    • ICT solutions have penetrated all areas of our daily lives. With the advent of Internet as well as broadband, wireless and mobile access, increasingly many applications rely upon communication capabilities, e.g. for data downloading or content streaming. Networks act as enablers for such applications. However, in case the enabler “network” does not work as expected, it turns into a disabler, hindering the application from working correctly: it hacks or hangs. In general, the user is neither informed about this explicitly, nor is (s)he told what to do. User reactions range from posing the question “What have I done wrong” via annoyance or ignorance to churn. It has becomes clear that customers wish clarity about (network) conditions affecting service performance. In the context of mobile services, the importance of taking care of user satisfaction with service provisioning as a whole has been realized. The corresponding Quality of Experience (QoE) concept covers end-to-end network Quality of Service (QoS) parameters, factors such as coverage or support, and subjective factors such as expectations of and experiences with the service. Quality feedback plays an important role for keeping QoS and QoE under control. The tutorial presents and discusses a couple of theses as well asrelated work and approaches to each of them: Thesis 1: Users do have – sometimes unconscious – expectations regarding ICT performance. Thesis 2: There is a need for more explicit feedback to make the user feel more confident. Thesis 3: The user needs to be relieved from decisions based on incomplete feedback. The tutorial concludes with a wish-list addressing low complexity, pro-active and reactive feedback; the Internet community; and, finally, researchers.     • This review considers current advances in tools to investigate the functional biology of Giardia, it's coding and non-coding genes, features and cellular and molecular biology. We consider major gaps in current knowledge of the parasite and discuss the present state-of-the-art in its in vivo and in vitro cultivation. Advances in in silico tools, including for the modelling non-coding RNAs and genomic elements, as well as detailed exploration of coding genes through inferred homology to model organisms, have provided significant, primary level insight. Improved methods to model the three-dimensional structure of proteins offer new insights into their function, and binding interactions with ligands, other proteins or precursor drugs, and offer substantial opportunities to prioritise proteins for further study and experimentation. These approaches can be supplemented by the growing and highly accessible arsenal of systems-based methods now being applied to Giardia, led by genomic, transcriptomic and proteomic methods, but rapidly incorporating advanced tools for detection of real-time transcription, evaluation of chromatin states and direct measurement of macromolecular complexes. Methods to directly interrogate and perturb gene function have made major leaps in recent years, with CRISPr-interference now available. These approaches, coupled with protein over-expression, fluorescent labelling and in vitro and in vivo imaging, are set to revolutionize the field and herald an exciting time during which the field may finally realise Giardia's long proposed potential as a model parasite and eukaryote.

  • Responsivity and noise equivalent power of a single cold-electron bolometer

    • We have developed a single-pixel capacitively coupled Cold-Electron Bolometer (CEB) and characterized it in the current-biased regime. The most attractive feature of the CEB is effective electron self-cooling of the absorber, which leads to a lower bolometer noise and higher dynamic range. The bolometer responsivity was measured by determining the voltage response to an applied power through the absorber from a heating current, modulated at frequencies from 35 Hz to 2 kHz. The optimum responsivity of 1.5 × 10 V/W was measured at a modulation frequency of 35 Hz. The noise equivalent power (NEP) was subsequently obtained from the estimated bolometer noise voltage with respect to the measured bolometer responsivity. The NEP of better 2 × 10 W/Hz was obtained for modulation frequencies greater than 100 Hz. The background power and the bolometer time constant were also estimated from the experimental results. The photon-noise-limited operation of CEB will dominate for a signal power of 10 fW and higher at frequency 80 GHz and higher. 10 −18 1/2

  • Electrical impedance myography combined with quantitative assessment techniques in paretic muscle of stroke survivors : Insights and challenges

    • Aging is a non-modifiable risk factor for stroke and the global burden of stroke is continuing to increase due to the aging society. Muscle dysfunction, common sequela of stroke, has long been of research interests. Therefore, how to accurately assess muscle function is particularly important. Electrical impedance myography (EIM) has proven to be feasible to assess muscle impairment in patients with stroke in terms of micro structures, such as muscle membrane integrity, extracellular and intracellular fluids. However, EIM alone is not sufficient to assess muscle function comprehensively given the complex contributors to paretic muscle after an insult. This article discusses the potential to combine EIM and other common quantitative methods as ways to improve the assessment of muscle function in stroke survivors. Clinically, these combined assessments provide not only a distinct advantage for greater accuracy of muscle assessment through cross-validation, but also the physiological explanation on muscle dysfunction at the micro level. Different combinations of assessments are discussed with insights for different purposes. The assessments of morphological, mechanical and contractile properties combined with EIM are focused since changes in muscle structures, tone and strength directly reflect the muscle function of stroke survivors. With advances in computational technology, finite element model and machine learning model that incorporate multi-modal evaluation parameters to enable the establishment of predictive or diagnostic model will be the next step forward to assess muscle function for individual with stroke.

    • Knowledge management technology is now ubiquitous in many businesses, yet it is much less common among local authorities. Following a "brainstorm meeting" in Lyon on knowledge management for regional planning and policymaking, a proposal was developed to set up a research agendum for future development of this topic. The main difficulty we identified is that regional knowledge encompasses many topics beyond business issues, especially those related to long-term sustainability. The goal of this paper is twofold, first to clarify the proper definitions and boundaries of regional knowledge and technology, and second, to identify the ways of using this technology effectively for governance and decision-making.

    • To examine whether free fatty acid receptor 4 (FFAR4) activation can protect against choroidal neovascularization (CNV), which is a common cause of blindness, and to elucidate the mechanism underlying the inhibition, we used the mouse model of laser-induced CNV to mimic angiogenic aspects of age-related macular degeneration (AMD). Laser-induced CNV was compared between groups treated with an FFAR4 agonist or vehicle, and between FFAR4 wild-type (Ffar4+/+) and knock out (Ffar4−/−) mice on a C57BL/6J/6N background. The ex vivo choroid-sprouting assay, including primary retinal pigment epithelium (RPE) and choroid, without retina was used to investigate whether FFAR4 affects choroidal angiogenesis. Western blotting for pNF-ĸB/NF-ĸB and qRT-PCR for Il-6, Il-1β, Tnf-α, Vegf, and Nf-ĸb were used to examine the influence of FFAR4 on inflammation, known to influence CNV. RPE isolated from Ffar4+/+ and Ffar4−/− mice were used to assess RPE contribution to inflammation. The FFAR4 agonist suppressed laser-induced CNV in C57BL/6J mice, and CNV increased in Ffar4−/− compared to Ffar4+/+ mice. We showed that the FFAR4 agonist acted through the FFAR4 receptor. The FFAR4 agonist suppressed mRNA expression of inflammation markers (Il-6, Il-1β) via the NF-ĸB pathway in the retina, choroid, RPE complex. The FFAR4 agonist suppressed neovascularization in the choroid-sprouting ex vivo assay and FFAR4 deficiency exacerbated sprouting. Inflammation markers were increased in primary RPE cells of Ffar4−/− mice compared with Ffar4+/+ RPE. In this mouse model, the FFAR4 agonist suppressed CNV, suggesting FFAR4 to be a new molecular target to reduce pathological angiogenesis in CNV. © 2020, Springer Nature B.V.
