocity and reducing the smoke concentration.

    • Liquid chromatography coupled tandem mass spectrometry (LC-MS/MS) is an important technique for detecting peptides in proteomics studies. Here, we present an open source software tool, termed IPeak, a peptide identification pipeline that is designed to combine the Percolator post-processing algorithm and multi-search strategy to enhance the sensitivity of peptide identifications without compromising accuracy. IPeak provides a graphical user interface (GUI) as well as a command-line interface, which is implemented in JAVA and can work on all three major operating system platforms: Windows, Linux/Unix and OS X. IPeak has been designed to work with the mzIdentML standard from the Proteomics Standards Initiative (PSI) as an input and output, and also been fully integrated into the associated mzidLibrary project, providing access to the overall pipeline, as well as modules for calling Percolator on individual search engine result files. The integration thus enables IPeak (and Percolator) to be used in conjunction with any software packages implementing the mzIdentML data standard. IPeak is freely available and can be downloaded under an Apache 2.0 license at https://code.google.com/p/mzidentml-lib/.

    • Dense alumina ceramics were additively manufactured efficiently through a 3D gel printing process. Hydroxyethyl cellulose (HEC) was applied to ensure the printability and rigid of the gel made from boehmite. A multi-step liquid desiccant drying method was implemented to improve the drying efficiency. The results showed that the solid loading and HEC addition were two useful parameters for adjusting the rheology properties of the gel to make it suitable for 3D printing. With polyethylene glycol(PEG) added as liquid desiccants, the printed bodies with section size of 10 mm could be dried within 26 h during which the deformation and crack formation was avoided despite a high linear shrinkage of 45 % was encountered. The successful preparation of dense monolithic alumna ceramics parts with an average grain size of 1 μm, 99 % of the theoretical density and a flexural strength of 380 ± 45 MPa indicated the potential of this process.

    • Human-centred thermal environment conditioning can guarantee thermal comfort needs of human occupants in their micro-environments by using localized heating/cooling devices. Meanwhile, less intensified thermal conditioning of unoccupied surrounding environments can achieve heating/cooling energy efficiency. The concept was originated from task/ambient conditioning, which was developed for personal comfort systems. Most of the localized heating/cooling devices are workstation based or chair based. Task conditioning would become more closely to targets (human bodies) by using special clothing materials or thermoelectric elements. From thermal physiological viewpoints, thermal stimulus to thermally sensitive body parts may generate better results for not only local thermal comfort but also for whole body thermal comfort. Thermal stimulus to the extremities (feet) has demonstrated good thermal comfort effects. Scandinavians are accustomed to outdoor harsh environments and prefer wearing heavy shoes' outdoors in winter. They have the habit of changing heavy shoes to sandals when entering offices, which give the opportunity to enhance localized heating effects of footwarmers by reducing shoes' thermal resistance. Climatic chamber tests with 32 Nordic subjects were performed under different indoor ambient heating temperatures with/without the aid of footwarmers. With footwarmers and sandals, indoor heating temperature at 16°C was acceptable, which achieved energy efficient thermal comfort.

  • Influence of relayed fans and low level exhausts on performance of attachment ventilation under heating mode

    • Compared with displacement ventilation, attachment ventilation characterized by high supply air momentum has the potential to generate warm air lake on the floor under winter heating mode. However, the airflow can not keep attaching to the floor and moves upwards by thermal buoyancy. The influence of relayed fans and low level exhausts on performance of attachment ventilation under heating mode was explored. One/two relayed fans and five dimensionless exhaust heights were investigated. Performance evaluating index include vertical temperature gradient, temperature efficiency, predicted mean vote (PMV), predicted percentage of dissatisfied (PPD) and Draught risk (DR). Simulation results showed that relayed fans and low level exhausts effectively increase the attachment length of warm air lake in winter. Operation of relayed fans reduces vertical temperature difference, maintaining the head and foot temperature difference less than 3 °C. Low level exhausts effectively increase occupant zone temperature, reduce airflow short circuit and improve temperature efficiency. The operation of relayed fans slightly increase the temperature efficiency under high level exhausts. Two relayed fans decrease the temperature efficiency under low level exhausts. Better performance can be achieved by using only one relayed fan.

    • Pure-red perovskite LEDs (PeLEDs) based on CsPb(Br/I)(3) nanocrystals (NCs) usually suffer from a compromise in emission efficiency and spectral stability on account of the surface halide vacancies-induced nonradiative recombination loss, halide phase segregation, and self-doping effect. Herein, a "halide-equivalent" anion of benzenesulfonate (BS-) is introduced into CsPb(Br/I)(3) NCs as multifunctional additive to simultaneously address the above challenging issues. Joint experiment-theory characterizations reveal that the BS- can not only passivate the uncoordinated Pb2+-related defects at the surface of NCs, but also increase the formation energy of halide vacancies. Moreover, because of the strong electron-withdrawing property of sulfonate group, electrons are expected to transfer from the CsPb(Br/I)(3) NC to BS- for reducing the self-doping effect and altering the n-type behavior of CsPb(Br/I)(3) NCs to near ambipolarity. Eventually, synergistic boost in device performance is achieved for pure-red PeLEDs with CIE coordinates of (0.70, 0.30) and a champion external quantum efficiency of 23.5%, which is one of the best value among the ever-reported red PeLEDs approaching to the Rec. 2020 red primary color. Moreover, the BS--modified PeLED exhibits negligible wavelength shift under different operating voltages. This strategy paves an efficient way for improving the efficiency and stability of pure-red PeLEDs.

  • Quantitative assessments of retinal macular structure among rural-dwelling older adults in China : a population-based, cross-sectional, optical coherence tomography study

    • ObjectivesTo quantitatively assess and compare retinal macular structures of rural-dwelling older adults in China using two different optical coherence tomography (OCT) scanners and to examine their associations with demographic, lifestyle, clinical and ocular factors.Design, setting and participants This population-based, cross-sectional study included 971 participants (age ≥60 years) derived from the Multimodal Interventions to Delay Dementia and Disability in Rural China study. We collected data on demographics, lifestyle factors, clinical conditions (eg, cardiovascular disease (CVD)) and ocular factors (eg, visual acuity and spherical equivalent). We used two models of spectral-domain OCT to measure macular parameters in nine Early Treatment Diabetic Retinopathy Study subfields. Data were analysed using the multiple general linear models.Results Spectralis OCT demonstrated higher macular thickness but a lower macular volume than Primus 200 OCT (p<0.05). Nasal quadrant of the inner and outer subfields was the thickest, followed by superior quadrant. Adjusting for multiple potential confounding variables, older age was significantly correlated with lower average inner and outer macular thicknesses and overall macular volume. Men had higher macular parameters than women. The presence of CVD was correlated with lower central macular thickness (β=−6.83; 95% CI: −13.08 to −0.58; p=0.032). Middle school or above was associated with higher average inner macular thickness (β=7.85; 95% CI: 1.14 to 14.55; p=0.022) and higher spherical equivalent was correlated with lower average inner macular thickness (β=−1.78; 95% CI: −3.50 to −0.07; p=0.042).Conclusions Macular thickness and volume assessed by Spectralis and Primus 200 OCT scanners differ. Older age and female sex are associated with lower macular thickness and volume. Macular parameters are associated with education, CVD and spherical equivalent.Trial registeration number MIND-China study (ChiCTR1800017758).

    • Loss of cell-cell adhesions is the indispensable first step for cancer cells to depart from the primary tumor mass to metastasize. Metastasis suppressor 1 (MTSS1) is frequently lost in metastatic tissues, correlating to advanced tumor stages and poor prognosis across a variety of cancers. Here we explore the anti-metastatic mechanisms of MTSS1, which have not been well understood. We found that MTSS1 is downregulated in NPC tissues. Lower levels of MTSS1 expression correlate to worse prognosis. We show that MTSS1 suppresses NPC cell migration and invasion in vitro through cytoskeletal remodeling at cell-cell borders and assembly of E-cadherin/beta-catenin/F-actin in adherens junctions. The I-BAR domain of MTSS1 was both necessary and sufficient to restore this formation of E-cadherin/beta-catenin/F-actin-mediated cell adherens junctions.

Assessing the surface quality of a machined part by characterizing subsurface microstructural alterations and residual stresses is essential for ensuring part performance and lifetime in aero-engines and power generators. This comparative study characterizes and analyzes subsurface microstructural alterations and residual stresses in Inconel 718 subjected to high-speed machining with PCBN and whisker-reinforced ceramic cutting tools. Effects of cutting tool materials and microgeometry on subsurface deformation, microstructural alterations, and residual stresses were investigated. Surface and subsurface regions of machined specimens were investigated using X-ray diffraction, electron channeling contrast imaging, and electron back-scatter diffraction to characterize microstructural alterations and measure deformation intensity and depth.     • Nickel-based superalloys are typical materials used in components of aeroengines and gas turbine machinery. The strength properties of these alloys at high temperatures are crucial not only to the performance (e.g., power generation efficiency, energy consumption, and greenhouse gas emissions) of aeroengines and industrial gas turbines, but also to machinability during component manufacturing. This study comparatively evaluated the surface integrity of two superalloys, AD730® and Inconel 718 (IN718), during high-speed finishing turning using cubic boron nitride (CBN) tools. IN718 is a conventional superalloy used for the hot section components of aeroengines and industrial gas turbines, while AD730® is a novel superalloy with enhanced high-temperature mechanical properties and good potential as a next-generation superalloy for these components. High-speed turning tests of two superalloys were conducted using a CBN cutting tool and jet stream cooling. The achieved surface integrity of the AD730® and IN718 superalloys was characterized and analyzed to assess the comparability of these alloys.

    • Amide proton transfer-weighted (APTw) MR imaging shows promise as a biomarker of brain tumor status. Currently used APTw MRI pulse sequences and protocols vary substantially among different institutes, and there are no agreed-on standards in the imaging community. Therefore, the results acquired from different research centers are difficult to compare, which hampers uniform clinical application and interpretation. This paper reviews current clinical APTw imaging approaches and provides a rationale for optimized APTw brain tumor imaging at 3 T, including specific recommendations for pulse sequences, acquisition protocols, and data processing methods. We expect that these consensus recommendations will become the first broadly accepted guidelines for APTw imaging of brain tumors on 3 T MRI systems from different vendors. This will allow more medical centers to use the same or comparable APTw MRI techniques for the detection, characterization, and monitoring of brain tumors, enabling multi-center trials in larger patient cohorts and, ultimately, routine clinical use.

    • Titania thin films were prepared on bare Si and Pt/Ti/SiO2/Si substrates by chemical bath deposition (CBD) from solutions of a titanium peroxo complex and subsequent calcinations at 350 and 700 degrees C, respectively. The CBD process allowed deposition on both uncoated and metal-coated Si substrates with the same deposition rate. Optimization of the annealing process yielded uniform and crack-free nanocrystalline anatase films. The influence of the film thickness, irradiation of visible light, measuring frequency, temperature and substrate on the dielectric properties will be discussed in the paper. Films with a final thickness of about 600 nm showed comparably high relative permittivity of 31.8 on silicon and of 52.7 on Pt/Ti/SiO2/Si substrates, respectively. The present route provides anatase thin films with higher dielectric constants than classical sol-gel routes and is therefore a promising candidate for potential applications in large scale integration.

    • The fatigue behavior of lead zirconate stannate titanate (PZST) ceramics prepared by spark plasma sintering (SPS) was investigated. Polarization and strain hysteresis loops were monitored. The material shows a high resistance to fatigue because of bipolar electric cycling. Both maximum strain and switchable polarization first show a fatigue stage 0 to 10(5) cycles and then a fatigue-free period up to 10(8) cycles. The maximum losses of maximum strain and switchable polarization are 18% and 10% of their initial values, respectively. The dominant fatigue mechanism is assigned to the pinning of domain walls by charged defects.

Compared with its counterpart densified by conventional sintering (CS), the SPS material shows a smaller remanent polarization and maximum strain as well as a higher coercive field. Electric fatigue in both materials was investigated. In contrast to CS samples, the SPS specimens show a lower resistance to bipolar electric cycling, characterized by a faster decrease in remanent polarization and maximum strain at cycle number below 10(6.5) and a subsequent slower reduction of the properties at high cycle numbers up to 10(8).

    • Lanthanum-doped lead zirconate stannate titanate ceramics were successfully compacted to full density by spark plasma sintering (SPS). SPS samples densified at 900degrees or 950degreesC exhibit nearly full density and fine grain size (about 300 nm). Compared with samples from conventional sintering (CS), SPS samples show larger permittivity accompanied by a deterioration in dielectric loss and special strain hysteresis loops similar to those of ferroelectric relaxors, with a diffuse AFE-FE phase transition and less field-induced longitudinal strain. The differences in the properties of SPS and CS materials are attributed to the variations of the resultant microstructures, especially the grain size, of the ceramics.

    • The pluripotency gene regulatory network of porcine induced pluripotent stem cells(piPSCs), especially in epigenetics, remains elusive. To determine the biological function of epigenetics, we cultured piPSCs in different culture conditions. We found that activation of pluripotent gene- and pluripotency-related pathways requires the erasure of H3K9 methylation modification which was further influenced by mouse embryonic fibroblast (MEF) served feeder. By dissecting the dynamic change of H3K9 methylation during loss of pluripotency, we demonstrated that the H3K9 demethylases KDM3A and KDM3B regulated global H3K9me2/me3 level and that their co-depletion led to the collapse of the pluripotency gene regulatory network. Immunoprecipitation-mass spectrometry (IP-MS) provided evidence that KDM3A and KDM3B formed a complex to perform H3K9 demethylation. The genome-wide regulation analysis revealed that OCT4 (O) and SOX2 (S), the core pluripotency transcriptional activators, maintained the pluripotent state of piPSCs depending on the H3K9 hypomethylation. Further investigation revealed that O/S cooperating with histone demethylase complex containing KDM3A and KDM3B promoted pluripotency genes expression to maintain the pluripotent state of piPSCs. Together, these data offer a unique insight into the epigenetic pluripotency network of piPSCs.
