  • Density-Dependent Effects of Simultaneous Root and Floral Herbivory on Plant Fitness and Defense

    • Plants are attacked by multiple herbivores, and depend on a precise regulation of responses to cope with a wide range of antagonists. Simultaneous herbivory can occur in different plant compartments, which may pose a serious threat to plant growth and reproduction. In particular, plants often face co-occurring root and floral herbivory, but few studies have focused on such interactions. Here, we investigated in the field the combined density-dependent effects of root-chewing cebrionid beetle larvae and flower-chewing pierid caterpillars on the fitness and defense of a semiarid Brassicaceae herb. We found that the fitness impact of both herbivore groups was independent and density-dependent. Increasing root herbivore density non-significantly reduced plant fitness, while the relationship between increasing floral herbivore density and the reduction they caused in both seed number and seedling emergence was non-linear. The plant defensive response was non-additive with regard to the different densities of root and floral herbivores; high floral herbivore density provoked compensatory investment in reproduction, and this tolerance response was combined with aboveground chemical defense induction when also root herbivore density was high. Plants may thus prioritize specific trait combinations in response to varying combined below- and aboveground herbivore densities to minimize negative impacts on fitness.

  • Do nitrogen and phosphorus additions affect nitrogen fixation associated with tropical mosses?

    • Tropical cloud forests are characterized by abundant and biodiverse mosses which grow epiphytically as well as on the ground. Nitrogen (N)-fixing cyanobacteria live in association with most mosses, and contribute greatly to the N pool via biological nitrogen fixation (BNF). However, the availability of nutrients, especially N and phosphorus (P), can influence BNF rates drastically. To evaluate the effects of increased N and P availability on BNF in mosses, we conducted a laboratory experiment where we added N and P, in isolation and combined, to three mosses (Campylopus sp., Dicranum sp. and Thuidium peruvianum) collected from a cloud forest in Peru. Our results show that N addition almost completely inhibited BNF within a day, whereas P addition caused variable results across moss species. Low N2 fixation rates were observed in Campylopus sp. across the experiment. BNF in Dicranum sp. was decreased by all nutrients, while P additions seemed to promote BNF in T. peruvianum. Hence, each of the three mosses contributes distinctively to the ecosystem N pool depending on nutrient availability. Moreover, increased N input will likely significantly decrease BNF associated with mosses also in tropical cloud forests, thereby limiting N input to these ecosystems via the moss-cyanobacteria pathway.

  • Copper Uptake and Its Effects on Two Riparian Plant Species, the Native Urtica dioica, and the Invasive Fallopia japonica

    • Copper accumulating in stream sediments can be transported to adjacent riparian habitats by flooding. Although being an essential element for plants, copper is toxic at high concentrations and restricts, among other things, plant growth. Besides copper, invasive plants, such as Fallopia japonica, which are known to be tolerant toward heavy metals, modify riparian habitats. If the tolerance of F. japonica is higher compared to native plants, this could accelerate invasion under high heavy metal stress. Therefore, we aimed to compare the effect of copper on two common riparian plants, the invasive F. japonica and the native Urtica dioica. We performed a pot experiment with a gradient from 0 to 2430 mg kg(-1) of soil copper. We hypothesized that (i) negative effects on plant growth increase with increasing soil copper concentrations with F. japonica being less affected and (ii) accumulating higher amounts of copper in plant tissues compared to U. dioica. In support of our first hypothesis, growth (height, leaf number) and biomass (above- and belowground) of F. japonica were impacted at the 810 mg kg(-1) treatment, while the growth of U. dioica was already impacted at 270 mg kg(-1). Due to 100% mortality of plants, the 2430 mg kg(-1) treatment was omitted from the analysis. In contrast, chlorophyll content slightly increased with increasing copper treatment for both species. While U. dioica accumulated more copper in total, the copper uptake by F. japonica increased more strongly after exposure compared to the control. In the 810 mg kg(-1) treatment, copper concentrations in F. japonica were up to 2238% higher than in the control but only up to 634% higher in U. dioica. Our results indicate that F. japonica might be able to more efficiently detoxify internal copper concentrations controlling heavy metal effects compared to the native species. This could give F. japonica a competitive advantage particularly in polluted areas, facilitating its invasion success.

  • Genome-Wide Association Analysis of Freezing Tolerance and Winter Hardiness in Winter Wheat of Nordic Origin

    • Climate change and global food security efforts are driving the need for adaptable crops in higher latitude temperate regions. To achieve this, traits linked with winter hardiness must be introduced in winter-type crops. Here, we evaluated the freezing tolerance (FT) of a panel of 160 winter wheat genotypes of Nordic origin under controlled conditions and compared the data with the winter hardiness of 74 of these genotypes from a total of five field trials at two locations in Norway. Germplasm with high FT was identified, and significant differences in FT were detected based on country of origin, release years, and culton type. FT measurements under controlled conditions significantly correlated with overwintering survival scores in the field (r <= 0.61) and were shown to be a reliable complementary high-throughput method for FT evaluation. Genome-wide association studies (GWAS) revealed five single nucleotide polymorphism (SNP) markers associated with FT under controlled conditions mapped to chromosomes 2A, 2B, 5A, 5B, and 7A. Field trials yielded 11 significant SNP markers located within or near genes, mapped to chromosomes 2B, 3B, 4A, 5B, 6B, and 7D. Candidate genes identified in this study can be introduced into the breeding programs of winter wheat in the Nordic region.

  • Novel Pathogen-Plant Host Interaction: Colletotrichum jiangxiense and Fraxinus americana L. (White Ash) in a Sentinel Garden in China

    • Fraxinus americana L. (white ash), a native North American tree commonly cultivated for its ornamental qualities, displayed symptoms of leaf spot disease in a sentinel garden located in Nanjing, Jiangsu, China, in 2022. This disease led to premature leaf shedding, adversely affecting the plant's growth and substantially diminishing its ornamental value. Potential fungal pathogens were isolated from the diseased leaves and the subsequent application of Koch's postulates confirmed the pathogenicity of the fungal isolates (BL-1, BL-2). Through a combination of multi-locus phylogenetic analysis, including ITS, ACT, ApMat, CAL, CHS-1, GAPDH, and TUB2, alongside morphological assessments, the fungus was conclusively identified as Colletotrichum jiangxiense. This represents the first record of C. jiangxiense affecting white ash, highlighting the important role of sentinel gardens in uncovering novel pathogen-plant host interactions.

    • Masterwort, Peucedanum ostruthium (L.) Koch, is an Apiaceae species originally native to the mountain areas of central and southern Europe. Written sources show that it was used in northern Europe. This study explores the cultivation history of masterwort and its past use in Sweden. Although only few details are known about the history of this taxon, it represents a cultural relict plant of an intentionally introduced species known in Sweden as early as the Middle Ages. In Sweden, the masterwort was mainly used as an ethnoveterinary herbal remedy from the seventeenth to nineteenth centuries. However, medicinal manuals, pharmacopoeias and some ethnographical records indicate that it was once also used in remedies for humans. Today, this species remains as a living biocultural heritage in rural areas, especially on the surviving shielings, which were once used as mountain pastures in Dalecarlia, and at former crofts that were inhabited by cattle owners in the forest areas of southern Sweden.

  • Calcium-Dependent Protein Kinase 5 (OsCPK5) Overexpression in Upland Rice (Oryza sativa L.) under Water Deficit

    • Water deficit significantly affects global crop growth and productivity, particularly in water-limited environments, such as upland rice cultivation, reducing grain yield. Plants activate various defense mechanisms during water deficit, involving numerous genes and complex metabolic pathways. Exploring homologous genes that are linked to enhanced drought tolerance through the use of genomic data from model organisms can aid in the functional validation of target species. We evaluated the upland rice OsCPK5 gene, an A. thaliana AtCPK6 homolog, by overexpressing it in the BRSMG Curinga cultivar. Transformants were assessed using a semi-automated phenotyping platform under two irrigation conditions: regular watering, and water deficit applied 79 days after seeding, lasting 14 days, followed by irrigation at 80% field capacity. The physiological data and leaf samples were collected at reproductive stages R3, R6, and R8. The genetically modified (GM) plants consistently exhibited higher OsCPK5 gene expression levels across stages, peaking during grain filling, and displayed reduced stomatal conductance and photosynthetic rate and increased water-use efficiency compared to non-GM (NGM) plants under drought. The GM plants also exhibited a higher filled grain percentage under both irrigation conditions. Their drought susceptibility index was 0.9 times lower than that of NGM plants, and they maintained a higher chlorophyll a/b index, indicating sustained photosynthesis. The NGM plants under water deficit exhibited more leaf senescence, while the OsCPK5-overexpressing plants retained their green leaves. Overall, OsCPK5 overexpression induced diverse drought tolerance mechanisms, indicating the potential for future development of more drought-tolerant rice cultivars.

  • In Vitro Propagation of the Blueberry 'Blue Suede (TM)' (Vaccinium hybrid) in Semi-Solid Medium and Temporary Immersion Bioreactors

    • The production of blueberries for fresh and processed consumption is increasing globally and has more than doubled in the last decade. Blueberry is grown commercially across a variety of climates in over 30 countries. The major classes of plants utilized for the planting and breeding of new cultivars are highbush, lowbush, half-high, Rabbiteye, and Southern highbush. Plants can be propagated by cuttings or in vitro micropropagation techniques. In vitro propagation offers advantages for faster generation of a large number of disease-free plants independent of season. Labor costs for in vitro propagation can be reduced using new cultivation technology and automation. Here, we test and demonstrate successful culture conditions and medium compositions for in vitro initiation, multiplication, and rooting of the Southern highbush cultivar 'Blue Suede (TM)' (Vaccinium hybrid).

  • Evaluation of the Carbohydrate Composition of Crabapple Fruit Tissues Native to Northern Asia

    • A comprehensive comparative analysis of the carbohydrate composition (soluble sugars and pectins) of fruit tissues of Malus baccata, Malus mandshurica, Malus chamardabanica, and Malus sachalinensis, characteristic of the vast territory of Eastern Siberia and the Far East, was carried out. It was shown that a large part of the soluble carbohydrates of the studied species were represented by transport sugars-sorbitol and sucrose. These compounds also provided the main variability in the carbohydrate composition of fruits in the studied material. The polymers pectins and protopectins isolated from the studied fruits were highly methoxylated (up to 60-70%), and their content averaged about 6% of dry weight. The greatest length of pectin polymers was found in the fruit tissues of M. chamardabanica and M. sachalinensis. Data on elemental analysis of fractions of pectins and protopectins of all studied species showed the absence of potentially toxic concentrations of heavy metals. Of note is the rather high content of calcium in both polymer fractions of the four studied species, while its content in protopectin is significantly higher. In addition, in all cases, the presence of low-molecular-weight oligosaccharide molecules with a low-dispersed linear structure was revealed in the tissues of the fruits. It is worth noting that the high content of ascorbic acid was observed in the fruits of all studied species. In addition to being of fundamental interest, information about the carbohydrate composition of the wild Malus species can be useful for apple breeding when choosing sources of genes underlying useful traits.

  • Melatonin Enhances the Photosynthesis and Antioxidant Enzyme Activities of Mung Bean under Drought and High-Temperature Stress Conditions

    • Mung bean, a legume, is sensitive to abiotic stresses at different growth stages, and its yield potential is affected by drought and high-temperature stress at the sensitive stage. Melatonin is a multifunctional hormone that plays a vital role in plant stress defense mechanisms. This study aimed to evaluate the efficiency of melatonin under individual and combined drought and high-temperature stress in mung bean. An experiment was laid out with five treatments, including an exogenous application of 100 & mu;M melatonin as a seed treatment, foliar spray, and a combination of both seed treatment and foliar spray, as well as absolute control (ambient condition) and control (stress without melatonin treatment). Stresses were imposed during the mung bean's reproductive stage (31-40 DAS) for ten days. Results revealed that drought and high-temperature stress significantly decreased chlorophyll index, Fv/Fm ratio, photosynthetic rate, stomatal conductance, and transpiration rate through increased reactive oxygen species (ROS) production. Foliar application of melatonin at 100 & mu;M concentration enhanced the activity of antioxidant enzymes such as superoxide dismutase, catalase, and ascorbate peroxidase and the concentration of metabolites involved in osmoregulation and ion homeostasis; thereby, it improves physiological and yield-related traits in mung bean under individual and combined stress at the reproductive stage.
