  • In vitro effects of the citrus flavonoids diosmin, naringenin and naringin on the hepatic drug-metabolizing CYP3A enzyme in human, pig, mouse and fish

    • Flavonoids are known to have effects on cytochrome P450 enzymatic activity. However, little effort has been made to examine species differences and the relevance of studies on mammalian and fish microsomes so that extrapolations can be made to humans. Therefore, the effects of several naturally occurring flavonoids on the activity of CYP3A-dependent 7-benzyloxy-4-trifluoromethylcoumarin O-debenzylase (BFCOD) were evaluated in human, pig, mouse, and juvenile rainbow trout sources of hepatic microsomes. Each was exposed to three concentrations (1, 10, and 100 mu M) of diosmin, naringin, and naringenin. Naringenin competitively inhibited BFCOD activity (K-i values were 24.6 mu M in human, 15.6 mu M in pig, and 19.6 mu M in mouse microsomes). In fish, BFCOD activity was inhibited in a noncompetitive manner (K-i = 7 mu M). Neither diosmin nor naringenin affected BFCOD activity in hepatic microsomes from the studied model organisms. These results suggest that dietary flavonoids potentially inhibit the metabolism of clinical drugs. (C) 2016 Elsevier Inc. All rights reserved.

  • Efficient Approximation of Values in Gain scheduled Routing

    • This paper shows how the cost of computation and memory of a previously proposed routing algorithm can be reduced. The routing algorithm, intended for communications networks carrying multiple call classes, is based on gain scheduling of precomputed relative value functions. Each relative value function is computed by solving a reinforcement learning problem, and defines a link admission control policy. We propose a method for automatically selecting points in a grid of per-class arrival intensities, for which relative value functions are computed. After construction of the grid, relative values are computed by interpolation. The numerical studies of routing in a network with two call classes show that less than 30 relative value functions are needed to avoid performance degradation.

    • Volume kinetics is a mathematical tool for macroscopic (whole-body) evaluation of the distribution and elimination of fluid given by intravenous infusion. Although the kinetic system has mostly been applied to crystalloid fluids, such as Ringer's solution, it has more recently been extended to glucose solution, which is characterized by interdependence between glucose and fluid kinetics. The elimination of glucose, as estimated by a one-compartment open model, serves as the driving force for cellular uptake of glucose and, by virtue of osmosis, of water. Key findings include the observation that the infused fluid, besides being accumulated in the cells, occupies a central body fluid space (VI), which is no larger than 3-4 L, and that the cellular hydration has a much longer time-course than the hydration of V-1. This explains the risk of hypovolemia associated with rapid infusion of 5% glucose; the dilution of V-1, which is quite substantial owing to the small size of this space at baseline, stimulates a brisk diuresis while the excess water is being "trapped" in the cells along with the glucose. Model linearity has been demonstrated for 2.5% glucose solution and this allows the construction of nomograms for administration of such fluid during surgery and critical illness.

  • Tron, ordet och nådegåvorna : en studie av Livets Ord i Tjeckien

    • In this thesis Word of Life (Slovo zivota) in the Czech Republic is studied from a sociological perspective with a special focus on the Swedish influence, especially the teachings of Ulf Ekman, on the Czech Word of Life-congregations. Another purpose is to examine if there also is space for domestic interpretations of the theology of the faith-movement and if there is any integration with Czech religious tradition. The study also poses the question if the Czech Word of Life-congregations stress the prosperity-theology and the importance of the spiritual gifts of grace. A parallel question is thus to find out if there has been any conflicts between Slovo zivota and other religious bodies or the society in the Czech Republic as has been apparent regarding Livets Ord in Sweden. The subjects in this thesis are discussed with the help of sociological theory, including the concept of charisma and church-sect typologies as well as textual analysis and field research. The study shows that even though the teachings of Ulf Ekman is still important within the Word of Life-congregations in the Czech Republic, the domestic religious traditions, mostly in the form of Husitism, are apparent. Other prominent features of these congregations are a strong focus on faith healing but also on religious tolerance together with an interest in ecumenical work with other churches and denominations in the country. The signs of striving for independence from Swedish influence are visible in the fact that the Czech Word of Life-congregations are forming a domestic variation of faith-theology where the national religious traditions are integrated with the theology usually connected with the faith-movement.
