    • The survival and function of cross-species (mouse-to-rat) grafts of fetal mesencephalic dopamine (DA) neurons, implanted as a cell suspension in the striatum of rats with lesions of the mesostriatal DA system, have been studied in animals with and without immunosuppression induced by Cyclosporin A (CyA). At 6 weeks after grafting 3 out of 7 non-CyA treated animals showed some degree of graft survival and variable functional compensation. In those three animals an average of 92 DA neurons per graft was counted. In the grafted animals treated with daily CyA injections, all grafts survived and produced partial or complete functional compensation, and they had an average of 557 DA neurons per graft. It is concluded that intracerebral graft survival and function can be greatly improved by CyA treatment and that the immunological protection of neural transplants in the brain is only partial.

  • A simple peer-to-peer algorithm for distributed optimization in sensor networks

    • We propose a distributed algorithm that solves a special class of optimization problems using only peer-to-peer communication. One application is parameter estimation problems in sensor networks. Current decentralized algorithms for solving this class of optimization problems typically rely on passing around a parameter estimate in a ring consisting of all network nodes. In our algorithm, which extends the randomized incremental subgradient method with fixed stepsize due to Nedic and Bertsekas, nodes maintain individual estimates and need to exchange information only with their neighbors. We establish approach of the solution to an interval around the optimum value. We illustrate the algorithm's performance, in terms of convergence rate and communication cost relative to alternative schemes, through several numerical examples.

  • Location of New Industries – The ICT-Sector 1990-2000

    • The overall purpose of this work is to study the location pattern of new industries and how it changes over time. With this objective as motivation, a set of 27 industries are classified as belonging to the ICT-sector (information and communication technology). The goods and services supplied by these industries were to a large extent new at the end of the 1980s. The paper outlines two interrelated models of vertical externalities to explain the location pattern of the industries in 1990 and 2000. The two externalities concern a firm’s input demand and its output demand. These models are introduced to illustrate how these externalities favour location in the largest functional urban regions. The same models predict that location in smaller regions is facilitated as demand grows, when internal scale economies (start-up costs) are not too strong. The empirical analyses apply a logit model to estimate location probabilities, which depend on the size and the diversity of a region’s economy.

    • DNA topoisomerase 1 (TOP11) inhibitors are mainstays of anticancer therapy. These drugs trap TOP1 on DNA, stabilizing the TOP1-cleavage complex (TOP1-cc). The accumulation of TOP1-ccs perturbs DNA replication fork progression, leading to DNA breaks and cell death. By analyzing the genomic occupancy and activity of TOP1, we show that cells adapt to treatment with multiple doses of TOP1 inhibitor by promoting the degradation of TOP1-ccs, allowing cells to better tolerate subsequent doses of TOP1 inhibitor. The E3-RING Cullin 3 ligase in complex with the BTBD1 and BTBD2 adaptor proteins promotes TOP1-cc ubiquitination and subsequent proteasomal degradation. NEDDylation of Cullin 3 activates this pathway, and inhibition of protein NEDDylation or depletion of Cullin 3 sensitizes cancer cells to TOP1 inhibitors. Collectively, our data uncover a previously unidentified NEDD8–Cullin 3 pathway involved in the adaptive response to TOP1 inhibitors, which can be targeted to improve the efficacy of TOP1 drugs in cancer therapy.

    • Monitoring vegetation gross primary production (GPP) is required for both carbon balance studies and early warning systems aiming to detect unfavorable crop and pasture conditions. This manuscript describes the assimilation of MODIS observations by a simple process model, fed by meteorological data (temperature, incident radiation and rainfall) and linked with a canopy reflectance model, to estimate GPP. GPP simulations are benchmarked against eddy covariance data collected in a semi-arid environment of a sparse Savanna in the Sudan.
