  • The Most Delicate Subject : A History of Sex Education Films in Sweden

    • The aim of this dissertation is to present a history of sex education films in Sweden, from their beginnings until the late 1970s, relating the films to the changing historical context of the Swedish welfare society. Within this framework, the dissertation explores two main sets of questions. First, utilizing a contextual understanding of genre, the dissertation analyzes how the films were produced, exhibited, and censored, what position and status they had, and where the borders were understood to be to kindred types of film, such as exploitation and pornography. Second, the norms and ideas about sexuality that the films construct are explored from a social constructionist perspective and put in historical context. Apart from the films themselves, the material used consists of censorship records, archival material, and articles and reviews from the daily press and the trade press.The study demonstrates that the sex education film has always been a delicate genre, and that this delicacy is related to the medium and to the fact that cinema is part of the commercial market. The sex education film came to Sweden through imported films during the silent era and was relatively common at Swedish movie theaters from the 1920s to the early 1970s. Very quickly, it became a contested genre thought to be in need of regulation, which can be seen in the censorship procedures. For instance, until the 1950s, screenings were often restricted through gender segregation. When film censorship was liberalized in the 1960s, sex education films could show more than before, and when the films in the sexually explicit Language of Love series (1969–1972) were released, their relationship to pornography became a central point of discussion. The changing views on using film in school sex education are also examined. Here, it is shown that if film was seen as a potentially helpful tool in the 1920s and 1930s, this view had changed into a more skeptical position by the early 1970s.In the analysis of the content of the films, only Swedish examples are studied. In the first group of theatrically shown films of this kind, which appeared in the 1940s and 1950s, sexuality is represented as problematic through the themes of venereal disease and abortion. Here, casual sex is seen as immoral and abortion is condemned. At the same time, the films display how traditional and modern views on sexuality and gender were under negotiation during this period. While reproduction was the dominating perspective in school films well into the 1970s, the Language of Love films display a radically different perspective on sex, not least because of their focus on sexual pleasure. However, while the films advocate a liberal idea about sexuality according to which science leads to liberation, a focus on romantic long-term relationships can also be noted. Throughout the period, there is an emphasis on rationality and science and on a secular perspective on sex in which love is seen as more important than matrimony. In this way, the films in general reflect a strong belief in modernity and progress that was characteristic of Sweden during the welfare era.

  • The Dual Memory Systems Model and it's implications for Technology Education

    • This paper examines the relationship between the explicit and the implicit memory and learning systems in terms of recent findings in neuropsychology and outlines the implications for technology education in terms of student learning. The psychological Dual Systems Theory situates the processing of perception, assessment, decision-making and action in two parallel but different cognitive systems. The Declarative/Explicit system is conscious, remembering facts and episodes; it is rational and logical but also limited in complexity and speed. Working Memory’s (WM) capacity, an “end station” of this system, is severely limited. Depending on modality between 4 and 9 variables or steps in a sequence may be handled at once. More will lead to cognitive loading and impasse. Since WM probably is our consciousness, we will not be able to analyse what was dropped out or what happened, we will just get confused. The narrow view of conscious vision, the slow speed and the problems with details and complexity in space and time makes this system less suitable for real time, real complexity work. The non conscious Implicit System on the other hand does not use Working Memory and will not be hampered by cognitive overload; it’s a pattern recognition system with very fast recognition of earlier encountered situations and objects. It has several important functions: 1- To direct conscious attention to what is important and relevant in a situation.2- To give a fast assessment, built upon somatic markers incorporated in memory.3- To start an automatic reaction to what is perceived, The lifespan of this memory system is long, and it seems very hard to erase or to change them. Most of what we usually refer to as skill seems to be connected to learning in this system. The Dual Systems model has recently been supported with neurophysiologic results showing, two different anatomical systems. The somatic markers have been identified, the secondary implicit vision system and concepts such as tacit knowing, automaticity, flow and intuition are starting to be understood. This model gives a new way of understanding what we do in technology education, gives clues of how to promote creativity, holistic learning, system thinking, forward reasoning and more.

  • Gene therapy for dopamine replacement.

    • Dopamine replacement for Parkinson's disease (PD) have seen three major iterations of improvements since the introduction of l-3,4-dihydroxyphenylalanine (l-DOPA) pharmacotherapy: dopamine receptor agonists, ex vivo gene transfer for cell transplantation and most recently in vivo gene therapy. In this chapter, we describe the principles behind viral vector-mediated enzyme replacement in PD. We focus on the enzymes involved in the dopamine synthesis and their internal regulation, the early experimental work on gene therapy using different viral vector types and selection of transgenes, and finally discuss the recently completed early phase clinical trials in PD patients.

    • Viral vector-mediated gene transfer utilizing adeno-associated viral vectors has recently entered clinical testing as a novel tool for delivery of therapeutic agents to the brain. Clinical trials in Parkinson's disease using adeno-associated viral vector-based gene therapy have shown the safety of the approach. Further efforts in this area will show if gene-based approaches can rival the therapeutic efficacy achieved with the best pharmacological therapy or other, already established, surgical interventions. One of the strategies under development for clinical application is continuous 3,4-dihydroxyphenylalanine delivery. This approach has been shown to be efficient in restoring motor function and reducing established dyskinesias in rats with a partial lesion of the nigrostriatal dopamine projection. Here we utilized high purity recombinant adeno-associated viral vectors serotype 5 coding for tyrosine hydroxylase and its co-factor synthesizing enzyme guanosine-5'-triphosphate cyclohydrolase-1, delivered at an optimal ratio of 5 : 1, to show that the enhanced 3,4-dihydroxyphenylalanine production obtained with this optimized delivery system results in robust recovery of function in spontaneous motor tests after complete dopamine denervation. We found that the therapeutic efficacy was substantial and could be maintained for at least 6 months. The tyrosine hydroxylase plus guanosine-5'-triphosphate cyclohydrolase-1 treated animals were resistant to developing dyskinesias upon peripheral l-3,4-dihydroxyphenylalanine drug challenge, which is consistent with the interpretation that continuous dopamine stimulation resulted in a normalization of the post-synaptic response. Interestingly, recovery of forelimb use in the stepping test observed here was maintained even after a second lesion depleting the serotonin input to the forebrain, suggesting that the therapeutic efficacy was not solely dependent on dopamine synthesis and release from striatal serotonergic terminals. Taken together these results show that vector-mediated continuous 3,4-dihydroxyphenylalanine delivery has the potential to provide significant symptomatic relief even in advanced stages of Parkinson's disease.

    • We used a single adeno-associated viral (AAV) vector co-expressing tyrosine hydroxylase (TH) and GTP cyclohydrolase 1 (GCH1) to investigate the relationship between vector dose, and the magnitude and rate of recovery in hemi-parkinsonian rats. Intrastriatal injections of >1E10 genomic copies (gc) of TH-GCH1 vector resulted in complete recovery in drug-naïve behavior tests. Lower vector dose gave partial to no functional improvement. Stereological quantification revealed no striatal NeuN+ cell loss in any of the groups, whereas a TH-GCH1 dose of >1E11 gc resulted in cell loss in globus pallidus. Thus, a TH-GCH1 dose of 1E10 gc gave complete recovery without causing neuronal loss. Safety and efficacy was also studied in non-human primates where the control vector resulted in co-expression of the transgenes in caudate-putamen. In the TH-GCH1 group, GCH1 expression was robust but TH was not detectable. Moreover, TH-GCH1 treatment did not result in functional improvement in non-human primates.

    • Preclinical efficacy of continuous delivery of 3,4-dihydroxyphenylalanine (DOPA) with adeno-associated viral (AAV) vectors has recently been documented in animal models of Parkinson's disease (PD). So far, all studies have utilized a mix of two monocistronic vectors expressing either of the two genes, tyrosine hydroxylase (TH) and GTP cyclohydrolase-1 (GCH1), needed for DOPA production. Here, we present a novel vector design that enables efficient DOPA production from a single AAV vector in rats with complete unilateral dopamine (DA) lesions. Functional efficacy was assessed with drug-induced and spontaneous motor behavioral tests where vector-treated animals showed near complete and stable recovery within 1 month. Recovery of motor function was associated with restoration of extracellular DA levels as assessed by online microdialysis. Histological analysis showed robust transgene expression not only in the striatum but also in overlying cortical areas. In globus pallidus, we noted loss of NeuN staining, which might be due to different sensitivity in neuronal populations to transgene expression. Taken together, we present a single AAV vector design that result in efficient DOPA production and wide-spread transduction. This is a favorable starting point for continued translation toward a therapeutic application, although future studies need to carefully review target region, vector spread and dilution with this approach.
