  • Socioeconomic differences in the progression of carotid atherosclerosis in middle-aged men and women with subclinical atherosclerosis in Sweden.

    • While the persistence of socioeconomic differences in cardiovascular disease (CVD) has been recognized for many years, less is known about whether socioeconomic factors are of importance to CVD before symptoms of the disease appear. In this study the associations among educational level, occupational status and progression of atherosclerosis were investigated in 1016 Swedish middle-aged men and women with signs of subclinical atherosclerosis, i.e., carotid plaque (defined as focal intima-media thickness (IMT) >1.2 mm). IMT in the common carotid artery (CCA) and in the carotid bifurcation area, as well as carotid plaque score, was determined by B-mode ultrasound. Results showed only weak associations between educational level, occupational status and age-, sex- and baseline IMT-adjusted progression of IMT in the CCA. However, in the age, sex- and baseline IMT-adjusted analyses, those in unskilled manual occupations showed a significantly higher yearly progression of carotid IMT in the bifurcation area compared to those in high- or medium-level non-manual occupations. Those with primary education tended to show a higher yearly progression of carotid IMT in the bifurcation area compared to those with completed secondary education. After adjustment for risk factors, the magnitude of these associations were somewhat attenuated. Similar patterns of associations were seen for the change of carotid plaque score. We conclude that low socioeconomic status (SES) is associated with progression of atherosclerosis in a middle-aged population with signs of subclinical atherosclerosis. Even though socioeconomic differences in cardiovascular risk factor levels could explain part of the found differences in progression rate in women, the mechanisms involved remain to be further established.

  • Aircraft Systems Modeling : Model Based Systems Engineering in Avionics Design and Aircraft Simulation

    • Aircraft developers like other development and manufacturing companies, are experiencing increasing complexity in their products and growing competition in the global market. One way to confront the challenges is to make the development process more efficient and to shorten time to market for new products/variants by using design and development methods based on models. Model Based Systems Engineering (MBSE) is introduced to, in a structured way, support engineers with aids and rules in order to engineer systems in a new way.In this thesis, model based strategies for aircraft and avionics development are studied. A background to avionics architectures and in particular Integrated Modular Avionics is described. The integrating discipline Systems Engineering, MBSE and applicable standards are also described. A survey on available and emerging modeling techniques and tools, such as Hosted Simulation, is presented and Modeling Domains are defined in order to analyze the engineering environment with all its vital parts to support an MBSE approach.Time and money may be saved by using modeling techniques that enable understanding of the engineering problem, state-of-the-art analysis and team communication, with preserved or increased quality and sense of control. Dynamic simulation is an activity increasingly used in aerospace, for several reasons; to prove the product concept, to validate stated requirements, and to verify the final implementation. Simulation is also used for end-user training, with specialized training simulators, but with the same underlying models. As models grow in complexity, and the set of simulation platforms is expanded, new needs for specification, model building and configuration support arise, which requires a modeling framework to be efficient.

    • Toll-like receptor (TLR) signaling is important for the induction of pro-inflammatory cytokines and interferon (IFN)-inducible genes in response to bacterial and viral challenge. Interleukin-1 receptor-associated kinase-1 (IRAK-1) is a signaling kinase situated downstream of the adapter protein myeloid differentiation factor 88 (MyD88) in the TLR intracellular signaling cascade and is required for normal signal transduction through this pathway. We investigated the importance of IRAK-1 in intestinal inflammation by using the dextran sulfate sodium (DSS)-colitis model. We show that IRAK-1 deficient mice are protected against systemic signs of inflammation, i.e., weight loss and spleen enlargement compared to wild-type controls irrespective of gender. However, IRAK-1(-/y) males but not IRAK-1(-/-) females display significant protection against colitis and thymic atrophy compared to wild-type mice. Our results indicate a gender specific effect of IRAK-1 in the DSS-induced colitis, an interesting finding since the Irak-1 gene is located on the X-chromosome and several inflammatory diseases have a gender dependent incidence.

    • Signalling downstream Toll-like receptors (TLR) is regulated at several levels in order to activate the immune response and prevent excessive inflammation. Altered intracellular signalling may be one reason that repeated stimulation of various TLRs results in hyporesponsiveness and cross-tolerance. We report that TLR cross-tolerance is inducible in the absence of interleukin-1 receptor-associated kinase-1 (IRAK-1) in peritoneal macrophages. Similar to wild-type macrophages, IRAK-1-deficient macrophages respond with decreased tumour necrosis factor (TNF) production to a secondary TLR stimulation, but in opposite to IRAK-1(+/+), IRAK-1(-/-) macrophages display increased interleukin (IL)-10 production at TLR restimulation. IRAK-1-deficient peritoneal macrophages have a defective TNF and IL-10 production in response to lipoteichoic acid stimulation as well as a defective IL-10-but a normal TNF production in response to high concentration of lipopolysaccharide. Our results demonstrate that IRAK-1 is not necessary for induction of TLR cross-tolerance as judged by TNF production.

  • Persistent organic pollutants in river food webs: influence of trophic position and degree of heterotrophy

    • We investigated how the degree of autotrophy/heterotrophy and organism trophic position influenced the bioaccumulation of polychlorinated biphenyls (PCBs) in 10 benthic river food webs consisting of terrestrial detritus, periphyton, invertebrates, and age-0 brown trout (Salmo trutta) in southern Sweden. Concentrations of PCBs increased with trophic position, estimated from delta(15)N and delta(13)C, on a dry weight basis (ng center dot g(-1) dry weight) but not on a lipid weight basis (ng center dot g(-1) lipid). PCB biomagnification factors between the first and second trophic levels (invertebrates/ periphyton and invertebrates/detritus) ranged between 0.3 and 2.3 and between the second and third levels (trout/invertebrates) between 0.3 and 2.0 on a lipid weight basis. The mean proportion of carbon ultimately derived from terrestrial sources, alpha, was 0.82 +/- 0.19 for invertebrates and 0.67 +/- 0.28 for trout. Contrary to our hypothesis, PCB concentrations in trout were positively related to alpha (r(2) = 0.58-0.77, p < 0.05). As alpha and the periphyton density (g C center dot m(-2)) in the rivers was positively related (r(2) = 0.88, p < 0.01), we propose that this relationship was due to an increased retention and exposure of PCBs to trout in rivers with low grazing pressure and high periphyton density.
