Complexation of aluminum with DNA.

- The extent of complexation of aluminum(III) with DNA (Calf thymus, Sigma type I) was estimated by means of two experimental techniques: potentiometric titration with a fluoride selective indicator electrode and dialysis followed by aluminum determination by graphite furnace AAS. Both types of experiments indicate that aluminum(III) is bound to DNA. The data are treated by assuming an ion exchange reaction with the phosphate diester groups. Using Rt to denote the concentration of these groups the values of log [AlMn-3R]/(Rt-3[AlMn-3R])[Al3+] decrease from approx. 7.6 to 5.6 when the concentration of sodium chloride is increased from 1 to 100 mM. In the pH range 4.5-5.5 the ion exchange constant increases approximately 0.5 log units. Dialysis gives lower values for the complex formation constant than potentiometry.

- The reactive triazine dye, Cibacron Blue F3G-A, has been covalently attached to microparticulate porous silica and used for the resolution of a number of complementary proteins by high-performance liquid affinity chromatography (HPLAC). Cibacron Blue F3G-A was converted to its 6-aminohexyl derivative by reaction with 1,6-diaminohexane and coupled to γ-glycidoxypropyltrimethoxysilane-activated porous silica (5 μm) to generate an adsorbent containing 5.5–6.7 μmol dye/g silica.Cibacron Blue F3G-A silica columns, in conjunction with on-line monitoring of protein concentration and enzyme activity, may be exploited for the high speed fully automated resolution of dehydrogenases, isoenzymes, kinases and other proteins such as pancreatic ribonuclease A from simple or complex mixtures. The examples demonstrate the versatility of HPLAC on silica-immobilised Cibacron Blue F3G-A.

Gas phase silylation, a rapid method for preparation of high-performance liquid chromatography supports

- A new method for preparing chromatographic supports is described. Porous silica was treated with gaseous silanes and gaseous triethylamine at high temperature and reduced pressure. The silylation procedure was rapid and gave supports with a covalently bound monolayer of aminopropyl-or epoxysilane. Diol silica prepared by gas phase silylation was compared with diol silica prepared in the aqueous or organic phase (toluene). Seven proteins were chromatographed and the protein recovery, plate number and peak asymmetry were calculated. Silica prepared by the gas phase silylation method showed improved performance. The separation of carbonhydrates on aminopropylsilica and the isolation of lactate dehydrogenase with nicotinamide-adenine dinucleotide-silica, prepared from gas phase silylated diol silica, are described.

High ambient concentrations of monoterpenes in a Scandinavian pine forest

- Total monoterpene levels of the order of 100 μg/m3 were assessed in the summer night air of a typical young planted forest of Scots pine (Pinus silvestris) in Sweden. These concentrations are much higher than those reported in previous studies of ambient monoterpenes. Predominant species were α-pinene and 3-carene. Camphene, β-pinene, myrcene, β-phellandrene and limonene were other prominent components determined. The monoterpenes were shown to originate almost exclusively from Scots pine and the proportions between them to be characteristic of this conifer species.

- Neuraminic acid derivatives were separated easily by liquid chromatography on amino phases and using acetonitrile/15 mM phosphate buffer, pH 5.2 as mobile phase. Detection at 200 nm allowed determination of substances in the nanomole range. All solvent systems were used isocratically at room temperature. The separation did not depend on ion-exchange but on a partition mechanism, the solutes being separated according to their polarity. Thus, it was possible to optimize the resolution by varying the stationary phase and the composition of the eluent.

- The effects of divalent metal ions on high affinity uptake glutamate and GABA were examined, using crude and purified synaptosomal fractions prepared from brains of DBA/2CBI. The uptake velocities of both amino acids are severely reduced in the presence of Cu2+, Fe2+ and Zn2+ but remain unaffected by Co2+.
