    • The erosion of Be and W marker layers was investigated using long-term samples containing marker layers during the second ITER-like wall discharge campaign 2013-2014 (ILW-2). The samples were mounted in Be coated Inconel tiles between the inner wall guard limiters (IWGL). They were analyzed using elastic backscattering (EBS) before and after exposure. All samples showed noticeable erosion. The results were compared to the data for Be and W erosion rates for the first 2011-2012 JET ITER-like wall (ILW-1) campaign, and to the data for C erosion during the 2005-2009 campaign when JET was operated with a carbon wall. The mean W erosion rates and the toroidal and poloidal distributions of the W erosion were nearly the same for the ILW-1 and ILW-2 campaigns. The mean erosion rate of Be during the ILW-2 campaign was smaller by a factor of about two compared to the ILW-1 campaign.

    • The increased interest in using monoclonal antibodies (mAbs) as a platform for biopharmaceuticals has led to the need for new analytical techniques that can precisely assess physicochemical properties of these large and very complex drugs for the purpose of correctly identifying quality attributes (QA). One QA, higher order structure (HOS), is unique to biopharmaceuticals and essential for establishing consistency in biopharmaceutical manufacturing, detecting process-related variations from manufacturing changes and establishing comparability between biologic products. To address this measurement challenge, two-dimensional nuclear magnetic resonance spectroscopy (2D-NMR) methods were introduced that allow for the precise atomic-level comparison of the HOS between two proteins, including mAbs. Here, an inter-laboratory comparison involving 26 industrial, government and academic laboratories worldwide was performed as a benchmark using the NISTmAb, from the National Institute of Standards and Technology (NIST), to facilitate the translation of the 2D-NMR method into routine use for biopharmaceutical product development. Two-dimensional H-1,N-15 and H-1,C-13 NMR spectra were acquired with harmonized experimental protocols on the unlabeled Fab domain and a uniformly enriched-N-15, 20%-C-13-enriched system suitability sample derived from the NISTmAb. Chemometric analyses from over 400 spectral maps acquired on 39 different NMR spectrometers ranging from 500 MHz to 900 MHz demonstrate spectral fingerprints that are fit-for-purpose for the assessment of HOS. The 2D-NMR method is shown to provide the measurement reliability needed to move the technique from an emerging technology to a harmonized, routine measurement that can be generally applied with great confidence to high precision assessments of the HOS of mAb-based biotherapeutics.

  • Erosion and deposition in the JET divertor during the first ILW campaign

    • Erosion and deposition were studied in the JET divertor during the first JET ITER-like wall campaign 2011 to 2012 using marker tiles. An almost complete poloidal section consisting of tiles 0, 1, 3, 4, 6, 7, 8 was studied. The data from divertor tile surfaces were completed by the analysis of samples from remote divertor areas and from the inner wall cladding. The total mass of material deposited in the divertor decreased by a factor of 4-9 compared to the deposition of carbon during all-carbon JET operation before 2010. Deposits in 2011 to 2012 consist mainly of beryllium with 5-20 at.% of carbon and oxygen, respectively, and small amounts of Ni, Cr, Fe and W. This decrease of material deposition in the divertor is accompanied by a decrease of total deuterium retention inside the JET vessel by a factor of 10 to 20. The detailed erosion/deposition pattern in the divertor with the ITER-like wall configuration shows rigorous changes compared to the pattern with the all-carbon JET configuration.

  • Efficacy and safety of sodium oxybate in alcohol-dependent patients with a very high drinking risk level

    • Medication development for alcohol relapse prevention or reduction of consumption is highly challenging due to methodological issues of pharmacotherapy trials. Existing approved medications are only modestly effective with many patients failing to benefit from these therapies. Therefore, there is a pressing need for other effective treatments with a different mechanism of action, especially for patients with very high (VH) drinking risk levels (DRL) because this is the most severely affected population of alcohol use disorder patients. Life expectancy of alcohol-dependent patients with a VH DRL is reduced by 22 years compared with the general population and approximately 90000 alcohol-dependent subjects with a VH DRL die prematurely each year in the EU (Rehm et al. 2018). A promising new medication for this population is sodium oxybate, a compound that acts on GABA(B) receptors and extrasynaptic GABA(A) receptors resulting in alcohol-mimetic effects. In this article, a European expert group of alcohol researchers and clinicians summarizes data (a) from published trials, (b) from two new-as yet unpublished-large clinical trials (GATE 2 (n = 314) and SMO032 (n = 496), (c) from post hoc subgroup analyses of patients with different WHO-defined DRLs and (d) from multiple meta-analyses. These data provide convergent evidence that sodium oxybate is effective especially in a subgroup of alcohol-dependent patients with VH DRLs. Depending on the study, abstinence rates are increased up to 34 percent compared with placebo with risk ratios up to 6.8 in favor of sodium oxybate treatment. These convergent data are supported by the clinical use of sodium oxybate in Austria and Italy for more than 25years. Sodium oxybate is the sodium salt of gamma-hydroxybutyric acid that is also used as a recreational (street) drug suggestive of abuse potential. However, a pharmacovigilance database of more than 260000 alcohol-dependent patients treated with sodium oxybate reported very few adverse side effects and only few cases of abuse. We therefore conclude that sodium oxybate is an effective, well-tolerated and safe treatment for withdrawal and relapse prevention treatment, especially in alcohol-dependent patients with VH DRL.
