  • In vitro phosphorylation of human complement factor C3 by protein kinase A and protein kinase C : Effects on the classical and alternative pathways

    • Complement factor C3, recently found to contain covalently bound phosphate, was phosphorylated in vitro by cyclic AMP-dependent protein kinase (protein kinase A) and Ca2+-activated, phospholipid-dependent protein kinase (protein kinase C). Both protein kinases phosphorylated the same serine residue(s) located in the C3a portion of the alpha-chain. In addition, protein kinase C phosphorylated the beta-chain to a lesser extent. Protein kinase A gave a maximal incorporation of 1 mol of phosphate/mol of C3 while that value with protein kinase C was 1.5 mol of phosphate/mol of C3. The velocity in pmol of [32P]phosphate/(min x unit kinase) was 20 times higher for protein kinase C than for protein kinase A although a 10 times lower ratio of protein kinase to C3 was used in the former case. The apparent Kmfor C3 was 2.6 µM when protein kinase C was used. The phosphorylated C3 was found to be more resistant to partial degradation by trypsin than unphosphorylated C3. It was also found that phosphorylation of C3 in the C3a portion of the alpha-chain inhibited both the classical and alternative complement activation pathways on an approximately stoichiometric basis.

  • Calpain processing of brain microtubules from the Atlantic cod, Gadus morhua.

    • Microtubules isolated from Atlantic cod (Gadus morhua) brains retained assembly competence and ultraculture, although treatment with rabbit calpain resulted in loss of MAPs. In addition, spirals and aberrant structures formed when calpain I was activated post assembly. No such effect was seen with calpain II. Soluble fractions from cod brain were found to contain proteolytic activity that could be blocked by exogenously added calpastatin. Calpain was also isolated from cod muscle tissue with 10 times less yield, compared to rabbit lung. On the basis of Ca(2+)-requirements for activation in the mM range, electrophoretic mobility, antigenicity and hydrophobicity, we conclude that the proteolytic activity was attributable to calpain II. There was no difference in effects of rabbit and cod calpain II on cod microtubule proteins, indicating that calpain is a conserved protein. Our results suggest that calpains might be involved in the Ca(2+)-dependent irreversible regulation of cod brain microtubules.

    • We postulate a new type of operator algebra with a non-abelian extension. This algebra generalizes the Kac-Moody algebra in string theory and the Mickelsson-Faddeev algebra in three dimensions to higher-dimensional extended objects (p-branes). We then construct new BRST operators, covariant derivatives and curvature tensors in the higher-dimensional generalization of loop space.

    • We study the cohomology of the Schwinger term arising in second quantization of the class of observables belonging to the restricted general linear algebra. We prove that, for all pseudodifferential operators in 3+1 dimensions of this type, the Schwinger term is equivalent to the ``twisted'' Radul cocycle, a modified version of the Radul cocycle arising in non-commutative differential geometry. In the process we also show how the ordinary Radul cocycle for any pair of pseudodifferential operators in any dimension can be written as the phase space integral of the star commutator of their symbols projected to the appropriate asymptotic component.

  • Mobile robot control with concurrent logic languages

    • Experiences from a project in which a small mobile robot is controlled by a concurrent logic language are described. The purpose of the project is to study what is required from the logic language, and to see what are the advantages of using such a language instead of more classical programming languages. It is found that real-time concepts like timers, sensors, actuators, feedback, exception handling, dynamic map construction and fully parallel communication between units can be easily accommodated in a simple framework based on a concurrent logic language. The simplicity with which this can be done is due to two factors: first, the symbolic processing ability, which enables clean handling of communication; second, the natural integration of parallel processing and synchronization into the language.
