  • Immune-related microRNAs in breast milk and their relation to regulatory T cells in breastfed children

    • BACKGROUND: The immunomodulatory capacity of breast milk may partially be mediated by microRNAs (miRNA), small RNA molecules that regulate gene expression on a post-transcriptional level and are hypothesized to be involved in modulation of immunological pathways. Here, we evaluate the expression of immune-related miRNAs in breast milk after pre- and postnatal supplementation with Limosilactobacillus reuteri and omega-3 (ω-3) polyunsaturated fatty acids (PUFAs), and the association to infant regulatory T cell (Treg) frequencies.METHODS: One-hundred and twenty women included in a double-blind, randomized, placebo-controlled allergy intervention trial received L. reuteri and/or ω-3 PUFAs daily from gestational week 20. Using Taqman qPCR, 24 miRNAs were analyzed from breast milk obtained at birth (colostrum) and after 3 months (mature milk) of lactation. The proportion of activated and resting Treg cells were analyzed in infant blood using flow cytometry at 6, 12, and 24 months.RESULTS: Relative expression changed significantly over the lactation period for most of the miRNAs; however, the expression was not significantly influenced by any of the supplements. Colostrum miR-181a-3p correlated with resting Treg cell frequencies at 6 months. Colostrum miR-148a-3p and let-7d-3p correlated with the frequencies of activated Treg cells at 24 months, as did mature milk miR-181a-3p and miR-181c-3p.CONCLUSION: Maternal supplementation with L. reuteri and ω-3 PUFAs did not significantly affect the relative miRNA expression in breast milk. Interestingly, some of the miRNAs correlate with Treg subpopulations in the breastfed children, supporting the hypothesis that breast milk miRNAs could be important in infant immune regulation.   • God innemiljö på svenska fartyg : Kartläggning av innemiljön och förslag på förbättringsåtgärder

    • Miljön ombord på fartyg är viktig för besättningens välbefinnande hälsa ocharbetseffektivitet. Inomhusluftens kvalitet och termisk komfort är viktiga delar avinnemiljön som inte har studerats så mycket på fartyg. Vid långa resor som vararveckor eller månader och kan passera flera klimatzoner finns inget sätt att byta miljöoch ingen utväg från fartyget, och då blir god innemiljön extra viktig eftersom denutgör både arbetsmiljö och boendemiljö (innemiljö). Syftet med denna studie var attkartlägga hur innemiljön på svenska fartyg ser ut, föreslå en enkel övervakningsmetodikför innemiljö samt föreslå förbättringsåtgärder där det behövs.Inom projektet har mätningar gjorts på nio fartyg under elva mätomgångar; ettfartyg undersöktes både vinter och sommar och ett annat fartyg före och efter byte avbränsle. Temperatur och relativ luftfuktighet som beskriver inneklimat, halterna avgasformiga luftföroreningarna koldioxid, kväveoxider, svaveldioxid, flyktiga organiskaämnen, polycykliska aromatiska kolväten, formaldehyd och partiklar (PM10, PM2.5 ochnanopartiklar) mättes i både maskinutrymmen och innemiljöer med diffusiva (passiva)provtagare och direktvisande instrument.Innemiljö och luftkvalitet på de undersökta fartygen var överlag god. Halter avluftföroreningar var under och mycket under både de hygieniska gränsvärdena förarbetsmiljö och rekommenderade riktvärden för god innemiljö. Även om det inteförekom hälsofarliga halter av luftföroreningarna, förekom stora skillnaderna i halterbåde inom och mellan fartyg. Principalkomponentanalys utpekade typ av bränsle somfrämsta indikator för luftkvalitet i maskinutrymmen samt bränslet och fartygensframdrivningssätt för andra innemiljöer på fartyget. Inom projektet har ”Indoor AirPollution Index” för fartyg utvecklats och använts för att rangordna fartygen medavseende på kvalitet av innemiljö.För att ytterligare höja kvaliteten på fartygens innemiljö, arbets- och boendemiljöhar följande förslag på tekniska och organisatoriska åtgärder för befintliga fartyg ochvid nybyggnation identifierats. För befintliga fartyg gäller framförallt kontroll ochunderhåll av ventilationssystem, ordning och reda särskilt i maskinutrymmen samt vidbehov användning av lämplig skyddsutrustning. Vid planering och design av nya fartygbör främst fartygets framdrivningssätt med tillhörande bränsle beaktas, eftersomanvändning av bränslen med bättre kvalitet än tjockolja också innebär förbättringar försåväl luftkvalitet som arbetsmiljö. Andra viktiga aspekter för upprätthållande av godinnemiljö är ventilationsanordning där man effektivt separerar avluftningar frånmaskinutrymmen från friskluftsintag till andra inneutrymmen samt utformning avarbetsplatser och innemiljöer med särskilt tonvikt på ett separat rengöringsrum imaskinutrymmen.

  • Argonaute, Vault, and Ribosomal Proteins Targeted by Autoantibodies in Systemic Lupus Erythematosus

    • OBJECTIVE: To expand, in an unbiased manner, our knowledge of autoantigens and autoantibodies in patients with systemic lupus erythematosus (SLE) and evaluate their associations with serological and clinical variables. METHODS: Human proteome arrays (> 21,000 proteins) were screened with serum from patients with SLE (n = 12) and healthy controls (n = 6) for IgG and IgA binding. Top hits were validated with 2 cohorts of patients with SLE (cohort 1, n = 49; cohort 2, n = 46) and other rheumatic diseases by ELISA. Clinical associations of the autoantibodies were tested. RESULTS: Ro60 was the top hit in the screen, and the 10 following proteins included 2 additional known SLE autoantigens plus 8 novel autoantigens involved in microRNA processing (Argonaute protein 1 [AGO1], AGO2, and AGO3), ribosomes (ribosomal protein lateral stalk subunit P2 and ovarian tumor deubiquitinase 5 [OTUD5]), RNA transport by the vault (major vault protein), and the immune proteasome (proteasome activator complex subunit 3). Patient serum contained IgG reactive with these proteins and IgA against the AGO proteins. Using the 95th percentile of healthy donor reactivity, 5-43% were positive for the novel antigens, with OTUD5 and AGO1 showing the highest percentages of positivity. Autoantibodies against AGO1 proteins were more prevalent in patients with oral ulcers in a statistically significant manner. IgG autoantibodies against AGO proteins were also seen in other rheumatic diseases. CONCLUSION: We discovered new autoantigens existing in cytosolic macromolecular protein assemblies containing RNA (except the proteasome) in cells. A more comprehensive list of autoantigens will allow for a better analysis of how proteins are targeted by the autoimmune response. Future research will also reveal whether specific autoantibodies have utility in the diagnosis or management of SLE.

  • Managing cross layer information in OMNeT++ simulations

    • This paper describes a general approach to dealing with cross layer information in OMNeT++-based network simulations. Rather than prescribe a specific cross layer architecture, this work is intended to address the software engineering problem of passing information between simulation modules in a sound way. An XML-based mechanism for passing information between modules using OMNeT++'s controlInfo data structure is presented. Because XML is an intermediate format for the OMNeT++ network description language (NED), it is further suggested that allowing NED files to be annotated with information about cross layer interactions will enhance readability and reuse-ability. A further goal of this paper is to initiate discussion and collaboration in the Swedish (and international) OMNeT++ community.

    • Interaction of macrophages with bacteria is a stimulus for production of cytokines such as IL-10 and IL-12. IL-12 stimulates T cell and natural killer (NK) cell cytotoxicity and interferon-gamma (IFN-gamma) production. IL-10 opposes the T cell-stimulating action of IL-12, decreases the release of proinflammatory cytokines from macrophages, and stimulates B cells. We have studied the capacity of human intestinal isolates from the three Lactobacillus species dominating on the human gastrointestinal mucosa, L. plantarum, L. rhamnosus and L. paracasei ssp. paracasei, to induce production of IL-10 and IL-12 from human blood mononuclear cells, or monocytes. Whole killed lactobacilli were potent stimulators of IL-12 over a wide range of bacterial concentrations. Lactobacillus paracasei gave the highest levels of IL-12 (1.5 ng/ml in response to 5 x 106 bacteria/ml), roughly 10 times more than obtained by stimulation with L. rhamnosus or L. plantarum. Escherichia coli induced on average < 50 pg/ml of IL-12 regardless of the bacterial concentration used. The secretion of free p40 subunit IL-12 followed the same pattern as the secretion of p70 (bioactive IL-12) with regard to the efficiency of different bacteria as stimulators. Escherichia coli was the most efficient trigger of IL-10 production, inducing 0.5 ng/ml IL-10 after stimulation with 5 x 106 bacteria/ml. Lactobacillus rhamnosus induced the highest levels of IL-10 among the lactobacilli (0.5 ng/ml) compared with 0.1 ng/ml evoked by L. plantarum or L. paracasei, but 10 times more bacteria were required for optimal stimulation than with E. coli. When neutralizing anti-IL-10 antibodies were added to the cultures, the IL-12-inducing capacity of L. rhamnosus was increased markedly, while that of E. coli remained low. The results show that mucosa-associated lactobacilli can be potent stimulators of IL-12, and thus potentially of cell-mediated immunity, if they pass over the gut epithelial barrier and interact with cells of the gut immune system.

  • Short-term effects and long-term changes of FUEL—a digital sports nutrition intervention on REDs related symptoms in female athletes

    • Female endurance athletes are at high risk for developing Relative Energy Deficiency in Sport (REDs), resulting in symptoms such as menstrual dysfunction and gastrointestinal (GI) problems. The primary aim of this study was to investigate effects of the FUEL (Food and nUtrition for Endurance athletes—a Learning program) intervention consisting of weekly online lectures combined with individual athlete-centered nutrition counseling every other week for sixteen weeks on REDs related symptoms in female endurance athletes at risk of low energy availability [Low Energy Availability in Females Questionnaire (LEAF-Q) score ≥8]. Female endurance athletes from Norway (n = 60), Sweden (n = 84), Ireland (n = 17), and Germany (n = 47) were recruited. Fifty athletes with risk of REDs (LEAF-Q score ≥8) and with low risk of eating disorders [Eating Disorder Examination Questionnaire (EDE-Q) global score <2.5], with no use of hormonal contraceptives and no chronic diseases, were allocated to either the FUEL intervention (n = 32) (FUEL) or a sixteen-week control period (n = 18) (CON). All but one completed FUEL and n = 15 completed CON. While no evidence for difference in change in LEAF-Q total or subscale scores between groups was detected post-intervention (BFincl < 1), the 6- and 12-months follow-up revealed strong evidence for improved LEAF-Q total (BFincl = 123) and menstrual score (BFincl = 840) and weak evidence for improved GI-score (BFincl = 2.3) among FUEL athletes. In addition, differences in change between groups was found for EDE-Q global score post-intervention (BFincl= 1.9). The reduction in EDE-Q score remained at 6- and 12- months follow-up among FUEL athletes. Therefore, the FUEL intervention may improve REDs related symptoms in female endurance athletes.   • Go get 'em tiger! When children are targeted by Criminal Law Policy not Social Law Policy

    • By drawing on Pierre Bourdieu’s view on the State, particularly the idea of the left hand and the right hand of the State, statutory reforms and policy in Sweden targeting youths committing crime will be analysed. While the left hand is constituted by, e.g., social welfare, education and health care, the right hand is constituted by, e.g., the justice system, the police and prisons. As will be discussed, one might argue that the handling of underage children committing crime have shifted from the left hand to the right hand of the State. In the Swedish context, it should be stressed that it is a continual development. One need to acknowledge that until the 1980s and 1990s, crime was generally framed as social problem with social causes. Since then, crime has been portrayed as a threat to the democratic society. More recently, on the policy level, crime is framed as a threat to the society and its functions. Consequently, criminal law policy is no longer viewed as a part of social policy. Instead, social policy has emerged as criminal law policy – but with other means. In other words, policies previously adopted within social law has shifted toward becoming criminal law. This particularly appears to be the case in adopted policy and statutory reforms concerning underage children. An illustrative example is the introduction of juvenile prisons for children between 15-17 years. The rationale behind the reform is that underage children committing crimes are a threat to us all, i.e., motivated by the “need to protect society”. This threat is deemed so severe that underaged children should be imprisoned. This marks a shift, since underage children committing crime previously have been handled within the Social Services, not the Swedish criminal justice system. Aside from elaborating the development outlined here, some tentative thoughts on possibilities and risks connected to the policies targeting underaged children will be discussed.
