  • Achievements and experiences from science–policy interaction in the field of air pollution : Synthesising 20 years of research and outreach,thinking about future needs

    • For 20 years, the Swedish Environmental Protection Agency together with the MISTRA research foundation have funded five air pollution research programmes with focus on producing knowledge that supports policy and emission control in national and international arenas. The research has been multidisciplinary and has included research on emissions, atmospheric transport and transformation processes, human health effects, ecosystem effects, and emission control strategies. Research has also been conducted on the interaction between air pollution and climate change.Over these years, the link between the research programmes and the development of emission control strategies and policies in Sweden, the EU, and the UNECE Air Convention has been of high importance. This report presents how the research programmes have created societal benefits through support for the development of air pollution policies and emission control measures. The report also identifies future research needs to ensure continued progress towards even better air quality for future generations.

  • Blood and cerebrospinal fluid neurofilament light differentially detect neurodegeneration in early Alzheimer's disease

    • Cerebrospinal fluid (CSF) neurofilament light (NfL) concentration has reproducibly been shown to reflect neurodegeneration in brain disorders, including Alzheimer's disease (AD). NfL concentration in blood correlates with the corresponding CSF levels, but few studies have directly compared the reliability of these 2 markers in sporadic AD. Herein, we measured plasma and CSF concentrations of NfL in 478 cognitively unimpaired (CU) subjects, 227 patients with mild cognitive impairment, and 113 patients with AD dementia. We found that the concentration of NfL in CSF, but not in plasma, was increased in response to Aβ pathology in CU subjects. Both CSF and plasma NfL concentrations were increased in patients with mild cognitive impairment and AD dementia. Furthermore, only NfL in CSF was associated with reduced white matter microstructure in CU subjects. Finally, in a transgenic mouse model of AD, CSF NfL increased before serum NfL in response to the development of Aβ pathology. In conclusion, NfL in CSF may be a more reliable biomarker of neurodegeneration than NfL in blood in preclinical sporadic AD.

  • Mineral paragenesis and sulphide trace element distribution in the metamorphosed Lovisa Zn-Pb deposit, Bergslagen (Sweden), as revealed by 3D X-ray tomography, ore petrography and LA-ICP-MS analysis

    • This study encompasses the ore mineralogy, textures and sulphide trace element chemistry of the Palaeoproterozoic Lovisa stratiform Zn-Pb deposit and the stratigraphically underlying Lovisa Fe Formation in the Bergslagen ore province (Sweden). We investigate the relative timing of formation and subsequent modifications of its ores in relation to the c. 1.87-1.80 Ga Svecokarelian orogeny. The Lovisa Zn-Pb deposit consists of several different ore types. The massive sphalerite-galena ore is distinctly deformed, exhibiting a multiple-scale "ball ore" texture with rounded silicate clasts within a deformed, fine-grained sulphide matrix. Underlying the massive ore is a locally folded, sphalerite-rich laminated ore, interpreted to represent a metamorphosed relict primary lamination. Several generations of sphalerite-galena fracture fillings and veins occur adjacent to the main ore zones and they cross-cut early ductile structures and metamorphic features. The trace element signatures of the sphalerite-galena infillings generally mimic those of the two main ore zones, thus supporting an origin by localised remobilisation of the primary sulphide ore and demonstrating limited trace element redistribution during this process. In contrast, discrete sulphosalt-rich fracture fillings cross-cutting earlier galena-chalcopyriterich fracture fillings and veinlets in the Lovisa Fe Formation suggest a significant but still relatively localised redistribution of metals. Trace element mapping of sulphides from the Lovisa Zn-Pb deposit reveals that inclusion-free overgrowths on pyrite crystals are locally Co-enriched compared to the cores, which resulted from the redistribution of Co during late metamorphic processes. Combined textural and geochemical evidence suggest that the originally syngenetic exhalative sulphide ore at Lovisa was locally strongly affected by polyphase deformation and remobilisation. This was initiated during the first stage of amphibolite facies grade regional metamorphism and deformation (D1, c. 1.87-1.85 Ga) but is mostly evident from the later stages (D2) and the evolution to retrograde and brittle conditions (c. 1.83-1.80 Ga and later).

  • Mellangrödor i växtföljden - för kolinlagring och effektivt kväveutnyttjande

    • Rapporten beskriver mellangrödors funktion för kolinlagring i jordbruksmark i relation till andra funktioner och miljörisker. Den består av en kunskapsgenomgång, en inventering av data från fältförsök samt en sammanfattning av odlares erfarenheter vid en workshop. Vår definition av mellangröda är: En gröda som har sin huvudsakliga tillväxt mellan två huvudgrödor, och där syftet med att odla mellangrödan kan variera.Mellangrödan ger kolinlagring, särskilt rötternaKolinlagringen av en mellangröda har uppskattats till ca 320 kg kol i medeltal per ha och år, men variationen är stor. Vi sammanställde resultat från fältförsök med mellangrödor under 30 år. För insådda gräsfånggrödor skiljer sig biomassaproduktionen i medeltal inte så mycket åt mellan olika regioner, medan variationen är större för eftersådda mellangrödor. Det saknas kunskap för att kunna ge konkreta råd om hur artval och jordbearbetningsstrategier m.m. för mellangröda kan användas för att optimera stabiliseringsprocesserna för kol i marken. Mellangrödans rötter bidrar med ca 2,5 gånger större inlagring än den ovanjordiska grönmassan. Mätningar vi funnit visar en stor variation i viktförhållandet mellan skott och rot, som beror på arters variation, men också på osäkerheter i provtagning och metoder.Utmaningar för odlarnaBåde försök och odlares erfarenheter vittnar om utmaningar med sådd av mellangrödor efter skörd, särskilt på jordar med hög lerhalt. Det handlar om att hinna så direkt efter huvudgrödans skörd för att lyckas med etableringen och för att utnyttja hösten för tillväxt. Att ge sådd av mellangrödan lika hög prioritet som en huvudgröda var ett råd som erfarna odlare poängterade vid en workshop. Det finns olika tankesätt och strategier kring val av arter, beroende på syftet med mellangrödan. Genom artblandningar kan man också kombinera funktioner. Det finns inga tydliga råd för arter som särskilt främjar kolinlagring utan det viktigaste är stor tillväxt, hög kol-kväve-kvot (C/N-kvot), stor rotbiomassa samt baljväxtinslag. Hushållning med kvävet En nordisk sammanställning visade att mellangrödor i medeltal minskade kväveläckaget med drygt 40 %. Tidpunkt för nedbrukning eller kemisk avdödning är metoder för att styra återmineraliseringen och kväveverkan i nästa gröda. För frostkänsliga mellangrödor med låg C/N-kvot finns en risk att kvävet frigörs så snabbt att det hinner utlakas innan våren. Under sådana förhållanden ökar också risken för lustgasemissioner. Mer kunskap behövs om hur kväveomsättning påverkas och hur lustgasemissioner kan minimeras, för att kunna anpassa mellangrödesystemen efter olika odlingskrav och klimatets förändring.

  • Tropical and Boreal Forest – Atmosphere Interactions : A Review

    • This review presents how the boreal and the tropical forests affect the atmosphere, its chemical composition, its function, and further how that affects the climate and, in return, the ecosystems through feedback processes. Observations from key tower sites standing out due to their long-term comprehensive observations: The Amazon Tall Tower Observatory in Central Amazonia, the Zotino Tall Tower Observatory in Siberia, and the Station to Measure Ecosystem-Atmosphere Relations at Hyytiäla in Finland. The review is complemented by short-term observations from networks and large experiments.The review discusses atmospheric chemistry observations, aerosol formation and processing, physiochemical aerosol, and cloud condensation nuclei properties and finds surprising similarities and important differences in the two ecosystems. The aerosol concentrations and chemistry are similar, particularly concerning the main chemical components, both dominated by an organic fraction, while the boreal ecosystem has generally higher concentrations of inorganics, due to higher influence of long-range transported air pollution. The emissions of biogenic volatile organic compounds are dominated by isoprene and monoterpene in the tropical and boreal regions, respectively, being the main precursors of the organic aerosol fraction.Observations and modeling studies show that climate change and deforestation affect the ecosystems such that the carbon and hydrological cycles in Amazonia are changing to carbon neutrality and affect precipitation downwind. In Africa, the tropical forests are so far maintaining their carbon sink.It is urgent to better understand the interaction between these major ecosystems, the atmosphere, and climate, which calls for more observation sites, providing long-term data on water, carbon, and other biogeochemical cycles. This is essential in finding a sustainable balance between forest preservation and reforestation versus a potential increase in food production and biofuels, which are critical in maintaining ecosystem services and global climate stability. Reducing global warming and deforestation is vital for tropical forests.

  • Comparing a pre-defined versus deep learning approach for extracting brain atrophy patterns to predict cognitive decline due to Alzheimer’s disease in patients with mild cognitive symptoms

    • Background: Predicting future Alzheimer’s disease (AD)-related cognitive decline among individuals with subjective cognitive decline (SCD) or mild cognitive impairment (MCI) is an important task for healthcare. Structural brain imaging as measured by magnetic resonance imaging (MRI) could potentially contribute when making such predictions. It is unclear if the predictive performance of MRI can be improved using entire brain images in deep learning (DL) models compared to using pre-defined brain regions. Methods: A cohort of 332 individuals with SCD/MCI were included from the Swedish BioFINDER-1 study. The goal was to predict longitudinal SCD/MCI-to-AD dementia progression and change in Mini-Mental State Examination (MMSE) over four years. Four models were evaluated using different predictors: (1) clinical data only, including demographics, cognitive tests and APOE ε4 status, (2) clinical data plus hippocampal volume, (3) clinical data plus all regional MRI gray matter volumes (N = 68) extracted using FreeSurfer software, (4) a DL model trained using multi-task learning with MRI images, Jacobian determinant images and baseline cognition as input. A double cross-validation scheme, with five test folds and for each of those ten validation folds, was used. External evaluation was performed on part of the ADNI dataset, including 108 patients. Mann-Whitney U-test was used to determine statistically significant differences in performance, with p-values less than 0.05 considered significant. Results: In the BioFINDER cohort, 109 patients (33%) progressed to AD dementia. The performance of the clinical data model for prediction of progression to AD dementia was area under the curve (AUC) = 0.85 and four-year cognitive decline was R2 = 0.14. The performance was improved for both outcomes when adding hippocampal volume (AUC = 0.86, R2 = 0.16). Adding FreeSurfer brain regions improved prediction of four-year cognitive decline but not progression to AD (AUC = 0.83, R2 = 0.17), while the DL model worsened the performance for both outcomes (AUC = 0.84, R2 = 0.08). A sensitivity analysis showed that the Jacobian determinant image was more informative than the MRI image, but that performance was maximized when both were included. In the external evaluation cohort from ADNI, 23 patients (21%) progressed to AD dementia. The results for predicted progression to AD dementia were similar to the results for the BioFINDER test data, while the performance for the cognitive decline was deteriorated. Conclusions: The DL model did not significantly improve the prediction of clinical disease progression in AD, compared to regression models with a single pre-defined brain region.

    • Increased cerebrospinal fluid neurofilament light (NfL) is a recognized biomarker for neurodegeneration that can also be assessed in blood. Here, we investigate plasma NfL as a marker of neurodegeneration in 13 neurodegenerative disorders, Down syndrome, depression and cognitively unimpaired controls from two multicenter cohorts: King's College London (n = 805) and the Swedish BioFINDER study (n = 1,464). Plasma NfL was significantly increased in all cortical neurodegenerative disorders, amyotrophic lateral sclerosis and atypical parkinsonian disorders. We demonstrate that plasma NfL is clinically useful in identifying atypical parkinsonian disorders in patients with parkinsonism, dementia in individuals with Down syndrome, dementia among psychiatric disorders, and frontotemporal dementia in patients with cognitive impairment. Data-driven cut-offs highlighted the fundamental importance of age-related clinical cut-offs for disorders with a younger age of onset. Finally, plasma NfL performs best when applied to indicate no underlying neurodegeneration, with low false positives, in all age-related cut-offs.

    • Blood biomarkers indicative of Alzheimer's disease (AD) pathology are altered in both preclinical and symptomatic stages of the disease. Distinctive biomarkers may be optimal for the identification of AD pathology or monitoring of disease progression. Blood biomarkers that correlate with changes in cognition and atrophy during the course of the disease could be used in clinical trials to identify successful interventions and thereby accelerate the development of efficient therapies. When disease-modifying treatments become approved for use, efficient blood-based biomarkers might also inform on treatment implementation and management in clinical practice. In the BioFINDER-1 cohort, plasma phosphorylated (p)-tau231 and amyloid-β42/40 ratio were more changed at lower thresholds of amyloid pathology. Longitudinally, however, only p-tau217 demonstrated marked amyloid-dependent changes over 4-6years in both preclinical and symptomatic stages of the disease, with no such changes observed in p-tau231, p-tau181, amyloid-β42/40, glial acidic fibrillary protein or neurofilament light. Only longitudinal increases of p-tau217 were also associated with clinical deterioration and brain atrophy in preclinical AD. The selective longitudinal increase of p-tau217 and its associations with cognitive decline and atrophy was confirmed in an independent cohort (Wisconsin Registry for Alzheimer's Prevention). These findings support the differential association of plasma biomarkers with disease development and strongly highlight p-tau217 as a surrogate marker of disease progression in preclinical and prodromal AD, with impact for the development of new disease-modifying treatments.

  • Islet Gene View-a tool to facilitate islet research

    • Characterization of gene expression in pancreatic islets and its alteration in type 2 diabetes (T2D) are vital in understanding islet function and T2D pathogenesis. We leveraged RNA sequencing and genome-wide genotyping in islets from 188 donors to create the Islet Gene View (IGW) platform to make this information easily accessible to the scientific community. Expression data were related to islet phenotypes, diabetes status, other islet-expressed genes, islet hormone-encoding genes and for expression in insulin target tissues. The IGW web application produces output graphs for a particular gene of interest. In IGW, 284 differentially expressed genes (DEGs) were identified in T2D donor islets compared with controls. Forty percent of DEGs showed cell-type enrichment and a large proportion significantly co-expressed with islet hormone-encoding genes; glucagon (GCG, 56%), amylin (IAPP, 52%), insulin (INS, 44%), and somatostatin (SST, 24%). Inhibition of two DEGs, UNC5D and SERPINE2, impaired glucose-stimulated insulin secretion and impacted cell survival in a human beta-cell model. The exploratory use of IGW could help designing more comprehensive functional follow-up studies and serve to identify therapeutic targets in T2D.

    • It is unknown how neuroinflammation may feature in the etiology of Alzheimer's disease (AD). We profiled acute phase response (APR) proteins (α1-antitrypsin, α1-antichymotrypsin, ceruloplasmin, complement C3, ferritin, α-fibrinogen, β-fibrinogen, γ-fibrinogen, haptoglobin, hemopexin) in CSF of 1291 subjects along the clinical and biomarker spectrum of AD to investigate the association between inflammatory changes, disease outcomes, and demographic variables. Subjects were stratified by Aβ42/t-tau as well as the following clinical diagnoses: cognitively normal (CN); subjective cognitive decline (SCD); mild cognitive impairment (MCI); and AD dementia. In separate multiple regressions (adjusting for diagnosis, age, sex, APOE-ε4) of each APR protein and a composite of all APR proteins, CSF Aβ42/t-tau status was associated with elevated ferritin, but not any other APR protein in CN and SCD subjects. Rather, the APR was elevated along with symptomatic progression (CN < SCD < MCI < AD), and this was elevation was mediated by CSF p-tau181. APOE ε4 status did not affect levels of any APR proteins in CSF, while these were elevated in males and with increased age. The performance of the APR in predicting clinical diagnosis was influenced by APOE ε4 status, sex, and age. These data provide new insight into inflammatory changes in AD and how this intersects with pathology changes and patient demographics.
