  • Imaging molecular structure through femtosecond photoelectron diffraction on aligned and oriented gas-phase molecules

    • This paper gives an account of our progress towards performing femtosecond time-resolved photoelectron diffraction on gas-phase molecules in a pump-probe setup combining optical lasers and an X-ray free-electron laser. We present results of two experiments aimed at measuring photoelectron angular distributions of laser-aligned 1-ethynyl-4-fluorobenzene (C8H5F) and dissociating, laser-aligned 1,4-dibromobenzene (C6H4Br2) molecules and discuss them in the larger context of photoelectron diffraction on gas-phase molecules. We also show how the strong nanosecond laser pulse used for adiabatically laser-aligning the molecules influences the measured electron and ion spectra and angular distributions, and discuss how this may affect the outcome of future time-resolved photoelectron diffraction experiments.

  • Genome-wide meta-analysis increases to 71 the number of confirmed Crohn's disease susceptibility loci

    • We undertook a meta-analysis of six Crohn's disease genome-wide association studies (GWAS) comprising 6,333 affected individuals (cases) and 15,056 controls and followed up the top association signals in 15,694 cases, 14,026 controls and 414 parent-offspring trios. We identified 30 new susceptibility loci meeting genome-wide significance (P < 5 × 10⁻⁸). A series of in silico analyses highlighted particular genes within these loci and, together with manual curation, implicated functionally interesting candidate genes including SMAD3, ERAP2, IL10, IL2RA, TYK2, FUT2, DNMT3A, DENND1B, BACH2 and TAGAP. Combined with previously confirmed loci, these results identify 71 distinct loci with genome-wide significant evidence for association with Crohn's disease.

  • X-Ray Diffraction from Isolated and Strongly Aligned Gas-Phase Molecules with a Free-Electron Laser

    • We report experimental results on x-ray diffraction of quantum-state-selected and strongly aligned ensembles of the prototypical asymmetric rotor molecule 2,5-diiodobenzonitrile using the Linac Coherent Light Source. The experiments demonstrate first steps toward a new approach to diffractive imaging of distinct structures of individual, isolated gas-phase molecules. We confirm several key ingredients of single molecule diffraction experiments: the abilities to detect and count individual scattered x-ray photons in single shot diffraction data, to deliver state-selected, e.g., structural-isomer-selected, ensembles of molecules to the x-ray interaction volume, and to strongly align the scattering molecules. Our approach, using ultrashort x-ray pulses, is suitable to study ultrafast dynamics of isolated molecules.

  • The REFUEL EU road map for biofuels in transport: Application of the project’s tools to some short-term policy issues

    • The current hot debate on biofuels calls for a balanced and realistic long-term strategy forbiofuels. The REFUEL project provides several ingredients for such a strategy. Analyses inthis project indicate that domestically produced biofuels can cover a significant share of EUfuel demand in the coming decades, with the EU-12 new member states and Ukraine asmost promising regions. This potential can be realised with residual streams and onexisting agricultural land, without conversion of e.g. nature reserves. Second generationbiofuels are essential for the long-term success of biofuels due to their superior performancein many ways. But generally, the key challenge for the near future would be how toenhance the development of biofuels in a responsible way, i.e. stimulating the productionchains with the best performance, and preventing negative impacts e.g., by paying carefulattention to possible system impacts of biofuel production such as indirect land usechanges and rising food prices. Finally, 2nd generation biofuels require specific policy: theprecursor role of 1st generation is overrated, both in technical terms as well as in their roleas market precursors. When it comes to synergies, 2nd generation biofuels might benefitmore from other developments in the energy sector, such as initiatives in co-firing ofbiomass for (heat and) power, than from 1st generation biofuels, also because of the publicresistance that the latter induce.

  • Genetic risk and a primary role for cell-mediated immune mechanisms in multiple sclerosis

    • Multiple sclerosis is a common disease of the central nervous system in which the interplay between inflammatory and neurodegenerative processes typically results in intermittent neurological disturbance followed by progressive accumulation of disability. Epidemiological studies have shown that genetic factors are primarily responsible for the substantially increased frequency of the disease seen in the relatives of affected individuals, and systematic attempts to identify linkage in multiplex families have confirmed that variation within the major histocompatibility complex (MHC) exerts the greatest individual effect on risk. Modestly powered genome-wide association studies (GWAS) have enabled more than 20 additional risk loci to be identified and have shown that multiple variants exerting modest individual effects have a key role in disease susceptibility. Most of the genetic architecture underlying susceptibility to the disease remains to be defined and is anticipated to require the analysis of sample sizes that are beyond the numbers currently available to individual research groups. In a collaborative GWAS involving 9,772 cases of European descent collected by 23 research groups working in 15 different countries, we have replicated almost all of the previously suggested associations and identified at least a further 29 novel susceptibility loci. Within the MHC we have refined the identity of the HLA-DRB1 risk alleles and confirmed that variation in the HLA-A gene underlies the independent protective effect attributable to the class I region. Immunologically relevant genes are significantly overrepresented among those mapping close to the identified loci and particularly implicate T-helper-cell differentiation in the pathogenesis of multiple sclerosis.

  • Elasticity in evaluations of criminal evidence: Exploring the role of cognitive dissonance

    • Purpose. Previous experiments have demonstrated asymmetrical scepticism in investigators' judgments of criminal evidence – evidence inconsistent (vs. consistent) with the dominant hypothesis about a case is judged as less reliable. In addition, some types of evidence (e.g., witness testimony) are more susceptible to asymmetrical scepticism than others (e.g., DNA evidence), indicating varying degrees of elasticity. This article proposes that inconsistent evidence arouses cognitive dissonance, and that the dissonance can be reduced through either asymmetrical scepticism (for high-elasticity evidence) or belief change (for low-elasticity evidence). The hypotheses are tested in two experiments. Methods. In both experiments, law students made a preliminary judgment about the guilt of a suspect in a homicide case, and subsequently received a piece of DNA or witness evidence which was either consistent or inconsistent with the preliminary judgment. The extent to which participants changed their guilt judgments, judged the additional evidence as reliable, and felt dissonance served as the main dependent variables. Results. Inconsistent (vs. consistent) evidence did arouse stronger dissonance, but only for witness (and not DNA) evidence. Experienced dissonance (Experiment 1) and dissonance reduction (Experiment 2) accounted for the effect of the evidence on changes in guilt judgments, but not for the effect on reliability judgments. The greatest dissonance reduction was observed among participants who received inconsistent witness evidence but did not change their guilt judgments accordingly. Conclusions. It appears that dissonance plays a significant, although complex, role in investigative judgments of guilt and reliability. Alternative dissonance-reducing mechanisms that can account for the findings and practical implications are discussed.

  • On the Ease of (Dis)believing: The Role of Accessibility Experiences in Credibility Judgments

    • Credibility judgments are common and consequential in many applied settings. Although much research has addressed human observers’ ability to discriminate true and deceptive statements, less is known about the psychological processes involved in such judgments. Here, it is proposed that the process of mustering evidence for or against credibility is reflected in a feeling-based form (ease-of-retrieval) and that such feelings can be used as a basis for credibility judgments. The results of an experiment show, as predicted, that the perceived ease with which participants could identify clues strongly influenced credibility judgments. Ironically, mustering more clues in support of a truthful account lowered credibility judgments; in contrast, mustering more clues in support of a deceptive account increased credibility judgments. Mediation analyses suggest that this is because participants relied on a feeling-based as opposed to content-based judgment strategy. Practical implications are discussed, and theoretical issues regarding the process of credibility judgment are raised.
