  • The correlation between gene expression of proinflammatory markers and bone formation during osseointegration with titanium implants

    • An in vivo interfacial gene expression model combined with biomechanical analysis was used in order to determine the relationship between the molecular events taking place during osseointegration and the biomechanical stability of the implant. Anodically oxidized and machined, threaded titanium implants were characterized topographically, chemically and ultrastructurally. The implants were inserted in rat tibiae and the implant bone torsion stability was evaluated. After measurements, the implants were removed and analyzed with qPCR. Results showed an increase in the breakpoint torque of 140%, 170% and 190%, after 6, 14, and 28 days, respectively, at the oxidized implants as compared to the machined. Gene expression analysis revealed higher expression of runt related transcription factor-2 (Runx2) (after 28 d), osteocalcin (OC) and tartrate resistant acid phosphatase (TRAP) (after 6, 14 and 28 d) and cathepsin K (CATK) (after 6 and 14 d) at the oxidized implants. On the other hand, machined implants were associated with higher expression of tumor necrosis factor-α (TNF-α) (after 6 and 28 d) and interleukin-1β (IL-1β) (after 6, 14 and 28 d) compared to the oxidized implants. In conclusion, the favorable cellular and molecular events at the oxidized implants were in parallel with significantly stronger bone anchorage during osseointegration.

  • The stimulation of an osteogenic response by classical monocyte activation.

    • The monocyte/macrophage system plays a central role in host defense, wound healing and immune regulation at biomaterial surfaces. Monocytes can be classically and alternatively activated, and can be stimulated differently in response to variations in biomaterial surface properties. In this study, human monocytes, cultured on polystyrene surfaces (Ps), were activated either classically, by lipopolysaccharide (LPS), or alternatively, by interleukin-4 (IL-4). Monocytes were also cultured on anodically oxidized (Ox) and machined (Ma) titanium surfaces, with and without LPS stimulation. Cells were cultured for 1 and 3 days and their conditioned media (CM) were collected. The osteogenic response of hMSCs to the monocyte CM was determined by analyzing the gene expression of key osteogenic markers. The CM from classically activated monocytes increased the hMSCs expression of runt-related transcription factor 2 (Runx2) and alkaline phosphatase (ALP). Furthermore, CM from monocytes cultured on Ox surface resulted in a modest increase of the expression of bone morphogenetic protein-2 (BMP-2). LPS stimulation of the surface-seeded monocytes overwhelmed the effect of the surface properties and resulted in significant upregulation of BMP-2 and Runx2 for all samples. The results show that human monocytes, cultured on different surfaces and/or under different activation pathways, communicate pro-osteogenic signals to hMSCs. The signals involve regulation of autologous BMP-2 in the hMSCs. The classical activation results in profound and prolonged osteogenic effect compared to the effect of the investigated surface properties.

  • Blood pressure and circulatory relationships with physical activity level in young normotensive individuals : IPAQ validity and reliability considerations

    • Physical activity (PA) reduces risk of cardiovascular diseases, including hypertension. However, the international physical activity questionnaire (IPAQ) relationships with blood pressure (BP) and flow (BF) and vascular resistance (VR) in healthy young individuals have not been studied. Therefore, BP, BF, and VR relationships with the IPAQ were evaluated in college normotensive students (18-23 yrs). Additionally, the IPAQ relationships with body fat (%BF), muscle mass (MM), body mass index (BMI), waist/hip (W/H) ratio, maximum walking distance in 6 min (6MWD), and handgrip strength (MHG) were examined to evaluate the questionnaire validity against fitness. Subsequently, the IPAQ was administered three times to examine its reliability. Walking, moderate, and total PAs correlated negatively with sysbolic blood pressure (SBP), diastolic blood pressure (DBP), and mean arterial pressure (MAP) (range: r = -3 to -0.5, p < 0.05). Additionally, all BP measures were greater in least physically active individuals. In a subgroup of 42 students, IPAQ sitting time correlated with BF (r = -0.3) and VR (r = 0.4). The intraclass correlation coefficient (ICC) for walking, moderate, vigorous, and total PAs and sitting time/week were, 0.97, 0.96, 0.97, 0.97, and 0.96, respectively. The males scored greater vigorous PA (p = 0.001) than the females, while moderate, walking, and total PAs were the same (p > 0.05). Additionally, vigorous PA correlated with %BF (r = -0.2), MM (r = 0.3), MHG (r = 0.3), and 6MWD (r = 0.3) and total PA correlated with MM (r = 0.2), MHG (r =0.2), and 6MWD (r = 0.3). The IPAQ association with the circulatory measures demonstrates PA importance for controlling BP and adds clinical value to the IPAQ. Additionally, the IPAQ is reliable, can discriminate between populations, and reasonably valid against health-related fitness.

    • There is an increasing amount of structure on the Web as a result of modern Web languages, user tagging and annotation, and emerg- ing robust NLP tools. These meaningful, semantic, annotations hold the promise to significantly enhance information access, by enhancing the depth of analysis of today’s systems. Currently, we have only started exploring the possibilities and only begin to un- derstand how these valuable semantic cues can be put to fruitful use. Unleashing the potential of semantic annotations requires us to think outside the box, by combining the insights of natural lan- guage processing (NLP) to go beyond bags of words, the insights of databases (DB) to use structure efficiently even when aggregating over millions of records, the insights of information retrieval (IR) in effective goal-directed search and evaluation, and the insights of knowledge management (KM) to get grips on the greater whole. This workshop aims to bring together researchers from these dif- ferent disciplines and work together on one of the greatest chal- lenges in the years to come. The desired result of the workshop will be to gain concrete insight into the potential of semantic an- notations, and in concrete steps to take this research forward; to synchronize related research happening in NLP, DB, IR, and KM, in ways that combine the strengths of each discipline; and to have a lively, interactive workshop where every participant contributes actively and which inspires attendees to think freely and creatively, working towards a common goal.

  • Jerky driving : an indicator of accident proneness?

    • This study uses continuously logged driving data from 166 private cars to derive the level of jerks caused by the drivers during everyday driving. The number of critical jerks found in the data is analysed and compared with the self-reported accident involvement of the drivers. The results show that the expected number of accidents for a driver increases with the number of critical jerks caused by the driver. Jerk analyses make it possible to identify safety critical driving behaviour or “accident prone” drivers. They also facilitate the development of safety measures such as active safety systems or advanced driver assistance systems, ADAS, which could be adapted for specific groups of drivers or specific risky driving behaviour.

  • From PDE3B to the regulation of energy homeostasis.

    • The incidence of obesity in the developed world is increasing at an alarming rate. Concurrent with the increase in the incidence of obesity is an increase in the incidence of type 2 diabetes. Cyclic AMP (cAMP) and cGMP are key second messengers in all cells; for example, when it comes to processes of relevance for the regulation of energy metabolism, cAMP is a key mediator in the regulation of lipolysis, glycogenolysis, gluconeogenesis and pancreatic β cell insulin secretion. PDE3B, one of several enzymes which hydrolyze cAMP and cGMP, is expressed in cells of importance for the regulation of energy homeostasis, including adipocytes, hepatocytes, hypothalamic cells and β cells. It has been shown, using PDE3 inhibitors and gene targeting approaches in cells and animals, that altered levels of PDE3B result in a number of changes in the regulation of glucose and lipid metabolism and in overall energy homeostasis. This article highlights the complexity involved in the regulation of PDE3B by hormones, and in the regulation of downstream metabolic effects by PDE3B in several interacting tissues.
