  • Injury-Induced Signalling in Arterial Smooth Muscle Cells

    • The vascular wall is an active, elastic and integrated organ made up of cellular and extracellular matrix (ECM) components. It is not a static organ; the components dynamically change and reorganize in response to physiological and pathological stimuli. Vascular injury induces a complex healing process, analogous to generalised wound healing. Subsequent to the inflammatory response, proliferation and migration of smooth muscle cells (SMCs) is initiated as well as reorganisation of the ECM. The primary function of SMCs in the normal media is contraction, and to be able to migrate and proliferate they must first change their phenotype. This thesis focuses on the underlying molecular mechanism involved in the SMC response to injury. The understanding of these mechanisms enables us to understand the overall SMC function in the vessel. It is based on signalling and extracellular matrix events following injury to cultured arterial smooth muscle cells. In the first paper, the cytotoxic effects of 7beta-hydroxycholestrol are characterized. We show that this natural compound induces Ca2+ oscillations, ERK1/2 activation followed by apoptosis in SMCs. In the next two studies, we analysed the involvement of ERK1/2 in SMC responses to mechanical injury. In paper II, intracellular Ca2+ release and tyrosine kinase activation was investigated. Immediately after injury, Ca2+ is released from thapsigargine sensitive Ca2+-pools and subsequent ERK1/2 phosphorylation is initiated. Our data indicate that injury-induced DNA synthesis is dependent on immediate phosphorylation of ERK1/2, and probably other tyrosine kinases, whilst the injury-induced migration is dependent on later phosphorylation events. Paper III suggests that osteopontin, a glycoprotein in the ECM, is a downstream target of ERK1/2 signalling. We found in paper IV that one part of the immediate SMC response-to-injury to be a generation of a novel dermatan sulphate-containing complex. Our data indicates that this mitogenic complex is formed/released from the ECM/cells as a response to injury. Due to the dermatan sulphate involvement in such a complex, this thesis last part describes biosynthetic differences between SMC phenotypes with regard to their proteoglycan profile.

  • Waste incineration : impact of input waste fuel composition on trace element distribution and chemical speciation in fly and bottom ashes

    • Incineration process for waste to energy is a widely developed method for solid waste management in Sweden and all over the world. In addition to its unique benefits of mass and volume reduction of waste, it can also provide electricity and/or heat. In Sweden 48% of household waste, 40% of industrial waste and 75% of recovered waste wood (RWW) (construction and demolition) are treated through incineration. In order to avoid environmental concerns and operational problems posed by incineration by-products (fly ash, bottom ash), it is important to understand that how trace element partitioning respond to the changes in waste composition. In this study, Influence of input waste fuel composition and chlorine content on trace element distribution in solid waste (RWW, industrial waste, RWW mixed with bark & sludge and mixed household & industrial waste) incineration fly and bottom ashes were studied. Further, chemical speciation of trace elements in resulting fly/bottom ashes was investigated by sequential extraction. Results indicate that Zn, Cu, Pb and Cr are the dominating trace elements in the waste fuel and ashes. Most part of high and medium volatile trace elements such as Cd, Pb, Zn, Sb and As were transferred to fly ash for pure RWW / industrial waste incineration in fluidized boilers. Overall, As and Cd seems to show increased transfer to fly ash with the increase in input content of these elements in the waste fuel, while low volatiles Cu and Cr showed decrease in transfer to fly ash with input content. Overall, Cd was partitioned mostly in the fly ash in all cases during incineration, most probably because of vaporization, condensation on fly ash particles and adsorption mechanisms. While 2/3 or more of Zn and Pb also transferred to fly ash. Low volatiles Cu and Cr stayed mainly in the bottom ash except in a few fluidized boilers operation that might be attributed to particle entrainment or turbulence. Increased chlorine in the waste fuel feed increased the trace element transfer to fly ash especially for Zn and Pb by forming their metal chlorides that are highly volatile. Sequential extraction results and risk assessment code showed that Zn and Pb in RWW, Cd in industrial waste, Pb, Cu and Cd in mixed household/industrial waste while Zn, Cd in mixed biofuel waste fly ash were posing high risk to the environment on utilization/landfilling. Further speciation results indicated that in RWW bottom ash, arsenic was mainly (around 50%) present in mobile fractions (ion exchangeable and acid soluble). While in mixed household & industrial waste bottom ash, 65% of Cu was bound to the oxidizable fraction that indicates the role of organic matter on leaching. Moreover, bottom ash from incineration of industrial waste or mixed household/industrial waste contained higher amounts (as compared to biofuel incineration bottom ashes) of trace elements such as that of Zn, Cu, Cr and Sb in labile fractions An increased input concentration of certain trace elements, such as Zn when firing pure RWW or mixed biofuel and chlorine while firing mixed waste or industrial waste, caused increased concentration in fly ash. It might also boost the deposition and corrosion problems. So it is suggested to keep input metal content and chlorine concentrations as low as possible

    • In Sweden female officers is a since long established fact. Today women make up about 38% of the prison officer work force. However, the distribution of sexes in the organisation is quite uneven between different types of prison wings and units, and at the in-group level there is an informal gendered division of labour going on. The article deals with how a gendered division of labour comes about as a result of socially established motives and notions among prison officers as well as prisoners about men’s and women’s different qualities in performing different types of work tasks. It is based on data from a minor interview study (2006), and a three-year multi-strategic research project containing qualitative case-studies and a nationwide survey (2007-2009). The article concludes that a great number of female officers contribute to a more prominent position for the rehabilitative side of prison work. However, the on-going gendered division of labour in and between wings and units is also one important factor behind sub-cultural differentiation and variations in work practices in the prison organization. A more balanced distribution of the sexes would stimulate rehabilitative work, and a more unified view of the occupational role throughout the whole organization.

  • Goal-directed arm movements in children with fetal alcohol syndrome : a kinematic approach

    • Background: Although many studies have documented deficits in general motor functioning in children with fetal alcohol syndrome (FAS), few have employed detailed measurements to explore the specific nature of such disabilities. This pilot study explores whether three-dimensional (3D) kinematic analysis may generate increased knowledge of the effect of intrauterine alcohol exposure on motor control processes by detecting atypical upper-limb movement pattern specificity in children with FAS relative to typically developing (TD) children.  Methods: Left and right arm and head movements during a sequential unimanual goal-directed precision task in a sample of children with FAS and in TD children were registered by an optoelectronic tracking system (ProReflex, Qualisys Inc.).  Results: Children with FAS demonstrated evidently poorer task performance compared with TD children. Additionally, analyses of arm movement kinematics revealed atypical spatio-temporal organization in the children with FAS. In general, they exhibited longer arm movement trajectories at both the proximal and distal level, faster velocities at the proximal level but slower at the distal level, and more segmented distal movements. Children with FAS also showed atypically augmented and fast head movements during the task performance.  Conclusions: Findings indicate neuromotor deficits and developmental delay in goaldirected arm movements because of prenatal alcohol exposure. It is suggested that 3D kinematic analysis is a valid technique for furthering the understanding of motor control processes in children with FAS/fetal alcohol spectrum disorders. A combination with relevant neuroimaging techniques in future studies would enable a more clear-cut interpretation of how atypical movement patterns relate to underlying brain abnormalities.

    • Congenital thrombocytopenic purpura (TTP) is a rare but serious condition. We present a case of a 29-year-old woman, diagnosed with this disease in adulthood. The episode that led to diagnosis was triggered by quetiapine. She presented with neurological symptoms and laboratory findings including low platelets and elevated creatinine. Interestingly, the signs of hemolysis were very subtle. Her symptoms were relieved by withdrawal of the medicine. The diagnosis was confirmed by very low ADAMTS13 activity, lack of antibodies against ADAMTS13 and the presence of a compound heterozygous ADAMTS13 mutation. Despite prophylactic plasma infusions, the patient developed a second episode of microangiopathy, leading to an extensive cerebral infarction. It is possible that even the latter episode was triggered by drugs. We suggest that the diagnosis of TTP should be considered in patients with neurological symptoms and unexplained thrombocytopenia.

  • Mechanisms of Blister Formation on Concrete Bridge Decks with Waterprooving Asphalt Pavement Systems

    • Bridge decks are commonly subjected to harsh environmental conditions that often lead to serious corrosion problems triggered by blisters under the hot mix asphalt bridge deck surfacing and secretly evolving during weather exposure until damage is often detected too late. Blisters may form under both the waterproofing dense mastic asphalt layer or under the waterproofing membrane which is often applied as additional water protection under the mastic asphalt (MA). One of the main technical issues is the formation of blisters under the membrane and asphalt-covered concrete structures caused by a complex mechanism governed by bottom-up pressure and loss of adhesion.A linear viscoelastic finite-element model was developed to simulate time-dependent blister growth in a dense mastic asphalt layer under uniformly applied pressure with and without temperature and pressure fluctuation. A finite element model was developed using ABAQUS with linear viscoelastic properties and validated with a closed form solution from first-order shear-deformation theory for thick plates. In addition, the blister test was conducted on different samples of MA in the laboratory and digital image correlation measurement technique was used to capture the three-dimensional vertical deflection of the MA over time. It was found that the blister may grow continuously under repeated loading conditions over subsequent days.With respect to blistering under waterproofing membranes, mechanical elastic modeling and experimental investigations were performed for three different types of membranes under in-plane stress state. The orthotropic mechanical behavior of a polymer modified bitumen membrane (PBM) was determined from biaxial test data. Finally, blister tests by applying controlled pressure between orthotropic PBMs and concrete plates were performed for studying the elliptical adhesive blister propagation using digital 3D image correlation. The energy calculated from elliptical blister propagation was found comparable to the adhesive fracture energy from standard peeling tests for similar types of PBMs. This indicates that the peeling test assists to evaluate and rank the adhesive properties of different types of membranes with respect to blister formation at room temperature without conducting time consuming and complicated pressurized blister propagation tests using digital 3D image correlation.
