    • The eruption of Mount Tambora (Indonesia) in April 1815 is the largest documented volcanic eruption in history. It is associated with a large global cooling during the following year, felt particularly in parts of Europe and North America, where the year 1816 became known as the "year without a summer". This paper describes an effort made to collect surface meteorological observations from the early instrumental period, with a focus on the years of and immediately following the eruption (1815–1817). Although the collection aimed in particular at pressure observations, correspondent temperature observations were also recovered. Some of the series had already been described in the literature, but a large part of the data, recently digitised from original weather diaries and contemporary magazines and newspapers, is presented here for the first time. The collection puts together more than 50 sub-daily series from land observatories in Europe and North America and from ships in the tropics. The pressure observations have been corrected for temperature and gravity and reduced to mean sea level. Moreover, an additional statistical correction was applied to take into account common error sources in mercury barometers. To assess the reliability of the corrected data set, the variance in the pressure observations is compared with modern climatologies, and single observations are used for synoptic analyses of three case studies in Europe. All raw observations will be made available to the scientific community in the International Surface Pressure Databank.

    • Immune responses need to be controlled tightly to prevent autoimmune diseases, yet underlying molecular mechanisms remain partially understood. Here, we identify biallelic mutations in three patients from two unrelated families in differentially expressed in FDCP6 homolog (DEF6) as the molecular cause of an inborn error of immunity with systemic autoimmunity. Patient T cells exhibit impaired regulation of CTLA-4 surface trafficking associated with reduced functional CTLA-4 availability, which is replicated in DEF6-knockout Jurkat cells. Mechanistically, we identify the small GTPase RAB11 as an interactor of the guanine nucleotide exchange factor DEF6, and find disrupted binding of mutant DEF6 to RAB11 as well as reduced RAB11+CTLA-4+ vesicles in DEF6-mutated cells. One of the patients has been treated with CTLA-4-Ig and achieved sustained remission. Collectively, we uncover DEF6 as player in immune homeostasis ensuring availability of the checkpoint protein CTLA-4 at T-cell surface, identifying a potential target for autoimmune and/or cancer therapy.

  • Limiting the impact of stealthy attacks on Industrial Control Systems

    • While attacks on information systems have for most practical purposes binary outcomes (information was manipulated/eavesdropped, or not), attacks manipulating the sensor or control signals of Industrial Control Systems (ICS) can be tuned by the attacker to cause a continuous spectrum in damages. Attackers that want to remain undetected can attempt to hide their manipulation of the system by following closely the expected behavior of the system, while injecting just enough false information at each time step to achieve their goals. In this work, we study if physics-based attack detection can limit the impact of such stealthy attacks. We start with a comprehensive review of related work on attack detection schemes in the security and control systems community. We then show that many of these works use detection schemes that are not limiting the impact of stealthy attacks. We propose a new metric to measure the impact of stealthy attacks and how they relate to our selection on an upper bound on false alarms. We finally show that the impact of such attacks can be mitigated in several cases by the proper combination and configuration of detection schemes. We demonstrate the effectiveness of our algorithms through simulations and experiments using real ICS testbeds and real ICS systems.

  • An Ocean-Colour Time Series for Use in Climate Studies : The Experience of the Ocean-Colour Climate Change Initiative (OC-CCI)

    • Ocean colour is recognised as an Essential Climate Variable (ECV) by the Global Climate Observing System (GCOS); and spectrally-resolved water-leaving radiances (or remote-sensing reflectances) in the visible domain, and chlorophyll-a concentration are identified as required ECV products. Time series of the products at the global scale and at high spatial resolution, derived from ocean-colour data, are key to studying the dynamics of phytoplankton at seasonal and inter-annual scales; their role in marine biogeochemistry; the global carbon cycle; the modulation of how phytoplankton distribute solar-induced heat in the upper layers of the ocean; and the response of the marine ecosystem to climate variability and change. However, generating a long time series of these products from ocean-colour data is not a trivial task: algorithms that are best suited for climate studies have to be selected from a number that are available for atmospheric correction of the satellite signal and for retrieval of chlorophyll-a concentration; since satellites have a finite life span, data from multiple sensors have to be merged to create a single time series, and any uncorrected inter-sensor biases could introduce artefacts in the series, e.g., different sensors monitor radiances at different wavebands such that producing a consistent time series of reflectances is not straightforward. Another requirement is that the products have to be validated against in situ observations. Furthermore, the uncertainties in the products have to be quantified, ideally on a pixel-by-pixel basis, to facilitate applications and interpretations that are consistent with the quality of the data. This paper outlines an approach that was adopted for generating an ocean-colour time series for climate studies, using data from the MERIS (MEdium spectral Resolution Imaging Spectrometer) sensor of the European Space Agency; the SeaWiFS (Sea-viewing Wide-Field-of-view Sensor) and MODIS-Aqua (Moderate-resolution Imaging Spectroradiometer-Aqua) sensors from the National Aeronautics and Space Administration (USA); and VIIRS (Visible and Infrared Imaging Radiometer Suite) from the National Oceanic and Atmospheric Administration (USA). The time series now covers the period from late 1997 to end of 2018. To ensure that the products meet, as well as possible, the requirements of the user community, marine-ecosystem modellers, and remote-sensing scientists were consulted at the outset on their immediate and longer-term requirements as well as on their expectations of ocean-colour data for use in climate research. Taking the user requirements into account, a series of objective criteria were established, against which available algorithms for processing ocean-colour data were evaluated and ranked. The algorithms that performed best with respect to the climate user requirements were selected to process data from the satellite sensors. Remote-sensing reflectance data from MODIS-Aqua, MERIS, and VIIRS were band-shifted to match the wavebands of SeaWiFS. Overlapping data were used to correct for mean biases between sensors at every pixel. The remote-sensing reflectance data derived from the sensors were merged, and the selected in-water algorithm was applied to the merged data to generate maps of chlorophyll concentration, inherent optical properties at SeaWiFS wavelengths, and the diffuse attenuation coefficient at 490 nm. The merged products were validated against in situ observations. The uncertainties established on the basis of comparisons with in situ data were combined with an optical classification of the remote-sensing reflectance data using a fuzzy-logic approach, and were used to generate uncertainties (root mean square difference and bias) for each product at each pixel.

    • To achieve the goals of substantial improvements in efficiency and emissions set by Flightpath 2050, fundamentally different concepts are required. As one of the most promising solutions, electrification of the aircraft primary propulsion is currently a prime focus of research and development. Unconventional propulsion sub-systems, mainly the electrical power system, associated thermal management system and transmission system, provide a variety of options for integration in the existing propulsion systems. Different combinations of the gas turbine and the unconventional propulsion sub-systems introduce different configurations and operation control strategies. The trade-off between the use of the two energy sources, jet fuel and electrical energy, is primarily a result of the trade-offs between efficiencies and sizing characteristics of these sub-systems. The aircraft structure and performance are the final carrier of these trade-offs. Hence, full design space exploration of various hybrid derivatives requires global investigation of the entire aircraft considering these key propulsion sub-systems and the aircraft structure and performance, as well as their interactions. This paper presents a recent contribution of the development for a physics-based simulation and optimization platform for hybrid electric aircraft conceptual design. Modeling of each subsystem and the aircraft structure are described as well as the aircraft performance modeling and integration technique. With a focus on the key propulsion sub-systems, aircraft structure and performance that interfaces with existing conceptual design frameworks, this platform aims at full design space exploration of various hybrid concepts at a low TRL level.

    • The aggregation of the most common natural cyclodextrins (α-, β-, and γ-) in aqueous solutions is addressed by studying the CD-CD interactions using deuterium relaxation rates for deuterium labeled CDs. Relaxation times (T1) and their corresponding relaxation rates (R1 = 1/T1) provide information about the rotational correlation times of CDs and serve as a proxy for solute-solute interactions. Measured T1's for α-, β-, and γ-CD at the lowest CD concentrations were in agreement with predictions of a hydrodynamic model for toroids, in particular with regard to the dependence of T1 on CD size. On the other hand, the dependence of T1's with respect to the increase in CD concentration could not be explained by hydrodynamic or direct interaction between CD molecules, and it is suggested that there is an equilibrium between monomeric and dimeric CD to account for the observed concentration dependence. No evidence in favor of large aggregates of CDs involving a non-negligible fraction was found for the investigated CDs.
