  • The plasma concentration of HDL-associated apoM is influenced by LDL receptor-mediated clearance of apoB-containing particles

    • ApoM is mainly associated with HDL. Nevertheless, we have consistently observed positive correlations of apoM with plasma LDL cholesterol in humans. Moreover, LDL receptor deficiency is associated with increased plasma apoM in mice. Here, we tested the idea that plasma apoM concentrations are affected by the rate of LDL receptor-mediated clearance of apoB-containing particles. We measured apoM in humans each carrying one of three different LDL receptor mutations (n = 9) or the apoB3500 mutation (n = 12). These carriers had increased plasma apoM (1.34 +/- 0.13 mu M, P = 0.003, and 1.23 +/- 0.10 mu M, P = 0.02, respectively) as compared with noncarriers (0.93 +/- 0.04 mu M). When we injected human apoM-containing HDL into Wt (n = 6) or LDL receptor-deficient mice (n = 6), the removal of HDL-associated human apoM was delayed in the LDL receptor-deficient mice. After 2 h, 54 +/- 5% versus 90 +/- 8% (P < 0.005) of the initial amounts of human apoM remained in the plasma of Wt and LDL receptor-deficient mice, respectively. Finally, we compared the turnover of radio-iodinated LDL and plasma apoM concentrations in 45 normocholesterolemic humans. There was a negative correlation between plasma apoM and the fractional catabolic rate of LDL (r = -0.38, P = 0.009). These data suggest that the plasma clearance of apoM, despite apoM primarily being associated with HDL, is influenced by LDL receptor-mediated clearance of apoB-containing particles.-Christoffersen, C., M.   • Oral Polio Vaccine Influences the Immune Response to BCG Vaccination. A Natural Experiment

    • Background: Oral polio vaccine (OPV) is recommended to be given at birth together with BCG vaccine. While we were conducting two trials including low-birth-weight (LBW) and normal-birth-weight (NBW) infants in Guinea-Bissau, OPV was not available during some periods and therefore some infants did not receive OPV at birth, but only BCG. We investigated the effect of OPV given simultaneously with BCG at birth on the immune response to BCG vaccine. Methods and Findings: We compared the in vitro and the in vivo response to PPD in the infants who received OPV and BCG with that of infants who received BCG only. At age 6 weeks, the in vitro cytokine response to purified protein derivate (PPD) of M. Tuberculosis was reduced in LBW and NBW infants who had received OPV with BCG. In a pooled analysis receiving OPV with BCG at birth was associated with significantly lower IL-13 (p = 0.041) and IFN-gamma (p = 0.004) and a tendency for lower IL-10 (p = 0.054) in response to PPD. Furthermore, OPV was associated with reduced in vivo response to PPD at age 2 months, the prevalence ratio (PR) of having a PPD reaction being 0.75 (0.58-0.98), p = 0.033, and with a tendency for reduced likelihood of having a BCG scar (0.95 (0.91-1.00), p = 0.057)). Among children with a scar, OPV was associated with reduced scar size, the regression coefficient being -0.24 (-0.43-0.05), p = 0.012. Conclusions: This study is the first to address the consequences for the immune response to BCG of simultaneous administration with OPV. Worryingly, the results indicate that the common practice in low-income countries of administering OPV together with BCG at birth may down-regulate the response to BCG vaccine.

  • Two type Ic supernovae in low-metallicity, dwarf galaxies : diversity of explosions

    • Aims: The aim of this paper is to discuss the nature of two type Ic supernovae SN 2007bg and SN 2007bi and their host galaxies. Both supernovae were discovered in wide-field, non-targeted surveys and are found to be associated with sub-luminous blue dwarf galaxies identified in SDSS images. Methods: We present BVRI photometry and optical spectroscopy of SN 2007bg and SN 2007bi and their host galaxies. Their lightcurves and spectra are compared to those of other type Ic SNe and analysis of these data provides estimates of the energetics, total ejected masses and synthesised mass of 56Ni. Detection of the host galaxy emission lines allows for metallicity measurements. Results: Neither SNe 2007bg nor 2007bi were found in association with an observed GRB, but from estimates of the metallicities of their host-galaxies they are found to inhabit similar low-metallicity environments as GRB associated supernovae. The radio-bright SN 2007bg is hosted by an extremely sub-luminous galaxy of magnitude MB = -12.4 ± 0.6 mag and an estimated oxygen abundance of 12+log(O/H) = 8.18 ± 0.17 (on the Pettini & Pagel 2004 scale). The early lightcurve evolution of SN 2007bg matches the fast-pace decline of SN 1994I giving it one of the fastest post-maximum decline rates of all broad-lined type Ic supernovae known to date and, when combined with its high expansion velocities, a high kinetic energy to ejected mass ratio (EK/Mej~2.7). We also show that SN 2007bi is possibly the most luminous type Ic known, reaching a peak magnitude of MR ~ -21.3 mag and displays a remarkably slow decline, following the radioactive decay rate of 56Co to 56Fe throughout the course of its observed lifetime. SN 2007bi also displays an extreme longevity in its spectral evolution and is still not fully nebular at approximately one year post-maximum brightness. From a simple model of the bolometric light curve of SN 2007bi we estimate a total ejected 56Ni mass of MNi = 3.5-4.5 M_ȯ, the largest 56Ni mass measured in the ejecta of a supernova to date. There are two models that could explain the high luminosity and large ejected 56Ni mass. One is a pair-instability supernova (PISN) which has been predicted to occur for massive stars at low metallicities. We measure the host galaxy metallicity of SN 2007bi to be 12+log(O/H) = 8.15 ± 0.15 (on the McGaugh 1991 scale) which is somewhat high to be consistent with the PISN model. An alternative is the core-collapse of a C+O star of 20-40 Mȯ which is the core of a star of originally 50-100 Mȯ.

  • The kinematics of the quadrupolar nebula M 1-75 and the identification of its central star

    • Aims. This paper investigates the kinematics and shaping of the multipolar nebula M 1-75, and briefly discusses the location and nature of its central star. Methods. Fabry-Perot data from GH alpha FAS on the WHT that samples the Doppler shift of the [NII] 658.3 nm line are used to study the dynamics of the nebula by means of a detailed 3D spatio-kinematical model. Multi-wavelength images and spectra from the WFC and IDS on the INT, as well as from ACAM on the WHT, allowed us to constrain the parameters of the central star. Results. The two pairs of lobes, angularly separated by similar to 22 degrees, were ejected simultaneously approx. similar to 3500-5000 years ago, at the adopted distance range from 3.5 to 5.0 kpc. The larger lobes show a slight degree of point symmetry. The formation of the nebula could be explained by wind interaction in a system consisting of a post-AGB star surrounded by a disc warped by radiative instabilities. This requires the system to be a close binary or a single star that engulfed a planet as it died. On the other hand, we present broad- and narrow-band images and a low S/N optical spectrum of the highly-reddened, previously unnoticed star that is likely the nebular progenitor. Its estimated V-I colour allows us to derive a rough estimate of the parameters and nature of the central star.
