  • Multi-ancestry genetic study of type 2 diabetes highlights the power of diverse populations for discovery and translation

    • We assembled an ancestrally diverse collection of genome-wide association studies (GWAS) of type 2 diabetes (T2D) in 180,834 affected individuals and 1,159,055 controls (48.9% non-European descent) through the Diabetes Meta-Analysis of Trans-Ethnic association studies (DIAMANTE) Consortium. Multi-ancestry GWAS meta-analysis identified 237 loci attaining stringent genome-wide significance (P < 5 x 10(-9)), which were delineated to 338 distinct association signals. Fine-mapping of these signals was enhanced by the increased sample size and expanded population diversity of the multi-ancestry meta-analysis, which localized 54.4% of T2D associations to a single variant with >50% posterior probability. This improved fine-mapping enabled systematic assessment of candidate causal genes and molecular mechanisms through which T2D associations are mediated, laying the foundations for functional investigations. Multi-ancestry genetic risk scores enhanced transferability of T2D prediction across diverse populations. Our study provides a step toward more effective clinical translation of T2D GWAS to improve global health for all, irrespective of genetic background. Genome-wide association and fine-mapping analyses in ancestrally diverse populations implicate candidate causal genes and mechanisms underlying type 2 diabetes. Trans-ancestry genetic risk scores enhance transferability across populations.

  • Interfacial defective Ti3+ on Ti/TiO2 as visible-light responsive sites with promoted charge transfer and photocatalytic performance

    • Defect sites on oxide semiconductors play a crucial role in promoting photocatalytiperformance and modulating the bandgap structure of photocatalysts. However, the role of interfacial coordinatively unsaturated defect sites between metal and oxide in photocatalysis is still under debate. So, we designed an experiment to probe the role of interfacial coordinatively unsaturated defect sites. In this work, a series of Ti/TiO2 photocatalysts with varying concentrations of interfacial Ti3+ sites were prepared through an epitaxial growth method under hydrothermal conditions. Through experimental and computational investigations, the roles of interfacial defect sites were discussed in detail. On the one hand, the interfacial coordinatively unsaturated Ti3+ sites could act as visible-light-responsive sites in photocatalytic reactions due to the overlap and hybridization of multiple electronic orbitals. On the other hand, the Ti/TiO2 interface exhibited a certain degree of metallic character near the Fermi level because of the partial delocalization and redistribution of electrons, facilitating the charge migration and separation across the metal-oxide interface. Consequently, the obtained Ti/TiO2 catalysts showed notably enhanced charge transfer efficiency and visible light photocatalytic activity compared to their pristine counterparts. This work may provide a new perspective to interfacial defect engineering in classic metal/oxide heterojunction photocatalysts and figure a more precise direction to synthesize higher effective photocatalysts for environmental governance.

  • Hydrogen-enhanced grain boundary vacancy stockpiling causes transgranular to intergranular fracture transition

    • The attention to hydrogen embrittlement (HE) has been intensified recently in the light of hydrogen as a carbon-free energy carrier. Despite worldwide research, the multifaceted HE mechanism remains a mat-ter of debate. Here we report an atomistic study of the coupled effect of hydrogen and deformation temperature on the pathway to intergranular fracture of nickel. Uniaxial straining is applied to nickel E5(210)[001] and E9(1-10)[22-1] grain boundaries with or without pre-charged hydrogen at various temperatures. Without hydrogen, vacancy generation at grain boundary is limited and transgranular frac-ture mode dominates. When charged, hydrogen as a booster can enhance strain-induced vacancy genera-tion by up to ten times. This leads to the superabundant vacancy stockpiling at the grain boundary, which agglomerates and nucleates intergranular nanovoids eventually causing intergranular fracture. While hy-drogen tends to persistently enhance vacancy concentration, temperature plays an intriguing dual role as either an enhancer or an inhibitor for vacancy stockpiling. These results show good agreement with recent positron annihilation spectroscopy experiments. An S-shaped quantitative correlation between the proportion of intergranular fracture and vacancy concentration was for the first time derived, highlight-ing the existence of a critical vacancy concentration, beyond which fracture mode will be completely intergranular.

  • Highly hydrophilic electrodeposited NiS/Ni3S2 interlaced nanosheets with surface-enriched Ni3+ sites as binder-free flexible cathodes for high-rate hybrid supercapacitors

    • In this study, nanostructured nickel sulfides (NiS, Ni3S2 and NiS/Ni3S2) were fabricated directly on the surface of flexible carbon fiber cloths by simply modifying the deposition parameters of pulse-reversal (PR) electrodeposition method and utilized as binder-free flexible electrodes for aqueous hybrid supercapacitors (SCs). X-ray photoelectron spectroscopy and contact angle measurement studies verifies that the surface of heterostructure NiS/Ni3S2 electrode has enriched Ni3+ sites and highly hydrophilic nature. Consequently, the heterostructure NiS/Ni3S2 electrode demonstrated superior rate capability than that of both single phase NiS and Ni3S2 electrodes. Additionally, the hybrid SC device based on the flexible NiS/Ni3S2 electrode delivered a capacity of 40.4 mAh g−1 at a current density of 2 A g−1 and representing a maximum energy density of 32.3 Wh kg−1 at an impressive power density of 1.6 kW kg−1. Furthermore, the device provided excellent electrochemical stability with a capacity retention of 86.2%, even after a 120-h floating test. Hence, the heterostructure NiS/Ni3S2 with interlaced nanosheets morphology should be considered as promising binder-free flexible electrode materials for next-generation energy storage applications.

    • Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection fatality rate (IFR) doubles with every 5 y of age from childhood onward. Circulating autoantibodies neutralizing IFN-α, IFN-ω, and/or IFN-β are found in ∼20% of deceased patients across age groups, and in ∼1% of individuals aged <70 y and in >4% of those >70 y old in the general population. With a sample of 1,261 unvaccinated deceased patients and 34,159 individuals of the general population sampled before the pandemic, we estimated both IFR and relative risk of death (RRD) across age groups for individuals carrying autoantibodies neutralizing type I IFNs, relative to noncarriers. The RRD associated with any combination of autoantibodies was higher in subjects under 70 y old. For autoantibodies neutralizing IFN-α2 or IFN-ω, the RRDs were 17.0 (95% CI: 11.7 to 24.7) and 5.8 (4.5 to 7.4) for individuals <70 y and ≥70 y old, respectively, whereas, for autoantibodies neutralizing both molecules, the RRDs were 188.3 (44.8 to 774.4) and 7.2 (5.0 to 10.3), respectively. In contrast, IFRs increased with age, ranging from 0.17% (0.12 to 0.31) for individuals <40 y old to 26.7% (20.3 to 35.2) for those ≥80 y old for autoantibodies neutralizing IFN-α2 or IFN-ω, and from 0.84% (0.31 to 8.28) to 40.5% (27.82 to 61.20) for autoantibodies neutralizing both. Autoantibodies against type I IFNs increase IFRs, and are associated with high RRDs, especially when neutralizing both IFN-α2 and IFN-ω. Remarkably, IFRs increase with age, whereas RRDs decrease with age. Autoimmunity to type I IFNs is a strong and common predictor of COVID-19 death.

  • Distinct germline genetic susceptibility profiles identified for common non-Hodgkin lymphoma subtypes

    • Lymphoma risk is elevated for relatives with common non-Hodgkin lymphoma (NHL) subtypes, suggesting shared genetic susceptibility across subtypes. To evaluate the extent of mutual heritability among NHL subtypes and discover novel loci shared among subtypes, we analyzed data from eight genome-wide association studies within the InterLymph Consortium, including 10,629 cases and 9505 controls. We utilized Association analysis based on SubSETs (ASSET) to discover loci for subsets of NHL subtypes and evaluated shared heritability across the genome using Genome-wide Complex Trait Analysis (GCTA) and polygenic risk scores. We discovered 17 genome-wide significant loci (P < 5 × 10−8) for subsets of NHL subtypes, including a novel locus at 10q23.33 (HHEX) (P = 3.27 × 10−9). Most subset associations were driven primarily by only one subtype. Genome-wide genetic correlations between pairs of subtypes varied broadly from 0.20 to 0.86, suggesting substantial heterogeneity in the extent of shared heritability among subtypes. Polygenic risk score analyses of established loci for different lymphoid malignancies identified strong associations with some NHL subtypes (P < 5 × 10−8), but weak or null associations with others. Although our analyses suggest partially shared heritability and biological pathways, they reveal substantial heterogeneity among NHL subtypes with each having its own distinct germline genetic architecture.

  • Genetic loci and prioritization of genes for kidney function decline derived from a meta-analysis of 62 longitudinal genome-wide association studies

    • Estimated glomerular filtration rate (eGFR) reflects kidney function. Progressive eGFR-decline can lead to kidney failure, necessitating dialysis or transplantation. Hundreds of loci from genome-wide association studies (GWAS) for eGFR help explain population cross section variability. Since the contribution of these or other loci to eGFR-decline remains largely unknown, we derived GWAS for annual eGFR-decline and meta-analyzed 62 longitudinal studies with eGFR assessed twice over time in all 343,339 individuals and in high-risk groups. We also explored different covariate adjustment. Twelve genomewide significant independent variants for eGFR-decline unadjusted or adjusted for eGFR- baseline (11 novel, one known for this phenotype), including nine variants robustly associated across models were identified. All loci for eGFR-decline were known for cross-sectional eGFR and thus distinguished a subgroup of eGFR loci. Seven of the nine variants showed variant- by-age interaction on eGFR cross section (further about 350,000 individuals), which linked genetic associations for eGFR-decline with agedependency of genetic cross- section associations. Clinically important were two to four-fold greater genetic effects on eGFR-decline in high-risk subgroups. Five variants associated also with chronic kidney disease progression mapped to genes with functional in- silico evidence (UMOD, SPATA7, GALNTL5, TPPP). An unfavorable versus favorable nine-variant genetic profile showed increased risk odds ratios of 1.35 for kidney failure (95% confidence intervals 1.03- 1.77) and 1.27 for acute kidney injury (95% confidence intervals 1.08-1.50) in over 2000 cases each, with matched controls). Thus, we provide a large data resource, genetic loci, and prioritized genes for kidney function decline, which help inform drug development pipelines revealing important insights into the age-dependency of kidney function genetics.

  • Diffusion-Controlled Lithium Trapping in Graphite Composite Electrodes for Lithium-Ion Batteries

    • Although graphite-based composite electrodes currently are widely used as negative electrodes in lithium-ion batteries due to their good cycle performances, improvements of their long-time cycling stability are still desirable. Herein, a series of lithium-metal half-cell experiments is performed to demonstrate that the diffusion-controlled lithium-trapping effect constitutes an additional, and so far, largely unrecognized, aging mechanism for graphite-based electrodes. This trapping effect, which stems from incomplete delithiation due to diffusion-controlled redistribution of intercalated lithium in graphite, is shown to account for around 30% of the total accumulated capacity loss during long-time cycling. The trapping effect is caused by the concentration gradients present at the end of the lithiation steps as these gradients result in lithium (i.e., coupled Li+ and e(-)) diffusion in the electrodes. As a result, a small fraction of the lithium becomes inaccessible on the timescale of the subsequent delithiation step. The results, however, also show that the inclusion of constant-voltage delithiation steps can increase the delithiation efficiency and decrease the influence of the lithium-trapping effect. This work consequently demonstrates that diffusion-controlled lithium-trapping effects need to be considered when trying to increase the lifetimes of graphite-based electrodes.

    • Chopper amplifiers for biopotential acquisition commonly suppress differential electrode DC offsets by using a DC-servo loop (DSL). However, the noise contribution of the DSL is always neglected in noise analysis. The noise introduced by the DSL, in particular at low frequencies, is of great importance in biosensor applications. This work presents the noise modeling of a current-balancing chopper amplifier with DSL and describes the effect of the DSL on the noise performance. Two different DSL implementations are analyzed. It is found that the exact placement of the chopper in the DSL has a strong impact in the noise performance; therefore, its placement can not be arbitrarily selected. A circuit topology to minimize its noise contribution is then proposed and verified by simulation.
