  • Population noise exposure modelling using large scale multi-agent simulation

    • Modelling the noise exposure for a given population is essential in order to assess its potential environmental health issues and to guide authorities in their urban planning or transportation policies. This paper introduces a new method for evaluating noise exposure of large-scale populations based on MATSim, an open- source agent-based transportation simulator. A list of agents, i.e. a synthetic population, is generated using available census and travel surveys, and are used in the transport simulation. The description of the agents includes their socio-professional profile. The resulting vehicle movements are then used in NoiseModelling, an open-source CNOSSOS-EU implementation for noise emission and propagation, to compute the noise levels every time step, typically every 15mn. Agents are then followed in their daily schedule and a noise level is assigned to them every time step. The result is a 24h noise level time history for every agent. Opportunities for analysis are discussed, such as the detailed population exposure assessment and the investigation of environmental justice issues. A case study is presented and some use cases are explored. Finally, a similar coupling for air pollutants (CO2, NOx, ...) using the HBEFA emission database and the SIRANE dispersion model is under development that will enable multi-exposure assessments.

    • Volatile impurities contained in polyethyleneimine (PEI), and identified as ethyleneimine dimers and trimers, are reported. These N-based molecules show a strong reducing character, as demonstrated by the change in electrical conductivity of organic (semi) conductors exposed to the PEI vapor. The results prove that electron transfer rather than a dipole effect at the electrode interface is the origin of the work-function modification by the PEI-based layers.

  • Improved production of Taxol ® precursors in S. cerevisiae using combinatorial in silico design and metabolic engineering

    • Background: Integrated metabolic engineering approaches that combine system and synthetic biology tools enable the efficient design of microbial cell factories for synthesizing high-value products. In this study, we utilized in silico design algorithms on the yeast genome-scale model to predict genomic modifications that could enhance the production of early-step Taxol® in engineered Saccharomyces cerevisiae cells. Results: Using constraint-based reconstruction and analysis (COBRA) methods, we narrowed down the solution set of genomic modification candidates. We screened 17 genomic modifications, including nine gene deletions and eight gene overexpressions, through wet-lab studies to determine their impact on taxadiene production, the first metabolite in the Taxol® biosynthetic pathway. Under different cultivation conditions, most single genomic modifications resulted in increased taxadiene production. The strain named KM32, which contained four overexpressed genes (ILV2, TRR1, ADE13, and ECM31) involved in branched-chain amino acid biosynthesis, the thioredoxin system, de novo purine synthesis, and the pantothenate pathway, respectively, exhibited the best performance. KM32 achieved a 50% increase in taxadiene production, reaching 215 mg/L. Furthermore, KM32 produced the highest reported yields of taxa-4(20),11-dien-5α-ol (T5α-ol) at 43.65 mg/L and taxa-4(20),11-dien-5-α-yl acetate (T5αAc) at 26.2 mg/L among early-step Taxol® metabolites in S. cerevisiae. Conclusions: This study highlights the effectiveness of computational and integrated approaches in identifying promising genomic modifications that can enhance the performance of yeast cell factories. By employing in silico design algorithms and wet-lab screening, we successfully improved taxadiene production in engineered S. cerevisiae strains. The best-performing strain, KM32, achieved substantial increases in taxadiene as well as production of T5α-ol and T5αAc. These findings emphasize the importance of using systematic and integrated strategies to develop efficient yeast cell factories, providing potential implications for the industrial production of high-value isoprenoids like Taxol®.

    • We extend an automatically generated bilingual Japanese-Swedish dictionary with new translations, automatically discovered from the multi-lingual online encyclopedia Wikipedia. Over 50,000 translations, most of which are not present in the original dictionary, are generated, with very high translation quality. We analyze what types of translations can be generated by this simple method. The majority of the words are proper nouns, and other types of (usually) uninteresting translations are also generated. Not counting the less interesting words, about 15,000 new translations are still found. Checking against logs of search queries from the old dictionary shows that the new translations would significantly reduce the number of searches with no matching translation.

  • Living through the experience of sibling sexual abuse : parents' perspectives

    • Sibling sexual abuse constitutes a substantial proportion of sexual abuse committed by young people. The consequences for victims of sibling abuse have been found as extensive as the consequences of abuse from adults. Sibling sexual abuse also affects the entire family. However, little research has investigated sibling sexual abuse and its consequences from a parents' perspective. This study examines the unique experiences of parents who faced disclosure of sibling sexual abuse. Five parents were interviewed, and the texts were analysed using interpretative phenomenological analysis. Four themes were identified: Parenting - insecurity and new parental challenges; Family - the loss of the family as a unit and the family as you knew it; The couple's life - the loss of the love relationship and the fight for keeping the bond; and Adaptation - a new way forward. The findings highlight the extensive consequences for families experiencing sibling abuse and the lack of support from professionals in dealing with the crisis. Clinical implications of the findings are discussed.PRACTICE IMPACT STATEMENTSibling sexual abuse can be experienced as a major crisis for all family members. This study illuminates the lived experiences of parents in managing sibling sexual abuse, the perceived impact on them as parents, and their relationships with each other and with their children, the child who sexually harmed, as well as harmed and non-abused children. The findings further underline the need of external support for affected families.

    • Increasing diversification, urbanization, economic restructuring, and distances, as well as declining economic dependence on forestry, are changing the characteristics of forest ownership and the conditions for environmental governance. Through an interview-based case study of Swedish forestry industrial actors, this article examined the organizational and governing aspects and implications of recent shifts by exploring the strategies and marketing/governing technologies of private/industrial forestry organizations. With a focus on local implementation, this study shows that forest owners are largely constructed, and engaged, as consumers (rather than, for example, as timber suppliers) and are governed, partly at a distance, through specific forms of guidance, technologies, and knowledge to overcome the lack of social and physical presence in the design and interaction of sale. This stresses the need to understand the role, function, and power of the forestry organizations and sales processes in research on environmental and forest policy implementation on multiple levels.
