    • The effect of the addition of chlorine and/or sulfur to the fuel on fly ash composition, deposit formation, and superheater corrosion has been studied during biomass combustion in a circulating fluidized-bed boiler. The chlorine (HCl (aq)) and sulfur (SO2 (g)) were added in proportions of relevance for the potassium chemistry. The composition of the bottom and the fly ashes was analyzed. Gas and particle measurements were performed downstream of the cyclone before the convection pass and the flue gas composition was recorded in the stack with a series of standard instruments and an FTIR analyzer. At the position downstream of the cyclone, a deposit probe was situated, simulating a superheater tube. Deposits on the probe and initial corrosion were examined. It is concluded that addition of sulfur and chlorine increases the formation of submicron particles leading to deposition of potassium sulfate and chloride. The results compare well with earlier work based on laboratory-scale experiments concerning effects of chlorine and sulfur on potassium chemistry.

  • STAT1 signaling is associated with acquired crossresistance to doxorubicin and radiation in myeloma cell lines

    • The myeloma cell line RPMI 8226/S and its doxorubicin resistant subline 8226/Dox40 were used as models to explore the potential importance of the STAT1 signaling pathway in drug and radiation resistance. The 40-fold doxorubicin resistant subline 8226/Dox40 was found to be crossresistant to single doses of 4 and 8 Gy of radiation. A genome-wide mRNA expression study comparing the 8226/Dox40 cell line to its parental line was performed to identify the underlying molecular mechanisms. Seventeen of the top 50 overexpressed genes have previously been implicated in the STAT1 signaling pathway. STAT1 was over expressed both at the mRNA and protein level. Moreover, analyses of nuclear extracts showed higher abundance of phosphorylated STAT1 (Tyr 701) in the resistant subline. Preexposure of the crossresistant cells to the STAT1 inhibiting drug fludarabine reduced expression of overexpressed genes and enhanced the effects of both doxorubicin and radiation. These results show that resistance to doxorubicin and radiation is associated with increased STAT1 signaling and can be modulated by fludarabine. The data support further development of therapies combining fludarabine and radiation.

    • Objectives: The purpose of this study was to design, validate, and optimize internally standardized real-time quantitative RT-PCR assays and to identify and avoid problems with assay reliability and examine the impact of an exogenous internal standard. Design and methods: The model system consisted of internally standardized quantitative real-time RT-PCR assays specific for PSA and hK2 mRNA based on time-resolved fluorometric detection of lanthanide chelates. Results: Reproducibility was best when large copy numbers (> 5000 per milliliter blood) were analyzed. Addition of an exogenous target-mimicking internal standard had no significant effect on the reproducibility of the method, but increased the calculated copy numbers by an average of 2-fold. Conclusions: We developed an internally standardized, specific and reproducible real-time RT-PCR analysis method for PSA and hK2 mRNA in circulating cells in the bloodstream. Both PSA and hK2 assays are sufficiently sensitive to detect two LNCaP cells per milliliter whole blood. (c) 2006 The Canadian Society of Clinical Chemists. All rights reserved.

  • Wind power development in Sweden : global policies and local obstacles

    • Recently, the Swedish government adopted a national planning goal of a yearly wind power generation of 10 TWh by 2015, implying a substantial increase from the current 0.6 TWh level. In this paper, we provide an economic assessment of the potential for future wind power investments in Sweden in close conjunction with an analysis of the legal, attitudinal and policy-related uncertainties that face a wind mill investor. It is shown that the economics of Swedish wind power is negatively affected by: (a) the lack of policy stability; (b) public criticism at the local level; and, in particular, (c) the legal provisions governing the assessment of the environmental impacts of wind mills and the planning procedures for mill location. While national and global energy policies as well as the general public point out wind power as particularly environmentally friendly, most of the objections to its expansion at the local level tend to have environmental origins. The interests of those who object to wind mill installations gain strong legal protection, and the municipal territorial planning monopoly in Sweden implies that it is hard to make national energy policy goals heard at the local implementation stage. Compared to its competitors, wind power is the technology that tends to have the most to loose from the risk and uncertainties created by this investment environment. The paper identifies and discusses a number of ways in which the national policy interests could be strengthened at the local level. We discuss the role of citizen participation, as well as solutions within the realms of the legal system. Moreover, since the diffusion of wind power encounters the most strident legal and attitudinal obstacles where it interferes with competing land uses, a move offshore appears to be an efficient strategy from the perspective of a wind mill investor. A stronger political commitment to wind power expansion in legal provisions as well as in the form of long-run stability in policy instrument implementation will probably be necessary to attain the 2015 policy goal.
