  • Contrasting Drought Propagation Into the Terrestrial Water Cycle Between Dry and Wet Regions

    • Drought's intensity and duration have increased in many regions over the last decades. However, the propagation of drought-induced water deficits through the terrestrial water cycle is not fully understood at a global scale. Here we study responses of monthly evaporation (ET) and runoff to soil moisture droughts occurring between 2001 and 2015 using independent gridded datasets based on machine learning-assisted upscaling of satellite and in-situ observations. We find that runoff and ET show generally contrasting drought responses across climate regimes. In wet regions, runoff is strongly reduced while ET is decoupled from soil moisture decreases and enhanced by sunny and warm weather typically accompanying soil moisture droughts. In drier regions, ET is reduced during droughts due to vegetation water stress, while runoff is largely unchanged as precipitation deficits are typically low in these regions and ET decreases are buffering runoff reductions. While these water flux drought responses are controlled by the large-scale climate regimes, they are additionally modulated by local vegetation characteristics. Land surface models capture the observed water cycle responses to drought in the case of runoff, but not for ET where the ET deficit (surplus) is overestimated (underestimated), related to a misrepresentation of the general soil moisture-evaporation interplay. In summary, our study illustrates how the joint analysis of machine learning-enhanced Earth observations can advance the understanding of global eco-hydrological processes, as well as the validation of land surface models.

  • Investigation of the inner structures around HD 169142 with VLT/SPHERE

    • We present observations of the Herbig Ae star HD 169142 with the VLT/SPHERE instruments InfraRed Dual-band Imager and Spectrograph (IRDIS) (K1K2 and H2H3 bands) and the Integral Field Spectrograph (IFS) (Y, J and H bands). We detect several bright blobs at similar to 180 mas separation from the star, and a faint arc-like structure in the IFS data. Our reference differential imaging (RDI) data analysis also finds a bright ring at the same separation. We show, using a simulation based on polarized light data, that these blobs are actually part of the ring at 180 mas. These results demonstrate that the earlier detections of blobs in the H and K-S bands at these separations in Biller et al. as potential planet/substellar companions are actually tracing a bright ring with a Keplerian motion. Moreover, we detect in the images an additional bright structure at similar to 93 mas separation and position angle of 355 degrees, at a location very close to previous detections. It appears point-like in the YJ and K bands but is more extended in the H band. We also marginally detect an inner ring in the RDI data at similar to 100 mas. Follow-up observations are necessary to confirm the detection and the nature of this source and structure.

  • Hypertension in stroke survivors and associations with national premature stroke mortality : data for 2·5 million participants from multinational screening campaigns

    • Background: Blood pressure control has a pivotal role in reducing the incidence and recurrence of stroke. May Measurement Month (MMM), which was initiated in 2017 by the International Society of Hypertension, is the largest global blood pressure screening campaign. We aim to compare MMM participants with and without a previous history of stroke and to investigate associations between national-level estimates of blood pressure management from MMM and premature stroke mortality. Methods: In this annual, global, cross-sectional survey, more than 2·5 million volunteers (≥18 years) from 92 countries were screened in May, 2017, and May, 2018. Three seated blood pressure readings and demographic, lifestyle, and cardiovascular disease data were collected. Associations between risk factors and stroke history were analysed with mixed-effects logistic regression, and associations between national-level estimates of blood pressure management and premature stroke mortality based on Global Burden of Disease data were investigated with linear regression. Findings: 2 222 399 (88·4%) of 2 515 365 participants had recorded data on a history of stroke, of whom 62 639 (2·8%) reported a previous stroke. Participants with a history of stroke had higher rates of hypertension (77·0% vs 32·9%, p<0·0001) and of treated (90·2% vs 57·0%, p<0·0001) and controlled (55·9% vs 32·4%, p<0·0001) hypertension than those without a history of stroke. A third of participants with a history of stroke had either untreated hypertension or treated but uncontrolled hypertension (blood pressure ≥140/90 mm Hg). Strong positive associations were found between national premature stroke mortality and mean systolic blood pressure (84·3 [95% CI 38·8 to 129·9] years of life lost [YLL] per 100 000 people per mm Hg increase) and the percentage of participants with raised blood pressure (49·1 [22·6 to 75·6] YLL per 100 000 people per 1% increase). Strong negative associations were found between national premature stroke mortality and the percentage of participants with hypertension on treatment (−21·0 [−33·0 to −8·9] YLL per 100 000 people per 1% increase) and with controlled blood pressure (−31·6 [−43·8 to −19·4] YLL per 100 000 people per 1% increase). Interpretation: Blood pressure control remains suboptimal worldwide among people with a history of stroke. National estimates of blood pressure management reflect national premature stroke mortality sufficiently to prompt policy makers to promote blood pressure screening and management.   • Hint of curvature in the orbital motion of the exoplanet 51 Eridani b using 3 yr of VLT/SPHERE monitoring

    • Context. The 51 Eridani system harbors a complex architecture with its primary star forming a hierarchical system with the binary GJ 3305AB at a projected separation of 2000 au, a giant planet orbiting the primary star at 13 au, and a low-mass debris disk around the primary star with possible cold and warm components inferred from the spectral energy distribution. Aims. We aim to better constrain the orbital parameters of the known giant planet. Methods. We monitored the system over three years from 2015 to 2018 with the Spectro-Polarimetric High-contrast Exoplanet REsearch (SPHERE) instrument at the Very Large Telescope (VET). Results. We measure an orbital motion for the planet of similar to 130 mas with a slightly decreasing separation (similar to 10 mas) and find a hint of curvature. This potential curvature is further supported at 3 sigma significance when including literature Gemini Planet Imager (GPI) astrometry corrected for calibration systematics. Fits of the SPHERE and GPI data using three complementary approaches provide broadly similar results. The data suggest an orbital period of 32(-9)(+17) yr (i.e., 12(-2)(+4), au in semi-major axis), an inclination of 133(-7)(+1)(4) deg, an eccentricity of 0.45(-0.15)(+0.10), and an argument of periastron passage of 87(-30)(+34) deg [mod 180 degrees]. The time at periastron passage and the longitude of node exhibit bimodal distributions because we do not yet detect whether the planet is accelerating or decelerating along its orbit. Given the inclinations of the orbit and of the stellar rotation axis (134-144 degrees), we infer alignment or misalignment within 18 degrees for the star-planet spin-orbit. Further astrometric monitoring in the next 3-4 yr is required to confirm at a higher significance the curvature in the motion of the planet, determine if the planet is accelerating or decelerating on its orbit, and further constrain its orbital parameters and the star-planet spin-orbit.

  • VLT/SPHERE astrometric confirmation and orbital analysis of the brown dwarf companion HR 2562 B

    • Context. A low-mass brown dwarf has recently been imaged around HR 2562 (HD 50571), a star hosting a debris disk resolved in the far infrared. Interestingly, the companion location is compatible with an orbit coplanar with the disk and interior to the debris belt. This feature makes the system a valuable laboratory to analyze the formation of substellar companions in a circumstellar disk and potential disk-companion dynamical interactions. Aims. We aim to further characterize the orbital motion of HR 2562 B and its interactions with the host star debris disk. Methods. We performed a monitoring of the system over similar to 10 months in 2016 and 2017 with the VLT/SPHERE exoplanet imager. Results. We confirm that the companion is comoving with the star and detect for the first time an orbital motion at high significance, with a current orbital motion projected in the plane of the sky of 25 mas (similar to 0.85 au) per year. No orbital curvature is seen in the measurements. An orbital fit of the SPHERE and literature astrometry of the companion without priors on the orbital plane clearly indicates that its orbit is (quasi-)coplanar with the disk. To further constrain the other orbital parameters, we used empirical laws for a companion chaotic zone validated by N-body simulations to test the orbital solutions that are compatible with the estimated disk cavity size. Non-zero eccentricities (>0.15) are allowed for orbital periods shorter than 100 yr, while only moderate eccentricities up to similar to 0.3 for orbital periods longer than 200 yr are compatible with the disk observations. A comparison of synthetic Herschel images to the real data does not allow us to constrain the upper eccentricity of the companion.

    • Objective: Neurofilament heavy-chain gene (NEFH) variants are associated with multiple neurodegenerative diseases, however, their relationship with ALS has not been robustly explored. Still, NEFH is commonly included in genetic screening panels worldwide. We therefore aimed to determine if NEFH variants modify ALS risk.Methods: Genetic data of 11,130 people with ALS and 7,416 controls from the literature and Project MinE were analysed. We performed meta-analyses of published case–control studies reporting NEFH variants, and variant analysis of NEFH in Project MinE whole-genome sequencing data.Results: Fixed-effects meta-analysis found that rare (MAF <1%) missense variants in the tail domain of NEFH increase ALS risk (OR 4.55, 95% CI 2.13–9.71, p < 0.0001). In Project MinE, ultrarare NEFH variants increased ALS risk (OR 1.37 95% CI 1.14–1.63, p = 0.0007), with rod domain variants (mostly intronic) appearing to drive the association (OR 1.45 95% CI 1.18–1.77, pMadsen–Browning = 0.0007, pSKAT-O = 0.003). While in the tail domain, ultrarare (MAF <0.1%) pathogenic missense variants were also associated with higher risk of ALS (OR 1.94, 95% CI 0.86–4.37, pMadsen–Browning = 0.039), supporting the meta-analysis results. Finally, several tail in-frame deletions were also found to affect disease risk, however, both protective and pathogenic deletions were found in this domain, highlighting an intricated architecture that requires further investigation.Interpretation: We showed that NEFH tail missense and in-frame deletion variants, and intronic rod variants are risk factors for ALS. However, they are not variants of large effect, and their functional impact needs to be clarified in further studies. Therefore, their inclusion in routine genetic screening panels should be reconsidered.

  • Interplay between BRCA1 and RHAMM Regulates Epithelial Apicobasal Polarization and May Influence Risk of Breast Cancer

    • Differentiated mammary epithelium shows apicobasal polarity, and loss of tissue organization is an early hallmark of breast carcinogenesis. In BRCA1 mutation carriers, accumulation of stem and progenitor cells in normal breast tissue and increased risk of developing tumors of basal-like type suggest that BRCA1 regulates stem/progenitor cell proliferation and differentiation. However, the function of BRCA1 in this process and its link to carcinogenesis remain unknown. Here we depict a molecular mechanism involving BRCA1 and RHAMM that regulates apicobasal polarity and, when perturbed, may increase risk of breast cancer. Starting from complementary genetic analyses across families and populations, we identified common genetic variation at the low-penetrance susceptibility HMMR locus (encoding for RHAMM) that modifies breast cancer risk among BRCA1, but probably not BRCA2, mutation carriers: n = 7,584, weighted hazard ratio ((w)HR) = 1.09 (95% CI 1.02-1.16), p(trend) = 0.017; and n = 3,965, (w)HR = 1.04 (95% CI 0.94-1.16), p(trend) = 0.43; respectively. Subsequently, studies of MCF10A apicobasal polarization revealed a central role for BRCA1 and RHAMM, together with AURKA and TPX2, in essential reorganization of microtubules. Mechanistically, reorganization is facilitated by BRCA1 and impaired by AURKA, which is regulated by negative feedback involving RHAMM and TPX2. Taken together, our data provide fundamental insight into apicobasal polarization through BRCA1 function, which may explain the expanded cell subsets and characteristic tumor type accompanying BRCA1 mutation, while also linking this process to sporadic breast cancer through perturbation of HMMR/RHAMM.

    • Recent observations of resolved cold debris discs at tens of au have revealed that gaps could be a common feature in these Kuiper-belt analogues. Such gaps could be evidence for the presence of planets within the gaps or closer in near the edges of the disc. We present SPHERE observations of HD 92945 and HD 107146, two systems with detected gaps. We constrained the mass of possible companions responsible for the gap to 1–2 MJup for planets located inside the gap and to less than 5 MJup for separations down to 20 au from the host star. These limits allow us to exclude some of the possible configurations of the planetary systems proposed to explain the shape of the discs around these two stars. In order to put tighter limits on the mass at very short separations from the star, where direct-imaging data are less effective, we also combined our data with astrometric measurements from Hipparcos and Gaia and radial-velocity measurements. We were able to limit the separation and the mass of the companion potentially responsible for the proper-motion anomaly of HD 107146 to values of 2–7 au and 2–5 MJup, respectively.

  • VLT/SPHERE exploration of the young multiplanetary system PDS70

    • Context. PDS 70 is a young (5.4 Myr), nearby (similar to 113 pc) star hosting a known transition disk with a large gap. Recent observations with SPHERE and NACO in the near-infrared (NIR) allowed us to detect a planetary mass companion, PDS 70 b, within the disk cavity. Moreover, observations in H-alpha with MagAO and MUSE revealed emission associated to PDS 70 b and to another new companion candidate, PDS 70 c, at a larger separation from the star. PDS 70 is the only multiple planetary system at its formation stage detected so far through direct imaging.Aims. Our aim is to confirm the discovery of the second planet PDS 70 c using SPHERE at VLT, to further characterize its physical properties, and search for additional point sources in this young planetary system.Methods. We re-analyzed archival SPHERE NIR observations and obtained new data in Y, J, H and K spectral bands for a total of four different epochs. The data were reduced using the data reduction and handling pipeline and the SPHERE data center. We then applied custom routines (e.g., ANDROMEDA and PACO) to subtract the starlight.Results. We re-detect both PDS 70 b and c and confirm that PDS 70 c is gravitationally bound to the star. We estimate this second planet to be less massive than 5 M-Jup and with a T-eff around 900 K. Also, it has a low gravity with log g between 3.0 and 3.5 dex. In addition, a third object has been identified at short separation (similar to 0.12 '') from the star and gravitationally bound to the star. Its spectrum is however very blue, meaning that we are probably seeing stellar light reflected by dust and our analysis seems to demonstrate that it is a feature of the inner disk. We cannot however completely exclude the possibility that it is a planetary mass object enshrouded by a dust envelope. In this latter case, its mass should be of the order of a few tens of M-circle plus. Moreover, we propose a possible structure for the planetary system based on our data, and find that this structure cannot be stable on a long timescale.
