  • Effects of SERM (selective estrogen receptor modulator) treatment on growth and proliferation in the rat uterus
  • 2003
  • In: Reproductive Biology and Endocrinology. - London, United Kingdom : BioMed Central (BMC). - 1477-7827. ; 1
  • Journal article (peer-reviewed)abstract
    • Background: Selective estrogen receptor modulators (SERMs) have been developed in order to create means to control estrogenic effects on different tissues. A major drawback in treatment of estrogen receptor (ER) positive breast cancer with the antagonist tamoxifen (TAM) is its agonistic effect in the endometrium. Raloxifene (RAL) is the next generation of SERMs where the agonistic effect on the endometrium has been reduced. Methods: The aim of the present study was to determine the effect of SERM treatment on the uterus, as assessed by proliferation markers and several factors involved in uterine growth. Ovariectomized (ovx) rats were treated with estradiol (E2), tamoxifen (TAM), RAL, ICI182780 (ICI) or vehicle (OVX-controls). We studied the effects on mRNA levels of the growth hormone (GH) receptor, insulin-like growth factor-I (IGF-I), ERalpha and ERbeta. In addition, by immunohistochemistry the proliferation markers PCNA and Ki-67, as well as ERalpha and ERbeta, were detected. Results: The uterine weight of the rats treated with E2 or TAM was increased as compared to OVX-controls. The uterine GH-receptor mRNA level was highest in the E2 treated animals. In ICI treated rats no GH-receptor mRNA could be detected. The IGF-I mRNA level increased 16-fold in uteri of the TAM treated group and 9-fold in the E2 treated rats as compared to OVX-controls. The ERalpha mRNA level was increased in the E2 treated rats, while the ERbeta mRNA level was increased after TAM treatment. The proliferation, as assessed by PCNA, was lowest in ICI treated animals. Conclusions: The uterine wet weight, the LE height and the GH-receptor mRNA levels showed similar patterns, indicating that GH is involved in the regulation of uterine weight. Tamoxifen, which has been related to increased incidence of endometrial carcinoma in women, dramatically increased IGF-I mRNA levels in rat uterus. Since proliferation was not higher in TAM and E2 treated rats than in OVX controls, this assay of simple, early proliferation does not give the full explanation of why TAM should enhance the risk of developing endometrial cancer.

  • Depositional history of the North Taymyr ice-marginal zone, Siberia - a landsystem approach
  • 2002
  • In: Journal of Quaternary Science. - : Wiley. - 1099-1417 .- 0267-8179. ; 17:4, s. 361-382
  • Journal article (peer-reviewed)abstract
    • The sediment-landform associations of the northern Taymyr Peninsula in Arctic Siberia tell a tale of ice sheets advancing from the Kara Sea shelf and inundating the peninsula, probably three times during the Weichselian. In each case the ice sheet had a margin frozen to its bed and an interior moving over a deforming bed. The North Taymyr ice-marginal zone (NTZ) comprises ice-marginal and supraglacial landsystems dominated by thrust-block moraines 2-3 km wide and large-scale deformation of sediments and ice. Large areas are still underlain by remnant glacier ice and a supraglacial landscape with numerous ice-walled lakes and kames is forming even today. The proglacial landsystem is characterised by subaqueous (e.g. deltas) or terrestrial (e.g. sandar) environments, depending on location/altitude and time of formation. Dating results (OSL, C-14) indicate that the NTZ was initiated ca. 80 kyr BP during the retreat of the Early Weichselian ice sheet and that it records the maximum limit of a Middle Weichselian glaciation (ca. 65 kyr BP). During both these events, proglacial lakes were dammed by the ice sheets. Part of the NTZ was occupied by a thin Late Weichselian ice sheet (20-12 kyr BP), resulting in subaerial proglacial drainage. • Effects of morphine analgesia in ventilated preterm neonates : primary outcomes from the NEOPAIN randomised trial
  • 2004
  • In: The Lancet. - 0140-6736 .- 1474-547X. ; 363:9422, s. 1673-82
  • Journal article (peer-reviewed)abstract
    • BACKGROUND: Opioid analgesia is commonly used during neonatal intensive care. We undertook the Neurologic Outcomes and Pre-emptive Analgesia in Neonates (NEOPAIN) trial to investigate whether pre-emptive morphine analgesia decreases the rate of a composite primary outcome of neonatal death, severe intraventricular haemorrhage (IVH), and periventricular leucomalacia (PVL) in preterm neonates.METHODS: Ventilated preterm neonates (n=898) from 16 centres were randomly assigned masked placebo (n=449) or morphine (n=449) infusions. After a loading dose (100 microg/kg), morphine infusions (23-26 weeks of gestation 10 microg kg(-1) h(-1); 27-29 weeks 20 microg kg(-1) h(-1); 30-32 weeks 30 microg kg(-1) h(-1)) were continued as long as clinically justified (maximum 14 days). Open-label morphine could be given on clinical judgment (placebo group 242/443 [54.6%], morphine group 202/446 [45.3%]). Analyses were by intention to treat.FINDINGS: Baseline variables were similar in the randomised groups. The placebo and morphine groups had similar rates of the composite outcome (105/408 [26%] vs 115/419 [27%]), neonatal death (47/449 [11%] vs 58/449 [13%]), severe IVH (46/429 [11%] vs 55/411 [13%]), and PVL (34/367 [9%] vs 27/367 [7%]). For neonates who were not given open-label morphine, rates of the composite outcome (53/225 [24%] vs 27/179 [15%], p=0.0338) and severe IVH (19/219 [9%] vs 6/189 [3%], p=0.0209) were higher in the morphine group than the placebo group. Placebo-group neonates receiving open-label morphine had worse rates of the composite outcome than those not receiving open-label morphine (78/228 [34%] vs 27/179 [15%], p<0.0001). Morphine-group neonates receiving open-label morphine were more likely to develop severe IVH (36/190 [19%] vs 19/219 [9%], p=0.0024).INTERPRETATION: Pre-emptive morphine infusions did not reduce the frequency of severe IVH, PVL, or death in ventilated preterm neonates, but intermittent boluses of open-label morphine were associated with an increased rate of the composite outcome. The morphine doses used in this study decrease clinical signs of pain but can cause significant adverse effects in ventilated preterm neonates.

  • Screening av Tetrabrombisfenol A
  • 2004
  • Reports (other academic/artistic)abstract
    • Tetrabrombisfenol A (TBBPA), CAS nr 79-94-7, är ett flamskyddsmedel med stor användning. Till största delen används det som reaktivt flamskyddsmedel, dvs. TBBPA binds in kemiskt i polymera material.

  • Modifying the cellular transport of DNA-based vaccines alters the immune response to hantavirus nucleocapsid protein
  • 2001
  • In: Vaccine. - 0264-410X .- 1873-2518. ; 19:28-29, s. 3820-3829
  • Journal article (peer-reviewed)abstract
    • Puumala virus is a member of the hantavirus genus (family Bunyaviridae) and is one of the causative agents of hemorrhagic fever with renal syndrome (HFRS) in Europe. A genetic vaccination approach was conducted to investigate if the immune response could be modulated using different cellular secretion and/or localisation signals, and the immune responses were analysed in BALB/c mice and in a bank vole infectious model. Rodents vaccinated with DNA constructs encoding the antigen fused to an amino-terminal secretion signal raised significantly higher antibody levels when compared to using constructs lacking secretion signals. Furthermore, the ratios of the IgG subclasses (IgG2a/IgG1) were raised by the use of cellular localisation signals, indicating a more pronounced Th1-type of immune response. The majority of the mice, or bank voles, immunised with DNA encoding a secreted form of the antigen showed a positive lymphoproliferative response and were protected against challenge with Puumala virus (strain Kazan-wt).

  • Causes behind a human cheese-borne outbreak of gastrointestinal listeriosis
  • 2004
  • In: Foodborne pathogens and disease. - New York, USA : Mary Ann Liebert. - 1535-3141 .- 1556-7125. ; 1:3, s. 153-159
  • Journal article (peer-reviewed)abstract
    • In a previous paper, we reported an outbreak of gastrointestinal listeriosis due to consumption of fresh cheese made from raw milk and manufactured on a summer farm. The aim of the present study was to investigate why the cheese harbored Listeria monocytogenes. To our knowledge, this is the first documented outbreak of listeriosis caused by raw milk cheese where the human epidemic strain has been cultured from a dairy animal, whose milk has been used for cheese production. The conditions on a summer farm can hardly fulfil the requirements for hygienic and strictly controlled conditions necessary for safe processing of fresh cheese.
