  • Disruption of STAT3 signalling promotes KRAS-induced lung tumorigenesis

    • STAT3 is considered to play an oncogenic role in several malignancies including lung cancer; consequently, targeting STAT3 is currently proposed as therapeutic intervention. Here we demonstrate that STAT3 plays an unexpected tumour-suppressive role in KRAS mutant lung adenocarcinoma (AC). Indeed, lung tissue-specific inactivation of Stat3 in mice results in increased KrasG12D-driven AC initiation and malignant progression leading to markedly reduced survival. Knockdown of STAT3 in xenografted human AC cells increases tumour growth. Clinically, low STAT3 expression levels correlate with poor survival and advanced malignancy in human lung AC patients with smoking history, which are prone to KRAS mutations. Consistently, KRAS mutant lung tumours exhibit reduced STAT3 levels. Mechanistically, we demonstrate that STAT3 controls NF-κB-induced IL-8 expression by sequestering NF-κB within the cytoplasm, thereby inhibiting IL-8-mediated myeloid tumour infiltration and tumour vascularization and hence tumour progression. These results elucidate a novel STAT3–NF-κB–IL-8 axis in KRAS mutant AC with therapeutic and prognostic relevance.

  • WorkflowDSL : Scalable Workflow Execution with Provenance for Data Analysis Applications

    • Data analysis projects typically use different programming languages (from Python for prototyping to C++ for support of runtime constraints) at their different stages by different experts. This creates a need for a data processing framework that is re-usable across multiple programming languages and supports collaboration of experts. In this work, we discuss implementation of a framework which uses a Domain Specific Language (DSL), called WorkflowDSL, that enables domain experts to collaborate on fine-tuning workflows. The framework includes support for parallel execution without any specialized code. It also provides a provenance capturing framework that enables users to analyse past executions and retrieve complete lineage of any data item generated. Graph database is used for storing provenance data. Advantages of usage of a graph database compare to relational databases are demonstrated. Experiments which were performed using a real-world scientific workflow from the bioinformatics domain and industrial data analysis models show that users were able to execute workflows efficiently when using WorkflowDSL for workflow composition and Python for task implementations. Moreover, we show that capturing provenance data can be useful for analysing past workflow executions.

    • Beyond fifth generation (5G) communication systems aim towards data rates in the tera bits per second range, with improved and flexible coverage options, introducing many new technological challenges in the fields of network architecture, signal pro- cessing, and radio frequency front-ends. One option is to move towards cell-free, or distributed massive Multiple-Input Multiple-Output (MIMO) network architectures and highly integrated front-end solutions. This paper presents an outlook on be- yond 5G distributed massive MIMO communication systems, the signal processing, characterisation and simulation challenges, and an overview of the state of the art in millimetre wave antennas and electronics.

    • This paper describes a model for mobile radio channels that includes consideration of directions of arrival and is thus suitable for simulations of the performance of wireless systems that use smart antennas. The model is specified for 13 different types of environments, covering macro- micro- and picocells. In this paper, a hierarchy of modeling concepts is described, as well as implementation aspects that are valid for all environments. The model is based on the specification of directional channel impulse response functions, from which the impulse response functions at all antenna elements can be obtained. A layered approach, which distinguishes between external (fixed), large-scale-, and small-scale- parameters allows an efficient parameterization. Different implementation methods, based on either a tapped-delay line or a geometrical model, are described. The paper also derives the transformation between those two approaches. Finally, the concepts of clusters and visibility regions are used to account for large delay and angular spreads that have been measured. In two companion papers, the environment-specific values of the model parameters are explained and justified.

    • A grid array antenna working around 145 GHz is proposed in this paper. The antenna is built on liquid crystal polymer (LCP) and designed for the D-band antenna-in-package application. The intrinsic softness of the LCP material is a limiting factor of the antenna's aperture size. A 0.5-mm-thick copper core is used to compensate. By doing this, the rigidness of the antenna is effectively improved, compared with an antenna without the copper core. Wet etching is used to realize the patterns on the top and bottom conductor. Compared with a low-temperature cofired ceramic counterpart, we obtain a considerable cost reduction with acceptable performance. The proposed antenna has an impedance bandwidth of 136-157 GHz, a maximum gain of 14.5 dBi at 146 GHz, and vertical beams in the broadside direction between 141 and 149 GHz. The fabrication procedures of the antennas are introduced, and a parametric study is carried out, which shows the antenna's robustness against fabrication tolerances, such as the not-well-controlled etching rate and the substrate surface roughness. This makes the antenna a promising solution for mass production.

    • Integration of a 140-GHz packaged low-temperature cofired ceramic (LTCC) antenna with a power detector is demonstrated under the concept of antenna-in-package. The detector is designed on an indium phosphide (InP) process. A grid array antenna in LTCC is designed to provide package for the detector. Coplanar ground-signal-ground (GSG) bond wires are used to connect the detector and the antenna. Parallel plate mode is observed in the InP substrate and absorbed by the LTCC substrate. The comparison between the measured responsivity of the power detector with and without the antenna indicates the acceptable insertion loss of the coplanar GSG bond wires transition. It shows a feasible solution for the D-band front-end integration and packaging.

  • Metallic 3-D Printed Antennas for Millimeter- and Submillimeter Wave Applications

    • This paper presents a study to use the metallic three dimensional (3-D) printing technology for antenna implementations up to 325 GHz. Two different printing technologies and materials are used, namely binder jetting/sintering on 316L stainless steel and selective laser melting (SLM) on Cu-15Sn. Phases, microstructure, and surface roughness are investigated on different materials. Balancing between the cost and performance, the manually polished Cu-15Sn is selected to develop a series of conical horn antennas at the E-(60-90 GHz), D-(110-170 GHz), and H-band (220-325 GHz). Good agreement is observed between the simulated and measured antenna performance. The antennas' impedance bandwidth (vertical bar S-11 vertical bar < -20 dB) cover the whole operational band, with in-band gain of > 22.5, > 22, and > 21.5 dBi for the E-, D-, and H-band antennas, respectively. Compared with the traditional injection molding and micromachining for metallic horn antenna implementation, the 3-D printed metallic horn antenna features environmental friendliness, low cost, and short turn-around time. Compared with the nonmetallic 3-D printed antennas, they feature process simplicity and mechanical robustness. It proves great potential of the metallic 3-D printing technology for both industrial mass production and prototyping.

  • An MPER antibody neutralizes HIV-1 using germline features shared among donors

    • The membrane-proximal external region (MPER) of HIV-1 envelope glycoprotein (Env) can be targeted by neutralizing antibodies of exceptional breadth. MPER antibodies usually have long, hydrophobic CDRH3s, lack activity as inferred germline precursors, are often from the minor IgG3 subclass, and some are polyreactive, such as 4E10. Here we describe an MPER broadly neutralizing antibody from the major IgG1 subclass, PGZL1, which shares germline V/D-region genes with 4E10, has a shorter CDRH3, and is less polyreactive. A recombinant sublineage variant pan-neutralizes a 130-isolate panel at 1.4 μg/ml (IC50). Notably, a germline revertant with mature CDR3s neutralizes 12% of viruses and still binds MPER after DJ reversion. Crystal structures of lipid-bound PGZL1 variants and cryo-EM reconstruction of an Env-PGZL1 complex reveal how these antibodies recognize MPER and viral membrane. Discovery of common genetic and structural elements among MPER antibodies from different patients suggests that such antibodies could be elicited using carefully designed immunogens.
