  • Catholicity Challenging Ethnicity : An Ecclesiological Study of Congregations and Churches in Post-apartheid South Africa

    • This thesis deals with the relationship between the catholicity of the Church and ethnicity. South African congregations and churches have been studied two decades after the advent of democracy to discern their ecclesiological practices. The Roman Catholic, Anglican, Methodist, Lutheran, and Apostolic Faith Mission churches, which are among the five large historical mainline churches in South Africa by membership, were selected.The first part of this thesis presents an analytical tool that has been derived from key documents of the Faith & Order movement and the World Council of Churches concerning the catholicity of the Church. An operative ecclesiology, originally proposed by Yves Congar and later developed by other theologians, has been used in order to reveal implicit and explicit ecclesiologies.The second part of the thesis tests the catholicity of the Church as described in the analytical tool against an operative ecclesiology in South African congregations and churches twenty years after the dismantling of apartheid. Interviews with church leaders and case studies among sixcongregations located in the three largest municipalities have been carried out. An established body of source material has been analysed through the analytical tool about catholicity.The most important conclusion generated by this thesis is that socio-economic conditions are significant ecclesiological structuring factors. Unequal power relationships, together with peoples’ geographic residential location, determined whether greater inter-ethnic interactions were possible. In general, reconciliation between people of diverse ethnicities was not sought or effected; but the churches’ organisations played a significant role in realising the catholicity of the Church.

    • OBJECTIVES: The study investigates the hypothesis that inflammation in myelofibrosis (MF) like in myeloma and lymphoma, may disturb iron distribution and contribute to anaemia.METHODS: A cross-sectional study of 80 MF and 23 ET patients was performed.RESULTS: About 35% of anaemic MF patients had functional iron deficiency (FID) with transferrin saturation <20 and normal or elevated S-ferritin (<500 µg/L). In ET, FID was rare. In MF patients with FID, 70.6% were anaemic, vs 29.4% in patients without FID (P = 0.03). Hepcidin was significantly higher in MF patients with anaemia, including transfusion-dependent patients, 50.6 vs 24.4 µg/L (P = 0.01). There was a significant negative correlation between Hb and inflammatory markers in all MF patients: IL-2, IL-6 and TNF-α, (P < 0.01-0.03), LD (P = 0.004) and hepcidin (P = 0.03). These correlations were also seen in the subgroup of anaemic MF patients (Table ). Tsat correlated negatively with CRP (P < 0.001). Symptom burden was heavier in MF patients with FID, and MPN-SAF quality of life scores correlated with IL-6 and CRP.CONCLUSIONS: The inflammatory state of MF disturbs iron turnover, FID is common and contributes to anaemia development and impairment of QoL. Anaemic MF patients should be screened for FID.

  • Christianity, Generation and Narrative : Religious Conversion and Change in Sidama, Ethiopia, 1974–2012

    • This study presents narratives concerning religious conversion and change in Evangelical-Pentecostal Christian communities in the Sidama zone, Southern Ethiopia. The study is based on narrative data collected through field research, which mainly focuses on the period 1974–2012. This data is supplemented with previous research and unpublished sources.The study discusses how narratives about religious conversion and change intersect with transformations in Ethiopian society during the period.  It describes and analyses narratives in order to show that individual narratives are part of collective stories. These stories become formative for common experiences and provide a distinct group identity as part of a social generation.The Ethiopian revolution in 1974 was a major turning point. The encounter with the revolutionary state resulted in narratives about challenges and persecution. On the other hand, this situation became the setting for mobility and connections, and created a communal identity between Christians.The stories about the period after the revolution in 1974 are conveyed in the present. After 1991, political transitions in Ethiopia created a new turning point which influenced narratives about religious conversion and change. The Christian communities experienced religious freedom and revitalisation, leading to growth and expansion. But the religious changes also led to competition and differences in and between Evangelical-Pentecostal communities. The changes created negotiation internally in Christian communities about denominational identities and about religious innovations. This setting intersected with an increase of new members who belonged to a younger emerging generation. This sets the study on the boundaries between generations, and where new narratives are renegotiated in the context of social change.The thesis consists of three parts; the first gives the reader an introduction to the research context of the study, a brief historical background and a discussion of methodology. The second part focuses on the period after the Ethiopian revolution in 1974. The last part discusses religious change in the Christian communities after the political transition in 1991, and into present.

  • Flow Cytometric Measurement of Blood Cells with BCR-ABL1 Fusion Protein in Chronic Myeloid Leukemia

    • Chronic myeloid leukemia (CML) is characterized in the majority of cases by a t(9;22)(q34;q11) translocation, also called the Philadelphia chromosome, giving rise to the BCR-ABL1 fusion protein. Current treatment with tyrosine kinase inhibitors is directed against the constitutively active ABL1 domain of the fusion protein, and minimal residual disease (MRD) after therapy is monitored by real-time quantitative PCR (RQ-PCR) of the fusion transcript. Here, we describe a novel approach to detect and enumerate cells positive for the BCR-ABL1 fusion protein by combining the in situ proximity ligation assay with flow cytometry as readout (PLA-flow). By targeting of the BCR and ABL1 parts of the fusion protein with one antibody each, and creating strong fluorescent signals through rolling circle amplification, PLA-flow allowed sensitive detection of cells positive for the BCR-ABL1 fusion at frequencies as low as one in 10,000. Importantly, the flow cytometric results correlated strongly to those of RQ-PCR, both in diagnostic testing and for MRD measurements over time. In summary, we believe this flow cytometry-based method can serve as an attractive approach for routine measurement of cells harboring BCR-ABL1 fusions, also allowing simultaneously assessment of other cell surface markers as well as sensitive longitudinal follow-up.
