  • Combined X-Ray Computed Tomography and X-Ray Fluorescence Drill Core Scanning for 3-D Rock and Ore Characterization : Implications for the Lovisa Stratiform Zn-Pb Deposit and Its Structural Setting, Bergslagen, Sweden
  • 2022
  • Ingår i: Economic geology and the bulletin of the Society of Economic Geologists. - : Society of Economic Geologists, Inc.. - 0361-0128 .- 1554-0774. ; 117:6, s. 1255-1273
  • Tidskriftsartikel (refereegranskat)abstract
    • We present the results of a pilot study that integrates automated drill core scanning technology based on simultaneous X-ray computed tomography (XCT) and X-ray fluorescence (XRF) analyses to provide high-spatial-resolution (<0.2 mm) information on 3-D rock textures and structures, chemical composition, and density. Testing of its applicability for mineral exploration and research was performed by scanning and analyzing 1,500 m of drill core from the Paleoproterozoic Lovisa stratiform Zn-Pb sulfide deposit, which is part of a larger mineral system also including Cu-Co and Fe-(rare earth element) mineralization, hosted by the highly strained West Bergslagen boundary zone in south-central Sweden. The obtained scanning data complements data derived from structural field mapping, drill core logs, and chemical analysis as well as from multiscale 3-D geologic modeling at Lovisa. Data integration reveals macroand mesoscopic folding of S-0/S-1 by asymmetric steeply SE-plunging F-2 folds and N-striking vertical F-3 folds. Stretching lineations, measured directly from the scanning imagery, trend parallel to F-2 fold hinges and modeled ore shoots at the nearby Hakansboda Cu-Co and Strassa and Blanka Fe deposits. The textural character of the Lovisa ore zones is revealed in 3-D by XCT-XRF scanning and highlight remobilization of Zn and Pb from primary layering into ductile and brittle structures. The downhole bulk geochemical trends seen in scanning and traditional assay data are generally comparable but with systematic variations for some elements due to currently unresolved XRF spectral overlaps (e.g., Co and Fe). The 3-D deformation pattern at Lovisa is explained by D-2 sinistral transpression along the West Bergslagen boundary zone in response to regional north-south crustal shortening at ca. 1.84-1.81 Ga. Local refolding was caused by D-3 regional east-west crustal shortening resulting in dextral transpression along the West Bergslagen boundary zone, presumably at ca. 1.80-1.76 Ga. Based on polyphase ore textures and modeled ore shoots aligned to F-2 fold hinges, we postulate that D-2 and D-3 transpressive deformation exerted both a strong control on ore remobilization and the resulting orebody geometries at Lovisa and neighboring deposits within the West Bergslagen boundary zone. We conclude that the combined XCT-XRF drill core scanning technique provides a valuable tool for 3-D ore and rock characterization, generating continuous downhole data sets, with the potential for increasing precision and efficiency in mineral exploration and mining.

  • Genome-wide association analyses for lung function and chronic obstructive pulmonary disease identify new loci and potential druggable targets.
  • 2017
  • Ingår i: Nature Genetics. - : Springer Science and Business Media LLC. - 1061-4036 .- 1546-1718. ; 49:3, s. 416-425
  • Tidskriftsartikel (refereegranskat)abstract
    • Chronic obstructive pulmonary disease (COPD) is characterized by reduced lung function and is the third leading cause of death globally. Through genome-wide association discovery in 48,943 individuals, selected from extremes of the lung function distribution in UK Biobank, and follow-up in 95,375 individuals, we increased the yield of independent signals for lung function from 54 to 97. A genetic risk score was associated with COPD susceptibility (odds ratio per 1 s.d. of the risk score (∼6 alleles) (95% confidence interval) = 1.24 (1.20-1.27), P = 5.05 × 10(-49)), and we observed a 3.7-fold difference in COPD risk between individuals in the highest and lowest genetic risk score deciles in UK Biobank. The 97 signals show enrichment in genes for development, elastic fibers and epigenetic regulation pathways. We highlight targets for drugs and compounds in development for COPD and asthma (genes in the inositol phosphate metabolism pathway and CHRM3) and describe targets for potential drug repositioning from other clinical indications.

  • Orgasm-Associated Urinary Incontinence and Sexual Life after Radical Prostatectomy.
  • 2011
  • Ingår i: The journal of sexual medicine. - : Oxford University Press (OUP). - 1743-6109 .- 1743-6095.
  • Tidskriftsartikel (refereegranskat)abstract
    • Introduction. Involuntary release of urine during sexual climax, orgasm-associated urinary incontinence, occurs frequently after radical prostatectomy. We know little about its prevalence and its effect on sexual satisfaction. Aim. To determine the prevalence of orgasm-associated incontinence after radical prostatectomy and its effect on sexual satisfaction. Methods. Consecutive series, follow-up at one point in calendar time of men having undergone radical prostatectomy (open surgery or robot-assisted laparoscopic surgery) at Karolinska University Hospital, Stockholm, Sweden, 2002-2006. Of the 1,411 eligible men, 1,288 (91%) men completed a study-specific questionnaire. Main Outcome Measure. Prevalence rate of orgasm-associated incontinence. Results. Of the 1,288 men providing information, 691 were sexually active. Altogether, 268 men reported orgasm-associated urinary incontinence, of whom 230 (86%) were otherwise continent. When comparing them with the 422 not reporting the symptom but being sexually active, we found a prevalence ratio (with 95% confidence interval) of 1.5 (1.2-1.8) for not being able to satisfy the partner, 2.1 (1.1-3.5) for avoiding sexual activity because of fear of failing, 1.5 (1.1-2.1) for low orgasmic satisfaction, and 1.4 (1.2-1.7) for having sexual intercourse infrequently. Prevalence ratios increase in prostate-cancer survivors with a higher frequency of orgasm-associated urinary incontinence. Conclusion. We found orgasm-associated urinary incontinence to occur among a fifth of prostate cancer survivors having undergone radical prostatectomy, most of whom are continent when not engaged in sexual activity. The symptom was associated with several aspects of sexual life. Nilsson AE, Carlsson S, Johansson E, Jonsson MN, Adding C, Nyberg T, Steineck G, and Wiklund NP. • GWAS Meta-Analysis of Suicide Attempt: Identification of 12 Genome-Wide Significant Loci and Implication of Genetic Risks for Specific Health Factors.
  • 2023
  • Ingår i: The American journal of psychiatry. - : American Psychiatric Association Publishing. - 1535-7228 .- 0002-953X. ; 180:10, s. 723-738
  • Tidskriftsartikel (refereegranskat)abstract
    • Suicidal behavior is heritable and is a major cause of death worldwide. Two large-scale genome-wide association studies (GWASs) recently discovered and cross-validated genome-wide significant (GWS) loci for suicide attempt (SA). The present study leveraged the genetic cohorts from both studies to conduct the largest GWAS meta-analysis of SA to date. Multi-ancestry and admixture-specific meta-analyses were conducted within groups of significant African, East Asian, and European ancestry admixtures. This study comprised 22 cohorts, including 43,871 SA cases and 915,025 ancestry-matched controls. Analytical methods across multi-ancestry and individual ancestry admixtures included inverse variance-weighted fixed-effects meta-analyses, followed by gene, gene-set, tissue-set, and drug-target enrichment, as well as summary-data-based Mendelian randomization with brain expression quantitative trait loci data, phenome-wide genetic correlation, and genetic causal proportion analyses.Multi-ancestry and European ancestry admixture GWAS meta-analyses identified 12 risk loci at p values <5×10-8. These loci were mostly intergenic and implicated DRD2, SLC6A9, FURIN, NLGN1, SOX5, PDE4B, and CACNG2. The multi-ancestry SNP-based heritability estimate of SA was 5.7% on the liability scale (SE=0.003, p=5.7×10-80). Significant brain tissue gene expression and drug set enrichment were observed. There was shared genetic variation of SA with attention deficit hyperactivity disorder, smoking, and risk tolerance after conditioning SA on both major depressive disorder and posttraumatic stress disorder. Genetic causal proportion analyses implicated shared genetic risk for specific health factors. This multi-ancestry analysis of suicide attempt identified several loci contributing to risk and establishes significant shared genetic covariation with clinical phenotypes. These findings provide insight into genetic factors associated with suicide attempt across ancestry admixture populations, in veteran and civilian populations, and in attempt versus death.

  • Meta-analysis of genome-wide association studies confirms a susceptibility locus for knee osteoarthritis on chromosome 7q22
  • 2011
  • Ingår i: Annals of the Rheumatic Diseases. - : BMJ. - 1468-2060 .- 0003-4967. ; 70:2, s. 349-355
  • Tidskriftsartikel (refereegranskat)abstract
    • Objectives Osteoarthritis (OA) is the most prevalent form of arthritis and accounts for substantial morbidity and disability, particularly in older people. It is characterised by changes in joint structure, including degeneration of the articular cartilage, and its aetiology is multifactorial with a strong postulated genetic component. Methods A meta-analysis was performed of four genome-wide association (GWA) studies of 2371 cases of knee OA and 35 909 controls in Caucasian populations. Replication of the top hits was attempted with data from 10 additional replication datasets. Results With a cumulative sample size of 6709 cases and 44 439 controls, one genome-wide significant locus was identified on chromosome 7q22 for knee OA (rs4730250, p = 9.2 x 10(-9)), thereby confirming its role as a susceptibility locus for OA. Conclusion The associated signal is located within a large (500 kb) linkage disequilibrium block that contains six genes: PRKAR2B (protein kinase, cAMP-dependent, regulatory, type II, beta), HPB1 (HMG-box transcription factor 1), COG5 (component of oligomeric golgi complex 5), GPR22 (G protein-coupled receptor 22), DUS4L (dihydrouridine synthase 4-like) and BCAP29 (B cell receptor-associated protein 29). Gene expression analyses of the (six) genes in primary cells derived from different joint tissues confirmed expression of all the genes in the joint environment.

  • Cortical thickness across the lifespan : Data from 17,075 healthy individuals aged 3-90 years
  • 2022
  • Ingår i: Human Brain Mapping. - : John Wiley & Sons. - 1065-9471 .- 1097-0193. ; 43:1, s. 431-451
  • Tidskriftsartikel (refereegranskat)abstract
    • Delineating the association of age and cortical thickness in healthy individuals is critical given the association of cortical thickness with cognition and behavior. Previous research has shown that robust estimates of the association between age and brain morphometry require large-scale studies. In response, we used cross-sectional data from 17,075 individuals aged 3-90 years from the Enhancing Neuroimaging Genetics through Meta-Analysis (ENIGMA) Consortium to infer age-related changes in cortical thickness. We used fractional polynomial (FP) regression to quantify the association between age and cortical thickness, and we computed normalized growth centiles using the parametric Lambda, Mu, and Sigma method. Interindividual variability was estimated using meta-analysis and one-way analysis of variance. For most regions, their highest cortical thickness value was observed in childhood. Age and cortical thickness showed a negative association; the slope was steeper up to the third decade of life and more gradual thereafter; notable exceptions to this general pattern were entorhinal, temporopolar, and anterior cingulate cortices. Interindividual variability was largest in temporal and frontal regions across the lifespan. Age and its FP combinations explained up to 59% variance in cortical thickness. These results may form the basis of further investigation on normative deviation in cortical thickness and its significance for behavioral and cognitive outcomes.

  • A Comparison of the Security of Windows NT and UNIX
  • 1998
  • Konferensbidrag (refereegranskat)abstract
    • This paper presents a brief comparison of two operating systems, Windows NT and UNIX. The comparison covers two different aspects. First, we compare the main security features of the two operating systems and then we make a comparison of a selection of vulnerabilities most of which we know have been used for making real intrusions. We found that Windows NT has slightly more rigorous security features than standard UNIX but the two systems display similar vulnerabilities. The conclusion is that there are no significant differences in the real level of security between these systems

  • Analysis of the Security of Windows NT
  • 1998
  • Rapport (refereegranskat)abstract
    • This paper presents an analysis of the security in Windows NT 4.0, working in both stand-alone and networking mode. The objective of the work was to find out how secure this operating system actually is. A technical overview of the system, and in particular its security features is given. The system security was analyzed and practical intrusion attempts were made in order to verify vulnerabilities or to find new ones. All vulnerabilities are described in detail and classified according to a classification scheme. A comparison to commonly known UNIX weaknesses was made. It revealed generic similarities between the two systems to a surprisingly high degree. Finally a number of recommendations are given. The paper concludes that there are ample opportunities to improve the security of Windows NT. We have reason to believe that it is probably not higher than that of UNIX
