  • Assignment of gene for coagulation factor V to chromosome 1 in man and to chromosome 13 in rat

    • Two different factor V cDNA fragments were used as hydridization probes in the chromosomal assignment of the human and rat factor V genes. A 1.6-kb EcoRI fragment was used as a hybridization probe to analyze a panel of human-rodent somatic cell hybrids. Cosegregation of factor V specific DNA restriction fragments with human chromosome 1 was observed. In addition, a panel of rat-mouse somatic cell hybrids was analyzed with another human factor V cDNA probe to localize the gene for rat coagulation factor V. In the rat, the gene for coagulation factor V was found to be located in chromosome 13. This is the first gene in the rat to be localized to chromosome 13.

  • Novel polyfucosylated N-linked glycopeptides with blood group A, H, X, and Y determinants from human small intestinal epithelial cells.

    • A novel type of N-linked glycopeptides representing a major part of the glycans in human small intestinal epithelial cells from blood group A and O individuals were isolated by gel filtrations and affinity chromatography on concanavalin A-Sepharose and Bandeiraea simplicifolia lectin I-Sepharose. Sugar composition, methylation analysis, 1H NMR spectroscopy of the underivatized glycopeptides and FAB-mass spectrometry and electron impact-mass spectrometry of the permethylated glycopeptides indicated a tri- and tetra-antennary structure containing an intersecting N-acetylglucosamine and an alpha (1----6)-linked fucose residue in the core unit for the majority of the glycans. In contrast to most glycopeptides of other sources, the intestinal glycopeptides were devoid of sialic acid, but contained 6-7 residues of fucose. The outer branches contained the following structures: Fuc alpha 1-2Gal beta 1-3GleNAc beta 1- (H type 1) Fuc alpha 1-2Gal beta 1-4GleNAc beta 1- (H type 2) Gal beta 1-4 (Fuc alpha 1-3)GlcNAc beta 1- (X) Fuc alpha 1-2Gal beta 1-4(Fuc alpha 1-3)GleNAc beta 1- (Y) GalNAc alpha 1-3(Fuc alpha 1-2)Gal beta 1-3GleNAc beta 1- (A type 1) GalNAc alpha 1-3(Fuc alpha 1-2)Gal beta 1-4GleNAc beta 1- (monofucosyl A type 2) GalNAc alpha 1-3(Fuc alpha 1-2)Gal beta 1-4 (Fuc alpha 1-3)GlcNAc beta 1- (trifucosyl A type 2) The blood group determinant structures were mainly of type 2, whereas glycolipids from the same cells contained mainly type 1 determinants. The polyfucosylated glycans represent a novel type of blood group active glycopeptides. The unique properties of the small intestinal glycopeptides as compared with glycopeptides of other tissue sources may be correlated with the specialized functional properties of the small intestinal epithelial cells.

  • Infection with delta agent in Sweden: introduction of a new hepatitis agent

    • To investigate the epidemiology of infection with delta (delta) agent in a Swedish city, 181 chronic carriers of hepatitis B surface antigen (HBsAg) and 599 patients with acute, self-limited hepatitis B were analyzed for delta antigen and antibody to delta antigen (anti-delta). The study covered the period from 1970 to 1981. The delta agent was found to have been introduced to this population in 1973. Markers of infection with delta agent were almost exclusively found in intravenous drug addicts and their close contacts. The proportion of drug addicts who were chronic HBsAg carriers with anti-delta increased with time and reached 72% in 1979-1981. An episode of acute hepatitis was frequently seen in connection with seroconversion to anti-delta. Among the domestic cases of acute, self-limited hepatitis, no simultaneous infections with hepatitis B virus and delta agent were found before 1975. From 1975 to 1980, between 18% and 44% of the drug addicts with acute hepatitis B were also infected with delta agent.

  • Evidence indicating trophic importance of IGF-I in regenerating peripheral nerves

    • The mechanisms influencing regeneration of peripheral nerves are incompletely known, but growth factors are supposed to play a key role. In the present study, we demonstrate, with the aid of immunohistochemical methods, that somatomedin C (Sm-C/insulin-like growth factor I/IGF-I) rapidly increased from low to high concentrations, reaching peak values in 2 weeks, in regenerating sciatic nerves of adult rats. In addition, IGF-I was demonstrated extracellularly, never observed in the control nerves. Reactive Schwann cells appeared to be the major source for IGF-synthesis. Higher concentrations were seen in tubulated nerves as compared to sutured ones. It is proposed that IGF-I exerts important growth supporting effects on regenerating peripheral nerves.

    • The utilization of multivariate statistical techniques is discussed with emphasis on the rather new method SIMCA, when applied to multielemental data. The procedures of scaling and normalizing are described. The data base used is from a project studying long range aerosol transport to southern Sweden. SIMCA reveals low variability in fine mode elemental composition in southerly air masses being clearly different from the elemental compositions found in northerly air masses.

  • PIXE in complex analytical systems for atmospheric chemistry

    • A schematic mechanistic box model presenting processes in atmospheric chemical cycles is used to present the main research objectives. From this the main use of PIXE within atmospheric chemistry is discussed. An analytic system giving molecular composition is necessary to fully understand and quantify the chemical and physical pathways described in the model for different compounds. Combining PIXE with complementary techniques can enhance knowledge of molecular composition. Therefore, techniques which are complementary to PIXE that give such important parameters as mass, soot, and major ions are discussed. Difficulties in efficiently combining and using the discussed analytical techniques in practice are recognized. The sampling substrates and sampling methods used are discussed and evaluated from this point of view. The use of thin Al foil as substrate and an electrostatic precipitator for aerosol sampling is suggested as one solution. The importance of developing methods for use of this analytical system of the different phases present in the atmosphere is stressed.

  • Intraneural edema following exposure to vibration

    • Peripheral neuropathy represents a well-known complication from long-term exposure to vibration. In the present study an experimental model is presented with the purpose of analyzing the formation of intraneural edema following vibration exposure. Vibration (82 Hz, peak-to-peak amplitude 0.21 mm) was induced in the hind limb of rats by the use of vibrating electric motors during 4 h/d for 5 d. Tracer techniques (with albumin Evans blue and horseradish peroxidase) were used to study the permeability of intraneural microvessels after the vibration exposure on day 5. It was found that the vibration trauma in this model induced epineurial edema in the sciatic nerve. It is hypothesized that the formation of intraneural edema may be an important pathophysiological factor in the occurrence of vibration-induced neuropathy.
