  • Robust estimation of bacterial cell count from optical density
  • 2020
  • Ingår i: Communications Biology. - : Springer Science and Business Media LLC. - 2399-3642. ; 3:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • A century of trends in adult human height
  • 2016
  • Ingår i: eLIFE. - 2050-084X. ; 5
  • Tidskriftsartikel (refereegranskat)abstract
    • Being taller is associated with enhanced longevity, and higher education and earnings. We reanalysed 1472 population-based studies, with measurement of height on more than 18.6 million participants to estimate mean height for people born between 1896 and 1996 in 200 countries. The largest gain in adult height over the past century has occurred in South Korean women and Iranian men, who became 20.2 cm (95% credible interval 17.522.7) and 16.5 cm (13.319.7) taller, respectively. In contrast, there was little change in adult height in some sub-Saharan African countries and in South Asia over the century of analysis. The tallest people over these 100 years are men born in the Netherlands in the last quarter of 20th century, whose average heights surpassed 182.5 cm, and the shortest were women born in Guatemala in 1896 (140.3 cm; 135.8144.8). The height differential between the tallest and shortest populations was 19-20 cm a century ago, and has remained the same for women and increased for men a century later despite substantial changes in the ranking of countries.

  • Rising rural body-mass index is the main driver of the global obesity epidemic in adults
  • 2019
  • Ingår i: Nature. - : Springer Science and Business Media LLC. - 0028-0836 .- 1476-4687. ; 569:7755, s. 260-4
  • Tidskriftsartikel (refereegranskat)abstract
    • Body-mass index (BMI) has increased steadily in most countries in parallel with a rise in the proportion of the population who live in cities(.)(1,2) This has led to a widely reported view that urbanization is one of the most important drivers of the global rise in obesity(3-6). Here we use 2,009 population-based studies, with measurements of height and weight in more than 112 million adults, to report national, regional and global trends in mean BMI segregated by place of residence (a rural or urban area) from 1985 to 2017. We show that, contrary to the dominant paradigm, more than 55% of the global rise in mean BMI from 1985 to 2017-and more than 80% in some low- and middle-income regions-was due to increases in BMI in rural areas. This large contribution stems from the fact that, with the exception of women in sub-Saharan Africa, BMI is increasing at the same rate or faster in rural areas than in cities in low- and middle-income regions. These trends have in turn resulted in a closing-and in some countries reversal-of the gap in BMI between urban and rural areas in low- and middle-income countries, especially for women. In high-income and industrialized countries, we noted a persistently higher rural BMI, especially for women. There is an urgent need for an integrated approach to rural nutrition that enhances financial and physical access to healthy foods, to avoid replacing the rural undernutrition disadvantage in poor countries with a more general malnutrition disadvantage that entails excessive consumption of low-quality calories.

  • Nanofibrillated cellulose composite hydrogel for the replacement of the Nucleus Pulposus
  • 2011
  • Ingår i: Acta Biomaterialia. - : Elsevier BV. - 1742-7061 .- 1878-7568. ; 7:9, s. 3412-3421
  • Tidskriftsartikel (refereegranskat)abstract
    • The swelling and compressive mechanical behavior as well as the morphology and biocompatibility of composite hydrogels based on Tween® 20 trimethacrylate (T3), N-vinyl-2-pyrrolidone (NVP) and nanofibrillated cellulose (NFC) were assessed in the present study. The chemical structure of T3 was verified by FTIR and 1H NMR and the degree of substitution (DS) was found to be around 3. Swelling ratios of neat hydrogels composed of different concentrations of T3 and NVP were found to range from 1.5 to 5.7 with decreasing concentration of T3. Various concentrations of cellulose nanofibrils (0.2 to 1.6 wt%) were then used to produce composite hydrogels that showed lower swelling ratios than neat ones for a given T3 concentration. Neat and composite hydrogels exhibited typical non-linear response under compression. All composite hydrogels showed an increase in elastic modulus compared to neat hydrogel of about 3 to 8-fold, reaching 18 kPa at 0% strain and 62 kPa at 20% strain for the hydrogel with the highest NFC content. All hydrogels presented a porous and homogeneous structure, with interconnected pore cells of around 100 nm in diameter. The hydrogels are biocompatible. The results of this study demonstrate that composite hydrogels reinforced with NFC may be viable as nucleus pulposus implant due to their adequate swelling ratio that may restore annulus fibrosus loading and their increased mechanical properties that could possibly restore the height of intervertebral discs.

  • Applied coronary physiology for planning and guidance of percutaneous coronary interventions. A clinical consensus statement from the European Association of Percutaneous Cardiovascular Interventions (EAPCI) of the European Society of Cardiology
  • 2023
  • Ingår i: EuroIntervention. - : Europa Digital & Publishing. - 1774-024X .- 1969-6213. ; 19:6, s. 464-
  • Tidskriftsartikel (refereegranskat)abstract
    • The clinical value of fractional flow reserve and non-hyperaemic pressure ratios are well established in determining an indication for percutaneous coronary intervention (PCI) in patients with coronary artery disease (CAD). In addition, over the last 5 years we have witnessed a shift towards the use of physio-logy to enhance procedural planning, assess post-PCI functional results, and guide PCI optimisation. In this regard, clinical studies have reported compelling data supporting the use of longitudinal vessel analysis, obtained with pressure guidewire pullbacks, to better understand how obstructive CAD contributes to myocardial ischaemia, to establish the likelihood of functionally successful PCI, to identify the presence and location of residual flow-limiting stenoses and to predict long-term outcomes. The introduction of new functional coronary angiography tools, which merge angiographic information with fluid dynamic equations to deliver information equivalent to intracoronary pressure measurements, are now available and potentially also applicable to these endeavours. Furthermore, the ability of longitudinal vessel analysis to predict the functional results of stenting has played an integral role in the evolving field of simulated PCI. Nevertheless, it is important to have an awareness of the value and challenges of physiology-guided PCI in specific clinical and anatomical contexts. The main aim of this European Association of Percutaneous Cardiovascular Interventions clinical consensus statement is to offer up-to-date evidence and expert opin-ion on the use of applied coronary physiology for procedural PCI planning, disease pattern recognition and post-PCI optimisation.

  • Preparation and characterization of water-redispersible nanofibrillated cellulose in powder form
  • 2010
  • Ingår i: Cellulose. - : Springer Science and Business Media LLC. - 0969-0239 .- 1572-882X. ; 17:1, s. 19-30
  • Tidskriftsartikel (refereegranskat)abstract
    • Water-redispersible, nanofibrillated cellulose (NFC) in powder form was prepared from refined, bleached beech pulp (RBP) by carboxymethylation (c) and mechanical disintegration (m). Two routes were examined by altering the sequence of the chemical and mechanical treatment, leading to four different products: RBP-m and RBP-mc (route 1), and RBP-c and RBP-cm (route 2). The occurrence of the carboxymethylation reaction was confirmed by FT-IR spectrometry and 13C solid state NMR (13C CP-MAS) spectroscopy with the appearance of characteristic signals for the carboxylate group at 1,595 cm-1 and 180 ppm, respectively. The chemical modification reduced the crystallinity of the products, especially for those of route 2, as shown by XRD experiments. Also, TGA showed a decrease in the thermal stability of the carboxymethylated products. However, sedimentation tests revealed that carboxymethylation was critical to obtain water-redispersible powders: the products of route 2 were easier to redisperse in water and their aqueous suspensions were more stable and transparent than those from route 1. SEM images of freeze-dried suspensions from redispersed RBP powders confirmed that carboxymethylation prevented irreversible agglomeration of cellulose fibrils during drying. These results suggest that carboxymethylated and mechanically disintegrated RBP in dry form is a very attractive alternative to conventional NFC aqueous suspensions as starting material for derivatization and compounding with (bio)polymers.

  • Reinforcing effect of carboxymethylated nanofibrillated cellulose powder on hydroxypropyl cellulose
  • 2010
  • Ingår i: Cellulose. - : Springer Science and Business Media LLC. - 0969-0239 .- 1572-882X. ; 17:4, s. 793-802
  • Tidskriftsartikel (refereegranskat)abstract
    • Bionanocomposites of hydroxypropyl cellulose (HPC) and nanofibrillated cellulose (NFC) were prepared by solution casting. The various NFC were in form of powders and were prepared from refined, bleached beech pulp (RBP) by mechanical disintegration, optionally combined with a pre- or post mechanical carboxymethylation. Dynamic mechanical analysis (DMA) and tensile tests were performed to compare the reinforcing effects of the NFC powders to those of their never-dried analogues. For unmodified NFC powders an inferior reinforcing potential in HPC was observed that was ascribed to severe hornification and reagglomeration of NFC. In contrast, the composites with carboxymethylated NFC showed similar behaviors, regardless of the NFC suspensions being dried or not prior to composite preparation. SEM characterization confirmed a homogeneous dispersion of dried, carboxymethylated NFC within the HPC matrix. These results clearly demonstrate that drying of carboxymethylated NFC to a powder does not decrease its reinforcing potential in (bio)nanocomposites.

  • Nitrate removal from water using a nanopaper ion-exchanger
  • 2016
  • Ingår i: Environmental Science. - : Royal Society of Chemistry (RSC). - 2053-1400 .- 2053-1419. ; 2, s. 117-124
  • Tidskriftsartikel (refereegranskat)abstract
    • Nitrates seriously affect drinking water quality. We herein present a process for the efficient removal of nitrates from water using a nanopaper ion-exchanger, which can be operated in flow-through conditions. The nanopaper ion-exchanger was produced from nanofibrillated cellulose obtained from fibre sludge, a paper-production waste stream, using a simple paper-making process. The cellulose nanofibrils were modified with quaternary trimethylammonium groups. The performance of these cationic nanopaper ion-exchangers was assessed with respect to their permeance and nitrate adsorption. Nitrates could be successfully captured onto the cationic nanopaper and thus rejected from contaminated water during dynamic filtration experiments. The ion-exchange nanopaper had adsorption capacities in the range of commercial available adsorbers but with the advantage of reduced contact time.

  • Prognostic impact of t(16;21)(p11;q22) and t(16;21)(q24;q22) in pediatric AML: a retrospective study by the I-BFM Study Group.
  • 2018
  • Ingår i: Blood. - : American Society of Hematology. - 1528-0020 .- 0006-4971. ; 132:15, s. 1584-1592
  • Tidskriftsartikel (refereegranskat)abstract
    • To study the prognostic relevance of rare genetic aberrations in acute myeloid leukemia (AML), such as t(16;21), international collaboration is required. Two different types of t(16;21) translocations can be distinguished: t(16;21)(p11;q22), resulting in the FUS-ERG fusion gene; and t(16;21)(q24;q22), resulting in RUNX1-core binding factor (CBFA2T3). We collected data on clinical and biological characteristics of 54 pediatric AML cases with t(16;21) rearrangements from 14 international collaborative study groups participating in the international Berlin-Frankfurt-Münster (I-BFM) AML study group. The AML-BFM cohort diagnosed between 1997 and 2013 was used as a reference cohort. RUNX1-CBFA2T3 (n = 23) had significantly lower median white blood cell count (12.5 × 109/L, P = .03) compared with the reference cohort. FUS-ERG rearranged AML (n = 31) had no predominant French-American-British (FAB) type, whereas 76% of RUNX1-CBFA2T3 had an M1/M2 FAB type (M1, M2), significantly different from the reference cohort (P = .004). Four-year event-free survival (EFS) of patients with FUS-ERG was 7% (standard error [SE] = 5%), significantly lower compared with the reference cohort (51%, SE = 1%, P < .001). Four-year EFS of RUNX1-CBFA2T3 was 77% (SE = 8%, P = .06), significantly higher compared with the reference cohort. Cumulative incidence of relapse was 74% (SE = 8%) in FUS-ERG, 0% (SE = 0%) in RUNX1-CBFA2T3, compared with 32% (SE = 1%) in the reference cohort (P < .001). Multivariate analysis identified both FUS-ERG and RUNX1-CBFA2T3 as independent risk factors with hazard ratios of 1.9 (P < .0001) and 0.3 (P = .025), respectively. These results describe 2 clinically relevant distinct subtypes of pediatric AML. Similarly to other core-binding factor AMLs, patients with RUNX1-CBFA2T3 rearranged AML may benefit from stratification in the standard risk treatment, whereas patients with FUS-ERG rearranged AML should be considered high-risk.

  • Cobalt selenium oxohalides : catalysts for water oxidation
  • 2014
  • Ingår i: Dalton Transactions. - 1477-9226 .- 1477-9234. ; 43:10, s. 3984-33989
  • Tidskriftsartikel (refereegranskat)abstract
    • Two new oxohalides Co4Se3O9Cl2 and Co3Se4O10Cl2 have been synthesized by solid state reactions. They crystallize in the orthorhombic space group Pnma and the monoclinic space group C2/m respectively. The crystal structure of the two compounds are made up of similar building blocks; Co4Se3O9Cl2 is made up of [CoO4Cl2], [CoO5Cl] and [SeO3] polyhedra and Co3Se4O10Cl2 is made up of [CoO4Cl2] and [SeO3] polyhedra. As several Co-containing compounds have proved to be good catalysts for water oxidation, the activities of the two new compounds were compared with the previously found oxohalide Co5Se4O12Cl2 in reference to CoO and CoCl2. The one electron oxidant Ru(bpy)33+ was used as oxidizing species in a phosphate buffer and it was found that the activities of the oxohalide species were in between CoO and CoCl2. The roles of Cl− and PO43− ions are discussed.

  • 2013
  • Ingår i: ACS Applied Materials and Interfaces. - : American Chemical Society (ACS). - 1944-8244 .- 1944-8252. ; 5:15, s. 7613-7620
  • Tidskriftsartikel (refereegranskat)abstract
    • To address brittleness of nanoclay hybrids of high inorganic content, ductile polymers (polyethylene oxide and hydroxyethyl cellulose) and montmorillonite (MTM) have been assembled into hybrid films using a water-based filtration process. Nacre-mimetic layered films resulted and were characterized by FE-SEM and XRD. Mechanical properties at ambient condition were studied by tensile test, while performance at elevated temperature and moisture conditions were evaluated by TGA, dynamic vapor sorption, and dynamic thermomechanical and hygromechanical analyses. Antiflammability and barrier properties against oxygen and water vapor were also investigated. Despite their high MTM content in the 60-85 wt % range, the hybrids exhibit remarkable ductility and a storage modulus above 2 GPa even in severe conditions (300 degrees C or 94% RH). Moreover, they present fire-shielding property and are amongst the best oxygen and water vapor barrier hybrids reported in the literature. This study thus demonstrates nanostructure property advantages for synergistic effects in hybrids combining inexpensive, available, and environmentally benign constituents.

  • Drying and Pyrolysis of Cellulose Nanofibers from Wood, Bacteria, and Algae for Char Application in Oil Absorption and Dye Adsorption
  • 2017
  • Ingår i: ACS Sustainable Chemistry and Engineering. - : AMER CHEMICAL SOC. - 2168-0485. ; 5:3, s. 2679-2692
  • Tidskriftsartikel (refereegranskat)abstract
    • Native cellulose nanofibers (CNF) constitute an abundant resource for pyrolysis leading to char materials offering a wide range of properties and application possibilities. With the aim to produce chars having large surface area and pore volume for dyes adsorption and oil absorption, respectively, we herein explore the slow pyrolysis process of cellulose nanofibers from wood, Cladophora algae and bacteria that were subjected to various drying routes. Whereas algae CNF with their large crystallites lead to high surface area (S-BET) substrates using conventional drying from aqueous suspension, CNF from wood having smaller crystallites requires drying from solvents to reach high S-BET substrates, which results in chars with a good adsorption capacity for both anionic and cationic dyes. Moreover, the porosity of the CNF substrate can be tuned via an ice-templating freeze-drying procedure reaching values as high as 99.7% and corresponding chars capable of absorbing 64-120 g g(-1) of various oils and organic solvents. Besides the absorption/adsorption properties of the chars, we report effects of CNF source and structure on the thermal properties assessed by thermogravimetric and thermomechanical analyses, differential scanning calorimetry, and mass spectrometry, and we identified over 20 decomposition products and 3 expansion events occurring during CNF pyrolysis.

  • 2017
  • Ingår i: Current Opinion in Biomedical Engineering. - : Elsevier BV. - 2468-4511. ; 2, s. 29-34
  • Forskningsöversikt (refereegranskat)abstract
    • Nanoscaled versions of cellulose viz. cellulose nanofibers (CNF) or cellulose nanocrystals (CNC) isolated from natural resources are being used extensively since the past decade in the biomedical field e.g. for tissue engineering, implants, drug delivery systems, cardiovascular devices, and wound healing due to their remarkable mechanical, chemical and biocompatible properties. In the recent years, 3D printing of nanocellulose in combination with polymers is being studied as a viable route to future regenerative therapy. The printability of nanocellulose hydrogels owing to their shear thinning behavior and the possibility to support living cells allows 3D bioprinting using nanocellulose, a recent development which holds tremendous potential.
