  • Genome and physiology of the ascomycete filamentous fungus Xeromyces bisporus, the most xerophilic organism isolated to date

    • Xeromyces bisporus can grow on sugary substrates down to 0.61, an extremely low water activity. Its genome size is approximately 22Mb. Gene clusters encoding for secondary metabolites were conspicuously absent; secondary metabolites were not detected experimentally. Thus, in its dry' but nutrient-rich environment, X.bisporus appears to have relinquished abilities for combative interactions. Elements to sense/signal osmotic stress, e.g. HogA pathway, were present in X.bisporus. However, transcriptomes at optimal (approximate to 0.89) versus low a(w) (0.68) revealed differential expression of only a few stress-related genes; among these, certain (not all) steps for glycerol synthesis were upregulated. Xeromyces bisporus increased glycerol production during hypo- and hyper-osmotic stress, and much of its wet weight comprised water and rinsable solutes; leaked solutes may form a protective slime. Xeromyces bisporus and other food-borne moulds increased membrane fatty acid saturation as water activity decreased. Such modifications did not appear to be transcriptionally regulated in X.bisporus; however, genes modulating sterols, phospholipids and the cell wall were differentially expressed. Xeromyces bisporus was previously proposed to be a chaophile', preferring solutes that disorder biomolecular structures. Both X.bisporus and the closely related xerophile, Xerochrysium xerophilum, with low membrane unsaturation indices, could represent a phylogenetic cluster of chaophiles'.

  • Fast-Time and Slow-Time Space-Time Adaptive Processing for Bistatic Radar Interference Suppression

    • In bistatic radar with transmitter and receiver geographically separated the interference from ground clutter and the direct path signal transmitter-receiver will be strong and must be suppressed in order to detect the target. We apply FT-STAP (Fast-Time Space Time Adaptive Processing) to the suppression, which is unusual, and compare with conventional ST-STAP (Slow-Time STAP) by simulations in order to see whether FT-STAP is an alternative to ST-STAP. The performance of FT-STAP is much worse than of ST-STAP. We give an explanation for this, which also may be the basis for large improvements of FT-STAP. Moreover, we suggest two new performance measures.

    • In this paper we propose a new change detection (CD) algorithm based on the Bayes theorem and probability assignments. Differently from any kind of likelihood ratio test (LRT) algorithms, the proposed algorithm does not present target alarms, but the probability of certain image position is a target position. In other words, the proposed method leads to quantitative estimates on the probability of a target at any pixel, whereas LRT algorithms can only be used as a figure of merit for any pixel to contain a target.

    • BackgroundPyometra often induces systemic inflammatory response syndrome (SIRS) and early diagnosis is crucial for survival. Chromogranin A (CgA) is a neuroendocrine secretory protein that is co-released with catecholamines from the adrenal medulla and sympathetic nerve endings. A prognostic value of CgA has been found in humans that are critically ill or that have SIRS associated with infection. CgA has not yet been studied in dogs with bacterial infection. The aim of the study was to investigate CgA, measured by Chromogranin A361-372 (Catestatin; Cst) and Chromogranin A17-38 (Vasostatin; VS) in healthy dogs and in dogs with pyometra.ResultsFifty dogs with pyometra, sampled prior to surgery and 64 healthy female dogs were included. In 19 pyometra cases, blood samples were also collected postoperatively. Concentrations of Cst and VS were measured in heparinised plasma and Cst also measured in EDTA plasma, by in-house radioimmunoassays. Student¿s t-test and Wilcoxon two-sample test was used to test for differences between dog groups. Pre- and postoperative samples in dogs with pyometra were analysed by paired t-test. Pearson correlation was used to investigate associations of laboratory variables and hospitalization. P < 0.05 was considered significant.Concentrations of Cst were decreased in pyometra dogs (mean ± SE, 1.01 ± 0.05 nmol/L) compared to healthy dogs (mean ± SE, 1.70 ± 0.03 nmol/L) (p ¿ 0.0001). VS concentrations did not differ significantly between dogs with pyometra (0.40 ± 0.04 nmol/L) and healthy dogs (0.42 ± 0.03 nmol/L). Mean ± SE pre- and postoperative concentration of Cst (1.0 ± 0.04 nmol/L and 0.9 ± 0.2 nmol/L) and VS (0.36 ± 0.04 nmol/L and 0.36 ± 0.04 nmol/L) in dogs with pyometra did not differ significantly. Neither Cst nor VS concentrations were associated with duration of hospitalization and were not significantly different in the four dogs with pyometra that had prolonged (¿3 d) postoperative hospitalization.ConclusionConcentrations of Cst, but not VS, were decreased in pyometra. Cst and VS concentrations before and after ovariohysterectomy did not differ significantly and were not associated with duration of hospitalization. Further studies are warranted to evaluate a possible diagnostic or prognostic value for Cst and VS.

    • SUMMARY:High-throughput genotyping and sequencing technologies facilitate studies of complex genetic traits and provide new research opportunities. The increasing popularity of genome-wide association studies (GWAS) leads to the discovery of new associated loci and a better understanding of the genetic architecture underlying not only diseases, but also other monogenic and complex phenotypes. Several softwares are available for performing GWAS analyses, R environment being one of them.RESULTS: We present cgmisc, an R package that enables enhanced data analysis and visualisation of results from GWAS. The package contains several utilities and modules that complement and enhance the functionality of the existing software. It also provides several tools for advanced visualisation of genomic data and utilises the power of the R language to aid in preparation of publication-quality figures.     • This paper presents an analysis of pre-filtered clutter VHF SAR images. The image data are reorganized into sub-vectors based on the observation of the image-pair magnitude samples. Based on this approach, we present a statistical description of the SAR clutter obtained by the subtraction between two real SAR images. The statistical analysis based on bivariate distribution data organized into different intervals of magnitude can be an important tool to further understand the properties of the backscattered signal, which can be a valuable premise for change detection processing.

    • Ultrawideband (UWB) synthetic aperture radar (SAR) as a tool for oil well perforation monitoring is experimentally investigated in this paper. Experiments are conducted on a lab prototype that emulates the small section of an oil well. Oil well perforations are scanned using the designed UWB radar system for different experimental conditions. High-resolution images in both down-range and cross-range are obtained by using UWB pulses and signal processing. High-quality images are constructed using SAR processing algorithm with the proposed modifications. Experiments are carried out for different perforation conditions such as open, partially clogged and fully clogged cases. It is shown by the experiments that the proposed radar technique and the data processing methods are capable of providing more revealing high-quality images. This approach can be applied effectively to monitor the oil well medium and detect well impairment locations.

    • We report results from high-resolution studies of D-5(+) cluster ion collisions with low-energy electrons performed in a heavy ion storage ring. Absolute dissociative recombination (DR) and dissociative excitation (DE) cross sections were determined for the energy range from 0.0005 to 20eV. The DR cross sections were exceedingly large at low energies, and DR resulted in efficient internal energy redistribution and pronounced fragmentation with two main product channels: D-2+3D (0.62 +/- 0.03) and 2D(2)+D (0.35 +/- 0.01). The DR and DE cross sections were comparable in the energy range from 0.2 to 20eV, which suggest that the two processes follow similar dynamics and are competing outcomes of the ion-electron interaction.     • Here, we discuss the potential role of capacitating epistasis in the genetic architecture of complex traits. Two alternative methods for identifying such gene-gene interactions in genetic association studies-mapping of variance controlling loci and the variance plane ratio (VPR) method-are introduced. An overview of the theoretical foundation of the methods is presented together with a discussion on their implementation and available software for performing these analyses. We conclude by highlighting a few examples of capacitating epistasis described in the literature and its potential impacts on the genetics of complex traits.
