    • A macrophage migration inhibitory factor (MIF), originally described as a product of activated lymphocytes, has been defined as a 12 kDa protein, expressed in a wide variety of tissues. Here MIF is identified as a phenylpyruvate tautomerase (EC 5.3.2.1) having p-hydroxyphenylpyruvate and phenylpyruvate as its natural substrates. The definition of MIF as an enzyme may yield insight into the mechanism of action of this proinflammatory and immunomodulating cytokine.

    • Background Global efforts to characterize diseases of poverty are hampered by lack of affordable and comprehensive detection platforms, resulting in suboptimal allocation of health care resources and inefficient disease control. Next generation sequencing (NGS) can provide accurate data and high throughput. However, shotgun and metagenome-based NGS approaches are limited by low concentrations of microbial DNA in clinical samples, requirements for tailored sample and library preparations plus extensive bioinformatics analysis. Here, we adapted molecular inversion probes (MIPs) as a cost-effective target enrichment approach to characterize microbial infections from blood samples using short-read sequencing. We designed a probe panel targeting 2 bacterial genera, 21 bacterial and 6 fungi species and 7 antimicrobial resistance markers (AMRs).Results Our approach proved to be highly specific to detect down to 1 in a 1000 pathogen DNA targets contained in host DNA. Additionally, we were able to accurately survey pathogens and AMRs in 20 out of 24 samples previously profiled with routine blood culture for sepsis.Conclusions Overall, our targeted assay identifies microbial pathogens and AMRs with high specificity at high throughput, without the need for extensive sample preparation or bioinformatics analysis, simplifying its application for characterization and surveillance of infectious diseases in medium- to low- resource settings.

Understanding how technology could be incorporated into solutions for sustainable tourism is a particularly interesting design challenge. This paper describes how we tried to meet such a challenge in an effort to help the residents of Split, Croatia enter into a dialogue with their local authorities about how to develop sustainable tourism within the specific socio-political constraints of their region.

  • The value of change: A scenario assessment of the effects of bioeconomy driven land use change on ecosystem service provision

    • Policy makers in Nordic countries envisage a developing bioeconomy as an important element in the transition towards a fossil-energy-free future. However, although the shape of such a bioeconomy is unclear, impacts on land use and land management are likely. To analyse the possible impact on ecosystem services value of this transition, we used five scenarios that describe possible socio-economic environments in the four Nordic countries in the year 2050. These five Nordic Bioeconomy Pathways (NBPs) have been labelled in brief as: 'sustainability first', 'conventional first', 'self-sufficiency first', 'city first' and 'growth first'. In this paper, we adapted an existing integrating framework of ecosystem service delivery to accommodate these NBPs and esti-mated economic value of all services for six study catchments across the Nordic countries: Odense (DK), Simojoki (FI), Haldenvassdraget (NO), Orrevassdraget (NO), Sa center dot vjaan (SE) and Vindela center dot lven (SE). We articulated the sce-nario storylines to a set of numerical attributes per NBP using input from secondary data sources and interviews with stakeholder representatives and local experts. We made land use change spatially explicit based on the catchment's physical characteristics and used links between catchment attributes and ecosystem services flow to estimate annual ecosystem services generation in biophysical and monetary units. Outcomes suggest: the value of active recreation increases more in 'sustainability first' and 'growth first' than in other scenarios; variations in total value are largest among catchments under 'city first'; the overall rank order of ecosystem services value within catchments largely remains unchanged under all NBPs. We conclude that outdoor recreation contributes a high benefit to society that is likely not adequately considered relative to provisioning services in current de-cision making, and that these benefits appear sensitive to how a bioeconomy will develop. Overall, the estimated summed value delivered in these catchments is highest under the 'sustainability first' and 'growth first' scenarios.

  • Does it really fit? : improve, find and evaluate garment fit

    • Clothes affect everyone; we wear them for all occasions; they silently communicate on our behalf, and they can enhance our level of confidence and comfort. For the garment to be comfortable, the garment fit has to be appropriate. Appropriate for the intended function and in line with the wearer’s preferences. For the garment to end up as an approved garment in the customer’s wardrobe, it has to be improved and evaluated many times over. The final evaluation to pass is when the customer finds the garment, tries it on and asks: Does it really fit? The common denominator for the studies included in this thesis is garment fit; the goal is to investigate some methods to improve, find and evaluate garment fit. To improve garment fit, two studies were done. One study is on improving garment fit with the help of a systematic model, based on anthropometric and garment numerical data; this is explored with the help of an experimental set up. The second study is on improving garment fit for the unique figure by offering made-to-measure garments, which is investigated with a structure of action research. An experimental strategy is used to find garments that fit, where the size and fit correspondence is compared between virtual and real garments. To tie everything together, variables for garment fit evaluation are identified with the help of a structured literature review and then analysed within each study. The result shows that the theoretical garment fit improves by using the systematic model. Both the overall accommodation for the target group increased as well as the fit value. The garment fit is improved for the unique figure; this is achieved through complex body measurements, invasive pattern modifications and garment make-up for fit evaluation. The accuracy for size selection based on virtual garments exceeded the one based on the more traditional key measurements. The variables involved in the garment fit evaluation can be divided into five areas: influencing factors, evaluations focus, resources, evaluators or fitting sessions.

    • Nucleic acids are fundamental molecules of living organisms functioning essentially as the molecular information carriers of life. From how an organism is built to how it responds to external conditions, all of it, can be found in the form of nucleic acid sequences inside every single cell of every life form on earth.Therefore, accessing these sequences provides key information regarding the molecular identity and functional state of any living organism, this is very useful for areas like biomedicine, where accessing and understanding these molecular signatures is the key to develop strategies to understand, treat and diagnose diseases.Decades of research and technological advancements have led to the development of a number of molecular tools and engineering technologies that allow accessing the information contained in the nucleic acids. This thesis provides a general overview of the tools and technologies available for nucleic acid analysis, and proposes an illustrative concept on how molecular tools and emergent technologies can be combined in a modular fashion to design methods for addressing different biomedical questions. The studies included in this thesis, are focused on the particular use of the molecular tools named: padlock and selector probes, rolling circle amplification, and fluorescence detection of single molecules in combination with microfluidics and portable microscopy. By using this combination, it became possible to design and demonstrate novel approaches for integrated nucleic acid analysis, inexpensive digital quantification, mobile-phone based diagnostics and the description of viral infections.These studies represent a step forward towards the adoption of the selected group of tools and technologies, for the design and building of methods that can be used as powerful alternatives to conventional tools used in molecular diagnostics and virology.
