  • Failed Blade Implant After 25 Years in Function: Case Description and Histologic Analysis.

    • This report describes a failed blade implant after 25 years in function in the left maxilla and the results of histologic analysis of peri-implant tissues. The blade was retrieved, and histologic findings revealed that it was not osseointegrated despite being successfully in function for so many years. The long-term function of blade implants without osseointegration can be attributed to biotolerance exhibited by the implant rather than actual osseointegration.

  • Micropatterned Scaffolds with Immobilized Growth Factor Genes Regenerate Bone and Periodontal Ligament-Like Tissues.

    • Periodontal disease destroys supporting structures of teeth. However, tissue engineering strategies offer potential to enhance regeneration. Here, the strategies of patterned topography, spatiotemporally controlled growth factor gene delivery, and cell-based therapy to repair bone-periodontal ligament (PDL) interfaces are combined. Micropatterned scaffolds are fabricated for the ligament regions using polycaprolactone (PCL)/polylactic-co-glycolic acid and combined with amorphous PCL scaffolds for the bone region. Scaffolds are modified using chemical vapor deposition, followed by spatially controlled immobilization of vectors encoding either platelet-derived growth factor-BB or bone morphogenetic protein-7, respectively. The scaffolds are seeded with human cells and delivered to large alveolar bone defects in athymic rats. The effects of dual and single gene delivery with and without micropatterning are assessed after 3, 6, and 9 weeks. Gene delivery results in greater bone formation at three weeks. Micropatterning results in regenerated ligamentous tissues similar to native PDL. The combination results in more mature expression of collagen III and periostin, and with elastic moduli of regenerated tissues that are statistically indistinguishable from those of native tissue, while controls are less stiff than native tissues. Thus, controlled scaffold microtopography combined with localized growth factor gene delivery improves the regeneration of periodontal bone-PDL interfaces.

    • Reliable prediction of evaporative losses from reservoirs is an essential component of reservoir management and operation. Conventional models generally used for evaporation prediction have a number of drawbacks as they are based on several assumptions. A novel approach called the co-active neuro-fuzzy inference system (CANFIS) is proposed in this study for the modeling of evaporation from meteorological variables. CANFIS provides a center-weighted set rather than global weight sets for predictor–predictand relationship mapping and thus it can provide a higher prediction accuracy. In the present study, adjustments are made in the back-propagation algorithm of CANFIS for automatic updating of membership rules and further enhancement of its prediction accuracy. The predictive ability of the CANFIS model is validated with three well-established artificial intelligence (AI) models. Different statistical metrics are computed to investigate the prediction efficacy. The results reveal higher accuracy of the CANFIS model in predicting evaporation compared to the other AI models. CANFIS is found to be capable of modeling evaporation from mean temperature and relative humidity only, with a Nash–Sutcliffe efficiency of 0.93, which is much higher than that of the other models. Furthermore, CANFIS improves the prediction accuracy by 9.2–55.4% compared to the other AI models.

    • Leptin and ghrelin are important regulators of energy homeostasis in mammals, whereas their physiological-roles in fish have not been fully elucidated. In the present study, the effects of leptin and ghrelin on adipogenesis, lipolysis and on expression of lipid metabolism-related genes were examined in rainbow trout adipocytes in vitro. Leptin expression and release increased from preadipocytes to mature adipocytes in culture, but did not affect the process of adipogenesis. While ghrelin and its receptor were identified in cultured differentiated adipocytes, ghrelin did not influence either preadipocyte proliferation or differentiation, indicating that it may have other adipose-related roles. Leptin and ghrelin increased lipolysis in mature freshly isolated adipocytes, but mRNA expression of lipolysis markers was not significantly modified. Leptin significantly suppressed the fatty acid transporter-1 expression, suggesting a decrease in fatty acid uptake and storage, but did not affect expression of any of the lipogenesis or beta-oxidation genes studied. Ghrelin significantly increased the mRNA levels of lipoprotein lipase, fatty acid synthase and peroxisome proliferator-activated receptor-beta, and thus appears to stimulate synthesis of triglycerides as well as their mobilization. Overall, the study indicates that ghrelin, but not leptin seems to be an enhancer of lipid turn-over in adipose tissue of rainbow trout, and this regulation may at least partly be mediated through autocrine/paracrine mechanisms. The mode of action of both hormones needs to be further explored to better understand their roles in regulating adiposity in fish. (C) 2015 Elsevier Inc. All rights reserved.
