  • Water Dissociation on Single Crystalline Anatase TiO2(001) Studied by Photoelectron Spectroscopy

    • The adsorption of water on the anatase TiO2(001)-(4 x 1) surface is studied using synchrotron radiation-excited core level photoelectron spectroscopy. The coverage-dependent adsorption of water at low temperature is monitored and compared to the sequence obtained after heating of a water multilayer. Two adsorption phases of submonolayer coverage can be defined: Phase 1 consists only of dissociated water, observed as OH-groups. This phase is found at low coverage at low temperature (190 K) and is the only state of adsorbed water above similar to 230 K. The saturation coverage of phase 1 is consistent with dissociation on the 4-fold-coordinated Ti ridge atoms of the (4 x 1) surface reconstruction. Phase 2 is found at higher coverage, reached at lower temperature. It consists of a mixture of dissociated and molecular water with a ratio of 1:1 at 170 K. The molecular water is found to bond to the hydroxyl groups. The hydroxyl coverage of phase 2 is approximately 2 times that of phase 1. The results suggest that the OH and H2O species of phase 2 are confined to the ridges of the surface.

    • A group of Swedish radiation oncologists and hospital physicists have estimated the number of patients in Sweden suitable for proton beam therapy in a facility where one of the principal aims is to facilitate randomized and other studies in which the advantage of protons can be shown and the magnitude of the differences compared with optimally administered conventional radiation treatment, also including intensity-modulated radiation therapy (IMRT) and brachytherapy, can be shown. The estimations have been based on current statistics of tumour incidence in Sweden, number of patients potentially eligible for radiation treatment, scientific support from clinical trials and model dose planning studies and knowledge of the dose-response relations of different tumours together with information on normal tissue complication rates. In Sweden, it is assessed that between 2200 and 2500 patients annually are eligible for proton beam therapy, and that for these patients the potential therapeutic benefit is so great as to justify the additional expense of proton therapy. This constitutes between 14-15% of all irradiated patients annually.

    • The two-tissue compartment model, including irreversible trapping in the second compartment (2TCM) is used to describe the kinetics of 5-Hydroxy-L-[beta-(11)C]-tryptophan ([(11)C]HTP), a radioligand used in positron emission tomography (PET) for probing the second enzymatic step in the biosynthesis of serotonin. In this study, we examined the capacity of the model to track pharmacological changes in this biological process. We also investigated the potential loss of [(11)C]HTP-derived radioactivity during a PET study, since loss should be negligible not to alter quantification. Six rhesus monkeys were investigated using bolus [(11)C]HTP/PET methodology before and after pharmacological intervention. The second enzymatic step in serotonin synthesis was inhibited using the aromatic L-amino acid decarboxylase inhibitor NSD1015 (10 mg/kg). The extent of [(11)C]-derived radioactivity loss from the brain was studied by inhibition of the enzyme responsible for formation of the tissue metabolite, monoamine oxidase A, using clorgyline (2 mg/kg). After NSD1015, the uptake of [(11)C]HTP-derived radioactivity was increased in all the investigated brain regions, while the parameter used to reflect decarboxylase activity, the net accumulation rate constant (K(acc)), was decreased by 37% in the striatum, compared with baseline. Pretreatment with clorgyline did not change the brain uptake of [(11)C]HTP-derived radioactivity or K(acc). This study demonstrates that the 2TCM for [(11)C]HTP/PET is able to detect changes occurring during alteration of the biological process (i.e., the conversion of HTP to serotonin). Elimination of the radiotracer metabolite [(11)C]HIAA from the brain may be considered negligible if the PET study is limited to 60 min.
