  • Proliferation of primitive myeloid progenitors can be reversibly induced by HOXA10

    • Recent studies show that several Hox transcription factors are important for regulation of proliferation and differentiation in hematopoiesis. Among these is HOXA10, which is selectively expressed at high levels in the most primitive subpopulation of human CD34(+) bone marrow cells. When overexpressed, HOXA10 increases the proliferation of early progenitor cells and can lead to the development of myeloid leukemia. To study the effects of HOXA10 on primitive hematopoietic progenitors in more detail, transgenic mice were generated with regulatable HOXA10 expression. The transgenic mouse model, referred to as tetO-HOXA10, contains the HOXA10 gene controlled by a tetracycline-responsive element and a minimal promoter. Thus, the expression of HOXA10 is inducible and reversible depending on the absence or presence of tetracycline or its analog, doxycycline. A retroviral vector containing the tetracycline transactivator gene (tTA) was used to induce expression of the HOXA10 gene In bone marrow cells from the transgenic mice. Reverse transcription-polymerase chain reaction analysis confirmed regulatable HOXA10 expression in several transgenic lines. HOXA10 induction led to the formation of hematopoietic colonies containing blastlike cells and megakaryocytes. Moreover, the induction of HOXA10 resulted in significant proliferative advantage of primitive hematopoietic progenitors (spleen colony-forming units [CFU-S-12]), which was reversible on withdrawal of induction. Activation of HOXA10 expression in tet0-HOXA10 mice will therefore govern proliferation of primitive myeloid progenitors in a regulated fashion. This novel animal model can be used to identify the target genes of HOXA10 and better clarify, the specific role of HOXA10 in normal and malignant hematopoiesis.

    • The resonant Auger decay spectra associated with doubly excited core-hole states near the N 1s threshold in the nitrogen molecule was measured. Electron-yield spectrum was obtained by setting a kinetic-energy window at 384 eV and scanning across the threshold region. At ground 410, 414, and 416 eV, photon energy, three dominant structures were observed. The increased signal-to-background ratio, intensity and resolution at third generation facilities were found to cast a different light on the dynamics of core photoionization in molecules.

    • OBJECTIVES: to compare the effect of surgery, exercise and simple observation on maximum exercise power in claudicants. Design: prospective, randomised study. METHODS: a total of 264 unselected claudicants were randomised to supervised exercise training, invasive treatment (open surgical or endovascular procedures) or observation. One year treatment outcomes were analysed on an intention to-treat basis. RESULTS: invasively treated patients showed a significant improvement in maximum walking power, stopping distance, post-ischaemic blood flow and big toe pressure at one year. Patients randomised to physical exercise training or to the control group did not improve in any outcome measure. CONCLUSION: invasive treatment increased walking capacity, leg blood pressure and flow. Supervised physical exercise training offered no therapeutic advantage compared to untreated controls.

    • Highlights are presented from studies of the electric field data from various regions along the CLUSTER orbit. They all point towards a very high coherence for phenomena recorded on four spacecraft that are separated by a few hundred kilometers for structures over the whole range of apparent frequencies from I mHz to 9 kHz. This presents completely new opportunities to study spatial-temporal plasma phenomena from the magnetosphere out to the solar wind. A new probe environment was constructed for the CLUSTER electric field experiment that now produces data of unprecedented quality. Determination of plasma flow in the solar wind is an example of the capability of the instrument.
