    • Soil microbial community structure is determined by environmental conditions and influenced by other factors, such as the intensity of the land use management. Studies addressing the effect of environmental factors and management on grassland soil microbial communities at the continental scale are missing, and the wide range of ecosystem services provided by these ecosystems are thus also wanting. To address this knowledge gap, this study presents data on grassland soil microbial communities along a pan-European agro-ecological gradient. The transect included five geographical locations (Sweden, Germany, Switzerland, Portugal mainland, Portugal Azores). At each location, soils were collected in two regions characterized by favourable and less favourable conditions for plant growth. In each of these ten regions, grasslands along a gradient of management intensity were selected, i.e. grassland under intensive, less intensive and extensive management. Phospholipid fatty acid analysis (PLFA) was used to characterize the microbial community structure (PLFA pattern) in relation to climatic and soil properties. Over the whole geographical range, the environmental properties determined the soil microbial community structure. In Sweden and Switzerland, the regional growth conditions had the strongest influence on the soil microbial communities, while in Germany, Portugal mainland and Azores the management intensity was more important. Splitting up this whole community response into individual groups reveals that, in general, saprotrophic fungal biomarkers were highest in extensively managed grasslands while bacterial biomarkers differed mainly between the regions. We conclude that at the transect level, climate and soil properties were the most important factors influencing soil bacterial community structure, while soil fungal groups were more responsive to grassland management intensity. Overall agricultural sustainability could benefit from informed soil health promoting management practices, and this study contributes to such knowledge, showing the importance of management for the soil microbial biomass and community structure.

  • A century of trends in adult human height

    • Being taller is associated with enhanced longevity, and higher education and earnings. We reanalysed 1472 population-based studies, with measurement of height on more than 18.6 million participants to estimate mean height for people born between 1896 and 1996 in 200 countries. The largest gain in adult height over the past century has occurred in South Korean women and Iranian men, who became 20.2 cm (95% credible interval 17.522.7) and 16.5 cm (13.319.7) taller, respectively. In contrast, there was little change in adult height in some sub-Saharan African countries and in South Asia over the century of analysis. The tallest people over these 100 years are men born in the Netherlands in the last quarter of 20th century, whose average heights surpassed 182.5 cm, and the shortest were women born in Guatemala in 1896 (140.3 cm; 135.8144.8). The height differential between the tallest and shortest populations was 19-20 cm a century ago, and has remained the same for women and increased for men a century later despite substantial changes in the ranking of countries.

  • A Genetic Mouse Model of Parkinson’s Disease Shows Involuntary Movements and Increased Postsynaptic Sensitivity to Apomorphine

    • Alpha-synuclein (SNCA) protein aggregation plays a causal role in Parkinson's disease (PD). The SNCA protein modulates neurotransmission via the SNAP receptor (SNARE) complex assembly and presynaptic vesicle trafficking. The striatal presynaptic dopamine deficit is alleviated by treatment with levodopa (L-DOPA), but postsynaptic plastic changes induced by this treatment lead to a development of involuntary movements (dyskinesia). While this process is currently modeled in rodents harboring neurotoxin-induced lesions of the nigrostriatal pathway, we have here explored the postsynaptic supersensitivity of dopamine receptor-mediated signaling in a genetic mouse model of early PD. To this end, we used mice with prion promoter-driven overexpression of A53T-SNCA in the nigrostriatal and corticostriatal projections. At a symptomatic age (18 months), mice were challenged with apomorphine (5 mg/kg s.c.) and examined using both behavioral and molecular assays. After the administration of apomorphine, A53T-transgenic mice showed more severe stereotypic and dystonic movements in comparison with wild-type controls. Molecular markers of extracellular signal-regulated kinase 1 and 2 (ERK1/2) phosphorylation and dephosphorylation, and Fos messenger RNA (mRNA), were examined in striatal tissue at 30 and 100 min after apomorphine injection. At 30 min, wild-type and transgenic mice showed a similar induction of phosphorylated ERK1/2, Dusp1, and Dusp6 mRNA (two MAPK phosphatases). At the same time point, Fos mRNA was induced more strongly in mutant mice than in wild-type controls. At 100 min after apomorphine treatment, the induction of both Fos, Dusp1, and Dusp6 mRNA was significantly larger in mutant mice than wild-type controls. At this time point, apomorphine caused a reduction in phospho-ERK1/2 levels specifically in the transgenic mice. Our results document for the first time a disturbance of ERK1/2 signaling regulation associated with apomorphine-induced involuntary movements in a genetic mouse model of synucleinopathy. This mouse model will be useful to identify novel therapeutic targets that can counteract abnormal dopamine-dependent striatal plasticity during both prodromal and manifest stages of PD.

  • Alternativa skötselmetoder för ängs- och betesmarker och användning av skördat växtmaterial

    • Ängs- och betesmarker utgör unika livsmiljöer och är mycket värdefulla för den biologiska mångfalden. I dagsläget är utmaningarna stora i Sverige för att kunna hävda ängs- och betesmarkerna med hög kvalitet, till stor del beroende på det minskande antalet betesdjur och minskat behov av fodermarker. Denna rapport sammanställer aktuell kunskap kring möjliga nya skötselmetoder för våra ängs- och betesmarker samt hur skördat material kan användas som en resurs i samhället. Sammanställningen bygger på vetenskaplig och populärvetenskaplig litteratur, författarnas erfarenheter inom området, samt information om pågående teknikutveckling och praktisk tillämpning. Utmaningarna för att utveckla hävden av ängs- och betesmarker består i huvudsak i att hitta rationella system, dels för hantering av betesdjur och dels för effektiv insamling och användning av biomassa. Vi har identifierat följande skötselmetoder och användningsområden som särskilt intressanta för vidare analyser: 1. Flexibel hävd genom till exempel extensivt bete, varierad hävd genom olika skötselmetoder olika år (hävdföljd), kombination av olika metoder genom till exempel maskinell skörd och bränning eller slåtter i kombination med bete. 2. Maskinanpassad hävd av skogsbeten och lövängar – kombinationen av ängsväxter och träd för såväl biologisk mångfald som kolinlagring och produktion av biomassa (både gräs och träd). 3. Småskalig och situationsanpassad teknik – slåtter och borttransport av biomassa från små och hinderrika fält genom en kombination av motormanuella och rationella lösningar. Detta kräver viss metodutveckling och logistik lämpad för avslagning, uppsamling och transport. 4. Samordning och utveckling av avsättning för biomassan för bioenergiändamål. Användning av ängsbiomassa som biogassubstrat har särskilt stor miljönytta och möjliggör effektivt resursutnyttjande genom att man både utvinner energi och en växtnäringsrik rötrest (biogödsel) samtidigt som markerna hävdas. Flera nya studier visar på små eller inga skillnader när slåtter genomförs med skärande eller klippande redskap (lie och knivslåtterbalk) jämfört med roterande redskap (rotorslåttermaskiner med vassa knivar eller röjsnöre). Denna kunskap öppnar upp för maskinell skötsel av slåtterängar som annars hotas av utebliven hävd.

    • Detta faktablad utgör slutrapport för projektet "Användning av outnyttjade gräsmarker för biodiversitet och bioenergi - nätverk och fältförsök", som har genomförts under 2011-2014. Projektet har fört samman många aktörer med intresse för jordbruk, naturvård och bioenergi, och visat att nyetablering och extensiv hävd av artrika vallar kan generera värdefulla synergieffekter mellan minskad klimatpåverkan, minskad övergödning och ett rikt odlingslandskap.

    • Transfer of fixed N from legumes to non-legume reference plants may alter the N-15 signature of the reference plant as compared to the soil N available to the legume. This study investigates how N transfer influences the result of N-15-based N-2 fixation measurements.We labelled either legumes or non-legumes with N-15 and performed detailed analyses of N-15 enrichment in mixed plant communities in the field. The results were used in a conceptual model comparing how different N transfer scenarios influenced the N-15 signatures of legumes and reference plants, and how the resulting N-2 fixation estimate was influenced by using reference plants in pure stand or in mixture with the legume.Based on isotopic signatures, N transfer was detected in all directions: from legume to legume, from legume to non-legume, from non-legume to legume, from non-legume to non-legume. In the scenario of multidirectional N transfer, N-2 fixation was overestimated by using a reference plant in pure stand.Fixed N transferred to neighbouring reference plants modifies the N-15 signature of the soil N available both to the reference plant and the N-2-fixing legume. This provides strong support for using reference plants growing in mixture with the legumes for reliable quantifications of N-2 fixation.

  • Host-specific competitiveness to form nodules in Rhizobium leguminosarum symbiovar viciae

    • Fabeae legumes such as pea and faba bean form symbiotic nodules with a large diversity of soil Rhizobium leguminosarum symbiovar viciae (Rlv) bacteria. However, bacteria competitive to form root nodules (CFN) are generally not the most efficient to fix dinitrogen, resulting in a decrease in legume crop yields. Here, we investigate differential selection by host plants on the diversity of Rlv. A large collection of Rlv was collected by nodule trapping with pea and faba bean from soils at five European sites. Representative genomes were sequenced. In parallel, diversity and abundance of Rlv were estimated directly in these soils using metabarcoding. The CFN of isolates was measured with both legume hosts. Pea/faba bean CFN were associated to Rlv genomic regions. Variations of bacterial pea and/or faba bean CFN explained the differential abundance of Rlv genotypes in pea and faba bean nodules. No evidence was found for genetic association between CFN and variations in the core genome, but variations in specific regions of the nod locus, as well as in other plasmid loci, were associated with differences in CFN. These findings shed light on the genetic control of CFN in Rlv and emphasise the importance of host plants in controlling Rhizobium diversity.
