  • Comparison of early stages of precipitation in Mo-rich and Mo-poor maraging stainless steels

    • The precipitation process during ageing in the Mo-poor alloy Carpenter 455 has been investigated by three-dimensional atom probe (3DAP) and energy-filtered transmission electron microscopy. The ageing treatment was performed at 475 degrees C for 5 min, 25 min, 1 h and 2 h. The precipitation of a mixed Ni-, Ti- and Cu-rich phase was observed after 25 min of ageing. A complete separation between Ni/Ti-rich and Cu-rich regions occurred within 1-2 h of ageing. After 2 h of ageing at 475 degrees C, coarsening of the eta-Ni3Ti precipitates was observed. Moreover, large Ni/Ti-rich and Cu-rich particles were observed in the lath boundaries after 2 h of ageing. The results were compared with the precipitation process in the Mo-rich alloy 1RK91.   • "I know that women don't like me!" Presuppositions in therapeutic discourse

    • One of the biggest problems concerning presuppositions has been correctly dealing with their sensitivity to the context, i.e. why inferences triggered by certain expressions do not project out in all linguistic environments, even though the triggering words preserve their semantic content in different settings. The answer which is of particular interest here goes along with the principles of the binding theory of presuppositions developed by van der Sandt (1992). According to this theory, presuppositions behave asanaphors and can be resolved in the same way at the level of discourse representation.This article contributes to a very scarce body of empirical work on presuppositions, as it scrutinizes examples of presuppositions that act like discourse anaphors in the context of three psychotherapeutic sessions. Such sessions can be analyzed in the same way as ordinary spoken discourse; however, the initial premise that the usage of presuppositions differs in this genre in comparison to daily interaction is confirmed. The results of both quantitative and qualitative analysis indicate that presuppositions are used for different strategic reasons in the two genres compared, which influences the way they should be interpreted and also their frequency.

  • Differential tissue-specific protein markers of vaginal carcinoma

    • The objective was to identify proteins differentially expressed in vaginal cancer to elucidate relevant cancer-related proteins. A total of 16 fresh-frozen tissue biopsies, consisting of 5 biopsies from normal vaginal epithelium, 6 from primary vaginal carcinomas and 5 from primary cervical carcinomas, were analysed using two-dimensional gel electrophoresis (2-DE) and MALDI-TOF mass spectrometry. Of the 43 proteins identified with significant alterations in protein expression between non-tumourous and tumourous tissue, 26 were upregulated and 17 were downregulated. Some were similarly altered in vaginal and cervical carcinoma, including cytoskeletal proteins, tumour suppressor proteins, oncoproteins implicated in apoptosis and proteins in the ubiquitin-proteasome pathway. Three proteins were uniquely altered in vaginal carcinoma (DDX48, erbB3-binding protein and biliverdin reductase) and five in cervical carcinoma (peroxiredoxin 2, annexin A2, sarcomeric tropomyosin kappa, human ribonuclease inhibitor and prolyl-4-hydrolase beta). The identified proteins imply involvement of multiple different cellular pathways in the carcinogenesis of vaginal carcinoma. Similar protein alterations were found between vaginal and cervical carcinoma suggesting common tumourigenesis. However, the expression level of some of these proteins markedly differs among the three tissue specimens indicating that they might be useful molecular markers.

  • Unexpected regulatory roles of TLR4 and TLR9 in experimental autoimmune encephalomyelitis

    • Innate immune mechanisms essential for priming encephalitogenic T cells in autoimmune neuroinflammation are poorly understood. Experimental autoimmune encephalomyelitis (EAE) is a IL-17-producing Th (Th17) cell-mediated autoimmune disease and an animal model of multiple sclerosis. To investigate how upstream TLR signals influence autoimmune T cell responses, we studied the role of individual TLR and MyD88, the common TLR adaptor molecule, in the initiation of innate and adaptive immune responses in EAE. Wild type (WT) C57BL/6, TLR-deficient and MyD88-deficient mice were immunized with myelin oligodendrocyte glycoprotein (MOG) in CFA. MyD88(-/-) mice were completely EAE resistant. Purified splenic myeloid DC (mDC) from MyD88(-/-) mice expressed much less IL-6 and IL-23, and serum and T cell IL-17 were absent. TLR4(-/-) and TLR9(-/-) mice surprisingly exhibited more severe EAE symptoms than WT mice. IL-6 and IL-23 expression by mDC and Th17 responses were higher in TLR4(-/-) mice, suggesting a regulatory role of TLR4 in priming Th17 cells. IL-6 expression by splenocytes was higher in TLR9(-/-) mice. Our data suggest that MyD88 mediates the induction of mDC IL-6 and IL-23 responses after MOG immunization, which in turn drives IL-17-producing encephalitogenic Th17 cell activation. Importantly, we demonstrate that TLR4 and TLR9 regulate disease severity in MOG-induced EAE.

    • Investigations of precipitation and its development during ageing at 475°C in four maraging steels with different Mo- and Si-contents have been conducted using high resolution techniques such as 3-dimensional atom probe and energy filtered electron microscopy. The precipitation sequence in the steels proved to be quite complex and involved formation of different phases one on the other. Presence of Mo in the steels contributed to a higher complexity of precipitation process and kept the interparticle distance being small up to 100h of ageing. Occurrence of Si promoted formation of Mo-rich precipitates. The influence of phase separation on the mechanical properties of the steels is discussed.
