    • HopQ is an outer-membrane protein of Helicobacter pylori that binds to human carcinoembryonic antigen-related cell-adhesion molecules (CEACAMs) with high specificity. We aimed to investigate fluorescence targeting of CEACAM-expressing colorectal tumors in patient-derived orthotopic xenograft (PDOX) models with fluorescently labeled recombinant HopQ (rHopQ). Western blotting, flow cytometry and ELISA were performed to determine the efficiency of rHopQ binding to CEACAMs. rHopQ was conjugated to IR800DyeCW (rHopQ-IR800). Nude mice received orthotopic implantation of colon cancer tumors. Three weeks later, mice were administered 25 μg or 50 μg HopQ-IR800 and imaged 24 or 48 h later. Intravital images were analyzed for tumor-to-background ratio (TBR). Flow cytometry and ELISA demonstrated binding of HopQ to CEACAM1, 3 and 5. Dose-response intravital imaging in PDOX models demonstrated optimal results 48 h after administration of 50 μg rHopQ-IR800 (TBR = 3.576) in our protocol. Orthotopic models demonstrated clear tumor margins of primary tumors and small regional metastases with a mean TBR = 3.678 (SD ± 1.027). rHopQ showed specific binding to various CEACAMs in PDOX models. rHopQ may be useful for CEACAM-positive tumor and metastasis detection for pre-surgical diagnosis, intra-operative imaging and fluorescence-guided surgery.

  • Towards a modest infrastructural imaginary? Sanitation in Kampala beyond the modern infrastructure ideal

    • The idea of the modern city continues to shape urban policies and practices, yet there has long been conflict over its meaning and relevance, particularly in southern cities. Alternative imaginaries, however, are partial and/or insufficiently detailed. In this paper, we first examine the modernness of modern urban infrastructure in the context of ongoing shifts away from modern ideals. Then, inspired by Gibson-Graham’s process of reading for difference, we purposefully look for fragments of what we call an emergent ‘modest sociotechnical imaginary’ in order to contribute to a fuller description of a non-modern imaginary. We examine policies and practices of sanitation in Kampala, where narratives make space for sanitation beyond the grid, seeking to improve sanitation without a predetermined teleological end. What this opening means, however, is the subject of ongoing contestations. Working between modern and anti-modern imaginaries means finding ways to choose technologies and legitimate, permit, govern, regulate, finance and distribute the costs and benefits of them with limited knowledge and control. In Kampala, we find evidence of both i) modest acceptance of limits and uncertainty and ii) efforts to layer modernist urges to create populations, know and control into emergent infrastructures. In this context, Kampala is not framed as an instantiation of a clearly defined alternative to the modern. Instead, we suggest the questions and contestations arising in Kampala are evidence of the coexistence of a modest imaginary, and that such examinations help us to deepen our understanding of ongoing struggles over what infrastructure is and ought to be.

  • Modelling and Optimization Methods of Integrated Production Planning for Steel Plate Mill with Flexible Customization

    • With diversified requirements and varying manufacturing environments, the optimal production planning encountered in a steel mill became more flexible and also complicated. The flexibility can provide the operators with auxiliary requirements through an implementable integrated production planning. In this paper, a mixed-integer nonlinear programming (MINLP) model is formulated for the optimal planning that incorporates various manufacturing constraints and the flexibility in a steel plate mill. Furthermore, two solution strategies are developed to overcome the weakness of solving MINLP problem directly. The first one is to transform the original MINLP formulation to an approximate mixed integer linear programming (MILP) using a classic linearization method. The second one is to decompose the original model using a branch-and-bound based iterative method. Computational experiments on various problem instances are presented in terms of the effectiveness and applicability. The result shows that the proposed second method performs better in computational efforts and solution accuracy.

    • The sorption of sodium silicate by synthetic magnetite (Fe3O4) at different pH conditions (pH 7-11) and initial silicate concentrations (1×10-3 and 10 x 10-3 mol L-1) was studied using in situ attenuated total reflection Fourier transform infrared (ATR-FTIR) spectroscopy. The analysis of infrared spectra of sodium silicate in solution as well as adsorbed on magnetite nano-particles clearly showed the evolution of different silicate species depending on pH and silica concentration. The silicate concentration studied (10 x 10-3 mol L-1 ) contained polymeric or condensed silicate species at lower pH as well as monomers at high pH, as evident from infrared spectra. Condensation of monomers resulted in an increased intensity of absorptions in the high frequency part (>1050 cm-1) of the spectral region, which contains information about both silicate in solution and sorbed silicate viz. 1300 cm-1-850 cm-1. In the pH range studied, infrared spectra of sorbed silicate and sorbed silicate during desorption both indicated the presence of different types of surface complexes at the magnetite surface. The sorption mechanism proposed is in accordance with a ligand exchange reaction where both monodentate and bidentate complexes could exist at low surface loading level, the relative proportion of the complexes being due to both pH and concentration in solution. Oligomerization occurred on the magnetite surface at higher surface loading.
