    • Meso-tetrakis (4-bromo-2-thiophene) and (5-bromo-2-thiophene) porphyrins were synthesized in 18% yield from freshly distilled pyrrole and the corresponding aldehydes in refluxing propionic acid. Reaction of both porphyrins with either Zn(OAc) 2 · 2H 2 O or Ag(OAc) afforded the corresponding metal complexes in 20% yield. The free bases and metal complexes were characterized by IR, NMR, EPR, MS and XPS. The crystal structure of 5,10,15,20(4-bromo-2-thiophene)porphyrinato Zn(II) was determined by X-ray diffraction and shows a 'zigzag-like' packing pattern due to S···S intermolecular interaction. Structural features of these molecules are discussed by DFT quantum chemical calculations. © 2008 Elsevier B.V. All rights reserved.

  • Microfluidic leukocyte isolation for gene expression analysis in critically ill hospitalized patients

    • BACKGROUND: Microarray technology is becoming a powerful tool for diagnostic, therapeutic, and prognostic applications. There is at present no consensus regarding the optimal technique to isolate nucleic acids from blood leukocyte populations for subsequent expression analyses. Current collection and processing techniques pose significant challenges in the clinical setting. Here, we report the clinical validation of a novel microfluidic leukocyte nucleic acid isolation technique for gene expression analysis from critically ill, hospitalized patients that can be readily used on small volumes of blood. METHODS: We processed whole blood from hospitalized patients after burn injury and severe blunt trauma according to the microfluidic and standard macroscale leukocyte isolation protocol. Side-by-side comparison of RNA quantity, quality, and genome-wide expression patterns was used to clinically validate the microfluidic technique. RESULTS: When the microfluidic protocol was used for processing, sufficient amounts of total RNA were obtained for genome-wide expression analysis from 0.5 mL whole blood. We found that the leukocyte expression patterns from samples processed using the 2 protocols were concordant, and there was less variability introduced as a result of harvesting method than there existed between individuals. CONCLUSIONS: The novel microfluidic approach achieves leukocyte isolation in <25 min, and the quality of nucleic acids and genome expression analysis is equivalent to or surpasses that obtained from macroscale approaches. Microfluidics can significantly improve the isolation of blood leukocytes for genomic analyses in the clinical setting. (c) 2008 American Association for Clinical Chemistry.

    • Aim. The objective of this study was to determine the prevalence and types of ST-segment changes of the fetal electrocardiogram (FECG) during labour in term fetuses born to mothers with diabetes mellitus (DM) or gestational diabetes. Methods. This was a retrospective case-control study involving populations from two multi centre trials: the Swedish Randomized Control Trial and the European Union ST-analysis (EU-STAN) trial. ST-segment changes were assessed in 104/309 cases and 207/468 controls from the Swedish and EU-STAN trials, respectively. Results. ST depression was present on the FECG in 22.1% of fetuses of mothers with DM compared to 12% of controls OR=2.6, 95% CI=1.4-4.7, p=0.002 after adjusting for trial, birth weight, and nulliparity. ST elevation was present in 47.1% of DM patients and 41.2% of controls (OR=1.4, 95% CI=0.9-2.3, p=0.18). Conclusion. ST depression on the FECG was significantly more prevalent in the fetuses of mothers with DM, probably not indicating hypoxia but an altered ability of the myocardium to respond to the stress of labour. Further studies into the mechanism of fetal compromise during diabetic labour, are required.
